Effects of Commiphora Myrrha (CM) on the Monosodium Urate (MSU)-induced Gout Model in Rats. (몰약이 Monosodium Urate로 유발된 백서의 통풍에 미치는 영향)

  • Objective : To identify the inhibitory effects of Commiphora Myrrha (CM) on monosodium urate (MSU)-induced gout model in rats. Materials and Methods: After pretreatment with CM-I (125mg/kg) or CM-II (50mg/kg) for 7 days followed by ones injection of MSU solution. the various indicators related to gout were measured on hematological and serum level including joint inflammation, Also, it was studied whether FBM directly inhibits the activity of xanthine oxidase in vitro. Results : As a result of this study, CM didn't show cytotoxicity in Jurkat cells, but it showed significant inhibition of activity of xanthine oxidase in vitro. CM slightly inhibited joint inflammation induced by MSU though not with statistical significance. CM partially decreased MSU-induced albumin, globulin, AST, ALT, BUN, creatinine. WBC, platelet count and ESR level and significantly decreased MSU-induced uric acid in serum. Conclusion : These results suggest that CM has therapeutic effects that are applicable to prevention and treatment of gout, and should be further investigated.

Anti-oxidization Effect of Extracts from Oriental Medicine and Cereal Medium Where Tricholoma matsutake Mycelia were Cultured

  • In order to explore the anti-oxidization effect of oriental medicine and cereal medium(OCM) where Tricholoma matsutake mycelia were cultured, measurement of hot water extract and UMPM(extraction method using ultra sonic waves, micro waves, micro bubble) extract, the total polyphenol content of crude polysaccharide from each extract, SOD-like activity, electron donating ability(EDA), xanthine oxidase inhibitory activity, and tyrosinase inhibitory activity was conducted. The total polyphenol content of each extract was found to be 16.36% for hot water extract(HE) group and 15.73% for UMPM extract(UE) group and the amount of crude polysaccharide precipitated into ethanol of extracts was found to be 8.79% for UMPM ethanol extract(UEE) group and 6.48% for hot water ethanol extract(HEE) group. As a result of measurement of SOD-like activity by concentration of each extract, it was found to be 96.17% for UE group, 91.23% for HE group, 91.33% for UEE group, and 87.11% for HEE group at 20 mg/mL. In the case of EDA, it was found to be 47.55% for UE group, 44.93% for HE group, 25.38% for UEE group, and 18.36% for HEE group. And in the cases of the rates of xanthine oxidase inhibitory activity and tyrosinase inhibitory activity, as the concentration of each extract increased, the inhibition rate increased accordingly. As a result of comparison between hot water extract method and UMPM extract method using extracts obtained from oriental medicine compound medium where Tricholoma matsutake mycelia were cultured, all of the extracts were judged to have a high anti-oxidization effect. In particular, UMPM extracts were found to have higher polyphenol content, SOD-like activity, EDA, xanthine oxidase inhibitory activity and tyrosinase inhibitory activity compared to hot water extract method. In this regard, extracts obtained from OCM where Tricholoma matsutake mycelia were cultured are considered to have high availability as functional material when and if they are prepared using UMPM extract method.

Anti-oxidant Activity of Ethanol Extracts from Fruits of Elaeagnus multiflora THUNB. during Maturation (성숙에 따른 뜰보리수(Elaeagnus multiflora THUNB.) 과실 에탄올 추출물의 항산화 효과)

  • This study was designed to analyze the election donating ability(EDA), superoxide dismutase(SOD)-like activity, nitrite scavenging ability and xanthine oxidase for food nutritional evaluation and excavation of functional materials in fruits of Elaeagnus multiflora THUNB. The electron donating ability of ethanol extracts in the 1.0 mg/ml concentration of extract was higher in extract of unripe fruits than others. The superoxide dismutase (SOD)-like activity was increased according to increasing of the extract concentration. The SOD-like activity was highest in the extract of unripe fruits among the extracts. The nitrite scavenging ability of unripe fruits ethanol extracts in 1.0 mg/ml of extraction solution in pH 1.2 was higher than extract of ripe and Ovenipe fruits. The nitrite scavenging ability was decreased according to increasing of pH. Also, inhibition of xanthine oxidase activity were higher in extracts of unripe fruits than those in extract of ripe and over-ripe fruits.

