Sea cucumbers contain more than 50% protein in their solid content, and they also possess various bioactive substances such as saponins and mucopolysaccharides. This study analyzed the activities of various enzymes derived from Bacillus and lactic acid bacteria and determined to degrade the components of sea cucumbers. Among the analyzed strains, B. subtilis K26 showed the highest activities in protease and xylanase and relatively high activity in cellulase. Accordingly, samples of sea cucumber and water were mixed in equal proportions, sterilized, and then fermented by inoculating them with B. subtilis K26. Following this, a higher amino acid content was observed between 1.5 and 7.5 hr, a lower residual solid content in this time, and a lesser fermentation odor. The saponin content in fermented sea cucumber powder extracted with butanol was measured to be 1.12 mg/g. The chondroitin sulfate content was evaluated to be 5.11 mg/g in raw sea cucumber. The total polyphenol content, flavonoid content, and antioxidant activities were 6.95 mg gallic acid equivalent/g, 3.69 mg quercetin equivalent/g, and 3.69 mg quercetin equivalent/g in raw sea cucumber, respectively. Moreover, the DNA damage protective effect of fermented sea cucumber extract was found to be concentration-dependent, with a very strong effect at very low concentrations. Overall, we suggest that sea cucumber fermented with B. subtilis K26 has a high potential as a food for inhibiting oxidation, enhancing immunity, and improving muscle function in the human body thanks to its high free amino acid content.
Sixty strains of Pleurotus ostreatus, white-rot fungi, were screened for production ability of their lignocellulolytic enzymes to selectively wood degradation. That results were shown that all of screened strains were produced lignocellulolytic enzymes on 2nd screening liquid culture medium. However, cellulase activity of selected six strains of P. ostreatus was low in avicel-yeast-peptone liquid culture medium. In the case of xylan degrading enzyme, No. 6 and No. 38 strains produced a xylanase(above 1.0U/ml) and a 1,4-${\beta}$-xylosidase (above 0.15 U/ml). Examination of the ligninolytic enzyme profiles of selected thirteen strains of the P. ostreatus, in the presence of Remazol Brilliant Blue R(RBBR), were observed that laccase(Lac) activity were earlier reached maximum level(0.8-2.0 U/ml) and then Mn-dependent peroxidase (MnP) were reached maximum level(0.5-1.5 U/ml) in glucose-yeast-peptone(GYP) medium. On the other hand, activity of lignin peroxidase(LiP) was not detected in this medium. I selected the No. 42 strain of P. ostreatus produced high levels of Mn-dependent peroxidase and laccase based on the screening method.
This study was conducted to evaluate the use of exogenous enzymes as a potential means of improving the ruminal digestion (i.e., degradability) of alfalfa hay and rice straw. Twenty six enzyme-additives were examined in terms of protein concentration and enzymic activities on model substrates. The exogenous enzymes contained ranges of endoglucanase, xylanase, ${\beta}$-glucanase, ${\alpha}$-amylase, and protease activities. Six of the enzyme additives were chosen for further investigation. The enzyme additives and a control without enzyme were applied to mature quality alfalfa hay substrate and subsequently incubated in rumen batch cultures. Five of the enzyme additives (CE2, CE13, CE14, CE19, and CE24) increased total gas production (GP) at 48 h of incubation compared to the control (p<0.05). The two additives (CE14 and CE24) having the greatest positive effects on alfalfa hay dry matter, neutral detergent fibre (NDF) and acid detergent fibre (ADF) degradability were further characterized for their ability to enhance degradation of low quality forages. The treatments CE14, CE24, a 50:50 combination of CE14 and CE24 (CE14+24), and control (no enzyme) were applied to mature alfalfa hay and rice straw. For alfalfa hay, application of the two enzyme additives, alone and in combination, increased GP compared to the control at 48 h fermentation (p<0.05), whereas only CE14 and CE14+24 treatments improved GP from rice straw (p<0.05). Rumen fluid volatile fatty acid concentrations throughout the incubation of rice straw were analyzed. Acetate concentration was slightly lower (p<0.05) for CE14${\times}$CE24 compared to the control, although individually, CE14 and CE24 acetate concentrations were not different from the control. Increases (p<0.05) in alfalfa hay NDF degradability measured at 12 and 48 h of incubation occurred only for CE14 (at 12 h) and for CE14+24 (at 12 and 48 h). Similarly, ADF degradability increased (p<0.05) with CE14 and CE14+24. As for rice straw, increased DM degradability was observed at 12 and 48 h of incubation for all enzyme treatments with an exception for CE14 at 12 h. The degradability of NDF was improved by all the enzyme treatments at either incubation time, while ADF degradability was only enhanced at 48 h. Overall, the enzymes led to enhanced digestion of mature alfalfa and there was evidence of improved digestibility of rice straw, an even lower quality forage.
