식이성 단백질 함량이 Methanethiol 투여한 흰쥐의 혈청 Xanthine Oxidase 활성에 미치는 영향 (Effect of Dietary Protein on the Serum Xanthine Oxidase Activity in Methanethiol-treated Rats)

  • 저 및 고단백식이로 성장한 흰쥐에 methanethiol 투여시 체중당 간무게 및 혈청 alanine aminotransferase활성의 증가율은 고단백식이군(HP군)보다 저단백식이군(LP군)이 높았으며, 이 때 혈청 보다 xanthine oxidase 활성도는 HP군보다 LP군이 높게 나타났다. 또한 혈청 중 요산함량 역시 HP군보다 LP군에서 높게 나타났으며, 간조직의 uricase 활성은 LP군이 HP군보다 높게 나타났다. 따라서 LP군이 HP군보다, methanethiol 투여로 인한 혈청 xanthine oxidase 활성이 높게 나타남은 식이중 단백함량을 낮출 때 methanethiol 에 의한 간독성이 심하게 나타나기 때문이며, 또한 단백영양 부족시 methanethiol 의 폭로가 고뇨산혈증을 유도함을 시사해 주고 있다.

한국산 감잎의 Polyphenol 화합물의 생리활성물질의 화학구조 및 효소저해효과 (Identification of Biologically Effect and Chemical Structure of Polyphenol Compounds from the Leaves of Korea Persimmon (Diospyrus kaki L. Folium))

  • The lyophilization of the solution extracted from 60 percent of acetone applied to persimmon leaves, the compounding process in accordance with the solution's concentration, and the gel filteration through Sephadex G-50 of biologically activated substances obstructing enzyme activity, such as tyrosinase, xanthine oxidase, and angiotesin converting enzyme (ACE) led to the assumption that polyphenol was the compound serving as biologically activated substances obstructing enzyme activity. Xanthine oxidase involved in pruine metabolism oxidizes hypoxanthine to xanthine and xanthine to uric acid. In the continuous study for natural compound, nine flavan-3-ols have been isolated from the persimmon leaves. The structures of (+)-catechin, (+)-gallocatechin, procyanidin B-1, pyrocyanidin C-1, prodelphinidin B-3, gallocatechin-$(4{\alpha}{\rightarrow}8)$-catechin, procyanidin B-7-3-O-gallate, procyanidin C-1-3'-3'-3'-O-trigallate and (-)-epigallocatechin-$(4{\alpha}{\rightarrow}8)$-epigallocatechin-$(4{\alpha}{\rightarrow}8)$-catechin were established by NMR and their inhibitory effect on xanthine oxidase activity was investigated. Procyanidin B-7-3-O-gallate, (-)-epigallocatechin-$(4{\alpha}{\rightarrow}8)$-epigallocatechin-$(4{\alpha}{\rightarrow}8)$-catechin and procyanidin C-1-3'-3'-3'-O-trigallate showed 94%, 90.69%, 80.90% inhibition at $100\;({\mu})M$ and inhibited on the angiotensin converting enzyme respectively. Procyanidin B-7-3-O-gallate and procyanidin C-1-3'-3'-3'-O-trigallate showed 66%, 63% inhibition at $100\;({\mu})M$ and inhibited on the xanthine oxidase competitively. Procyanidin C-1-3'-3'-3'-O-trigallate showed 70% inhibition at $100\;({\mu})M$ and inhibited on the tyrosinase competitively.

Antioxidant, Anti-Melanogenic and Anti-Wrinkle Effects of Phellinus vaninii

  • In this study, the antioxidant, anti-xanthine oxidase, anti-melanogenic and anti-wrinkle effects of methanol (ME) and hot water (HE) extracts from the fruiting bodies of Phellinus vaninii were investigated. The 1,1-diphenyl-2-picryl-hydrazyl free radical scavenging activity of 2.0 mg/mL HE (95.38%) was comparable to that of butylated hydroxytoluene (96.97%), the reference standard. The hydroxyl radical scavenging activities of ME (98.19%) and HE (97.55%) were higher than that of butylated hydroxytoluene (92.66%) at 2.0 mg/mL. Neither ME nor HE was cytotoxic to murine melanoma B16-F10 cells at 25-750 ㎍/mL. Although the xanthine oxidase (XO) inhibitory effects of ME and HE were significantly lower than that of allopurinol, the values were higher than 84 percent. The in vitro tyrosinase inhibitory activities of ME and HE were comparable to kojic acid at 2.0 mg/mL. The cellular tyrosinase and melanin synthetic activities of ME and HE on B16-F10 melanoma cells at 500 ㎍/mL were higher than arbutin, indicating that the inhibitory effects of arbutin on the tyrosinase and melanin synthesis were higher than those of ME and HE. The collagenase inhibitory activity of HE was comparable to EGCG at 2.0 mg/mL, however, the elastase inhibitory activity of ME and HE was lower than EGCG at the concentration tested. The study results demonstrated that the fruiting bodies of Ph. vaninii possessed good antioxidant, anti-xanthine oxidase, cell-free anti-tyrosinase, cellular anti-tyrosinase, anti-collagenase, and moderate anti-elastase activities, which might be used for the development of novel anti-gout, skin-whitening, and skin anti-wrinkle agents.

