Comparison of Biological Activity between Nelumbo nucifera G. Extracts and Cosmetics Adding Nelumbo nucifera G. (백련(Nelumbo nucifera G.) 추출물 및 화장품에 첨가 시 생리활성 비교)

  • The solvent extracts of Nelumbo nucifera G. were investigated for antioxidant activities, whitening and anti-wrinkle effects to apply as a functional ingredient in cosmetic products. For their industrial application, the cosmetic products were also prepared with advanced formulation techniques such as W/O/W multiple emulsion. Total phenolic and flavonoids contents increased in Nelumbo nucifera G.-Leaf (NN-L). The electron donating ability of Nelumbo nucifera G.-Flower (NN-F) or Nelumbo nucifera G.-Leaf (NN-L) extracts were above 85% at a concentration of 500 ppm. The superoxide dismutase (SOD)-like activity of Nelumbo nucifera G. (NN-L) extracts was about 60% at a concentration of 1,000 ppm. The xanthine oxidase inhibitory effect of NN-L extract was higher than that of NN-F and NN-S extracts. The tyrosinase inhibitory effect, which is related to skin-whitening, was 36% in NN-F at 1,000 ppm. For anti-wrinkle effect, the elastase inhibition activity of NN-L was about 30% at 1,000 ppm. The results of stability test showed that W/O/W multiple emulsion (ME) containing Nelumbo nucifera G. extracts. The electron donating ability of the ME containing NN-F and NN-L were about 60% at a concentration of 100 ppm. The superoxide dismutase (SOD)-like activity of the ME containing NN-L was 30% at 1,000 ppm. The tyrosinase inhibitory effect, which is related to skin-whitening, was 34% in the ME containing NN-F at 1,000 ppm. In anti-wrinkle effect, the elastase inhibition activity of the ME containing NN-L was about 55% at 1,000 ppm.

Effect of Melothria heterophylla Extract on Expression of Matrix Metalloproteinase-1 in Human Skin Fibroblasts (백렴 추출물의 Matrix Metalloproteinase-1 발현 저해 효과)

  • Matrix metalloproteinases (MMPs) are known to play an important role in photoaging by mediating the degradation of extracellular matrix proteins. In this study, to develop a n ew anti-aging agent, we investigated the antioxidant activity and the inhibitory effect of Melothria heterophylla extract on expression of MMP-1 in UVA-irradiated human dermal fibroblasts and MMP-1 activity. The M.heterophylla extract was found to scavenge radicals and reactive oxygen species (ROS) with the $SC_{50}$ values of $13{\mu}g/ml$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and $20{\mu}g/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. UVA-induced MMP-1 expression was reduced about 80% by $100{\mu}g/ml$ of the M.heterophylla extract but MMP-1 Mrna expression was not inhibited. Therefore, we conclude that the M.heterophylla extract significantly inhibits MMP-1 expression at the protein level. Also, the M.heterophylla extract inhibited MMP-1 activity in a dose dependent manner. From these results, we suggest that the M.heterophylla extract can be used as a new anti-aging agent by antioxidant activity, regulation of UVA-induced MMP-1 production, and inhibition of MMP-1 activity.

Effects of the Grapevine Shoot Extract on Free Radical Scavenging Activity and Inhibition of Pro-inflammatory mediator Production in RAW264.7 Macrophages (포도나무가지 추출물의 프리라디칼 소거 작용 및 염증 발현 매개인자 생성 억제 효과)

  • Free radical scavengers or quenching agents for reactive oxygen species (ROS) present in consumable fruits, vegetables, and beverages have received considerable attention as potential antioxidants, and thus uses for treatment of several human diseases. In this study, grapevine shoot extract (GSE) containing high concentration of resveratrol and viniferine was evaluated for antioxidant potential and inhibition of pro-inflam-matory mediator production. Utilizing 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity and xanthine oxidase (XOD) inhibition assay the GSE showed inhibitory effects of DPPH radical scavenging and XOD activity with the $IC_{50}$/ values of 34.5 and 155 $\mu\textrm{g}$/ml, respectively. In addition, GSE also exhibited the inhibition of prostaglandin E$_2$ (PGE$_2$) and nitric oxide (NO) production in lipopolysaccharide (LPS)-induced mouse macrophage RAW264.7 cells with the $IC_{50}$/ value of 6.4 and 14.5 $\mu\textrm{g}$/ml, respectively. This result suggests that grapevine shoot extract has the potential activity as a natural antioxidant or antiinflammatory agent.

