  • Leucas aspera Link. (Lamiaceae) is an important medicinal plant in indigenous systems of medicine in India and commonly known as 'Thumbai'. It has various ethnomedicinal values as various traditional communities find diverse medicinal properties. It is used as antipyretic, stimulant, expectorant and diaphoretic drug. The present communication deals with the detailed pharmacognostical evaluation of whole plant of L. aspera collected from five different geographical zones of the country-Uttar Pradesh, Orissa, Karnataka, West Bengal and Gujarat. The botanical characters and TLC fingerprint profile of all the samples were quite similar but some variations were observed in physicochemical parameters. However, some microscopical characters and TLC profile can be used as diagnostic characters for identification of L. aspera, for example amphistomachic leaves and two types of trichomes-abundant, non glandular, uniseriate, 1-3 celled and few glandular, 2-5 celled stalk with rounded tip. Presence of some components at $R_fs-0.56$, 0.65 and 0.76 under UV 366 and at $R_fs-0.31$, 0.43, 0.60, 0.76 and 0.82 under visible light after derivetization in TLC profile may also be used as diagnostic character.

Effects of Pre-wilting Treatment and Formic Acid Addition on Quality of Silage Made from Spring Sown Oat Crops (봄재배 연맥의 Silage 제조 이용시 예건 및 Formic Acid 처리에 의한 품질개선 효과)

  • Spring sown oat crops were grown during early March to late June in 1993~1994. The crops were harvested at different stage of maturity to determine the yield performance, chemical components and net energy value of the plants for silage utilization. The pattern of dry matter accumulation, absolute growth rate(Abs. GR), leaf weight ratio(LWR), stalk diameter(SD) and other growth parameters were analysed to evaluate the suitability of the plant materials for ensiling. The effect5 of wilting treatment and formic acid addition on the quality of silage was also disscused during the experiments. Spring sown oat crops was evaluated as a suitable materials for whole plant silage making. The best qualitative oat silage was obtained at the stage of soti dough to hard dough, when the plant reached 32.2~35.7% in dry matter concentration. Spring sown oat produced 9.43 ton in DM basis, 5.79 ton in TDN and 57.550 MJ-NEL/ha in net energy lactation. Early harvesting of the oat corps needed wilting treatment of the material before ensiling. Under pre-witing treatment (1/2 day) and formic acid addition(0.3%) silage quality point of oat crops harvested at the bloom stage was improved from 58 to 84 point.

Bulk 건조엽의 편평엽 발생양상과 이화학적 특성

  • The occurrence and physico-chemical properties of flat leaves found in 1997 curing trial related to the prevention of occurrence of flat leaves at bulk curing of flue-cured tobacco were investigated and compared with those of normal leaves. Cured leaves of NC82 were separated into 4 classes of none, slight, fair and severe symptoms of flat leaf by the percentage of flattened parts to whole leaf area. The flat leaves were mostly found among the leaves of lower stalk position, and it was estimated that growth rate of lower leaves also influenced on the occurrence of flat leaves. In chromatic aberration of cured leaf, flat leaves showed remarkably lower b and L values than in those of normals but there was no difference in a value. On the other hand, in chemical analyses of flat leaf samples, nicotine, total sugar, ether extract and total nitrogen contents were decreased with the degree of flat symptoms. In physical properties, filling capacity of cured leaves was decreased with the degree of flat symptoms comparing with those of normal leaves, while shatter index was showed a reverse tendency, and then within the same leaf, flat parts were decreased in total sugar, ether extract and filling capacity compared with those of normal ones, but remarkably increased in shatter index, and there was no difference in nicotine and total nitrogen contents.

Morphological and Histochemical Study on the Anterior Tentacular Antenna of a Korean Slug, Incilaria fruhstorferi (한국산 산민달팽이 ( Incilaria fruhstorferi ) 전촉각의 형태 및 조직화학적 연구)

  • A morphological andk histochimical study on the amterior tintacular antenna of Korean sulg, Incilaria fruhstorferi was conducted under the light microscopic observations. The histological sturctures of the antenna were apparently divided into three parts such as the epithelium, the connective tissues and the muscular layers. The cells forming the antenna were classified into several types on the basis of their morphological and histochemical characteristics. The simple columnar epithelium cotering the whole antenna was composed of supporting cells, sensory neurons and type-a clear cells. The connective tissue was consisted of dispersed large cells, type-b clear cells and 7 types of secretory cills such as type-A, type-B, type-F, thpe-G, type-H, type-J and type-K. The large cells found in the form of group situated only in the stalk of the antenna. The large cells possessed relatively small nuclei as compared with their cytoplasm. The cytoplasm positively reacted upon alcian blue, and the nucleus was PASpositive. The type-a and type-b clear cells which were irregular in shape showed no evident reaction against various stains employed in the present study. The secrtory cells were observed mainly in the connective tissues and in the muscular layers. Histochemical components of the type-A, type-B and K were identified as acid mucopolysaccharides and those of type-F and H were neutral mucopolysaccharides. The muscular layders supporting the epithelium possessed the type-B and F secretory cells which were also observed in the connective tissues.

