Effects of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini Extracts on Melanin Synthesis in Melanoma Cells (애기수영, 방가지똥 및 암대극 추출물이 Melanoma Cell에서 멜라닌 합성에 미치는 영향)

  • In this study, we investigated the effect of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini extracts on tyrosinase activity and melanin production as natural products of whitening functional cosmetics. To measure the melanin production, 50, 100, $200{\mu}g/mL$ of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini extracts were treated on ${\alpha}-MSH$ treated B16F10 melanoma cells, respectively. Melanin contents in ${\alpha}-MSH$ treated B16F10 melanoma cells were decreased by 41.5, 51.11, and 61% in $200{\mu}g/mL$ treatment compared to none treatment, respectively. In addition, the intracellular tyrosinase activity was decreased after treatments with all extracts. Furthermore, $100{\mu}g/mL$ of Euphoibia jolkini extract was decreased 81.5% of melanin production in B16F10 melanoma cells. When the three extracts were compared, Euphoibia jolkini extract was considered to be the most functional material for whitening effect.

Antimelanogenic of Artemisia fukudo Makino Extract in Melanoma Cells (큰비쑥 추출물의 멜라닌 생성 억제 효과)

  • Melanin is one of the most important factors affecting skin color. Melanogenesis is the bioprocess of melanin production by melanocytes in the skin and hair follicles and is mediated by several enzymes, such as tyrosinase, tyrosinase related protein (TRP)-1, and TRP-2, MITF. In this study, we investigated the effect of Artemisia fukudo Makino extracts on tyrosinase activity and melanin production as natural products of whitening functional cosmetics. Melanin content in murine B16F10 melanoma cells were decreased by Artemisia fukudo Makino extracts in a dose-dependently. In addition, the inhibition of tyrosinase activity of Artemisia fukudo Makino extracts showed to decrease tyrosinase activity as the concentration of ${\alpha}-MSH$ was increased. Furthermore, western blot analysis revealed that Artemisia fukudo Makino extracts significantly downregulated the expression of tyrosinase, TRP-1 which treat of ${\alpha}-MSH-induced$ melanogenesis in murine B16F10 melanoma cells. As a result, Artemisia fukudo Makino extract showed functionalities as an effective whitening agent to inhibit melanin formation.

Inhibitory Efficacy of Dioscoreae Rhizoma on MITF, TRP-1, TRP-2, Tyrosinase, PKA and ERK Expression in Melanoma Cells (B16F10) (산약의 멜라노마 세포(B16F10)에서 MITF, TRP-1, TRP-2, Tyrosinase, PKA, ERK 발현 억제 효과)

  • Objectives : The purpose of this study was to research the whitening effects and developing by cosmetics of the extract fromDioscoreae Rhizoma, which is one of the most popular health-promoting herb in herbal medications.Methods : We performed tyrosinase inhibition assay, reverse transcription-polymerase chain reaction (RT-PCR) and western blot for whitening effects. Also we measured MTT assay for cell viability.Results : The results were obtained as follows : For whitening effect, tyrosinase inhibition rate of extract fromDioscoreae Rhizomashowed more than 42.28% at 1,000 ㎍/㎖ concentration. Cell toxicity effect on melanoma cells (B16F10) of extract fromDioscoreae Rhizomashowed 81.97% with toxicity at 50 ㎍/㎖ concentration. So we were measured at a concentrations of 5, 10 and 50 ㎍/㎖ in all experiments involving cell. In addition, whitening related mRNAs including microphthalmia associated transcription factor (MITF), tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2), tyrosinase were reduced byDioscoreae Rhizoma. We also foundDioscoreae Rhizomatransiently decreased protein kinase A (PKA) which is known to be upstream to the down regulation of MITF and tyrosinase. But phosphorylation of extracellular signal related kinase (pERK) were increased byDioscoreae Rhizoma. These results imply thatDioscoreae Rhizomadecrease melanogenesis via ERK activation and subsequent down regulation of MITF and tyrosinase.Conclusions : Therefore, all these findings suggested the potent usage ofDioscoreae Rhizomaas materials of functional cosmetics by confirming whitening activity related with melanin content.

Effects of the Sciripi rhizoma on Antioxidation and Melanogenesis (삼릉추출물이 항산화와 멜라노제네시스에 미치는 영향)

  • Whitening effect, which decreases the skin pigmentation, is the one of important targets in cosmetics. This study was investigated the effects of Scirpi rhizoma on ant ioxidation and melanogenesis. S.rhizoma is a rhizome of Scirpus fluviatilis G. a perennial Cyperaceae species of wide occurrence in Asia, Europe, Africa and North America. S.rhizoma shown scavenging activities of free radicals and reactive oxygen species (ROS) with the IC50 of 638${\mu}g/ml$ against 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and 21.7${\mu}g/ml$ against superoxide radicals in the xanthine/xanthine oxidase system, respectively. S.rhizoma treatment (48 h) suppressed the biosynthesis of melanin up to 27% and reduced tyrosinase activity up to 31% at 100${\mu}g/ml$ in B16 melanoma cells. S.rhizoma was also able to significantly inhibit tyrosinase and TRP-1 expres- sion in protein level. These results suggest that S.rhizoma inhibited melanin biosynthesis by regulating tyrosinase activity and expression in B16 melanoma cells. Therefore S.rhizoma may be useful as new whitening agent due to the antioxidant effect and the inhibitory effect against melanogenesis.

