Study on the sexing of preimplantation mouse embryo exposed to H-Y ntisera I. Sexing of mouse embryos by cytolytic assay (H-Y항체에 의한 생쥐초기배의 성판별에 관한 연구 I. 세포발육능검사에 의한 성판별)

  • There studies were conducted using inbred ICR mice to examine the sex of preimplantation mouse embryo. The morphological normality of mice embryos treated with the culture medium containing rat H-Y antiserum(10%, v/v) plus complement(20%,v/v) was observed and also the sexing of embryos was investigated by chromosomal analysis. The results obtained were summarized as follows: 1. The viability of preimplantation mouse embryos, which were incubated in vitro with different media condition, was scored 68.9-85.5% in control group. However, 151 embryos normally developed up to blastocyst and 160 embryos were retarded growth or destroyed out of total 311 embryos treated in the medium containing H-Y antiserum(10%, v/v) plus complement(20%,v/v). 2. H-Y antiserum was prepared from inb red rats (Wistar and Donryu strain) with different immunization times (4, 5 and 6th) to examine the specific titer of embryos by the number of immunization. Precentage of normally developed embryos incubated either in the medium containing the antiserum of Wistar plus complement or Donryu plus complement was revealed 50.9, 47.4 and 50.0% (4, 5 and 6th immunization and 47.8, 41.2 and 48.7%, respectively. 3. Twenty two females and five males were identified out of fourty-eight normally developed embryos incubated in the medium containing H-Y antiserum plus complement by chromosomal analysis.

Effect of Marine Protein Supplementation on Lipid Profile of Growing Rats Compared to Soybean Protein and Casein

  • Although beneficial effects of dietary plant proteins on lipid metabolism are well documented, not much information exists on the influence of different seafood proteins on the lipid metabolism. The present study evaluated the effect of 2 marine proteins (tuna protein and scallop ovary proteins) in comparison to casein and soy protein in male Wistar rats. The concentration of total lipids in the plasma of rats fed experimental diets was significantly lower from that of control (278.2 mg/dL) group (p<0.05); and, the liver lipid content was not significantly different (p>0.05). Fecal excretion of cholesterol and bile acids was significantly higher in marine proteins and soy protein fed groups compared to casein only fed control (6.1 and 6.4 mg/day, respectively) group (p<0.05). No significant differences were observed in the mRNA concentrations of different transcriptional factors (p>0.05).

The Effect of Cyclosporin A and Methylprednisolone on the Epithelial Regeneration of the Allograft Rat Tracheal Segments (Cyclosporin A와 methylprednisolone이 이인자형이식된 쥐기관의 상피조직재생에 미치는 영향에 대한 연구)

  • This study examined the effects of cyclosporin A [CsA] and methylprednisolone[MP] on the viability of the devascularized trachea after heterotopic transplantation. Fourty-two tracheal segments were harvested from 21 donor Wistar rats. Those tracheal segments were heterotopically implanted into the abdomen of recipient rats after wrapping in omentum. Heterotopical implantation was performed in six groups of rats:Group I was Wistar syngeneic controls, and five groups of Sprague Dawley recipients, receiving no immunosuppression[Group II], CsA alone[Group III, V], and CsA in combination with MP[Group IV, VI]. After 14 days, the tracheal segments were histologically evaluated.Epithelial thickness and the degree of epithelial regeneration were significantly different between group I and group II, III, VI, VI [p< 0.05]. There were significant differences in the epithelial thickness between group II, III, IV and group V, VI. In the degree of epithelial regeneration, there were significant differences in group II, group III-IV, and group V-VI. Without immunosuppression there was virtually no epithelium, whereas low-dose immunosuppression yielded intermediated viability, and with high dose CsA and MP we observed improved tracheal viability. Our results suggest that optimal combination of CsA and MP may improve the viability in heterotopic tracheal allografts.