Continuous Production of Phalaenopsis Clones by Basal Shoot Culture (호접란 줄기기저부 절편배양을 통한 조직배양묘의 연속생산)

  • This study was conducted to establish a practical masspropagation system of Phalaenopsis clones from basal shoot segments. The frequency of PLB (protocorm like body) induction was compared with various explants. Basal shoot segments showed the most successful result of 45%, while root tips, stalk node segments, stalk leaves and mature leaves represented low frequency (below 5%). The PLB induction ratio in the culture of basal shoot segments was examined with 11 different Phalaenopsis varieties, and the majority of varieties, including pink flower lines, showed an about 30% rate of PLB formation. Especially, when whole basal shoot parts without cutting were inoculated onto PLB induction medium, giant PLB was induced from explant. This giant PLB was green color and big in size compared with normal PLB. When dissected giant PLB segments inoculated onto PLB multiplication medium, only normal size of PLBs were induced from them. PLBs induced by basal shoot culture were transferred onto proliferation medium and then shooting medium, from which normal plants were formed. Therefore, this culture method is considered as effective and practical protocol for Phalaenopsis mericlone production. In addition, it is suggested that clones of an infinite number can be produced consecutively by this culture system through repeated cycles of PLB induction and proliferation using the basal shoot segment of flask plant.

Gray Mold of Kalanchoe (Kalanchoe blossfeldiana) Caused by Botrytis cinerea in Korea (Botrytis cinerea에 의한 칼란코에 잿빛곰팡이병)

  • In April of 2002, the gray mold disease occurred severely on kalanchoe (Kalanch oe blossfeldiana) grown in a nursery in Daesan-myon, Changwon City, Korea. The lesions were started with water-soaked spot and the leaves discolored from the tip and became gray or dark and gradully expanded to whole leaf. The conidia and mycelia of pathogen appearedd on flower, flower stalk and stem. Conidia were gray, 1-celled, mostly ellipsoid or ovoid in shape and were 8~17 ${\times}$5~12${\mu}m$ in size. Conidiophores were 13~34${\mu}m$ in size. The sclerotia were formed abundantly on potato-dextrose agar. The optimum temperature for sclerotial formation was $20^{\circ}C$. Pathogenicity of the causal organism was proved on Kalanchoe blossfeldiana according to Koch,s postulate. The causal organism was identified as Botrytis cinerea based on mycological characteristics. This is the first report on gray mold of Kalanchoe(Kalanchoe blossfeldiana) caused by Botrytis cinerea in Korea.

Nitrogen Use and Yield of Silage Corn as Affected by Hairy Vetch(Vicia villosa Roth) Soil-incorporated at Different Time in Spring

  • Winter green manure crops including legume increase grain yield of subsequent crop and substitute N fertilizer requirement with organic-No Hairy vetch grows vigorously and can provide N-rich green manure for corn with its soil incorporation after wintering. But, grain yield of corn as succeeding crop would be reduced if its planting time is delayed until late spring. This experiment was carried out to find the proper incorporation time of hairy vetch green manure and planting time of subsequent corn in cropping system with winter hairy vetch(green manure)-summer corn. Hairy vetch was incorporated into soil at a ten-day interval between April 10 and May 10 and corn was planted at 5 days after each hairy vetch incorporation. Soil nitrate concentration on April 10 and 20 in hairy vetch plot was slightly lower than that at winter fallow. Above-ground dry matter and organic-N of hairy vetch increased linearly with delayed hairy vetch incorporation time from April 10 to May 10. Average dry matter and organic-N produced by hairy vetch were 5.7 ton/ha and 248 kgN/ha, respectively. Corn growth and yield decreased as delayed corn planting time after May in spite of increasing dry matter and N-yield of hairy vetch. Nitrogen concentration of corn grain, stalk and whole plant at harvest were the highest in May 5 planting, but total N-uptake of May 5 planting were not different from that of April 25 planting because of lower grain yield. It was concluded that the proper incorporation time of hairy vetch and corn planting time were April 20 and April 25, respectively, because grain yield was the highest and corn could use hairy vetch-N effectively to produce dry matter.

