Benzyl Alcohol Oxidation over H5PMo10V2O40 Catalyst Chemically Immobilized on Sulfur-containing Mesoporous Carbon (황이 포함된 중형기공성 탄소에 화학적으로 고정화된 H5PMo10V2O40 촉매 상에서 Benzyl Alcohol 산화반응)

  • $H_5PMo_{10}V_2O_{40}$ ($PMo_{10}V_2$) catalyst chemically immobilized on sulfur-containing mesoporous carbon (S-MC) was prepared, and it was applied to the benzyl alcohol oxidation reaction. S-MC was synthesized by a templating method using SBA-15 and p-toluenesulfonic acid as a templating agent and a carbon precursor, respectively. S-MC was then modified to have a positive charge, and thus, to provide sites for the immobilization of $PMo_{10}V_2$. By taking advantage of the overall negative charge of $[PMo_{10}V_2O4_{40}]^{5-}$, $PMo_{10}V_2$ catalyst was immobilized on the S-MC support as a charge matching component. It was revealed that $PMo_{10}V_2$ species were finely and molecularly dispersed on the S-MC via chemical immobilization. In the vapor-phase oxidation of benzyl alcohol, $PMo_{10}V_2$/S-MC catalyst showed higher conversion of benzyl alcohol and higher yield for benzaldehyde and benzoic acid than unsupported $PMo_{10}V_2$ catalyst. The enhanced catalytic performance of $PMo_{10}V_2$/S-MC was due to fine dispersion of $PMo_{10}V_2$ species on the S-MC via chemical immobilization.

Progesterone and Estrogen Levels in Holstein Blood and Milk Following Artificial Insemination and Embryo Transfer (인공수정 및 수정란이식 후 젖소의 혈액과 유즙에서 Progesterone과 Estrogen 농도 변화와 수태율과의 상관관계)

  • Early pregnancy diagnosis of bovine is an essential component for efficient reproductive plan in farms because long term of non-pregnancy results in economic losses by failure of offspring production and low milk yield in dairy cattle. The major steroid hormones related with reproduction are known to be progesterone and estrogen in bovine pregnancy. To evaluate detection level of hormones in milk, plasma and milk progestrone and estrogen of Holstein cows was analyzed during artificial insemination (AI) and embryo transfer (ET). Progesterone concentration at 21 days postestrus was significantly different in plasma and milk between pregnant and non-pregnant cows. Estrogen concentration at estrus was higher in pregnant recipients than that in non-pregnant recipients. To analyze correlation between hormone levels and conception rates in Holstein, the conception and return rates were checked following AI, and the returned cows were on the track of pregnancy after consecutive AI. Pregnant cows following first AI were considered as high conception group while pregnant cows following third AI were rated as low conception group. Proportion of high and low conception groups in this study was 78.2% and 9.1%, respectively. Hormone analysis indicated that high conception group had higher estrogen level during estrus than low conception group ($26.45{\pm}3.32$ vs $19.017{\pm}2.97$). Progesterone level was not different between high and low conception groups during estrus but increased significantly after 21 days postestrus (21 day: $4.95{\pm}1.12$ vs $0.95{\pm}0.23$, 35 day: $12.47{\pm}3.82$ vs $2.41{\pm}1.21$). In conclusion, the pattern of progesterone and estrogen secretion in Holstein milk samples could be a good candidate for early pregnancy detection and selection of recipients during ET.

  • [ ${\beta}-Glucan$ ], one of the cell wall components, is most plentiful polysaccharides in cell wall and has several advantages in immune system. In yeast ${\beta}-glucan$ is mainly contained in the yeast cell wall, and thus it is important to produce high levels of cell mass for the mass production of yeast ${\beta}-glucan$. The best carbon and nitrogen sources on cell mass production were high fructose syrup and yeast extract. Response surface methodology (RSM) was very potential tool for the optimization of process factor and medium component. It was applied to estimate the effects of medium components on the production of cell mass. Optimal concentrations of high fructose syrup and yeast extract by response surface methodology were 8.0% (v/v) and 5.2% (w/v), respectively and the cell mass predicted was $17.0\;g/{\ell}$ at 20 h of cultivation.

Design of a pen-shaped input device using the low-cost inertial measurement units (저가격 관성 센서를 이용한 펜 형 입력 장치의 개발)

  • In this paper, we present a pen-shaped input device equipped with accelerometers and gyroscopes that measure inertial movements when a user writes on 2 or 3 dimensional space with the pen. The measurements from gyroscope are integrated once to find the attitude of the system and are used to compensate gravitational effect in the accelerations. Further, the compensated accelerations are integrated twice to yield the position of the system, whose basic concept stems from the field of inertial navigation. However, the accuracy of the position measurement significantly deteriorates with time due to the integrations involved in recovering the handwriting trajectory This problem is common in the inertial navigation system and is usually solved by the periodic or aperiodic calibration of the system with external reference sources or other information in the filed of inertial navigation. In the presented paper, the calibration of the position or velocity is performed on-line and off-line. In the on-line calibration stage, the complementary filter technique is used, where a Kalman filter plays an important role. In the off-line calibration stage, the constant component of the resultant navigational error of the system is removed using the velocity information and motion detection algorithm. The effectiveness and feasibility of the presented system is shown through the experimental results.