Effects of ANTORIN R-10 on Ovarian Morphology, Follicular Development and Serum Estradiol Level in Immature Wistar Rats (미성숙 Wistar 랫트에서 ANTORIN R-10이 난소형태, 난포발육 및 혈중 호르몬 농도에 미치는 영향)

  • This study evaluated the effect of ANTORIN R-10(pFSH), a commercially available follicle stimulating hormone on ovarian morphology, on follicular development and serum estradiol levels in rats. Immature female Wistar S/T rats(27 day old; 80-100 g B.wt) maintained under controlled environmental conditions($22{\pm}2^{\circ}C$; 50% humidity; 12 h light/12 h dark cycle) with free access to standard laboratory feed and tap water were utilized. Animals were allowed to acclimatize to the new environment for at least 2 weeks before being included in the experiment. Rats were randomly allotted to 5 groups(Control, SL 0.1AU, SH 0.2AU, TL 0.1AU and TH 0.2AU). ANTORIN R-10 was subcutaneously injected twice daily for 3 days. Twenty hours after hormone treatment, blood was collected to estimate the serum estradiol $17-\beta$ concentration. Immediately, all rats were sacrificed and the ovarian morphology, ovary weight and number of follicles were recorded. Ovaries were fixed for histomorphological examination. Higher standard and treatment groups were significantly increased on ovary weight and the number of follicles more than 1mm compared with lower standard and treatment. However, no difference revealed between standard and treatment groups. ANTORIN R-10 was similar effects of follicles development and maturation compared with House standard FSH.

Preliminary Mechanistic Study on the Trachea Smooth Muscle Relaxant Activity of Aqueous Leaf Extract of Tridax Procumbens in Male Wistar Rats

  • Objectives: Aqueous leaf extract of Tridax procumbens (ALETP) has potent relaxant activity. However, this relaxant activity in respiratory smooth muscle remains uninvestigated. This study investigates the effect of ALETP on the contractile activity of tracheal smooth muscle (TSM) in adult male Wistar rats. Methods: Twelve male Wistar rats divided into 2 groups and were treated with either 100 mg/kg of ALETP (ALETP treatment group) or vehicle (distilled water; control group) through oral gavage for 4 weeks. Dose responses of TSM from the 2 groups to acetylcholine (10-9 to 10-5 M), phenylephrine (10-9 to 10-5 M), and potassium chloride (KCl; 10-9 to 10-4 M) were determined cumulatively. Furthermore, cumulative dose responses to acetylcholine (10-9 to 10-5 M) after pre-incubation of TSM with atropine (10-5 M), L-NAME (10-4 M), indomethacin (10-4 M), and nifedipine (10-4 M), were determined. Results: Treatment with ALETP substantially inhibited TSM contraction stimulated by cumulative doses of acetylcholine, phenylephrine, and KCl. Furthermore, preincubation of TSM from the 2 groups in atropine significantly inhibited contractility in TSM. Incubation in L-NAME and indomethacin also significantly inhibited contractility in TSM of ALETP-treated rats compared to that of controls. Contractile activity of the TSM was also inhibited significantly with incubation in nifedipine in ALETP-treated rats. Conclusion: ALETP enhanced relaxant activity in rat TSM primarily by blocking the L-type calcium channel and promoting endothelial nitric oxide release. ALETP contains agents that may be useful in disorders of the respiratory tract.

Synergistic interaction between acetaminophen and L-carnosine improved neuropathic pain via NF-κB pathway and antioxidant properties in chronic constriction injury model

  • Background: Inflammation is known to underlie the pathogenesis in neuropathic pain. This study investigated the anti-inflammatory and neuroprotective mechanisms involved in antinociceptive effects of co-administration of acetaminophen and L-carnosine in chronic constriction injury (CCI)-induced peripheral neuropathy in male Wistar rats. Methods: Fifty-six male Wistar rats were randomly divided into seven experimental groups (n = 8) treated with normal saline/acetaminophen/acetaminophen + L-carnosine. CCI was used to induce neuropathic pain in rats. Hyperalgesia and allodynia were assessed using hotplate and von Frey tests, respectively. Investigation of spinal proinflammatory cytokines and antioxidant system were carried out after twenty-one days of treatment. Results: The results showed that the co-administration of acetaminophen and L-carnosine significantly (P < 0.001) increased the paw withdrawal threshold to thermal and mechanical stimuli in ligated rats compared to the ligated naïve group. There was a significant (P < 0.001) decrease in the levels of nuclear factor kappa light chain enhancer B cell inhibitor, calcium ion, interleukin-1-beta, and tumour necrotic factor-alpha in the spinal cord of the group coadministered with acetaminophen and L-carnosine compared to the ligated control group. Co-administration with acetaminophen and L-carnosine increased the antioxidant enzymatic activities and reduced the lipid peroxidation in the spinal cord. Conclusions: Co-administration of acetaminophen and L-carnosine has anti-inflammatory effects as a mechanism that mediate its antinociceptive effects in CCI-induced peripheral neuropathy in Wistar rat.

