황토로부터 분리한 Bacillus licheniformis의 항진균 chitinase 생산과 효소 특성 (Production and Characterization of Antifungal Chitinase of Bacillus licheniformis Isolated from Yellow Loess)

  • In this study, we isolated two novel chitinase producing bacterial strains from yellow loess samples collected from Jullanamdo province. The chitinase producing bacteria were isolated based on the zone size of clearance in the chitin agar plates. Both of them were gram positive, rod ($2{\sim}3{\times}0.3{\sim}0.4{\mu}m$), spore-forming, and motility positive. They were facultative anaerobic, catalase positive and hydrolyzed starch, gelatin, and casein. From the 16s rRNA gene sequence analysis, the isolates were labeled as Bacillus licheniformis KYLS-CU01 and B. licheniformis KYLS-CU02. The isolates showed higher extracellular chitinase activities than B. licheniformis ATCC 14580 as a control. The optimum temperature and pH for chitinase production were $40^{\circ}C$ and pH 7.0, respectively. Response Surface Methodology (RSM) was used to optimize the culture medium for efficient production of the chitinase. Under this optimal condition, 1.5 times higher chitinase activity of B. licheniformis KYLS-CU02 was obtained. Extracellular chitinases of the two isolates were purified through ammonium sulfate precipitation and anion-exchange DEAE-cellulose column chromatography. The specific activities of purified chitinase from B. licheniformis KYLS-CU01 and B. licheniformis KYLS-CU02 were 7.65 and 5.21 U/mg protein, respectively. The molecular weights of the two purified chitinases were 59 kDa. Further, the purified chitinase of B. licheniformis KYLS-CU01 showed high antifungal activity against Fusarium sp.. In conclusion, these two bacterial isolates can be used as a biopesticide to control pathogenic fungi.

중국 황토고원지구의 물침식과 대책 (Water Erosion and Its Combating Measures in Loess Plateau, China)

  • Water erosion is progressing in the Loess Plateau, especially in gullies, and the sediment runoff to the Yellow River amounts to 975 million tons every year. Natural factors for water erosion include climate, soil, geological feature, terrain and vegetation. Many development projects due to the increasing population reduced the forest coverage ratio to 10%, and 200 million people in the downstream area are suffering from the damage during intense rainfall. Accordingly, the Chinese government is continuously trying to efficiently prevent the erosion by establishing measures for water erosion, including fish-scale pits, terrace technique, and check dams.

천연 황색안료 염색한지의 패션소재 적용 가능성 평가에 관한 연구 (Properties of Hanji with natural pigment dyeing for use as a fashion material)

  • This study analyzes and compares Hanji made with loess to Hanji made with kaolin, two yellow-based inorganic pigments, in terms of its physical properties, optical properties, and color fastness to light with the aim of using it as a fashion material. Hanji made by adding inorganic pigments showed an approximately 20% retention ratio on average. This figure was similar to those of loess and kaolin. Physical properties were analyzed, with the following results. A higher amount of additives lowered the apparent density and increased thickness and bulk. In general, inorganic pigment-added Hanji had lower tensile strength, bursting strength, and folding endurance compared to non-additive Hanji. The analysis of optical properties showed a lower brightness index for Hanji made with inorganic pigments compared to non-additive Hanji. When comparing the two inorganic pigments, the brightness of Hanji made with kaolin was higher. Regarding color fastness to light, loess showed level 4 and kaolin showed level 5 when 25% inorganic pigments on pulp were added to Hanji. Thus, Hanji made by adding inorganic pigments during the manufacturing process may perform well as materials for fashion because the additives enhanced both the color fastness to light and the bulk while maintaining the strength. In addition, Hanji dyed with inorganic pigments may have the potential to serve as materials for the fashion industry while still retaining the characteristics of Hanji.

