상지(桑枝) 목초액이 호흡기 객담 과다분비에 미치는 영향 (Effect of Wood Vinegar Produced from Morus alba on Hypersecretion of Airway Mucus)

  • Objectives : In this study, the author tried to investigate whether wood vinegar produced from Morus alba (MA) significantly affects the increase in airway epithelial mucosubstances and hyperplasia of tracheal goblet cells of rats, and in vitro airway mucin secretion and PMA- or EGF- or TNF-alpha-induced MUC5AC mucin production / gene expression from human airway epithelial cells. Materials and Methods : For the in vivo experiment, the author induced hypersecretion of airway mucus and goblet cell hyperplasia by exposure of rats to SO2 over 3 weeks. Effect of orally-administered MA over 2 weeks on increase in airway epithelial mucosubstances from tracheal goblet cells of rats and hyperplasia of goblet cells were assessed using histopathological analysis after staining the epithelial tissue with alcian blue. For the in vitro experiment, confluent RTSE cells were chased for 30 min in the presence of MA to assess the effect of MA on mucin secretion by enzyme-linked immunosorbent assay (ELISA). Also, effects of MA on PMA- or EGF- or TNF-alpha-induced MUC5AC mucin production and gene expression from human airway epithelial cells (NCI-H292) were investigated. Confluent NCI-H292 cells were pretreated for 30 min in the presence of MA and treated with PMA (10 ng/ml), EGF (25 ng/ml) or TNF-alpha (0.2 nm) for 24 hrs, to assess both effects of MA on PMA- or EGF- or TNF-alpha-induced MUC5AC mucin production by enzyme-linked immunosorbent assay (ELISA) and gene expression by reverse transcription-polymerase chain reaction (RT-PCR). Possible cytotoxicities of MA in vitro were assessed by examining LDH release from RTSE cells and the rate of survival and proliferation of NCI-H292 cells. In vivo liver and kidney toxicities of MA were evaluated by measuring serum GOT/GPT activities and serum BUN/creatinine concentrations of rats after administering MA orally. Results : 1. MA decreased the amount of intraepithelial mucosubstances of rats exposed to sulfur dioxide inhalationally. 2. MA decreased in vitro mucin secretion from cultured RTSE cells. 3. MA significantly inhibited PMA-, EGF-, and TNF-alpha-induced MUC5AC mucin productions and the expression levels of MUC5AC mRNA from NCI-H292 cells. 4. MA did not show either in vitro or in vivo hepatic or renal toxicities. Conclusion : The results from this study suggests that MA can regulate the secretion, production and gene expression of airway mucin observed in diverse respiratory diseases accompanied by mucus hypersecretion and does not show in vivo toxicity to liver and kidney functions after oral administration. Effects of MA should be further studied using animal experimental models that simulate the diverse pathophysiology of respiratory diseases via future research.

소나무 우량 가계별 기초재질 변이 - 가도관, 마이크로피브릴 경사각, 수지구 분포 및 전건비중을 중심으로 - (Variation of Material Properties of Korean Red Pine of Superior Families - Tracheid Length, Microfibril Angle, Resin Canal and Specific Gravity -)

  • 재질이 우수하고 균일한 목재를 생산하기 위해서는 목부조직의 재질과 구성세포의 변동패턴 및 생장 기구에 대한 이해가 필요하며, 이러한 요소들이 우량종자 생산을 위한 육림기술 지침으로 고려되어야 한다. 본 연구에서는 소나무 우량가계의 차대 검정림에서 가계별 재질특성에 영향하는 인자 중 섬유장, 비중, 마이크로피브릴 경사각, 수지구 분포비율 등을 조사하여 다음과 같은 결과를 얻었다. 방사방향 가도관 길이 변이는 강원26, 30, 90, 56호 및 충남 4호는 $11{\sim}13$년 사이에서 가도관 길이 신장률이 1% 이하로 안정되었으며, 13년을 기점으로 매우 빠르게 성숙재로 전환 되었다. 전건비중은 $0.35{\sim}049$의 범위에 있었으며, 가계별로는 강원26호가 0.35로 가장 작고, 강원57호가 0.49로 가장 컸으며, 각 가계 간 차이가 큰 것으로 나타났다. $mm^2$당 수평수지구는 강원79호가 1.1개로 가장 적고, 강원83호는 1.7개로 많았으며, 가계별 차이가 인정되었다. 성숙재의 평균 마이크로피브릴 경사각은 $9.0{\sim}28.6^{\circ}$의 범위 내에 속하며 강원74호가 $9.0^{\circ}$로 가장 작고, 충남4호가 $28.6^{\circ}$로 가장 크며 가계 간 차이가 큰 것으로 나타났다.