Colorimetric and Chemical Characteristics of Grey Leaf Tobacco (Grey 증상 잎담배의 색채 및 화학성분 특성)

  • This experiment was conducted to get the fundamental information about the difference in color and some chemical components of grey flue-cured tobacco cultivated in paddy field to compare with normal tobacco. Cured leaves of NC82 were separated with visual characters into three classes of slight, fair and severe symptoms of grey leaf by percentage of grey parts to whole leaf area, and compared to normal ones. Occurrence of grey leaves tended to be higher in lower stalk position than in upper ones. Cl, Mn and Fe content of grey leaves was much higher when it compared with normal leaves, and within the same leaf, grey parts were increased in Fe, Mn and Cl content than in those of normal parts but there was no appearent difference in total sugar, nicotine and Ca contents. Chemical analysis of grey leaf samples indicated that they contained more Fe, Mn and Cl contents than those of normal leaves. These tendencies in chemical traits were accordance with the degree of grey symptoms. Grey leaves showed remarkably lower b*value and slightly lower L*value than those of normal leaves, while a*value was mostly higher in grey leaves. In general, grey appearence of grey flue-curd tobacco seemed to be from less yellowish color, poorer brightness with more brownish or reddish color.

Occurrence of Sword bean Scab Caused by Cladosporium cucumerinum in Korea

  • A black scab disease occurred on sword bean (Canavalia gladiata) in plastic film houses around Chinju area during the spring season of 1999. The disease started from flower bud, then moved to flower stalk, pod, petiole, cirrus, stem and leaves. The lesions started with small dark brown spots then were gradually expanded. Severely infected plants reached 37.4% of whole plant covered with scab. Numerous conidia were produced on the diseased flower disk, pod, floral axis, stem and leaves. Most of the conidia were appeared to be readily dispersed in the air, but the mycelia were not suggested causing of sooty mold by ectoparasitism. A fungus was isolated from the diseased stem, and inoculated to healthy plants to satisfy the Koch's postulates and proved the fungus was the causal agent of the disease. The isolated fungus grew on potato dextrose agar, forming greenish black to pale brown colonies. Conidia were ellipsoidal, fusiform or subspherical, mostly one-celled but occasionally septated. The conidia were $3.9{\sim}34.1{\times}2.7{\sim}5.1\;{\mu}m$ in size and formed in long branched chains on the erected conidiophores which were pale olivaceous brown and variable in length between $7.2{\sim}210.7\;{\mu}m$ in size. Ramoconidia were $7.6{\sim}29.2{\times}3.2{\sim}14.4\;{\mu}m$ in size. The fungus was identified as Cladosporium cucumerinum based on the above morphological characteristics. The optimum temperature for mycelial growth and conidial formation was about 15 to $25^{\circ}C$. Cladosporium scab of sword bean caused by the fungi has not been reported in Korea previously.

An Aspect of Occurrence and Chemical Properties of Grey Leaf Tobacco (Type III) (연초 III형 Grey엽의 발생양상과 화학성분 특성)

  • The occurrence and chemical properties of grey tobacco leaves (Type III) found in 1995 crop of flue-cured tobacco cultivated in paddy field were investigated to compare with those of normal leaves. Cured leaves of NC82 were separated by visual characters into 3 classes of slight, fair and severe symptoms of grey leaf by the percentage of grey parts to whole leaf area. Number of samples classified with discoloring portion was the order of greyish brown > reddish brown > greyish yellow, respectively. Grey leaves of this type were mostly found among the leaves harvested from upper stalk position, and it was estimated that growth rate of upper leaves also influenced on the occurrence of grey leaves. Grey leaves showed remarkably lower b* and L* values than those of normal leaves, while a* value was mostly higher in grey leaves. These tendencies in chromatic aberration showed more remarkable difference in the degree of grey symptoms. Chemical analyses of grey leaf samples indicated that they contained less total nitrogen and nicotine, and more total sugar and starch than those of normal leaves. In chemical traits, these tendencies were accordance with the degree of grey symptoms, and within the same leaf, grey parts were decreased in total nitrogen and ether extract content compared with those of normal parts, but there was no difference in nicotine and Cl contents.
