  • The key targets of endocrine disruptors are nuclear hormone receptors, which bind to steroid hormones and regulate their gene transcription. A yeast-based steroid hormone receptor gene trascription assay was previously developed for the evaluation of chemicals with endocrine modulating activity. The yeast transformants used in this assay contain the human estrogen receptor along with the appropriate steroid response elements upstream of the $\beta$-galactosidase reporter gene. We tried to evaluate several natural and synthetic steroids of their potential to interact directly with the steroid receptor. Some putative endocrine disruptors, including nonylphenol, are weakly estrogenic. But the combined treatment oj these chemicals with di-(2-ethylhexyl)phthalate (DEHP) significantly increased the $\beta$-galactosidase activity in the yeast transformant. These results suggest that we also have to consider the synergistic effects of endocrine disruptors. In this study, we showed that yeast-based bioassay is a valuable tool for screening potential endocrine disruptors and quantitative determination of estrogenicity. And the possibility that the estrogen receptor binds multiple environmental chemicals adds another level of complexity to the interaction between the endocrine disruptors and the human hormone system.

An Efficient Method for the Extraction of Astaxanthin from the Red Yeast Xanthophyllomyces dendrorhous

  • This study investigated an efficient method for the extraction of astaxanthin from the red yeast Xanthophyllomyces dendrorhous. The extraction process comprised three steps: 1) cultivating the yeast; 2) treating the yeast culture suspension with microwaves to destroy the cell walls and microbodies; and 3) drying the yeast and extracting the astaxanthin pigment using ethanol, methanol, acetone, or a mixture of the three as the extraction solvent. Ultimately, various treatment tests were performed to determine the conditions for optimal pigment extraction, and the total carotenoid and astaxanthin contents were quantified. A frequency of 2,450 MHz, an output of 500 watts, and irradiation time of 60 s were the most optimum conditions for yeast cell wall destruction. Furthermore, optimal pigment extraction occurred when using a cell density of 10g/l at $30^{\circ}C$ over 24 h, with a 10% volume of ethanol.

Nutritional Value of Candida utitis for Rotifer and Larval Flounder Paralichthys olivaceus

  • Baker's yeast, Saccharomyces cerevisiae, has been widely used as a food organism for rotifers used in the larval production of marine fish. However, the nutritional value of the yeast is relatively poor compared with that of the marine alga Chlorella. We examined the nutritional value of another yeast, Candida utilis, and whether its food value could be increased through manipulation such as a cell wall treatment. Candida utilis and S. cerevisiae and their manipulated varieties were assessed with regard to the growth and nutrition of the rotifer Brachianus plicatilis. Larvae of the flounder Paralichthys alivaceus were cultured with rotifers fed on the yeast species, and the dietary value of the rotifers for the larvae was examined. Rotifers that were fed C. utilis grew faster than those provided with S. cerevisiae. Rotifers grew slightly faster on manipulated yeast than on non-manipulated yeast varieties. Of the two yeast species, C. utilis had better dietary value for rotifers. Flounder larvae cultured with rotifers that had fed on C. utilis displayed better growth and survival ($\%$) than did those cultured with rotifers that had fed on S. cerevisiae. Although the manipulated variety of C. utilis was better than the non-manipulated variety in terms of rotifer growth, the flounder larvae survived ($\%$) and grew better when they were fed rotifers that had eaten non-manipulated C. utilis. However, the nutritional value of this yeast species was still lower than that of Chlorella.

Yeast Biomass Production from Concentrated Sugar Cane Stillage Using a Thermotolerant Candida rugosa

  • Concentrated Brazillian sugar cane stillage was used as a substrate for the yeast biomass production using Candida rugosa isolated from East Africa. At the optimum stillage concentration of 10% dry matter, biomass production was 20.4 g/l and COD reduction rate was 41%. The specific growth rate of the yeast was 0.17 $h^{-1}$ and the corresponding productivity 0.91 g $l{-1} h^{-1}$ in the batch fermentation was observed at $40{\circ}^C$.

