The protective mechanism of Rubus coreanus compound on Cisplatin-induced nephrotoxicity

  • This study is investigated the effect of Rubus coreanus MIQ. against nephrotoxicity induced by cisplatin in rat. We examined the potency of the extract of R. coreanus fruits by the activity-guided fractionation. The EtOAc- and BuOH fraction, niga-ichigoside F$_1$and 23-hydroxytormentic acid showed significant protective effects as lipid peroxidation in renal tissue and was was not affect the activity of xanthine oxidase and aldehyde oxidase by cisplatin-induced nephrotoxicity. The concentration of glutathione in renal tissue was decreased by cisplatin-induced nephrotoxicity, but was improved by pretreatment of R. coreanus compounds especially in butanol, ethylacetate fraction and 23-hydroxytormentic acid.

  • In the presence of dithionite, two kinds of molybdothiol complexes, particularly isolated Mo-cysteine complexes, used as models for xanthine oxidase or aldehyde oxidase exhibited catalytic activity on the reduction of nitrofurazone to its amino derivative. Of the two Mo-cysteine complexes, the activity of oxo-bridged one was apparently greater than that of sulfido-bridged one. The promoting effect was hardly shown by added cofactors or their replacements of the enzymes. The catalyzed reduction is considered to take place by consecutive direct two-electron transfer mechanism from catalytically active reduced form of the molybdothiol complexes to nitrofurazone and the probable intermediates.

Superoxide Dismutase Mimetic Activity of Cu(II)-Salicylic Acid Analogs

  • The superoxide dismutase (SOD)-mimetic activities of copper complexes of a series of salicylic acid (SA) analogs were tested and compared to the activity of bovine erythrocyte SOD using ferricytochrome c reduction assay. Stability constants of copper complexes were measured potentiometrically using SCOGS2 program. In the presence of 10 g/l albumin, all the copper complexes lost their SOD mimetic activities. Multiple regression analysis was employed for the statistical comparisons between the SOD mimetic activity and their physicochemical properties. Correlation exists for the SOD mimetic activity and steric parameter $(E_s)$ and/or electronic parameter $({\Sigma}{\sigma})$ in xanthine/xanthine oxidase (XOD) system, demonstrating that E, plays a key role in SOD activity whereas ${\Sigma}{\sigma}$ influences it to a lesser extent. The protective effect of copper complexes against membrane damage was measured by counting D-glucose released frm $EG_s$. D-glucose and XOD were entrapped within $EG_s$ and acetaldehyde was used as a substrate for XOD. In this membrane model system using $EG_s$, hydrophobic parameter $({\Sigma}{\pi})$ is of most importance, producing parabolic equation while $E_s$, and ${\Sigma}{\sigma}$ appear to playa minor role in protection against D-glucose release. In summary, to design an efficient SOD mimetic, stability, steric factor, lipophilicity and redox potential should be considered.

Effects of Artemisia selengensis Methanol Extract on Ethanol-Induced Hepatotoxicity in Rat Liver (쑥(물쑥)추출물이 에탄올에 의한 흰쥐의 간 손상에 미치는 영향)

  • This study was done to investigate the effects of Artemisia selengensis methanol extract on ethanol-induced hepatotoxicty in rat liver. Sprague-Dawley(SD) rats weighing about 150g were divided into the following 4 groups : control group(CON), Astemisia selengensis methanol extract administered group(ASE), ethanol adminstered group(ETH) and Artemisia selengenis methanol extract and ethanol administered group(ASA). Ethanol and Artemisia selengenis methanol extract were administered orally by 5m1/kg and 200mg/kg body weight per day for 6weeks, respectively. Body weight, daily food intake and percent liver weight per body weight were significantly changed by ethanol administration in comparison to control group. The activities of serum alanine aminotransferase(ALT), asparate aminotransferase(AST), and hepatic TBA-reactants increased by ethanol were decreased significantly by Artemisia selengensis methanol extract compared with ethanol group. It was also obseued that superoxide dismutase, catalase and glutathione peroxidase were not changed by Artemisia selengensis methanol extract, whereas hepatic xanthine oxidase activity was inhibitied by Artemisia selengensis methanol extract as compared to ethanol group. The glutathione contents in liver decreased by ethanol adminstration, but glutathione levels increased in ASA compared with ethanol group. These results suggest that Artemisia selengenis methanol extract have a possible protective effect on the ethanol-induced hepatotoxicity in rat liver.