점봉산 거제수나무에서 분리한 차가버섯의 배양특성 (Cultural characteristics of Inonotus obliquus isolated from Betula costata at Mt. Jumbong in Korea)

  • 차가는 배양균사로부터 위염, 위궤양 및 여러 가지 암에 효력을 나타내는 것으로 알려진 베풍긴(befungin) 약제의 생산을 위하여 사용되고 있다. 차가는 자작나무 줄기에서 주로 생산되는 것으로 알려져 있는데, 우리나라의 경우 자작나무 자생지는 해발고도 1,100m 이상에만 나타난다. 하지만, 우리는 점봉산의 거제수나무에서 차가버섯을 채집할 수 있었으며, 스트렙토마이신을 첨가한 PDA 배지를 이용하여 균주를 분리할 수 있었다. 차가 균은 5개 부위 중 차가 균으로 만연된 나무조직에서 분리될 뿐 다른 부분에서는 분리가 어려웠다. 균핵에서 확인한 포자는 $6.0{\sim}10.0{\times}4.5{\sim}6.0{\mu}m$ 크기를 나타내었고, 균사 굵기는 $2.5{\sim}5.0{\mu}m$ 이었다. 실내 배양에서 균총은 불규칙한 모양을 띄며, 시간이 경과할수록 균총의 중간 중간이 노란색으로 변색되었다. 약 20일이 경과하면 노란색으로 변색된 부분에서 적갈색의 강모(setae)를 확인할 수 있으며, 강모의 크기는 $4{\sim}6{\times}100{\sim}420{\mu}m$ 이었다. 차가의 균사생장에 가장 좋은 배지는 OA(oatmeal agar)이었다. 적합한 온도 범위는 $25{\sim}30^{\circ}C$이었으며, 균사생장에 적합한 pH는 8.0~9.5로 비교적 높은 산도를 좋아하는 것으로 나타났다.

증기 전처리 및 2단 증해 시스템에 의한 닥 인피부의 펄프화 특성 (Pulping Properties of Bast Fibers of Paper Mulberry by Pre-steaming and 2-stage Cooking System)

  • The traditional Hanji-making was confronted with lots of industrial disadvantages and economic problems, due to the original hand-made process. Recently, the studies on the automation of overall Hanji manufacturing process is carried out by applying the commercial chemical pulping method in order to expand industrial application or efficiency of non-wood fibrous materials. However, the application of commercial pulping methods to the bast tissues of paper mulberry leads to the chemical and mechanical deterioration of cellulosic fibers. In this study, the optimal cooking method using the bast parts of paper mulberry produced by an auto-scraping device was applied to minimize the damage of fiber strength for the paper yarn manufacture. The pre-steaming treatment and alkaline pulping systems were evaluated in removal efficiency of lignin and pectin materials within the bast tissue of paper mulberry. With the application of pre-steaming treatment and 2 stage pulping system using potassium carbonate and then sodium hydroxide, kappa values were decreased two times more in lignin removal than the single stage of pulping method. It was also identified from SEM images and ATR-FTIR spectra that the pectin components within cellular structure of bast tissue were easily removed and the debarked bast parts by a auto-scraping device were easily defiberized by 2-stage pulping sequence using potassium carbonate/sodium hydroxide pulping system.

침엽수재(針葉樹材)의 1차조직(次組織)과 2차조직(次組織)의 미세구조(微細構造) (Fine Structure of the Primary and the Secondary Tissues in a Coniferous Wood)

  • X선회절법(線回折法)을 이용(利用)해 소나무(Pinus densiflora)의 1차조직(次組織)과 2차조직(次組織)의 미세구조(微細構造)의 차(差)를 검토(檢討)했다. 그 결과(結果)는 다음과 같다. 1. 1차조직(次組織)과 2차조직(次組織)에서 (200)면(面)과 (004)면(面)의 면간격(面間隔)은 차이(差異)가 없었다. 2. 1차조직(次組織)과 2차조직(次組織)의 상대결정화도(相對結晶化度)는 각각(各各) 23%와 35%로 다소(多少) 차이(差異)가 있었다. 3. 미결정(微結晶)의 폭(幅)은 1차조직(次組織)과 2차조직(次組織)과의 차이(差異)가 없었다. 4. 1차조직(次組織)의 미결정(微結晶)의 배향(配向)은 무배향(無配向)을 나타냈지만 2차조직(次組織)의 미결정(微結晶)은 섬유축(纖維軸)에 $20{\sim}25^{\circ}$ 의 배향도(配向度)를 나타냈다.

  • 잣나무털녹병 (Korean white pine blister rust) 이병목(罹病木)의 해부학적(解剖學的) 진단(診斷)을 위(爲)하여, 이병부(罹病部) 수피(樹皮) 혹(或)은 목질부(木質部)의 면도날 절편(切片)을 Chloroform 1 : methanol 2 의 혼합액(混合液)에 24시간(時間) 침지(浸漬)하고, lactophenol cotton blue를 modify 한 액(液)(lactic acid 20gm, phenol crystal 20gm, cotton blue 0.05gm, 60% EtOH 100ml)에 2~5분(分) 염색(染色)하고 60~70% EtOH에 2~3분(分) 씻은 다음 lactophenol로 mount하여 검경(檢鏡)하였다. 기주조직중(寄主組織中)에 분기만연(分技蔓延)한 본병원균(本病原菌) Cronartium ribicola의 비교적(比較的) 넓은 균사(菌絲)와 곤봉상(棍棒狀) 혹(或)은 만곡(彎曲)한 흡기(吸器)가 청염(靑染)됨으로서, 잣나무의 세포조직(細胞組織)과 용이(容易)하게 식별(識別)할 수 있었다. 수피부(樹皮部)에서는 균사(菌絲) 발달(發達)이 양호(良好)하나 목질부(木質部)에서는 방사조치(放射組織)의 투심종단면(透心縱斷面)에서의 관찰(觀察)이 용역(容易)하였고 타(他) 목질부(木質部) 조직(組織)에서는 거의 관찰(觀察)할 수 없었다.