효소 분해법에 의한 맥주효모 추출물의 제조 (Production of Brewer's Yeast Extract by Enzymatic Method)

  • Cell lytic enzyme, 5'-phosphodiesterase, and AMP-deaminase were used to produce yeast extract as a natural seasoning from beer yeast cells. Prior to the addition of cell lytic enzyme, heat treatment was performed to increase the cell wall degradation` the optimum condition of the cell lytic enzyme was 50C at pH 7.0. The production yields by the enzymatic method and conventional autolysis method were 42% and 35%, respectively. The total quantity of 5'-nucleotides, GMP and IMP, produced by enzymatic method was increased by 45% than that by the conventional method. Futhermore, the operation time of enzymatic method was only 6.5 hrs, significantly reduced from 24 hrs of the conventional method.

  • 두유박의 단백질 소재화를 위하여 셀룰레이즈(cellulase), 단백질 분해효소, 코지(koji), 효모 등의 복합처리에 의한 가용화 방법이 고형분 및 단백질 회수율, 아미노산, 유기산 조성에 미치는 영향을 고찰하였다. 셀룰레이즈와 단백질 분해효소를 복합 사용하여 두유박을 가수분해하는 것이 높은 고형분 회수율(80.2%) 및 단백질회수율(83.5%)을 얻을 수 있었고, 코지와 효모로 처리할 경우 유기산의 함량을 증대시킬 수 있었으며 양조간장 특유의 맛과 향을 내는데 효과적이었다. 특히 셀룰레이즈로 처리한 가수분해물에서 유기산 함량이 높았다. 유리 아미노산 함량은 코지보다 단백질 분해효소로 분해한 경우에서 높았으며 셀룰레이즈 처리는 유리 아미노산 함량에 큰 영향을 미치지 않았다. 두유박 가수분해물에 대한 관능검사 결과 코지로 가수분해한 시료가 관능적으로 우수하였으나 전체적인 기호도는 대조구로 사용된 양조간장에 비해 낮았다.

Effects of Various Sources and Levels of Chromium on Performance of Broilers

  • Three hundred and twenty four one day old mixed sex broiler chicks were assigned at random into 9 treatment groups. The experimental design was a 3${\times}$3 factorial arrangement. During the starter period (week 1-3), chicks were fed ad libitum. A cornsoybean meal based diet contained 23% crude protein, 3,200 kcal/kg metabolizable energy (NRC, 1994), and supplemented with organic or inorganic forms of chromium. Two organic chromium products, chromium yeast (Cr-Yeast from Alltech Biotechnology Corporation Limited) and chromium picolinate (Cr-Pic) were supplemented at the rate of 200, 400 and 800 ppb. One inorganic product, chromium chloride, was supplemented at the rate of 200, 400 and 800 ppb. During the finishing period (week 4-7), the corn-soybean meal based diet contained 20% crude protein, 3,200 kcal/kg metabolizable energy (NRC, 1994), and the same levels of chromium as in the starter period were added. No significant difference was observed among treatment groups in average daily gain, feed intake, body weight gain, feed conversion ratio and mortality. The carcass percentage of broilers receiving 200 and 400 ppb organic chromium (Cr-Yeast or Cr-Pic) was significantly increased (p<0.01). In addition, the supplementation of organic chromium reduced (p<0.05) breast meat fat content but increased breast meat protein content. The addition of chromium in the diet had no effect on boneless breast, skinless boneless breast, boneless leg, skinless boneless leg but reduced percentage of sirloin muscle. Total cholesterol and triglycerides were reduced by organic Cr supplementation. Supplementation with 200 and 400 ppb of both Cr-Yeast and Cr-Pic showed the lowest total cholesterol. The effects of type of Cr on HDL and LDL were variable, however, LDL increased with increasing level of Cr supplementation. This trial indicates that organic chromium tended to improve growth performances and carcass composition, reduced total cholesterol and triglycerides. The optimum level of organic chromium supplementation was at 200 ppb.

