오미자에서 분리한 새로운 Chitin Synthase II 저해제인 Gomisin N 과 Wuweizisu C (Gomisin N and Wuweizisu C, new Inhibitors for Chitin Systhase II from Schizandra chinensis)

  • To search for new chitin biosynthesis inhibitors from natural sources, several higher plants were examined the inhibitory activity against chitin synthase IIby enzymatic assay. Among them, the extract of Schizandra chinensis strongly showed the inhibitory activity against chitin synthase II. Gomisin N and wuweizisu C were isolated from Schizandra chinensis and showed $IC_50$ value of $62.4{\;}\mu\textrm{g}/ml$ and $19.2{\;}\mu\textrm{g}/ml$, respectively. Activities of these compounds were more stronger than that of polyxin D. However, gomisin N and wuweizisu C showed weakly antifungal activities against various human pathogens.

Capillary-GC(FID)에 의한 오미자 Lignan 성분의 정량 (Determination of Lignan Compounds in Fruits of Schisandra chinensis BAILLON by Capillary-GC(FID))

  • 무주산 오미자로부터 lignan 화합물인 deoxyschizandrin, gomisin N, schizandrin, gomisin A, wuweizisu C, angeloygomisin H 및 tigloylgomisin H를 분리 동정하고 capillary-GC (FID)를 이용하여 이들 화합물의 정량성을 조사하였다. 이 때 lignan 화합물의 동정에는 GC/MS (Finigan MAT 212; El, 70eV), 1H-NMR (Bruker FT; 300MHz) 및 IR (Perkin Elmer 599B)을 이용하였으며, lignan 화합물의 정량에는 FID 및 integrator(Hewlett-Packard 3393A)가 부착된 GC (Hewlett-Packard 5890A)를 이용하였다. 한편, GC column은 SPB-1 fused silica capillary$(0.25mm\;ID{\times}30m,\;Supelco)$를 사용하였고 column oven의 온도는 $200^{\circ}C$부터 $300^{\circ}C$까지 분당 $4^{\circ}C$씩 승온하였으며, carrier gas는 $N_2$, 1.0ml/min(split ratio=40 : 1)을 사용하였다. 각 lignan 화합물의 정량 가능농도범위는 deoxyschizandrin과 wuweizisu C가 $2{\sim}500ppm$이었고 gomisin N, schizandrin, gomisin A, angeloylgomisin H 및 tigloygomisin H 가 $5{\sim}500ppm$이었으며 무주산 오미자의 lignan성분 함량은 schizandrin이 6.50mg/g으로 가장 높았고 gomisin N, gomisin A, wuweizisu C, angeloylgomisin H, deoxyschizandrin, tigloylgomisin H의 순으로 낮은 경향이었다.

Lignans from the Fruits of Schizandra chinensis and Their Inhibitory Effects on Dopamine Content in PC12 Cells

  • Five lignans including gomisin N (1), wuweizisu C (2), gomisin L1 (3), (+)-deoxyschizandrin (4), and gomisin J (5) have been isolated from the fruits of Schizandra chinensis. The structures of the isolated compounds were elucidated by analyzing MS and NMR spectra. Effects of the compounds isolated in this study on the dopamine content in PC12 cells were investigated to evaluate their inhibitory effectiveness. The gomisin N, wuweizisu C, and gomisin J showed 25.4%, 39.8%, and 35.1 %, respectively, inhibition effect on dopamine content in PC12 cells at the concentration of $50\;{\mu}g/ml$.

HPLC에 의한 오미자 리그난 성분의 정량분석 (Quantitative Determination of Lignans from Schizandra chinensis by HPLC)

  • A reversed phase HPLC method was established for quantitative determination of lignans including schizandrin (1), gomisin A (2), deoxyschizandrin (3), gomisin N (4), and wuweizisu C (5) from Schizandra chinensis using a binary gradient of acetonitrile and water as a mobile phase with UV detection at 254nm. In result, contents of schizandrin (1), gomisin A (2), deoxyschizandrin (3), gomisin N (4), and wuweizisu C (5) in Schizandra chinensis from Moonkyung were 8.41${\pm}$0.30, 3.01${\pm}$0.13, 1.06${\pm}$0.05, 7.69${\pm}$0.30, and 1.68${\pm}$0.06 mg/g, respectively.

  • GC/MS를 이용하여 오미자의 chloroform-methanol 추출물로부터 11종의 lignan 성분을 동정하였다. GC column은 SPB-1 fused silica capillary $(0.25mm\;ID{\times}30m,\;Supelco)$를 사용하였고 column oven의 온도는 $200^{\circ}C$부터 $300^{\circ}C$까지 분당 $4^{\circ}C$씩 승온하였으며 MS의 ionization voltage는 70eV (El mode)이었다. 동정된 lignan 성분은 gomisin J, deoxyschizandrin, gomisin N, schizandrin, wuweizisu C, gomisin A, angeloylgomisin H, tigloylgomisin H, angeloylgomisin Q, gomisin B 및 benzoylgomisin H이었으며, GC chromatogram 상에서 이들 화합물의 분리상태는 양호하였다.

