Inhibition of The Stem Cell Factor-Induced Migration of Mast Cells by Dexamethasone

  • Mast cells accumulation can be causally related with several allergic inflammations. Previous work has demonstrated that glucocorticoids decreased tissue mast cell number and stem cell factor (SCF)-induced migration of mast cells required p38 mitogen-activated protein kinase (MAPK) activation. In the present study, we investigated the effects of dexamethasone on SCF-induced migration of rat peritoneal mast cells (RPMCs). SCF significantly induced migration of RPMCs at 4 h. Dexamethasone dose-dependently inhibited SCF-induced migration of RPMCs (about 90.1% at 100 nM, P<0.05). MAPK p38 inhibitor, SB203580 (20 ${\mu}$M) also inhibited the SCF-induced migration. The ability of SCF to enhance morphological alteration and F -actin formation was also abolished by treatment of dexamethasone. Dexamethasone inhibited SCF-induced p38 MAPK activation to near basal level and induced the MKP-1 expression. In addition, SCF-induced inflammatory cytokine production was significantly inhibited by treatment of dexamethasone or SB203580 (p<0.01). Our results show that dexamethasone potently regulates SCF -induced migration, p38 MAPK activation and inflammatory cytokine production through expression of MKP-l protein in RPMCs. Such modulation may have functional consequences during dexamethasone treatment, especially mast cell-mediated allergic inflammation disorders.

Migration of Radiative Gas Giants with GIZMO

  • A gas giant formed in a massive protoplanetary disk via gravitational instability migrates inward due to its gravitational interaction with the disk. Planet migration occurs in various ways depending on the disk structure and internal processes, but previous studies only considered quantitative radiative feedback resulting from mass accretion onto the planet. Allowing for accretion feedback, we perform three-dimensional hydrodynamic simulations with GIZMO to investigate orbital evolution of giant planets in a protoplanetary subject to -cooling. This work shows a planet gains mass due to accretion and gradually opens a gap as it moves inward. The migration in the end halts when the planet clears the gas around its orbit. A more massive planet grows its mass faster and migrates more rapidly, stalling at an orbit farther away from the protostar. Models with a cold disk readily construct a circumplanetary disk and result in high mass growth of the planet. Accretion feedback, in general, reduces the rate of the planet growth and delays migration. We discuss our results with GIZMO in comparison with the previous results with different codes.

TRAIL Suppresses Human Breast Cancer Cell Migration via MADD/CXCR7

  • Background: Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) can specifically induce apoptosis limited to various cancer cells, so this reagent is considered a promising medicine for cancer therapy. TRAIL also exerts effects on non-apoptotic signals, relevant to processes such as metastasis, autophagy and proliferation in cancer cells. However, the mechanisms of TRAIL-regulated non-apoptotic signals are unclear. The purpose of this study was to investigate MADD/CXCR7 effects in TRAIL-mediated breast cancer cell migration. Materials and Methods: The ability of MADD/CXCR7 to regulate MVP signaling in TRAIL-mediated breast cancer cells migration was evaluated by transwell migration assay, quantitative RT-PCR, Western blotting and knock down experiments. Results: In this study, we found that treatment with TRAIL resulted in induced expression levels of MADD and CXCR7 in breast cancer cells. Knock down of MADD followed by treatment with TRAIL resulted in increased cell migration compared to either treatment alone. Similarly, through overexpression and knockdown experiments, we demonstrated that CXCR7 also positively regulated TRAIL-inhibited migration. Surprisingly, knock down of MADD lead to inhibition of TRAIL-induced CXCR7 mRNA and protein expression and overexpression of CXCR7 lead to the reduction of MADD expression, indicating that MADD is an upstream regulatory factor of TRAIL-triggered CXCR7 production and a negative feedback mechanism between MADD and CXCR7. Furthermore, we showed that CXCR7 is involved in MADD-inhibited migration in breast cancer cells. Conclusions: Our work defined a novel signaling pathway implicated in the control of breast cancer migration.

Negotiations in the Gendered Experiences of Transpinay Entertainers in Japan

  • Among Filipino entertainers in Japan, trans women (transgender women) or transpinay (Filipino trans woman) entertainers remain understudied compared to cisgender women. Though the number of entertainers has declined, transpinay entertainers remain relevant as transgender issues continue to be salient globally. This study explains the gendered experiences of the transpinay migrants, particularly in entertainment work and their relationships, which are different from cisgender Filipino women entertainers' experiences. Using grounded theory and drawing on concepts of performativity in interactions to analyze the narratives of transpinay entertainers, I delve into how transpinay entertainers negotiate their gender and migrant identities as they establish professional and personal relationships. Moreover, the transpinays' entertainment work is a significant contributing factor to their sense of belonging in Japan, as they form relationships with colleagues, clients, and partners who support them and, thereby, sustain their lifestyles as trans women. The transpinay entertainers' flows of migration between Japan and the Philippines reveal that they embrace various aspects of social remittances and use them to their advantage to create and enhance their transpinay identity in Japan. By examining the transpinays' migrant experiences, this study aims to elucidate the gendered experiences of transpinay entertainers, which involve significant negotiations in their migration pathways notably different from cisgender Filipino women entertainers, reveal resilience.