  • 효모의 종류에 따라 세포벽의 composition이 다른 것으로 알려져 있으나 이에 대한 체계적인 연구는 아직 이루어지지 않고 있다. 본 연구에서는 한국 유전자은행 (KCTC)과 한국미생물 보존센터 (KCCM)에서 구입한 15종류의 효모를 $Glucanex^{(R)}$ 200G로 처리한 후 생균수를 측정하여 상대적인 저항성을 비교하였다. 그 결과 Trigonopsis variabilis나 Sporidiobolus pararoseus는 $\beta$-glucanase에 대한 저항성이 높았으며 Pichia stiptis나 Filibasidium cpasuligenum은 $\beta$-glucanase에 대한 저항성이 낮았다. $\beta$-glucan의 함량이 높은 세포는 높은 농도의 $\beta$-glucanase 농도에서도 저항성을 가지므로 이를 바탕으로 여러 효모의 세포벽의 상대적인 $\beta$-glucan 함량을 추정할 수 있다.

  • Metal ions are essential to life. However, some metals such as mercury are harmful, even when present at trace amounts. Toxicity of mercury arises mainly from its oxidizing properties. Ionizing radiation (IR) is an active tool for destruction of cancer cells and diagnosis of diseases, etc. IR induces DNA double strand breaks in the nucleus, In addition, it causes lipid peroxidation, ceramide generation, and protein oxidation in the membrane, cytoplasm and nucleus. Yeasts have been a commonly used material in biological research. In yeasts, the physiological response to changing environmental conditions is controlled by the cell types. Growth rate, mutation and environmental conditions affect cell size and shape distributions. In this work, the effect of IR and mercury chloride (II) on the morphology of yeast cells were investigated. Saccharomyces cerevisiae cells were treated with IR, mercury chloride (II) and IR combined with mercury chloride (II). Non-treated cells were used as a control group. Morphological changes were observed by a scanning electron microscope (SEM). The half-lethal condition from the previous experimental results was used to the IR combined with mercury. Yeast cells were exposed to 400 and 800 Gy at dose rates of 400Gy $hr^{-1}$ or 800 Gy $hr^{-1}$, respectively. Yeast cells were treated with 0.05 to 0.15 mM mercury chloride (II). Oxidative stress can damage cellular membranes through a lipidic peroxidation. This effect was detected in this work, after treatment of IR and mercury chloride (II). The cell morphology was modified more at high doses of IR and high concentrations of mercury chloride(II). IR and mercury chloride (II) were of the oxidative stress. Cell morphology was modified differently according to the way of oxidative stress treatment. Moreover, morphological changes in the cell membrane were more observable in the frequently stress treated cells than the temporarily stress treated cells.

Aerobic Stability and Effects of Yeasts during Deterioration of Non-fermented and Fermented Total Mixed Ration with Different Moisture Levels

  • The present experiment evaluated the influence of moisture level and anaerobic fermentation on aerobic stability of total mixed ration (TMR). The dynamic changes in chemical composition and microbial population that occur after air exposure were examined, and the species of yeast associated with the deterioration process were also identified in both non-fermented and fermented TMR to deepen the understanding of aerobic deterioration. The moisture levels of TMR in this experiment were adjusted to 400 g/kg (low moisture level, LML), 450 g/kg (medium moisture level, MML), and 500 g/kg (high moisture level, HML), and both non-fermented and 56-d-fermented TMR were subjected to air exposure to determine aerobic stability. Aerobic deterioration resulted in high losses of nutritional components and largely reduced dry matter digestibility. Non-fermented TMR deteriorated during 48 h of air exposure and the HML treatment was more aerobically unstable. On dry matter (DM) basis, yeast populations significantly increased from $10^7$ to $10^{10}cfu/g$ during air exposure, and Candida ethanolica was the predominant species during deterioration in non-fermented TMR. Fermented TMR exhibited considerable resistance to aerobic deterioration. Spoilage was only observed in the HML treatment and its yeast population increased dramatically to $10^9cfu/g$ DM when air exposure progressed to 30 d. Zygosaccharomyces bailii was the sole yeast species isolated when spoilage occurred. These results confirmed that non-fermented and fermented TMR with a HML are more prone to spoilage, and fermented TMR has considerable resistance to aerobic deterioration. Yeasts can trigger aerobic deterioration in both non-fermented and fermented TMR. C. ethanolica may be involved in the spoilage of non-fermented TMR and the vigorous growth of Z. bailii can initiate aerobic deterioration in fermented TMR.