도라지담배 각초 및 연기중의 Lignan화합물 정량 (Quantitative Analysis of Lignan Compounds in Shred and Smoke of 'Balloon-Flower' Cigarettes)

  • Deoxyschizandrin, gomisin N, schizandrin, wuweizisu C, gomisin A and angeloylgomisin H were isolated from fruits of Schisandra chinensis BAILLON, and the lignan contents in shred and smoke of 'Balloon-Flower' cigarettes were quantitatively analyzed by capillary-GC(FID). The GC column was SPB-1 fused silica capillay (0.25mm id$\times$30m, Supelco) and the column oven temperature was programmed from 20$0^{\circ}C$ to 30$0^{\circ}C$ at the rate of 4$^{\circ}C$/min. In the shred of 'Balloon-Flower' cigarettes deoxyschizandrin. gomisin N, schizandrin, gomisin A and angeloylgomisin H were detected and schizandrein contets were the highest among them, 22.77$\mu\textrm{g}$/cig., gomisin N, schizandrin and gomisin A were 0.023, 0.054, 0.0849 and 0.167$\mu\textrm{g}$/cig. respectively, and angeloylgomisin H was not detected.

Analysis of Dibenzocyclooctadiene Lignans in Omija Wine and Cheong by Liquid Chromatography-Tandem Mass Spectrometry

  • Schisandra chinensis is a traditional herbal medicine that is widely spread in Korea, Japan, and China. The fruits of S. chinensis Bailon, known as omija in Korea, have traditionally been used for the treatment of coughs, fatigues, and insomnia. Up to now, there have been several reports for the identification of major lignan compounds and their quantitation in S. chinensis extracts. To the best of our knowledge, however, there is no report on the analysis of lignans in omija wine and omija cheong (sugared omija or omija sugar syrup). In the present study, seven dibenzocyclooctadiene lignans (gomisin A, gomisin B, gomisin C, gomisin N, schisandrin, deoxyschisandrin, and wuweizisu C) in omija wine and omija cheong were analyzed and quantitated using liquid chromatography-tandem mass spectrometry. Among seven lignans, pharmacologically active gomisin A, schisandrin, and deoxyschisandrin, which are major components in fruits of S. chinensis, were the most abundant lignans in omija wine and cheong. The content of lignan in omija wine was in the order: schisandrin > gomisin A > deoxyschisandrin > gomisin N > gomisin B > gomisin C > wuweizisu C. The concentration of deoxyschisandrin and gomisin N in omija wine was approximately 2.0- and 6.0-fold higher than for omija cheong. Additionally, this study provided a systematic identification of lignans in omija wine and cheong and indicated that the omija wine and cheong might be of value for their dietary application.

오미자로부터 분리된 화합물의 암세포 증식 억제 효과 (The Antiproliferative Effects of Compounds Isolated from Schisandra chinensis)

  • 오미자 추출물의 n-hexane 분획으로부터 open column chromatography, 분취용 HPLC 등 여러 가지 크로마토그라피를 이용하여 총 15종의 화합물을 분리하였다. 분리된 화합물들은 각각의 기기분석 데이터를 문헌치와 비교하여, 각각 wuweizisu C (1), gomisin N (2), deoxyschisandrin (3), gomisin A (4), schisandrin (5), chamigrenal (6), schisanlactone D (7), methylgomisin O (8), angeloylgomisin O (9), (-)-gomisin $L_2$ (10), schisandronic acid (11), (-)-gomisin $L_1$ (12), (+)-gomisin $K_3$ (13), gomisin J (14), tigloylgomisin H (15)으로 동정하였다. 이들 중 methylgomisin O (8)는 이 식물에서 처음으로 분리되었다. 분리된 화합물들은 HL-60 (human leukemia), HeLa (human cervical carcinoma) 및 MCF-7 (breast cancer cells) 세포주에 대하여 세포독성 실험을 실시하였다. 그 결과 화합물 7, 8 및 9는 HL-60 세포주에 대하여 각각 7.37, 6.60 및 $8.00{\mu}M$$IC_{50}$로 비교적 강한 세포독성 효과를 나타냈다. 화합물 6은 MCF-7에 대하여 $IC_{50}$ $30.50{\mu}M$로 적정한 세포독성을 나타냈다. 그리고 화합물 8은 HeLa cells에 대하여 $IC_{50}$ $1.46{\mu}M$로 비교적 강한 세포 독성으로 나타냈다.