An Analysis of the Gyungokgo's Ingredients and a Comparison Study on Anti-oxidation Effects According to the Kinds of Extract (경옥고(瓊玉膏)의 성분 분석 및 추출물별 항산화(抗酸化) 효능 비교)

  • Objectives : To estimate the value of the Gyungokgo as therapeutic agent preventing against aging with an analysis of the ingredients and the bio-activating effects by enzymologic methods. Methods : A quantitative analysis of general ingredients' of the Gyungokgo's extract was done first. The effects on electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, whitening effect have been investigated in the physiological activity measurement of function experiment. Results : The contained hydrolyzed amino acid is Valine, Aspartic acid, Arginine, Isoleucine and the contained free amino acid is Arginine, Phenylalanine, Valine, Glycine. The derivative of free amino acid is Phosphoserine, Carnocine, ${\gammer}$-Aminoisobutyric acid. And the Gyungokgo contains 14 species of minerals, K>Na>Ca>Mg>Fe>Al>Mn. Then, to assure of the Gyungokgo's anti-oxidation, these following subjects -polyphenol, electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, tyrosinase inhibation- are analyzed and show high activity especially the most in chloroform extracts, (every ingredients written by the order of high amount) Conclusions : The Gyungokgo contains many materials functioning as anti-oxidation, neurotransmitter, anti-fatigue and immune agent.

An analysis of the Gongjindan's ingredients and its efficacy on anti-oxidation (공진단(拱辰丹)의 성분 분석 및 갱산화(坑酸化) 작용에 미치는 영향)

  • Objectives : For the congenital feeble, the Gongjindan is useful medicine. The experiment is to estimate the value of the Gongjindan as therapeutic agent preventing against aging with an analysis of the ingredients and the bio-activating effects by enzymologic methods. Methods : General ingredients' of the Gongjindan's extract were analyzed first and the quantitative analysis of a reducing sugar, a soluble protein, an amino acid and minerals was made. The Gongjindan, which is extracted, concentrated, and freeze drying with water, ethanol and chloroform, have got applied for the experiment. The effects on electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, whitening effect have been investigated in the physiological activity measurement of function experiment. Results : The contained amino acid, in order of high amount, is Arginine, Alanine, Glutamic acid, Proline and the contained free amino acid is Glutamic acid, Leucine, Lysine, Phenylalanine. The derivative of free amino acid is ${\gamma}-Aminoisobutyric$ acid, Phosphoserine, Taurine. And the Gongjindan is containing 13 species of minerals in order of Ca>K>Na>Mg>Fe>AI>Mn. Then, to assure of the Gongjindan's capability of anti-oxidation, these following subjects-polyphenol, electronic donating ability, SOD-like activity, nitric oxide inhibition, xanthine oxidase inhibition, tyrosinase inhibition- are analyzed and show high activity especially the most in ethanol extracts. Conclusion : With this analysis of ingredients, the Gongjindan is containing many materials functioning as anti-oxidation, anti-aging, anti-fatigue, neurotransmitter and immune agent. Moreover, In every water, ethanol, chloroform extracts, the Gongjindan's capability of anti-oxidation is confirmed so that we can apply to patients' treatment clinically.

Anti-oxidant and Anti-inflammation Activities of Prunus persica Flos (도화(桃花, Prunus persica Flos)의 항산화 및 항염증 활성)

  • The solvent extracts of Prunus persica Flos were investigated for the activities of anti-oxidant and anti-inflammation to apply as a functional ingredient for cosmetic products. The electron donating ability of both ethanol (PPE) or acetone (PPA) extracts of P. persica Flos was above 90.0% at the concentration of 500ppm. The superoxide dismutase (SOD)-like activity of P. persica Flos extracts (PPE, PPA) were approximately 40.0% at 1,000 ppm. The xanthine oxidase inhibitory effect of P. persica Flos extracts (PPE, PPA) was approximately 30.0% at 1,000 ppm and equivalent to that of ascorbic acid. Hyaluronidase inhibition activity related to the anti-inflammation effect was 35.0% with the treatment of P. persica Flos extracts (PPW, PPE, PPA) at 1,000 ppm, respectively. In the experiment of anti-inflammation effect, P. persica Flos extracts (PPW, PPE, PPA) inhibited the generation of nitric oxide. In the antimicrobial activity test against the human skin-resident microflora such as Staphylococcus epidermidis and Propionibacterium acnes, a clear zone was identified from 4mg/disc in P. persica Flos (PPE) extract.

