처치온도에 따른 초미분화 산양삼의 홍삼화 연구

  • Objectives : The aim of this study was to find optimal conditions for producing red ginseng from cultivated wild ginseng using the Turbo Mill. Methods : Characteristics of powdered cultivated wild ginseng based on various temperature settings of the Turbo Mill were observed, and changes in the content was measured by HPLC for various ginsenosides. Results : 1. The diameter of cultivated wild ginseng powder ground by the Turbo Mill was around 10${\mu}m$. 2. As the temperature rose, pressure, Specific Mechanical Energy(SME), and density decreased, whileas Water Solubility Index(WSI) increased. 3. As the temperature rose, super fine powder showed tendency to turn into dark brown. 4. Measuring content changes by HPLC, there was no detection of ginsenoside Rg3 and ginsenosideRg1, Rb1, and Rh2 concentrations decreased with increase in temperature. Conclusions : Super fine powder of cultivated wild ginseng produced by the Turbo Mill promotes easy absorption of effective ingredients by breaking the cell walls. Using this mechanism to produce red ginseng from cultivated wild ginseng, it yielded less than satisfactory results under the current experiment setup. Further researches are needed to verify more suitable condition for the production of red ginseng.

  • Objectives : This study was carried out to examine protective effect of wild ginseng extract on HepG2 human hepatoma cell line against tert-Butyl hydroperoxide (t-BHP)-induced oxidative damage. Methods : To evaluate protective effect of wild ginseng extract against t-BHP induced cytotoxicity, LDH level and activity of glutathione peroxidase and reductase were measured. Gene expression was also measured using DNA microarray. Results : Wild ginseng extract showed a significant protective effect against t-BHP-induced cytotoxicity in HepG2 cell line. It is not, however, related with the activities of glutathione peroxidase and glutathione reductase. Analysis of gene expression using DNA chip, demonstrated that 28 genes were up-regulated in t-BHP only group. Five genes - selenoprotein P, glutathione peroxidase 3, sirtuin 2, peroxiredoxin 2, serfiredoxin 1 homolog - may be related with the protective effect of wild ginseng extract. Conclusions : Based on the results, a protective effect of wild ginseng extract against t-BHP-induced oxidative damage in HepG2 cell line is not associated with the activities of glutathione peroxidase and glutathione reductase, but with the expression of selenoprotein P, glutathione peroxidase 3, sirtuin 2, peroxiredoxin 2, and serfiredoxin 1 homolog.

Monitoring of diseases in wild marine fish stocks collected in June 2006 by a trawl in the Southern Korean Waters (2006년 6월에 트롤어법으로 어획된 남해안 자연산어류에 대한 질병 모니터링)

  • Viral and bacterial pathogens of the wild marine fishes were monitored in 176 wild fish and 15 wild shrimp from 13 and 1 species, respectively, which were captured by a trawl net in the southern sea of Korea during June 2006. Viral pathogens that are common etiologically agents to cultured fish in Korea were not isolated. One and 5 bacterial strains were affiliated to the genus Proteus and Pseudomonas, respectively, but these bacteria do not seem to be associated with mortality of aquacultural fish. An extended monitoring on wild marine fishes were necessary for identification of agents responsible for the cultured fish infections.

Fermentation Characteristics of Wild Grape (Vitis amurensis) Wine Prepared with Reed (Phragmites communis) Root (갈대뿌리를 첨가하여 제조한 산머루주의 발효 특성)

  • This study investigated the fermentation characteristics of wild grape(Vitis amurensis) wine prepared with reed(Phragmites communis) root. The reed root was added to the fermentation of wild grape wine in the quantities of 0.5%, 1.0% and 2.0%. An addition of reed root during yeast fermentation of wild grape wine decreased the acidity and increased pH of fermentation culture, thus enhancing the consumption of sugar and production of ethanol. Especially, the addition of 2% reed root led to production of about 13% ethanol in 4 day of fermentation time. The sensory evaluation showed that the wild grape wine fermented with 2.0% reed root was most acceptable.

  • For this paper, a study targeting 9 forest patches in Dangjin to suggest for the wild birds, was conducted. The correlation between the emergence of wild birds and the forest vegetation was analyzed, and the patterns of forest vegetation for the improvement of species diversity were suggested by classifying wild birds and foraging guild by the type of forest vegetation. As for the characteristics of the wild birds emerged, more species and bigger population appeared in the vegetation type of mixed stand forest where the natural deciduous broadleaf trees and evergreen needleleaf trees coexisted in the large scale vegetation area. Thus, it was known that the advent of summer migratory birds and resident birds were affected the most by forest patches and vegetation types. As for foraging guild, the larger the forest paches the more the species and the population of the wild birds inhabiting in shrubs and bushes.