Biomechanical Analysis of Different Thoracolumbar Orthosis Designs using Finite Element Method (유한요소 해석을 이용한 정형용 흉·요추 보조기의 형태에 따른 생체역학적 분석)

  • Thoracolumbar orthosis has been used for the rehabilitation of the patients with senile kyphosis. Recently, a number of different thoracolumbar orthosis designs have been introduced but its biomechanical effectiveness still remain unknown. In this study, we compared the pressure distribution on the surface of the trunk and stresses on the orthosis in relation to changes in connecting frame designs (Type 1, one-connecting frame type; Type 2, two-connecting frame type; Type 3, all-in-one type) using finite element (FE) models under different motions of the trunk. The results showed that Type 3 distributed the pressure on the trunk most evenly followed by Type 2 and Type 1 and the difference between Type 1 and Type 2 was negligible. ROM was limited most effectively by Type 3 ($8.5{\sim}9.4^{\circ}$), followed by Type 2 ($11.3{\sim}13.9^{\circ}$) and Type 1 ($12.1{\sim}15.4^{\circ}$). The ratio between the peak von Mises stress and yield strength of each material remained less than 20% regardless of orthosis type indicating low likelihood of component failure. In conclusion, our study found that all-in-one type of orthosis was the most effective design for the conservative treatment of spinal deformity in terms of function and comfort.

Characteristics of Microwave-Assisted Extraction for Grape Seed Components with Different Solvents (추출용매에 따른 포도씨 성분의 마이크로웨이브 추출 특성)

  • Microwave energy (2450 MHz) was applied to extract health-effective component (HEC) from grape seed. Three different solvents approved for grape seed extract such as water, ethanol and acetone, were confirmed in their microwave-heating properties and by which extraction efficiencies were determined. Microwave-assisted extraction (MAE) was performed under different parameters; microwave power : (0, 50, 100 and 150), time : (1, 3, 5, 7, and 9 min), the sample to solvent ratio : (1:20, 1:10, 1:5 and 1:2.5), and particle size(whole, 20, 40 and 60 mesh) and the subsequent extracts were used for determining their physicochemical properties, such as total yield : (TY), total phenolics : (TP), catechin content : (CC), electron donating ability : (EDA), and browning color. The heating properties of solvent demonstrated the optimal ranges of microwave and time as 100 W and 2 to 6 min. The TY and HFC content were higher with increasing powers in water and ethanol solvent, while HFC content were lower in acetone at over 100 W. The longer of extraction time up to 5 min, the higher extraction efficiency. Based upon the overall MAE efficiency and solvent recovery, it was found optimal to use 10 times volume of ethanol for 10 mesh of seed particle at 100 W.

Variation of Seed Component Contents in Wild Soybean(Glycine soja Sieb. & Zucc.) (야생콩(Glycine soja Sieb. & Zucc.)의 종실성분 함량변이)

  • Studies were conducted to develop new breeding resources using wild soybean (Glycine soja Sieb. & Zucc.) species. In 132 wild soybeans, the average crude protein content was $45.4\%$, ranged from $37.4\%\;to\;50.2\%$ and average crude oil content was $9.3\%$ showing lower than cultivated soybean. The average total amino-acids content was $19,214\;{\mu}g/g$, containing $252\;{\mu}g/g$ of methionine and $103\;{\mu}g/g$ of cysteine, a sulfur containing amino-acid. In ratio of individual amino-acids, the glutamic acid, methionine and cystein content was $16.1\%,\;1.3\%,\;and\;0.5\%$, respectively. The correlation between total amino-acid and sulfur containing amino-acid was not significant. Total isoflavone concentration was, on average, $1,105\;{\mu}g/g$ in wild soybean germplasm. As a result of comparison with total isoflavone and genestein contents, significant positive correlations were observed between total isoflavone and genistein concentration.

Identification of the quantitative trait loci for breaking and bending types lodging resistance in rice, using recombinant inbred lines derived from Koshihikari and a strong culm variety, leaf star