수성접착제를 이용한 목조주택 내벽용 황토 마감재의 물성 및 친환경성 (Loess(Yellow Soil) Finishing Materials Using Water-based Adhesive for Wooden Construction Indoor Wall)

  • 현대인들은 실내에서 보내는 시간이 하루 중 90% 달하는 것을 간주할 때 실내의 공기는 현대에게 중요한 요소임을 알 수 있다. 1960년대 이후로 산업이 급격히 발전하면서 건축 재료는 천연재료에서 화학물질을 기본으로 하는 재료로 변하게 되고 에너지 절약의 대책으로 건물의 기밀도를 높임으로써 건축자재에서 발생하는 유해물질이 현대인에게 악영향을 미치기 시작했고 이런 요소들이 근원이 되어 새집증후군을 유발하게 되었다. 생활수준이 높아지면서 사람들은 건강에 대한 관심도 높아지게 되고 보다 유익한 친환경 건축자재를 요구하게 되었다. 본 연구에서는 황토, 물, 수지, 경화제, 무기물 충전제를 이용한 친환경 황토 마감재를 개발하였고 한국형 목조 주택 내장벽에 사용할 수 있는 최적의 비율을 연구하였다. 그리고 이런 황토 마감재가 실내벽에 사용 시 요구물성인 내잔갈림성, 내세척성, 내충격성, 부착강도를 측정하였고 황토마감재의 특성인 원적외선 실험과 포름알데히드, TVOC 방산 농도를 통하여 친환경성에 대한 평가를 하였다.

다층 도플러 유속계(ADCP)를 이용한 황토 살포 해역의 플랑크톤 평가 (Distribution of Zooplankton by ADCP's Echo Intensity in the Coastal Water used Yellow Loess)

  • 우리나라에서는 적조가 발생하면 적조발생 해역에 황토를 살포하고 있다. 황토가 살포되는 해역은 대부분 가두리 양식장 주변이며 매년 권장 살포량이상으로 대량 살포되고 있다. 본 연구에서는 지금까지 적조가 발생하여 매년 황토가 살포되어온 해역과 황토가 살포되지 않은 해역을 대상으로 추계(2008년 10월)와 춘계(2009년 4월)에 플랑크톤 네트에 의한 채집과 ADCP에 의한 체적산란강도를 계측하여 플랑크톤 분포특성과 플랑크톤의 분포밀도를 조사하였다. 황토를 살포하지 않은 해역에서 채집된 생물의 종수 및 단위체적당 개체수는 높았으나, 황토를 살포한 해역에서는 낮았다. 각 정점에서 채집한 플랑크톤의 종별, 체장별 개체수와 분포밀도의 평균치를 이용한 체적산란강도 $SV_c$와 ADCP에 의해 계측된 체적산란강도 $SV_m$를 비교해 본 결과, 황토를 살포하지 않은 해역의 $SV_c$$SV_m$는 거의 일치하였으나, 황토를 살포한 해역의 $SV_m$$SV_c$보다 4.3 dB 높았다. 즉 황토를 살포한 해역에서는 부유물이 ADCP에 크게 영향을 미치고 있음을 확인하였다. ADCP에 의해 계측된 체적산란강도의 수평분포도에서 체적산란강도는 황토를 살포한 해역이 황토를 살포하지 않은 해역보다 높았으며, 봄철이 가을철보다 높았다. 또한 체적산란강도 추정에 ADCP를 이용하면 채집에 의한 분포밀도의 과대 또는 과소평가를 방지할 수 있음을 확인할 수 있었다.