경사계를 이용한 토립자 유출 관련 피해 시공 관리 사례 연구 (Case Study of Construction Management in Damage due to Soil Particle Migration Using Inclinometer Incremental Deflection)

  • Excavation works of cylindrical shafts and tunnels for the construction of a variety of infrastructures have been frequently going on in the urban areas. When ground excavations of cylindrical shafts and shallow tunnels proceed in the ground condition of high water level and silt particle component, ground water drawdown involving soil particle migration causes loosening of ground around tunnels and shafts, causes settlement and deformation of ground. Damages due to ground sinking and differential settlement can occur in the adjacent ground and structures. The extent and possibility of damage relevant to ground water drawdown and soil particle migration can't be so precisely expected in advance that we will face terrible damages in case of minor carefulness. This paper introduces two examples of construction management where using incremental deformation graph of inclinometer, we noticed the possibility of soil migration due to ground water drawdown in the excavation process of vertical shaft and shallow tunnel, analysed a series of measurement data in coupled connection, properly prepared countermeasures, so came into safe and successful completion of excavation work without terrible damages. The effort of this article aims to improve and develop the technique of design and construction in the coming projects having similar ground condition and supporting method.

Dexamethasone Disrupts Cytoskeleton Organization and Migration of T47D Human Breast Cancer Cells by Modulating the AKT/mTOR/RhoA Pathway

  • Background: Glucocorticoids are commonly co-administered with chemotherapy to prevent drug-induced allergic reactions, nausea, and vomiting, and have anti-tumor functions clinically; however, the distinct effects of GC on subtypes of tumor cells, especially in breast cancer cells, are still not well understood. In this study, we aimed to clarify the effect of GC on subtypes of T47D breast cancer cells by focusing on apoptosis, cell organization and migration, and underluing molecular mechanisms. Materials and Methods: The cell scratch test was performed to observe the cell migration rate in T47D cells treated with dexamethasone (Dex). Hoechst and MTT assays were conducted to detect cell survival and rhodamine-labeled phalloidin staining to observe cytoskeleton dynamics. Related factors in the AKT/mTOR pathway were determined by Western blotting. Results: Dex treatment could effectively inhibit T47D breast cancer cell migration with disruption of the cytoskeletal dynamic organization. Moreover, the effect of Dex on cell migration and cytoskeleton may be mediated by AKT/mTOR/RhoA pathway. Although Dex inhibited T47D cell migration, it alone may not induce cell apoptosis in T47D cells. Conclusions: Dex in T47D human breast cancer cells could effectively inhibit cell migration by disrupting the cytoskeletal dynamic organization, which may be mediated by the AKT/mTOR/RhoA pathway. Our work suggests that glucocorticoid/Dex clinical use may prove helpful for the treatment of breast cancer metastasis.

  • Protactinium contamination is a mayor issue in the thorium fuel cycle. We investigate, in this work, the consequences of Pa incorporation in vacancy defects and interstitials in Th, ThC and ThN. We calculate charge transfers and lattice distortions due to these incorporations as well as migration paths and energies involved in the diffusion of Pa.

  • Southeast Asia accounts for nearly a tenth of total worldwide cross-border movements of migrant workers. Cambodia, Indonesia, Laos, Myanmar, and Philippines make up the sending countries while Malaysia, Singapore, and Thailand are the major destinations. Migrant worker movements are predominantly in production process and low- to medium-skilled sectors. It is not unusual for irregular or undocumented movements to take place. In not a few instances, migrants work under harsh and exploitative conditions. In recent years, however, ASEAN has taken steps to manage labor migration at the regional level. The paper argues that ASEAN has not managed these cross-border labor flows as well as it should particularly in terms of protecting and promoting the human rights of migrants. It will be difficult to establish the genuine building blocks for a regional human rights mechanism unless there is a diffusion of alternative universal norms and standards to what ASEAN already embodies. As long as states resist any attempt to weaken or question or deligitimize their capacity to determine who gets to enter, stay, and leave their jurisdictions, it will be difficult to establish an effective migrant rights framework for the region.

  • 본 논문에서는 네트워크 트래픽 감지 및 이동 에이전트의 이주 노드들에 대한 최적 경로 탐색을 통해 분산 환경에서의 효율적인 작업 처리 능력을 가진 CORBA 기반의 이동 에이전트 모델(CMAN:CORBA-based Mobile Agent Model)을 설계한다. 기존 이동 에이전트 모델은 사용자로부터 다양한 자임을 부여받게 돨 경우, 실행모듈 크기의 증가로 인해 네트워크 부하 및 트래픽을 가중시키고, 사용자에 의한 수동적인 호스트(이하 노드) 라우팅 스케줄 지정에 따라 노드 이주 수행 시 많은 트래픽이 발생학 경우, 트래픽으로 인한 많은 노드 순회 검색 시간 비용이 소요된다. 따라서, 본 논문에서는 능동적인 라우팅 스케줄지정에 따라 특정한 상황에 동적으로 대처하여 에이전트의 이주 신뢰성을 보장하고 최적 경로 탐색을 통해 에이전트의 순회 작업 처리 시간을 최소화할 수 있는 새로운 이동 에이전트 모델을 설계한다. 제안된 모델은 확장된 형태의 MAFFinder를 통해 능동적으로 이주 노드들의 라우팅 스케줄을 지정하고, 에이전트 크기로 인한 네트워크 부하를 감소하기 위해 기존의 이동 에이전트 객체를 CORBA의 분산 객체 형태에 기반하여 에이전트 호출 모듈만을 포함한 이동 에이전트와 작업 실행 모듈을 가진 푸시 에이전트로 분리한다. 노한, 이주 노트의 최적 경로 탐색을 통해 이동 에이전트의 순회 작업 처리 시 소요되는 시간을 단축시킨다