A Study on Antimutagenic acitivities of from Puerariae Flos Extracts (갈화(葛花)의 항돌연변이(抗突然變異) 활성(活性)에 관한 연구(硏究))

  • Antigenotoxicity test (SOS chromotest), antimutagenecity test (Ames test), and antioxidant test (NBT method and xanthine-xanthine oxidase method) were carried out using water-soluble and methanolic extracts from Puerariae Flos. Against the mutagens MNNG and NQO, antigenotoxic activity of methanolic extracts were much more effective than that of water-soluble ones. When the methanolic extract was added to the certain concentration $(100{\mu}{\ell}/tube)$, antigenotoxic acivity against the mutagen MNNG was enhanced. Contrary to the water-soluble extract, the methanolic extract showed high antigenotoxicity against the mutagen NQO with increment of the extract. Against the mutagen MNNG with Ames test, antimutagenic activity of the methanolic extract at $300{\mu}{\ell}/tube$ was 96% as an inhibition ratio of revertant forming CFU/plate. The antioxidant activity of water-soluble extract was comparatively higher than that of the methanolic one.

Physiological Activities of Korean and Chinese Viola mandshurica Extracts (국내산과 중국산 자화지정(Viola mandshurica) 추출물의 생리활성)

  • This study was carried out to compare the physiological activities of the extracts from Korean and Chinese Viola mandshurica W. Baker. The water extract from leaves of Chinese V. mandshurica exhibited the highest extraction yields of 30.45 g/100 g and the highest content of total flavonoids as 102.30 mg/g. Also, its ethanol extract showed the best content of polyphenol compounds as 136.16 mg/g. The leaf extract of Korean V. mandshurica produced higher electron donating abilities (EDA) of 92.69% (KLW) and 93.61% (KLE) than the other fractions. The strongest SOD-like activity was shown in the ethanol extract from Korean leaves of 17.28% at the concentration of 1.0 mg/mL. The nitrite scavenging abilities (NSA) of the leaf extracts of V. mandshurica from China were intense over 50% at pH 1.2 and 3.0. In the results of inhibitory rates of xanthine oxidase (XO), both ethanol extracts from Korean and Chinese leaves were higher than the other fractions as 98.67% and 93.80% respectively. Effect of tyrosinase inhibition was the highest in the water extract (45.04%) of Chinese leaves, followed by its ethanol extract (31.36%). The results of EDA, SOD-like activity and XO inhibition of the leaf extracts from Korean V. mandshurica were higher than those of Chinese, on the other hand, determinations on total polyphenol contents, NSA and tyrosinase inhibition were higher in those of Chinese.

Antioxidative Activities of Syneilesis palmata Extracts (우산나물(Syneilesis palmata) 추출물의 항산화 활성)

  • This study was conducted to examine the antioxidant activities of the extracts from aerial parts and roots of Syneilesis palmata. The ethanol extract of aerial parts showed the highest content of flavonoid compounds (31.72 mg/g), and the ethanol extract of roots has the highest content of total polyphenol compounds (68.11 mg/g). The water extract of S. palmata roots showed the highest xanthine oxidase inhibition of 99.29% and the ethanol extract of aerial parts showed 98.48% at 1.0 mg/mL. The ethanol extract of roots showed the highest value of nitrite scavenging ability of 70.89% at pH 1.2, SOD-like activity of 13.06% and electron donating ability of 98.58% at 1.0 mg/mL concentration. The effect of tyrosinase inhibition was found at only the ethanol extracts of roots (9.33%). We found that root extracts contain abundance polyphenol compounds and their antioxidant activities were greater than those of the extracts of aerial part.

Dendropanax morbifera and Rubus coreanus Miq. Extracts Inhibits the Formation of Uric Acid Crystal by Reducing Xanthine Oxidase Activity

  • Uric acid is the end product of purine metabolism in human body, originating from hypoxanthine after enzyme catalysis by Xanthine oxidase (XOD). Hyperuricemia results as a result of either over-generation of uric acid or a reduction in its excretion. In silico modelling methods such as Absorption, Distribution, Metabolism, Excretion and Toxicity (ADMET) prediction, Autodock 4.2.6 program were used to study the potential inhibitory compounds of XOD. Also we investigated the inhibition of XOD activity by using the extracts of Dendropanax morbifera and Rubus coreanus Miq spectrophotometrically. According to ADMET data, several compounds from D. morbifera and R. coreanus plants, were found to be more potent in inhibiting the XOD activity than allopurinol. XOD inhibitory activity is evaluated by quantifying the formation of uric acid by measuring the absorbance at 290 m ($A_{290}$).D. morbifera extract inhibited XOD activity at $250{\mu}g/ml$, however the extracts from R. coreanus has inhibited XOD activity at $25{\mu}g/ml$. The major compound of R. coreanus, ellagic acid significantly increased the inhibition rate from $9{\mu}g/ml$ and showed a 71% suppression rate at $15{\mu}g/ml$. Finally, these results suggested a potential inhibitory activities of the extracts from D. morbifera and R. coreanus Miq, but further research is needed to validate to ensure their safe usage as drug.