Influence of Heat Treatment on the Physicochemical Property and Mineral Composition of Various Processed Salts

  • The effects of heat treatment on the physicochemical properties and mineral composition of sun-dried salt were investigated. The salts parched at high temperature were appeared the higher alkalinity and the lower oxidation-reduction potential (ORP) than the samples without heat treatment. The commercial salts (bamboo salt and yellow loess salt) and the sun-dried salt parched at high temperature had relatively higher sodium ion content (418-450 ppm) compared to that (418.0 ppm) of refined salt. The increase of calcium ion occurred in the salts parched at high temperature compared to the sun-dried salt without heat treatment, but the magnesium ion was vice versa. The commercial salt, yellow loess salt had highest turbidity (0.973) whereas sun-dried salt showed lowest level (0.097) among the tested samples. Turbidity of heat treatment samples decreased as solubility increased. The maximum concentration of dialyzed salt was reached after 4 hr regardless of various processed salts, but those had no difference significantly among the tested samples. The X-ray diffraction patterns of the parched sun-dried salts showed different peak intensity with common salts, and they were similar to the patterns of oxide salts, especially MgO. The maximum value (2.56%) of MgO appeared in the sun-dried salt parched at $1,400^{\circ}C$.

Clay-based Management for Removal of Harmful Red Tides in Korea: A Multi-perspective Approach

  • Periodically, harmful algal blooms (HABs) have occurred, with impacts on various areas including public health, tourism, and aquatic ecosystems, especially aquacultured and caged fisheries. To prevent or manage invasions of HABs into fish farms on an emergency basis, many methods have been proposed. Frequently over the past 30 years in coastal countries, treatments of clay and clay mixed with polyaluminum chloride (PAC) and chitosan have been tested for HAB-removal effectiveness in both the laboratory and the field. In Korea, yellow loess clay (hwangto) has been dispersed using electrolytic clay dispensers, both to decrease the amount of yellow loess clay's usage in containers and enhance HAB-removal efficiency. However, this emergency method has limitations, among which is the requirement for more effective controlling agents for field applications. Thus, in this paper, we review technologies for clay-based red tides prevention and control and their limitations, and, further, introduce next-generation algicidal technologies for the emergency protection of fish farms.

황토나노입자를 부착한 황토이불 사용이 수면장애에 미치는 효과 (Effect of Loess Bedding with Loess Nanoparticles on Sleep Disorder)

  • 배경: 황토면 침구의 사용이 불면증 환자에 미치는 연구는 보고되지 않았다. 목적: 본 연구에서는 원적외선을 방사하는 황토면 침구를 불면증을 겪는 15명에게 사용하게 한 후에 '삶의 질' 및 '수면의 질'에 어떠한 변화를 일으키는지를 연구하였다. 방법: 실험군에는 황토면 침구 그리고 대조군에는 일반 황색면을 사용하게 한 다음에 대상자들의 'WHO의 삶의 질'에 대한 설문 양식으로 연구하였다. 결과: 전체 삶의 질 평가는 대조군은 사전과 사후의 변화가 유의성이 없었고, 실험군에서는 사전과 사후 간의 변화에는 유의한 차이로 개선되었다. 실험군의 '신체적 변화'는 황토면 침구를 사용하여 유의성이 있게 호전되었다. '수면시간'은 실험군의 실제 수면시간이 5.21시에서 7.2시간으로 유의하게 증가하였고, 수면의 질도 유의성이 있게 증가하였다. '수면의 기간'은 실험군에서 사전보다 사후에 유의하게 길어졌다. 습관적 수면 실험군에서는 사전보다 사용 후에는 유의하게 높아졌다. '수면 방해'는 실험군의 사전보다 사후에는 유의하게 낮아졌다. '수면 약물 이용' 및 '낮시간 실험군'에서는 사용하기 전보다 사용 후에는 유의하게 수면 효과가 개선되었다. '사회성 척도, 환경변화 척도'와 '삶의 질 척도'의 실험군에서는 사전과 사후 간에는 편차가 유의하게 개선되었다. '삶의 질 간'의 상관관계는 실험군에서는 14개의 항목에서 Pearson 상관관계분석에서 유의 확률 p < .05 및 p < .01에서도 상관성이 높게 나타났다. '주간 졸음, 우울증 및 불안 척도 변화'는 실험군에서는 사전과 사후 간에는 유의한 차이로 개선되었다. 예후조사에 의하면 실험에서 황토면 침구사용 중 및 사용 후에 마음과 신체의 편안함을 느끼었고, 수면의 질도 좋았다고 평가하였다. 결론: 수면 및 '삶의 질'에 대한 전체 평가는 일반 황색면을 사용한 대조군에서는 사전과 사후 간에는 유의한 차이가 없었으나, 황토면 침구를 사용한 실험군에서는 유의한 차이를 보이었다. 실험군의 전체적인 삶의 질은 황토면 침구를 사용하여 증가하는 것으로 나타났다.