Effects of Dietary Vitamin E on the Lipid Peroxidation in Rats Fed Caffeine (식이성 비타민 E가 카페인 투여 흰쥐의 체내 지질과산화에 미치는 영향)

  • This experiment was undertaken to investigate the effect of dietary vitamin E on the lipid peroxidation in the caffeine-fed rats. Male Sprague-Dawley rats were fed for 6 weeks diets containing one of three levels of dietary vitamin E(30, 500 or 1,000mg/kg diet) with 1,000mg caffeine per liter in drinking water but control group was only given without caffeine. Net weight gain food intake caffeine intake and hematocrit were not significantly different. In rats fed caffeine liver lipid peroxidation level was slightly increased but significantly lower in rats fed 1, 000mg vitamin E/kg diet than in those fed 500mg vitamin E/kg diet. Glutathione peroxidase activity in rats fed caffeine was higher than in the control And the higher dietary vitamin E level the lower glutathione peroxidase activity. Catalase activity was significantly increased in the caffeine-fed rats. however xanthine oxidase activity was not afexcted in all experimental groups.

Effects of Jingansikpung-tang and Gamijingansikpung-tang Water Extract on the Cultured Spinal Sensory Neurons (진간식풍탕 및 가미진간식풍탕 추출물이 배양 척수감각신경세포에 미치는 영향)

  • To evaluate the mechanism of oxidative damage by Xanthine oxidase(XO) and hypoxanthine(HX)-induced oxygen radicals, XTT assay was carried out. Neurofilament EIA and PKC activity were measured to evaluate the protective effect of Jingansikpung-tang(JST) and Gamijingansikpung-tang(GJST) water extract on cultured spinal sensory neurons damaged by XO/HX, after the cultured mouse spinal sensory neurons were preincubated with various concentrations of JST and GJST water extract for 3 hours prior to exposure of XO/HX. The results were XO/HX decreased significantly, in proportion to concentration and exposed time, the survival rate of the cultured mouse sensory neurons on XTT assay. And in proportion to concentration and exposed time on cultured spinal sensory neurons, XO/HX showed the quantitative decrease of neurofilament by EIA, increase of PKC activity, but JST and GJST showed the neuroprotective effects against decrease of neurofilament and increase of PKC activity by XO/HX. From the above results, it is concluded that XO/HX have a neurotoxic effect on cultured spinal sensory neurons and the herbs water extract, such as JST and GJST prevent the toxicity of XO/HX effectively.

Antioxidant and Anti-inflammatory Effects of Fractions from Pruni persicae Flos (도화(桃花, Pruni persicae Flos) 분획물의 항산화 및 항염증 효과)

  • Objectives : The solvent extracts of Pruni persicae Flos were investigated for the activities of anti-oxidant and anti-inflammatory effects to apply as a functional ingredient for cosmetic products. Methods : In this study, the fractions of P. persicae Flos were extracted with 70.0% acetone and purified using Sephadex LH-20 column chromatography. As a result, eight fractions were isolated. We performed MTT assay, total polyphenol contents, DPPH free radical scavenging assay, SOD-like activity, xanthine oxidase inhibition assay, astringent activity assay, hyaluronidase inhibition assay and the production of nitric oxide. Results : For anti-oxidant effects, the electron donating ability of fraction (Fr.) 2-5, Fr.-8 isolated from P. persicae Flos was above 90.0% at 100 ppm respectively. The superoxide dismutase (SOD) - like activity of Fr.-5 isolated from P. persicae Flos was 92.1% at 1,000 ppm. The xanthine oxidase inhibitory effect of Fr.-6 isolated from P. persicae Flos was about 83.3% at 1,000 ppm. Hyaluronidase inhibition activity related to the anti-inflammation effect was 94.0% for Fr.-4 isolated from P. persicae Flos at 500 ppm. In the anti-inflammation effect, the Fr.-4 isolated from P. persicae Flos inhibited the generation of nitric oxide. Conclusions : All these findings suggested that the fractions of P. persicae Flos has a great potential as a cosmeceutical ingredient with a anti-oxidant and anti-inflammatory effects.