  • It is difficult to find the penetrating foreign bodies in the head and neck area only with history taking and physical examinations. One of the most important things is to detect the precise location of foreign bodies or possibly remained materials. The detection of wooden foreign bodies is important because they can cause phlegmon formation and because wood is often contaminated by Clostridium tetani bacteria. CT has proved to be an expedient method for detecting foreign bodies of various materials in soft tissues, but the wooden foreign body is often misinterpreted as a gas bubble in soft tissue. We have experienced a case of wooden foreign body which has penetrated through nasal dorsum and remained for 4 months. It had been initially misinterpreted as longstanding inflammatory lesion and osteitis of maxilla and nasal bone.

Pentachlorophenol 대사물과 세포내 거대분자물의 반응에 관한 연구 (Covalent Interactions of Reactive Pentachlorophenol Metabolites with Cellular Macromolecules)

  • Pentachlorophenol(PCP) which ks widely used in wood preservation, pulp and paper mills, has led to a substantial envirortmental contamination. To get the reliable data for the effective health risk assessment with PCP, covalent binding potential of PCP to cellular macromolecules and glutathione(GSH) was investigated after intraperitoneal administration of $^{14}C-PCP$ to rats. PCP metabolites were able to bind covalently to serum albumin and hepatic protein in a dose- and time-dependent manner. Hepatic protein adducts of PCP metabolites were increased as a function of cytochrome P-450 activities, whereas, albumin adducts significantly decreased. Covalent binding of PCP metabolites with DNA or hemoglobin was not observed. GSH levels in liver tissue decreased over 12hrs, however, the level was recovered after 48hrs. Tetrachloro-1,4-benzoquinone (1,4-TCBQ), one of the most reactive PCP metabolites, conjugated with GSH very rapidly. Base on our results, we could conclude that PCP metabolized to reactive electrophilic metabolites by cytochrome P-450 isoenzymes and conjugated rapidly with neighboring protein or nonprotein sulfhydryl before reacting with DNA or hemoglobin. We propose that albumin adducts and mercapturic acids of PCP metabolites can be used good biomarker of recent PCP exposure.

닥 펄프 제조공정 자동화에 따른 단위공정 폐수의 특성 (Characteristics of wastewater from unit systems of automative process for manufacture of paper mulberry pulp fibers)

  • Paper mulberry fibers have been used as fibrous raw materials for manufacturing traditional handmade paper, hanji for a long time. Compared to wood pulp fibers, pulp fibers from mulberry bast tissue have some benefits in physical and chemical properties due to their high D.P (degree of polymerization), M.W (molecule weight) and long fiber length. Specially, Korean handmade hanji shows outstanding characteristics in mechanical tensile strength, folding endurance, flexibility and long sustainable conservation properties. Therefore, hanji is widely applied to daily supplies, hygienic goods, medical supplies, clothing industries and so on. Recently, the potential demand of mulberry pulp fibers is more and more increased on the strength of high application fields. This study was focused on the possibility of wastewater recycling in unit operation systems for the development of automated mass production line. The properties and environmental loads of wastewater from debarking, cooking, bleaching and screening process were analyzed by means of COD, conductivity, turbidity and solid materials. The wastewater from debarking and cooking process was comparatively high in pollution load, and would be treated by additional approaches of chemico-physical method.

포항 분지의 제3기 장기층군에서 산출된 규화목의 산출상태와 구성광물 (The Mode of Occurrence and Composing Minerals of Petrified Woods from the Tertiary Janggi Group of Pohang Basin in Korea)

  • 경북 포항의 장기층군에서 산출된 규화목의 산출상태로부터 판단하면 규화목은 모두 이지성 화석임을 지시한다. 장기층군에는 규화목과 함께 탄화목, 목탄도 발견되며, 일부의 규화목에는 껍질부분에 갈탄화된 유기물이 일부 남아있는 것도 있다. 규화목의 X선 회절 분석으로부터 Opal-CT, Opal-C, 석영+크리스토발라이트의 3가지 유형이 확인되었다. 이러한 광물의 존재는 규화작용이 비교적 낮은 온도에서 일어났음을 지시한다. 실리카($SiO_2$)의 주공급원은 응회암으로 추정된다. 규화목의 구성광물의 종류와 조직 보존 상태로부터 판단하면 규화작용은 주로 치환에 의해 일어난 것으로 추정된다.