  • To develop rice cultivars with increased biomass and grain yield, superior lodging resistance is an essential trait. The new breeding approach can be adopted for the improvement of stem lodging resistance by enhancing culm strength. The resistance to breaking type lodging is attributed to bending moment of basal culm (M), which is composed of the section modulus (SM) and bending stress (BS). The resistance to the bending type lodging is attributed to flexural rigidity (FR) of stem, which is composed of the secondary moment of inertia (SMI) and Young's modulus (YM). Starch and cell wall components such as cellulose, hemicellulose and lignin also play a significant role in physical strength of culm, and thus affect lodging. Leaf Star has a superior lodging resistance due to its thick and stiff culm because of its high M and FR compared with Koshihikari. Furthermore, Leaf Star contains high densities of hemicellulose, cellulose and low lignin density in culm compared with Koshihikari. In this study, we performed QTL analysis for these traits associated with culm strength, using 94 recombinant inbred lines (RILs, $F_8$), derived from a cross between Leaf Star and Koshihikari. The SM in the RILs showed a continuous distribution. QTLs for SM were detected on chrs.2, 3 and 10. Leaf Star alleles increased SM on chrs. 2 and 3, but Koshihikari allele increased on chr.10. These QTLs overlapped with those QTLs identified using backcrossed inbred line derived from a cross between Chugoku 117 and Koshihikari, the parents of Leaf Star. The FR in Leaf Star was higher than that in Koshihikari due to the larger SMI and YM. 3 QTLs for SMI were detected on chrs.2, 3 and 10. Leaf Star alleles increased SMI on chrs.2 and 3, and Koshihikari alleles increased on chr.10. One QTL on chr.3 and two QTLs on chr.5 for hollocelulose content were detected with Leaf Star alleles contribution. Moreover, two QTLs were detected for hemicellulose density on chrs.3 and 5. Leaf Star allele increased hemicellulose density on chr.5, and Koshihikari allele increased on chr.3. Furthermore, two QTLs for cellulose density were detected on chr.5, and one QTL on chr.2. For starch content, one QTL on chr.3 and two QTLs on chr.5 with Leaf Star alleles contribution were detected. TULK-6 carrying a chromosome segment of Leaf Star on chr.5 in the Koshihikari genetic background showed higher densities of starch and hemicellulose than those in Koshihikari. These results suggest that the detected QTLs for culm strength could be utilized for the improvement of lodging resistance in rice by marker-assisted selection.

Selective Ni Recovery from Spent Ni-Mo-Based Catalysts (니켈-몰리브데늄 성분계 폐촉매로부터 니켈의 선택적 회수)

  • The objective of this study is to optimize the leaching conditions of sequential leaching and extracting processes for selective Ni recovery from spent Ni-Mo-based catalyst. The selective Ni recovery process consists of two processes of leaching and extracting process. In this 2-step process, Ni component is dissolved from solid spent Ni-Mo-based catalyst into leaching agent in leaching process and sequentially extracted to Ni complex with an extracting agent in the extracting process. The solutions of nitric acid ($HNO_3$), ammonium carbonate ($(NH_4)_2CO_3$) and sodium carbonate ($Na_2CO_3$) were used as a leaching agent in leaching process and oxalic acid was used as an extracting agent in extracting process. $HNO_3$ solution is the most efficient leaching agent among the various leaching agent. Also, the optimized leaching conditions for the efficient and selective Ni recovery were the leaching temperature of $90^{\circ}C,\;HNO_3$ concentration of 6.25 vol% and elapsed time of 3 h. As a result, Nickel oxalate having the highest yield of 88.7% and purity of 100% was obtained after sequentially leaching and extracting processes under the optimized leaching conditions.

Metabolic Engineering of the Brassinosteroid Biosynthetic Pathways (Brassinosteroid의 대사공학)

  • Sterols play two major roles in plants: a bulk component in biological membranes and precursors of plant steroid hormones. Physiological effects of plant steroids, brassinosteroids (BRs), include cell elongation, cell division, stress tolerance, and senescence acceleration. Arabidopsis mutants that carry genetic defects in BR biosynthesis or its signaling display characteristic phenotypes, such as short robust inflorescences, dark-green round leaves, and sterility. Currently there are more than 100 dwarf mutants representing 7 genetic loci in Arabidopsis. Mutants of 6 loci, dwf1/dim1/cbb1, cpd/dwf3, dwf4, dwf5, det2/dwf6, dwf7 are rescued by exogenous application of BRs, whereas bri1/dwf2 shares phenotypes with the above 6 loci but are resistant to BRs. These suggest that the 6 loci are defective in BR biosynthesis, and the one locus is in BR signaling. Biochemical analyses, such as intermediate feeding tests, examining the levels of endogenous BR, and molecular cloning of the genes revealed that dwf7, dwf5, and dwf1 are defective in the three consecutive steps of sterol biosynthesis, from episterol to campesterol via 5-dehydroepisterol. Similarly, det2/dwf6, dwf4, and cpd/dwf3 were Shown to be blocked in $D^4$ reduction, 22a-hydroxylation, and 23 a-hydroxylation, respectively. A signaling mutant bri1/dwf2 carries mutations in a Leucine-rich repeat receptor kinase. Interestingly, the bri1 mutant was shown to accumulate significant amount of BRs, suggesting that signaling and biosynthesis are dynamically coupled in Arabidopsis. Thus it is likely that transgenic plants over-expressing the rate-limiting step enzyme DWF4 as well as blocking its use by BRI1 could dramatically increase the biosynthetic yield of BRs. When applied industrially, BRs will boost new sector of plant biotechnology because of its potential use as a precursor of human steroid hormones, a novel lead compound for cholesterol-lowering effects, and a various application in plant protection.