Reactive Oxygen Species Mediates Lysophosphatidic Acid-induced Migration of SKOV-3 Ovarian Cancer Cells (SKOV-3 난소암 세포주에서 lysophosphatidic acid 유도 세포의 이동에 있어 활성산소의 역할)

  • Cell motility plays an essential role in many physiological responses, such as development, immune reaction, and angiogenesis. In the present study, we showed that lysophosphatidic acid (LPA) modulates cancer cell migration by regulation of generation of reactive oxygen species (ROS). Stimulation of SKOV-3 ovarian cancer cells with LPA strongly promoted migration. but this migration was completely blocked by pharmacological inhibition of phosphatidylinositol 3-kinase (PI3K)/Akt signaling pathway. Inhibition of the ERK pathway had no effect on migration. Stimulation of SKOV-3 ovarian cancer cells with LPA significantly induced the generation of ROS in a time-dependent manner. LPA-induced generation of ROS was significantly blocked by pharmacological inhibition of PI3K or Akt, but inhibition of the ERK signaling pathway had little effect. LPA-induced generation of ROS was blocked by pretreatment of SKOV-3 ovarian cancer cells with an NADPH oxidase inhibitor, whereas inhibition of xanthine oxidase, cyclooxygenase, or mitochondrial respiratory chain complex I had no effect. Scavenging of ROS by N-acetylcysteine completely blocked LPA-induced migration of SKOV-3 ovarian cancer cells. Inhibition of NADPH oxidase blocked LPA-induced migration whereas inhibition of xanthine oxidase, cyclooxygenase, or mitochondrial respiratory chain complex I did not affect LPA-induced migration of SKOV-3 ovarian cancer cells. Given these results, we suggest that LPA induces ROS generation through the PI3K/Akt/NADPH oxidase signaling axis, thereby regulating cancer cell migration.

Anti-oxidation and Anti-aging Effect of Mixed Extract from Korean Medicinal Herbs (한약재 복합 추출물이 항산화 및 항노화에 미치는 영향)

  • Objectives : The present study was designed to investigate effects of mixed extract from korean medicinal herbs (MIX) on oxidation/reduction reaction-related and aging-related enzyme in vitro. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase inhibition assay. Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. The MIX showed 97% inhibition of collagenase activity, and 64% inhibition of elastase activity at 1 mg/ml concentration of MIX, next only to positive control, which indicate good efficacy for anti-wrinkle ingredient. Also it's treatment showed 34% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of MIX. Antioxidant activities were determined by DPPH radical scavenging, xanthine oxidase (XO) inhibiting activity and SOD-like activity. Also these scavenging, XO-inhibiting and SOD-like activities were measured in 91%, 80%, and 63% inhibition, respectively, at a treated dose of 1 mg/ml, compare to control. Conclusions : These results suggest that possibility of mixed korean medicinal herbs as a functional ingredient for anti-wrinkle and whitening, anti-oxidation and anti-aging cosmetic formula.

Cosmeceutical Activities of Doinseunguitang and Its Composition (도인승기탕 및 그 구성약재의 화장품약리활성)

  • Objectives : Cosmeceutical activities such as anti-oxidative and anti-aging effects of the Doinseunguitang and its composition, a traditional prescription, were evaluated. Methods : We performed MTT assay, melanin synthesis inhibition assay, DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay, astringent activity assay, tyrosinase inhibition assay, elastase and collagenase inhibition assay. Results : The results were obtained as follows : DPPH free radical scavenging of water extract Doinseunguitang (DISG) and ethanol extract DISG was 60% and 50% at 1,000 ppm. Xanthine oxidase inhibition effect of ethanol extract showed more than 80% at 500 ppm. Tyrosinase inhibition and inhibition melanin synthesis activities were measured in 40% and 50% at 1,000 ppm and 100 ppm. Elastase and collagenase inhibition rate of ethanol extracts DISG 40% and 80% at 1,000 ppm. It was concluded that compositive ingredients (Persicae Seman, Glycyrrhizae Radix, Cinnamomi Ramulus, Rhei Rhizoma) influenced the most results. Conclusions : The results indicated that, ethanol extract which is superior in its anti-oxidative and anti-aging effects is useful to be applied in herbal cosmetic industry.