시판 과일 주스와 식초의 유기산 함량과 Xanthin Oxidase 저해 활성 비교 (Comparison of Organic Acid Contents and Xanthine Oxidase Inhibitory Activities of Commercial Fruit Juices and Vinegars)

  • 시판 과일 주스와 발효식초의 총 페놀 함량, 플라보노이드함량, 유기산 함량, 그리고 xanthine oxidase(XO) 저해 활성을 비교하였다. 과일 주스와 식초에 함유된 총 페놀(mg gallic acid equivalent/100 mL)과 플라보노이드(mg catechin equivalent/100 mL) 함량은 각각 82.8~157.6 mg과 15.7~85.8 mg으로 측정되었다. 주스류에서는 사과 주스, 식초류에서는 흑초가 각각 가장 높은 총 페놀 함량과 플라보노이드를 함유하였다. 유기산의 함량 중 citric acid, malic acid, ascorbic acid는 주로 과일 주스에서 검출되었다. Acetic acid가 주성분인(688.3~4,413.7 mg/100 mL) 식초류 중에서 흑초는 acetic acid뿐 아니라 citric acid, malic acid, tartaric acid를 함유하여 유기산의 분포가 가장 다양하였다. XO 저해 활성은 과일 주스보다 식초가 평균 34.2% 더 높게 측정되었으며, 흑초는 64.9%로 가장 높은 저해 활성을 나타내었다. 특히 유기산의 종류 중에서 아세트산의 함량과 XO 저해 활성 간에 높은 정의 상관성이 존재하여($R^2=0.7192$) 식초류의 XO 저해 활성의 주요 지표물질로 나타났다.

외적요인으로서의 Chlorambucil 투여가 흰쥐 심근 노화에 미치는 영향 II. 세포화학 및 생화학적 연구 (Effect of Chlorambucil as Extrinsic Factor on Aging Process of Rat Heart II. Cytochemical and Biochemical Study)

  • The present study was investigated to elucidate the effects of chlorambucil the heart tissue of various-aged rats. The male rats ranging from 3 to 36 months were used. The cytochemical and biochemical changes in myocardium of the rats were studied in the aspect of free radical roles in aging process. With the goals of evaluating the potential roles of free radicals in aging process, evidence was shought for alterations of myocardial lipid peroxide levels in control and chlorambucil treated rats. The result are summarized as follows: 1. Cytochemical studies showed that the activities of $Mg^{++}$-ATPase and succinic dehydrogenase increased with age. However, these enzyme activities were decreased with treatment of chlorambucil, when compared with control group. Interestingly it was observed that chlorambucil treatment increased the activity of acid phosphatase from 6 months upto 18 months, and decreased after 18 months. 2. The lipid peroxide level in myocatdium was increased with age; chlorambucil-treated group was higher than that of control group. 3. Age-dependent increase in activities of monoamine oxidase, xanthine oxidase and catalase was observed. But the increase of catalase activity was higher than that of monoamine oxidase and xanthine oxidase activity in control group. However, in chlorambucil-treated group, age-dependent decrease of these enzyme activities was observed, and catalase activity was more significant particularly with regard to other enzymes. In consequently, the morphological alterationsof myocardium due to chlorambucil treatment was exclusively observed. We demonstrate that this alteration is occured by lipid peroxidation upon chlorambucil treatment.

갈화(葛花)의 항돌연변이(抗突然變異) 활성(活性)에 관한 연구(硏究) (A Study on Antimutagenic acitivities of from Puerariae Flos Extracts)

  • Antigenotoxicity test (SOS chromotest), antimutagenecity test (Ames test), and antioxidant test (NBT method and xanthine-xanthine oxidase method) were carried out using water-soluble and methanolic extracts from Puerariae Flos. Against the mutagens MNNG and NQO, antigenotoxic activity of methanolic extracts were much more effective than that of water-soluble ones. When the methanolic extract was added to the certain concentration $(100{\mu}{\ell}/tube)$, antigenotoxic acivity against the mutagen MNNG was enhanced. Contrary to the water-soluble extract, the methanolic extract showed high antigenotoxicity against the mutagen NQO with increment of the extract. Against the mutagen MNNG with Ames test, antimutagenic activity of the methanolic extract at $300{\mu}{\ell}/tube$ was 96% as an inhibition ratio of revertant forming CFU/plate. The antioxidant activity of water-soluble extract was comparatively higher than that of the methanolic one.