  • Oriental cosmetics make the skin radiant by supplement and circulate qihyul(氣血). Oriental cosmetics aim at the radiant skin based on inner health. It accords the new trend of 21th century, well-being and naturalism. Recently a lot of new oriental cosmetics have been developed and enjoyed great popularity. The research of oriental cosmetics elucidated the active compounds having whitening effect and wrinkle cure effect. And the anti-acne and anti-atopy cosmetics have been researched. The development of oriental cosmetics should be based on oriental medical theory.

Chemical Stability and Whitening Activity of Mixture of Functional Ingredients (미백 기능성 성분의 혼합물에 대한 미백 활성 및 안정성에 관한 연구)

  • Recently, arbutin, oil soluble licorice extact (GLY), ascorbyl glucoside (AA2G), and ethyl ascorbyl ether (EAE) have been widely used as functional whitening ingredients. To Investigate which combination between the above agents could be more effective for whitening effect, tyrosinase activity and MSH-induced melanin production in B-16 melanoma cells were investigated. Both GLY and arbutin dose-dependently inhibited purified tyrosinase activity. The inhibitory effects of GLY with AA2A or EAE on Drosinase activity were more potent than those of GLY alone, whereas that of arbutin with other ingreadients did not show those effects. In MSH-induced melanin production in B-16 melanoma cells, the mixture of Gly and EAE more significantly reduced melanin formation than Gly alone. Stability of mixture of GLY, arbutin, AA2A and EAE exposed at the temperature of $25^{\circ}C\;or\;45^{\circ}C$ for 30 days were also investigated. All of the combinations of whitening agents did not show any critical changes in their composition stability. These data suggest that the combination of GLY and vitamin C derivatives such as AA2G and EAE may be useful for the promotion of whitening effect.

The Antioxidant and Skin Whitening Effect of Withania somnifera (Winter Cherry) (윈터체리 추출물의 항산화 및 미백 개선 효과)

  • Withania somnifera has been used in folk medicine to treat various ailments for centuries. In this studies to investigate the whitening effect of Withania somnifera extracts as an active ingredient for whitening cosmetics, the antioxidant capacity and the effects of Withania somnifera extracts on melanogenesis in B16-F10 melanoma cells were identified. Withania somnifera extracts significantly reduced both tyrosinase activity and melanin content in a concentration-dependent manner. Furthermore, it was found that Withania somnifera extracts decreased ${\alpha}-MSH$ (melanocyte-stimulating hormone)-induced tyrosinase activity and MITF(microphthalmia associated transcription factor) protein expression. These data indicate that Withania somnifera extracts attenuate ${\alpha}$-MSH-stimulated melanin synthesis by modulating MITF expression and that they may be a useful therapeutic agent for treating hyperpigmentation and an ingredient of whitening cosmetics.

Inhibition of Tyrosinase by Metabolites Originating from Thrichoderma atroviride (Thrichoderma atroviride 배양액의 tyrosinase 억제제에 관한 연구)

  • In today's society, functional whitening cosmetics are important to beauty. Fungi are known to produce a variety of whitening-related metabolites. In this study, we searched for tyrosinase inhibitors with metabolic products derived from Trichoderma atroviride supernatant in order to apply a material for whitening functional cosmetics. In addition, the inhibitory effect was compared to arbutin, which has already been approved as a whitening raw material by the Korea Ministry of Food and Drug Safety (KMFDS). The metabolites from the T. atroviride supernatant showed higher tyrosinase inhibitory activity than that of arbutin. Some of the tyrosinase inhibitors were stable to heat, whereas some were unstable. The heat unstable material was exhibited in the case of samples treated with little amounts, such as 0.02~0.2%. They were very unstable in acidic and alkali pHs, especially under acidic conditions. However, it was found that a weakly-acidic to neutral pH range was the optimal working pH, especially neutral pH. Since the activity of the inhibitory substances in the T. atroviride supernatant was maintained regardless of proteinase K treatment, it was assumed that the metabolites, but not the bioactive peptides, were involved in the activity. In summary, we propose that the metabolites derived from T. atroviride supernatant have strong potential as whitening raw material.

  • Melanin biosynthesis inhibitors are useful not only for the materials used in cosmetics as skin-whitening agents but also for the remedy of hyperpigmentation. In order to find the new skin-whitening compounds from the natural products. screening of tyrosinase inhibitory activity in vitro has been carried out.