Antioxidant Activities of Extracts from Fermented Black Jujube (발효 흑대추 추출물의 항산화 효과)

  • This study was conducted to analyze the antioxidant activities of fermented black jujube and to compare these with those of dried jujube, for the development of functional materials. The antioxidative activities of dried jujube and fermented black jujube extracts were analyzed by electron-donating ability (EDA) using 1,1-diphenyl-2-picryl hydrazyl (DPPH), superoxide-dismutase-(SOD)-like activity by pyrogallol, nitrite-scavenging ability, and xanthin oxidase. The yield of the fermented black jujube extracts was higher than that of the dried jujube extracts, and that of the ethanol extracts was higher than that of the hot-water extracts. The total phenol contents of the hot-water extracts from fermented black jujube were higher. The EDA values of the hot-water and ethanol extracts from fermented black jujube and dried jujube increased with an increase in extract concentration, and were about 85% in a $1000{\mu}g/mL$ extract concentration. The SOD-like activity increased with an increase in extract concentration. The SOD-like activity of the hot-water extract from fermented black jujube was higher than that of the other extracts. The nitrite-scavenging ability at pH 1.2 of the hot-water extracts from dried jujube was higher than that of the other extracts. The xanthine oxidase inhibitory activities of the hot-water and ethanol extracts from fermented black jujube were higher than those of the other extracts, and increased along with the concentrations of the extracts.

  • It was attempted to observe the effect of garlic on the hepatic purine metabolizing enzymes in this study. The activities of adenosine deaminase, guanase, and uricase in rats were not changed significantly following the feeding of 5% garlic juice. Whereas, garlic juice inhibited significantly the hepatic xanthine oxidase activity compared to control group with the lapse of treated-period. The orate level of serum in rats was significantly decreased by the treatment of garlic juice. The above inhibitory effect of garlic was greater in boiled garlic juice than fresh garlic juice-treated group. These results indicated that, according to the chemical properties of allicin which is unstable in heat, other components than allicin in garlic may regulate the hepatic purine metabolizing enzymes.

Screening of Biological Activities of the Extracts from Basil (Ocimum basilicum L.) (Basil(Ocimum basilicum L.) 추출물의 생리활성 탐색)

  • Physiological functionalities of water and ethanol extracts from Basil were determined. The concentration of total phenolic compounds of the water and ethanol extracts were $286.0\;{\mu}g/ml$, $250.0\;{\mu}g/ml$. Antioxidant activities of Basil extracts were determined using 2,2'-azinobis (3-ethyl benzothiazoline-6-sulfonic acid) radical cations (ABTS), 2,2-diphenyl-1-picryl hydrazyl radicals (DPPH), antioxidant protection factor and thiobarbituric acid reactive substances. The total antioxidant activities of Basil extracts using ABTS were 96.8% in the water extracts and 94.7% in the ethanol extract, DPPH were 87.0%, 93.9%, PF were 0.69, 1.16 and TBARS were $0.2{\times}10^{-3}\;{\mu}M,\;0.6{\times}10^{-3}\;{\mu}M$. Angiotensin converting enzyme inhibitory activity and xanthine oxidase inhibitory activity of Basil were higher in ethanol extracts (99.7%, 100.0%) than those of water extracts (39.9%, 54.7%). Phenolic profiles in Basil extracts were analyzed using HPLC. The result was that among the 6 phenolics, rosemarinic acid was the highest in ethanol extracts.

Antioxidant activities and physiological properties of Euphorbia humifusa extracts prepared using different solvents (추출용매에 따른 비단풀 추출물의 항산화 및 생리활성)

  • This study was conducted to investigate the antioxidant activities and physiological properties of Euphorbia humifusa extracts prepared using three different solvents (water, ethanol, and methanol). The highest total polyphenol content (293.25 mg/100 g) and total flavonoid content (21.05 mg/100 g) were observed in the methanol extract. The content of substances related to proanthocyanidin were highest in the water extract (8.42 mg/100 g), followed by methanol (5.70 mg/100 g) and ethanol (5.39 mg/100 g) extracts. The DPPH and ABTS radical scavenging activities of the methanol extract were 91.72% and 85.83%, respectively, at 50 mg% concentrations, which were higher than those of the other extracts. The extract reducing power decreased in the following order: ethanol > methanol > water. The methanol extract had relatively high antioxidant activity. The ${\alpha}-glucosidase$ and xanthine oxidase inhibitory activities of the methanol extract at a concentration of 10 mg% were somewhat higher than the other extracts. HMG-CoA reductase inhibitory activity in the water extract was slightly higher than in the methanol and water extracts. These results indicated that Euphorbia humifusa extracts were a high-value food ingredient due to their antioxidant activities and nutritional value.