  • Scanning electron microscopic observation of the oviductal epithelium was carried out on the virgin white Wistar rat during normal sexual cycle, and obtained the results as follows; Rat oviductal epithelium during the normal sexual cycle generally consisted of two types of cells: ciliated and non-ciliated cell. The ciliated cells had cilia which was measured $0.2{\mu}$ in width and $3-5{\mu}$ in length. According to the length of microvilli and the distribution in each segment, the non-ciliated cell was further subdivided into four types of cells; NC-I, NC-II, NC-III, and NC-IV. The morphologic characteristics of each type of cells were also ascertained by SEM observations.

  • This study observes the change of small intestine mucosa paneth cell by changing the amount of radiation to rat. It uses the rat(Wistar) of 250-300g as the experimental animal and irradiation equipment is Gammacell 3000Elan System. and the irradiation is conducted for 500Rad group for 34sec., 1000rad for 68sec., and 1500Rad for 102sec. once on the whole body of each group, eachgroup is anesthetized with ether after 24hours. its small intestine is extrated and then it is observed by transmission electronic microscopy. The experimental results are as follows : 1. 500 Rad Group The Slightly elongated form of mitochondria and rough endoplasmic reticulum are observed in 500 Rad group. 2. 1000 Rad Group Golgi apparatus is appeared as the extended plasmodium, secretory granules exist only external membrane due to the self-fusion, the number of mitochondria that are changed as L-type are reduced, rough endoplasmic reticulum is distributed with the expanded form. 3. 1500 Rad Group The number of Golgi apparatus and granules is remarkably reduced, mitochondria is changed into C-type and free ribosomes can be observed instead of the reduction of rough endoplasmic reticulum.

  • This study was to investigate the biomechanical characteristics of isoflavone-treated ovariectomized growing rat's femur. 4-week-old Wistar (female rats were randomly divided into 5 groups and assigned to sham(SH) group, and four ovariectomized groups consisting of one ovariectomized(OVX) group and genisetin(10 mg/kg b.w. )(OVX+G), daidzein(10 mg/kg b.w.)(OVX+D), and 17 $\beta$-estradiol(10 $\mu$g/kg b.w.)(OVX+ES) in AIN-76 modified diet for 8 weeks. The OVX groups gained more body weight than the SH group. Femoral weight was increased by feeding genistein and estradiol, whereas femoral length among groups was not significantly different. The breaking farce, stiffness, deformation, and energy values of the OVX group were lower than those of other groups. The stress, strain, and Young's modulus values of the OVX group were higher than those of other groups.

Altered Cardiac $Na^{+}$,$K^{+}$-ATPase Activity in Prehypertensive Spontaneously Hypertensive Rat

  • $Na^{+}$,$K^{+}$-ATPase activity, $Na^{+}$-dependent phosphorylalion, and [$^3$〕ouabain binding in sarcolemma prepared from 4 week old spontaneously hypertensive rat(SHR) ventricles were compared to the same parameters in sarcolemma from age matched normotensive Wistar-Kyoto(WKY) rat ventricles to examine whether the reduced myocardial $Na^{+}$-pump activity in SHR is an inherited enzymatic defect or a second phenomenon due to sustained hypertension. The total body weights, ventricular weights, and blood pressures were the same for SHR and WKY. No significant differences in sarcolemmal protein content and protein recovery were noted between the two pups. Sarcolemma isolated from SHR ventricles showed sigificantly less $Na^{+}$,$K^{+}$-ATPase activity and number of phosphorylation sites when compared to snrcolemma 1mm the WKY ventricles. Equilibrium binding of [$^3H$〕ouabain and the turnovcr number of myocardial $Na^{+}$,$K^{+}$-ATPast however, wee the same for both groups. These results. indicate that the low affinity(${\alpha}$, or ${\alpha}_1$) isoform for ouabain is reduced in SHR compared to WKY but that the high affinity(${\alpha}$+, or ${\alpha}_2$) isoform is the same in ventricles of SHR and WKY. The reduced amount of at isoform of the $Na^{+}$,$K^{+}$-ATPasc in prehypertensive SHR ventricles may play some pole in the development of hypertension.