Comparative Study on the Runoff Process of Granite Drainage Basins in Korea and Mongolia

  • Dissected erosional surfaces are widely distributed in the western part of Korea (e.g. Icheon, Chungju, Jecheon, Seosan). The deposits with thickness of less than 2m occur on the smooth bedrock surface are composed of poorly sorted subangular gravels with less than 20cm diameter. However, only weathered mantle of granites without the gravel layer are observed at some outcrops. The results of grain size analysis of deposits of Icheon district revealed that the characteristic of the gain size distribution is very similar with the results of sheetflood deposits presented by Blair (1999) in the Death Valley. Loess layer with buried soil layers of MIS7 covers the sheetflood deposits. The loess layer implies that the sheetflood deposits occurred before MIS7 based on the typical Loess sequences presented by Naruse et al.(2003). On the other hand, the climate of Korean Peninsula in MIS2 was very dry and cold (Yoon and Hwnag, 2003) by pollen analysis. This is because Yellow Sea was completely emerged during the MIS2(e.g. Sau\ito, 1998). So, it is thought that the climate in Korean Peninsula of not only MIS2 but also other glacial ages such as MIS8 was similar with present Mongolian climates. Tanaka et al.(2005) pointed out that Hortonian overlandflow occurs in grass vegetated granite basin in Mongolia. Therefore, dissected piedmont gentle slopes in the western Korea were possibly formed by sheetflood erosion during probably MIS8 as pediment widely distributed in Mongolia.

Identification of Source Locations for Atmospheric Dry Deposition of Heavy Metals during Yellow-Sand Events in Seoul, Korea in 1998 Using Hybrid Receptor Models

  • Elemental dry deposition fluxes were measured using dry deposition plates from March to June 1998 in Seoul, Korea. During this spring sampling period several yellow sand events characterized by long-range transport from China and Mongolia impacted the area. Understanding the impact of yellow-sand events on atmospheric dry deposition is critical to managing the heavy metal levels in the environment in Korea. In this study, the measured flux of a primarily crustal metal, Al and an anthropogenic metal, Pb was used with two hybrid receptor models, potential source contribution function (PSCF) and residence time weighted concentration (RTWC) for locating sources of heavy metals associated with atmospheric dry deposition fluxes during the yellow-sand events in Seoul, Korea. The PSCF using a criterion value of the 75th percentile of the measured dry deposition fluxes and RTWC results using the measured elemental dry deposition fluxes agreed well and consistently showed that there were large potential source areas in the Gobi Desert in China and Mongolia and industrial areas near Tianjin, Tangshan, and Shenyang in China. Major industrial areas of Shenyang, Fushun, and Anshan, the Central China loess plateau, the Gobi Desert, and the Alaskan semi-desert in China were identified to be major source areas for the measured Pb flux in Seoul, Korea. For Al, the main industrial areas of Tangshan, Tianjin and Beijing, the Gobi Desert, the Alashan semi-desert, and the Central China loess plateau were found to be the major source areas. These results indicate that both anthropogenic sources such as industrial areas and natural sources such as deserts contribute to the high dry deposition fluxes of both Pb and Al in Seoul, Korea during yellow-sand events. RTWC resolved several high potential source areas. Modeling results indicated that the long-range transport of Al and Pb from China during yellow-sand events as well as non yellow-sand spring daytimes increased atmospheric dry deposition of heavy metals in Korea.