Antioxidant Properties and Total Phenolic Contents of Cherry Elaeagnus (Elaeagnus multiflora Thunb.) Leaf Extracts

  • In Korea and China, cherry elaeagnus (Elaeagnus multiflora Thunb.) has been used traditionally to treat cough, diarrhea, itching, and foul sores. Therefore, in this study, the ethanol and water extracts of cherry elaeagnus leaves were examined for their antioxidant activities. The ethanol extract of the cherry elaeagnus leaves contained more phenolics than the water extract. All the cherry elaeagnus leaf extracts had higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability than ascorbic acid at concentrations of $250-1,000\;{\mu}g/mL$. The ethanol extract also showed higher superoxide dismutase (SOD)-like activity compared to the water extract. Furthermore, the SOD-like activity of the ethanol extract amounted to 89% of that of ascorbic acid at a concentration of $500\;{\mu}g/mL$. The nitrite scavenging ability and xanthine oxidase inhibitory (XOI) activity of the ethanol extract were higher than those of the water extract. In particular, the ethanol extract had higher XOI activity than ascorbic acid at a concentration of $1,000\;{\mu}g/mL$.

The Effect of Zinc Levels on Free Radical Generating System in Cadmium Treated Rats (아연수준이 카드뮴을 투여한 흰쥐의 항산화 효소게에 미치는 영향)

  • This study was conducted to investigate the effect of dietary zinc (Zn) and/or cadmium (Cd) on hepatic microsomal and cytosol enzyme activities. Male Spraque-Dawley rats (110$\pm$10g ) received zinc (0, 30 and 300 ppm/) and Cd-treated groups were administered oral intubation with Cd chloride (5.0mg/kg of body weight 0 at the same time once a week. The effect of Cd on the activity of hepatic cytochromep-450 , xanthine oxidase(X. O) and superoxide dismutase (SOd) was studied in rats. Cd oral intubation resulted in a decrease in cytochrome P-450 content and SOD activity whereas a significant increase in the X.O. activity was observed was observed . Intake of excessive Zn led to an increased activity of microsomal alcohol dehydrogenase (ADH) , whereas Zn deficiency group led to a decreased group. The mechanism by which Zn induces the decreasing of Cd toxicity in rats, seems to rely on the protection of the enzyme systems P-450, ADH, aldehyde dehydrogenase (ALDH) and X.O. in the liver, possibly by forming non-toxic Cd metallothionein. These results indicate that Zn and Cd regulation might occur via inhibitory protein component of the $H_2O$$_2$ -generator system.

산삼과 산양삼 추출물의 항암 및 항산화 효능

  • Objectives : The aim of this study was to verify anti-cancer and anti-oxidant efficacies of Korean wild ginseng and cultivated wild ginseng of Korea and China. Methods : For the measurement of anti-oxidation, SOD-like activity was evaluated using xanthine oxidase reduction method under in vitro environment. Subcutaneous and abdominal cancer were induced using CT-26 human colon cancer cells for the measurement of growth inhibition of cancer cells and differences in survival rate. Results : 1. Measurement of anti-oxidant activity of ginseng, Chinese and Korean cultivated wild ginseng, and natural wild ginseng samples showed concentration dependent anti-oxidant activity in HX/XOD system. Anti-oxidant activity showed drastic increase at 1mg/ml in all samples. 2. For the evaluation of growth inhibition of cancer cells after hypodermic implantation of CT-26 cancer cells in the peritoneal cavity of mice, Chinese and Korean cultivated wild ginseng and natural wild ginseng groups showed significant inhibition of tumor growth from the 12th day compared to the control group. Similar inhibitory effects were also shown on the 15th and 18th days. But there was no significant difference between the experiment groups. 3. For the observation of increase in survival rate of the natural wild ginseng group, CT-26 cancer cells were implanted in the peritoneal cavity of mice.