피부 화상으로 유도된 간 손상에서 Allopurinol의 효과 (Allopurinol Decreases Liver Damage Induced by Dermal Scald Burn Injury)

  • 화상에 의한 피부 손상이 간에 미치는 영향을 알아보기 위하여 흰쥐를 이용하여 피부 화상을 유도한 다음 각각 5시간, 24시간 후 생화학적 정량법과 형태학적 관찰을 통해 간 조직 손상의 발병기전을 검토해 보고자 하였다. 흰쥐의 화상 유발은 등쪽면의 털을 깎고(total burn surface area $20\sim25%$) $100^{\circ}C$ 물로 10초간 흡입손상 없이 피부 화상만을 가하였다. 생화학적 정량으로는 혈청 내 xanthine oxidase(XO)와 aniline aminotransferase (ALT)의 활성변화, 그리고 혈장 단백질 함량 변화를 측정하였고, 형태학적 관찰은 혈액 중 다형핵 백혈구 수의 산정과 간 세포의 미세구조 변화를 관찰하였다. 실험 결과, 화상 후 혈청 내 XO의 활성 증가(P<0.01)와 함께 체중 당 간 무게(p<0.05)와 혈청 내 ALT의 활성이 증가되었다. 화상 직후 allopurinol의 복강투여로 XO활성, 간 무게, 그리고 ALT의 활성은 모두 감소되었다. 화상 손상에 의한 간 조직의 미세구조적 변화로는 소포체 종창, 리보솜 탈락, 지방소적의 축적, 그리고 담모세관과 세포간질의 확장이 관찰되었다. 뿐만 아니라 염증세포인 호중구의 침윤과 함께 혈관 내피세포의 손상, 쿠퍼세포의 활성화, 그리고 미세융모의 손상들이 관찰되었다. 또한 혈 중 다형핵 백혈구의 수적인 변화에서 화상 유발 5시간 후에 현저히 감소되어 내부 장기에 호중구 침윤의 가능성을 알 수 있었다. 하지만 allopurinol의 투여로 이러한 미세구조의 변화를 예방할 수 있는 것으로 나타났다. 이러한 결과들을 종합해 볼 때, 피부 화상으로부터 간 손상을 유발하는데 있어서 혈 중 XO의 활성증가가 매우 핵심적인 역할을 담당하는 것으로 사료되었다.

우피소근(牛皮消根)의 알칼로이드 분획이 과산화지질 생성에 미치는 영향 (Effect of Alkaloidal Fraction from Cynanchi Radix on Lipid Peroxidation)

  • The crude alkaloidal fraction of the root of Cynanchum caudatum Max.(Asclepiadaceae) was tested for the effects on the activities of free radical generating enzymes and the formation of lipid peroxide. Aldehyde oxidase was strongly inhibited to about 90% of the activity by treating 1.0 mg/ml of alkaloidal fraction, corresponding to competitive inhibition. Moreover, the formation of lipid peroxide which causes damage of cell membrane was reduced in proportion to the increasing alkaloid concentration. However, xanthine oxidase of which structure and function are similar to those of aldehyde oxidase was not inhibited by the alkaloidal fraction.

대추잎 추출물의 생리활성 작용 (Physiological Activity of Zizyphus jujuba Leaf Extracts)

  • This study was designed to investigate the possible utilization of Zizyphus jujuba leaves as a source of functional ingredients. The physiological activity of different solvent fractions prepared from ethanol extract of Zizyphus jujuba leaves were analyzed. Xanthine oxidase inhibitory effect was very high in all fractions except chloroform fraction. The very high electron donating ability was observed in the ethylacetate fraction and the effect was similar to 0.1% tocopherol. Nitrite scavenging effect of all fractions was more than 40% even at low concentration of 1mg/ml and was increased with increasing concentration. Angiotensin I converting enzyme inhibitory activity was appeared in ethyl acetate and chloroform fractions only at high concentraton.

수산생물자원 추출물의 과산화지질생성의 억제효과 (Screening of Marine Natural Products on Inhibitory Effect of the Formation of Lipid Peroxidation)

  • The methanolic extracts of some marine natural products were tested for investigating the effects on the formation of lipid peroxide and the activities of free radical generating enzymes. The methanolic extracts of Styela clava, Ecklonia stolonifera, Pachymeniopsis elliptica and Hypnea charoides which decreased the formation of lipid peroxide inhibited the activity of xanthine oxidase about 41, 20, 20 and 21% by adding $100\;{\mu}g/ml$ of each methanolic extracts, respectively. However, the four extracts didn't inhibit the activity of aldehyde oxidase.