Weaning is the most stressful event for nursery pigs because they are moved from familiar to unfamiliar environments. In addition, weaned pigs have immature digestive and immune systems. This situation makes weaned pigs susceptible to diseases and makes the absorption of nutrients from diets difficult. A feed approach, such as dietary enzyme supplementation, can be considered a solution. This study investigated the effects of dietary enzyme cocktail on diarrhea and immune responses of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.05% enzyme cocktail (Cocktail; combination of xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Incidence of diarrhea, packed cell volume (PCV), white blood cells (WBC) count, and immunoglobulin content were measured. A significantly lower incidence of diarrhea (p < 0.05) was observed in the Cocktail group as compared with the CON group. The Cocktail group also showed a decreased PCV (p < 0.1) on d 3 after weaning than the CON group. However, no differences were observed for number of WBC and contents of immunoglobulin G, M, and A between the Cocktail and CON groups. Consequently, inclusion of an enzyme cocktail in diets for weaned pigs had a positive influence on gut health by reducing the incidence of diarrhea in the present study.
Soybean, one of most widely used swine feed component in the world, contains non-starch polysaccharides (NSP). The digestive system of weaned pigs is not yet fully developed, and thus weaned pigs cannot easily digest diets based on corn and soybean meal. Dietary exogenous enzymes supplementation has been intensively investigated to assist digestion of anti-nutritional factors, such as NSP. This experiment was conducted to investigate the effects of dietary enzyme cocktail on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.05% enzyme cocktail (Cocktail; mixture of xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Growth performance, morphology of ileum, apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of dry matter, crude protein, and energy of weaned pigs were measured. No significant differences (p > 0.05) were observed for growth performance for the duration of the experimental period, and morphology of ileum, and nutrient digestibility between CON and Cocktail treatment groups. Therefore, the results from the current study indicated that enzyme cocktail supplementation in diets had no influence on growth performance, intestinal morphology, and nutrient digestibility of weaned pigs.

This study was conducted to investigate both plant growth-promoting and plant disease- controlling activities of bacterial strains isolated from soil. All the isolated strains were able to grow at various temperatures. All the strains, except ANG40, showed antagonistic effects against various phytopathogenic fungi. This antagonism can be ascribed to the production of siderophores and antibiotic substances. In addition, all the strains showed abilities such as nitrogen fixation, phosphate solubilization, and siderophore production. These results suggest that nitrogen, phosphorus, and iron can be converted into forms that can be easily absorbed by the plants for their growth. Analysis of the growth-promoting properties revealed that ANG51 produced 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase and indole-3-acetic acid (IAA) both of which are related to ethylene production. In contrast, the other strains were found to have only IAA-producing ability. Therefore, this study suggests that Pseudomonas geniculata ANG3, Exiguobacterium acetylicum ANG40, and Burkholderia stabilis ANG51, which were selected through analysis of comparative advantages for both plant growth promotion and disease-controlling activity, may be used as biological agents.
This experiment was carried out to investigate the effect of dietary supplementation of complex probiotics and enzyme on air quality in finishing pig building and the performance of finishing pigs. A total 117 crossbred $[(Landrace{\times}Yorkshire){\times}Duroc]$ pigs were randomly arranged into nine groups and assigned to three treatments. Pigs were fed a basal diet supplemented with 0, 0.1% level of probiotics and 0.1% level of complex probiotics and enzyme until the market weight for 42 days of the experimental period. Ammonia and hydrogen sulfide concentrations in the finishing pig building were significantly (p<0.05) decreased by dietary supplementation of complex probiotics and enzyme compared with those of control, however, indoor carbon dioxide concentration was not affected by dietary supplementation of probiotics or complex probiotics and enzyme. Average daily feed intake and feed conversion ratio were significantly improved (p<0.05) with dietary supplementation of 0.1% complex probiotics and enzyme; however, average daily gain was not affected by dietary supplementation of probiotics or complex probiotics and enzyme. In conclusion, the results obtained from this experiment suggest that the dietary supplementation of complex probiotics and enzyme for finishing pigs may improve air quality in the finishing pig building and the performance.