Fatty Acid and Amino Acid Compositions of Dodok (Codonopsis laceolata) (더덕의 지방산 및 아미노산 조성)

  • In the present study, proximate compostions, fatty acid and amino acid compositions of wild and cultivated Dodok (Codonopsis laceolata B. et H.) were analysed. The contents of crude ash and crude protein were higher in the cultivated Dodok than in the wild Dodok. The main fatty acids in the total lipid, free lipid and bound lipid of wild and cultivated Dodok were linoleic acid, palmitic acid and followed by linolenic acid. In the case of wild Dodok, numerous unknown peaks were appeared significantly. The amino acids analyzed in wild aid cultivated Dodok were lysine, histidine, arginine, aspartic acid, threonine, serine, glutamic acid, proline, glycine, alanine, valine, methionine, isoleucine, leucine, tyrosine and phenylalanine. Arginine was the predominant amino acid in both wild and cultivated Dodok.

New polymorphic microsatellite markers in the Korean mi-iuy croaker, $Miichthys$ $miiuy$, and their application to the genetic characterization of wild and farmed populations

  • Eighteen new polymorphic microsatellite markers were developed for the Korean mi-iuy croaker ($Miichthys$ $miiuy$, Perciformes, Sciaenidae), and allelic variability was compared between a wild population in Mokpo, Korea, and a hatchery population in Tongyeong, Korea. All loci were amplified readily and demonstrated allelic variability, with the number of alleles ranging from 5 to 37 in the wild population, and from 4 to 12 in the farmed population. The average observed and expected heterozygosities were estimated, respectively, to be 0.74 and 0.78 in the hatchery population samples, and 0.79 and 0.86 in the wild samples. These results indicate lower genetic variability in the hatchery population compared with the wild population, and significant genetic differentiation between the wild population and the hatchery samples ($F_{ST}$=0.058, P<0.001). These microsatellite loci may be valuable for future population genetic studies, monitoring changes in the genetic variation within stocks in a commercial breeding program, conservation genetics, and molecular assisted selective breeding of the mi-iuy croaker in the future.

Manufacture and Quality Characteristics of Wild Grape Wine (머루 발효주의 제조 및 품질 특성)

  • To develop a new functional wine, manufacture and quality characteristics of wild grape wine were investigated. Ethanol contents of wild grape wine was 11.0%, and total acidity and volatile acid content were 0.63% and 0.0402%, respectively. Antioxidant activity of the wild grape wine which is related in anti-aging functionality, was 41.6%. From this results, we concluded that the wild grape wine is a high valuable wine.

Genetic Variation in Korean Populations of Wild Radish, Raphanus sativus var.hortensis f. raphanistroides (Brassicaceae)

  • Raphanus sativus L. var. hortensis f. raphanistroides (wild radish: Brassicaceae), a herbaceous perennial, occurs only on beaches in East Asia. Genetic diversity and population structure of seven Korean populations were investigated using starch gel electrophoresis. Although the Korean populatins are small, isolated with patchy distribution, the population maintain a moderate level of genetic diversity; the mean percentage fo polymorphic loci was 51.4%, mean number of alleles per locus was 1.84, and mean expected heterozygosity was 0.116. A combination of animal-outcrossing breeding system, wide geographical distribution, restricted ecological distribution, and a propensity for high fecundity may in part be explanatory factors contributing the moderate level of genetic diversity within populations. An overall excess of homozygotes relative to Hardy-Weinberg expetations (mean FISa=0.116) indicates that consanguineous mating occur within wild radish populations, leading to a family structure within a circumscribed area. Although population of wild radish experience a limited gene flow, only 5% of the total genetic variation found in Korean wild radish populations examined is due to differences among populations (mean GST=0.052). This value is considerably lower than the mean values of species with similar life history and ecological characteristics. However, significant differences were found in allele frequencies between populations for all polymorphic loci (P<0.01). It is supposed that directional selection toward genetic uniformity (similar gene frequencies) in a relatively homogenous habitat is thought to be operated among Korean wild radish populations.

Optimal Amount of Seeding and Wild Herbaceous Plants for the Rooftop Revegetation (옥상녹화용 야생초본류와 적정 파종량에 관한 연구)

  • The purpose of this study is to select the appropriate wild herbaceous plants on the rooftop. For the purpose of the experiments, 14 wild herbaceous plants were chosen. As the results of the experiments, the wild herbaceous plants growing on the culture soil(perlite) Among the 14 kinds of wild herbaceous plants, such as Chrysanthemum boreale, Taraxacum mongolicum, Aster Koraiensis, Aster yomena, Oenothera odorata, Oenothera lamarckiana, Patrinia scabiosaefolia showed good effects on growth of above ground parts. Therefore, these plants will effective for the rooftop revegetation. Callistephus chinensis and Lotus corniculatus var. Japonicus showed good height growth, wherase covering rate was worst. Chrysanthemum boreale, Platycodon grandiflorum, Patrinia scabiosaefolia were plant height, wherase covering rate was not good . For the expected number of seedings more than 3,000 per square meter, many seedings got withered to death while the survivors were suppressed to grow slow in the early stage due to the densityproblem.