Flavonoid compounds show several biological activities and generally exist in the forms of glycones linking sugar moiety to main structure. Flavonoid glycones such as naringin and hesperidin in korean citrus peel are slower absorbed and consequently less active than their aglycone, naringenin and hesperetin, respectively. Therefare to increase the content of flavonoid aglycone in korean citrus peel, we used commercial carbohydrate-hydrolysing enzymes, AMG 300 L, Pectinex 100 L, and Viscozyme for transforming flavonoid glycones to aglycones. Optimal conditions of enzyme reaction were pH 5.0-7.0, $5\%$ enzyme, and 24-48 hrs. The content of naringenin and hesperetin as flavonoid aglycones in untreated citrus peel is $100\~200\;ng/g$ of dried citrus peel. In case of enzyme-treated citrus peel the content of naringenin and hesperetin increased to $1,539\∼6,674\;ng/g\;and\;1,974\∼8,906\;ng/g$ of dried citrus peel, respectively. Finally the content of flavonoid aglycones could be extracted to 10-80 times. Now enzyme-treated citrus peel may be applied to use for functional food because of its higher flavonoid aglycones as more active compounds.

This study was carried out to examine the antagonistic effect against phytopathogenic fungi of isolated strains from soil samples collected from Busan, Changwon, and Jeju Island: Botrytis cinerea, Colletotrichum acutatum, Corynespora cassiicola, Fusarium sp., Rhizoctonia solani, Phytophthora capsici, and Sclerotinia sclerotiorum. According to results of our studies, isolated strains showed an antagonistic effect against phytopathogenic fungi. Such an antagonistic effect against phytopathogenic fungi is seen due to the production of siderophores, antibiotic substances, and extracellular amylase, cellulase, protease, and xylanase enzyme activities. Extracellular enzymes produced by isolated strains were significant, given that they inhibited the growth of phytopathogenic fungi by causing bacteriolysis of the cell wall of plant pathogenic fungi. This is essential to break down the cell wall of plant pathogenic fungi and thus help plant growth by converting macromolecules, which cannot be used by the plant for growth, into small molecules. In addition, they are putative candidates as biological agents to promote plant growth and inhibit growth of phytopathogenic fungi through nitrogen fixation, indole-3-acetic acid production, siderophore production, and extracellular enzyme activity. Therefore, this study suggests the possibility of using Bacillus subtilis ANGa5, Bacillus aerius ANGa25, and Bacillus methylotrophicus ANGa27 as new biological agents, and it is considered that further studies are necessary to prove their effect as novel biological agents by standardization of formulation and optimization of selected effective microorganisms, determination of their preservation period, and crop cultivation tests.
Effects of increasing dietary rice straw on chewing activity, ruminal fermentation, and fibrolytic enzyme activity in growing goats were investigated in a $4{\times}4$ Latin Square experiment. The goats were offered four diets with an increasing proportion of rice straw (i.e. 0.05, 0.10, 0.15 and 0.20, respectively, on dry matter basis). Increasing level of rice straw increased ($P_{linear\;effect}$ <0.05) the time spent on eating, ruminating, and chewing. The ruminal pH and acetate: propionate ratio were increased ($P_{linear\;effect}$ <0.05), while the $NH_3$-N concentration was decreased ($P_{linear\;effect}$ <0.01). Increasing level of rice straw in the diet increased ($P_{linear\;effect}{\leq}0.01$) molar proportion of acetate and isovalerate, and decreased ($P_{linear\;effect}$ <0.01) molar proportion of propionate. The CMCase, xylanase and cellobiase activities in the rumen were decreased ($P_{linear\;effect}$ <0.05) with increasing level of dietary rice straw, whereas the avicelase activity was increased ($P_{linear\;effect}$ <0.01). In summary, increased level of rice straw elevated the dietary neutral detergent fibre (NDF) content in the diet and had a great impact on chewing activity and ruminal fermentation.
