Whitening Effect of Androsace umbellata Extract (봄맞이 추출물의 미백 효능 연구)

  • The purpose of this study is to identify the whitening effect of Androsace umbellata extract. To discover the anti-pigmentation effective, we investigated Androsace umbellata extract on tyrosinase and melanogenesis inhibition. As results, it reduced tyrosinase activity and melanin contents in B16F1 melanoma cells in a dose-dependent manner with decreased to about 32% at a concentration of $25{\mu}g/mL$. To reveal how it works in inner-cellular level, we performed Western blot method. We found out that it also inhibited the protein expression in tyrosinase, tyrosinase related protein 1 (TRP-1), and microphthalmia associated transcription factor (MITF) in melanocytes. Therefore, we successfully identified the whitening effect of Androsace umbellata extract, and this finding suggested that Androsace umbellata extract is a considerable potent cosmetics ingredient for whitening. Based on this, we anticipated further researches about Androsace umbellata extract for gene levels and additional mechanisms to develop not only for functional cosmetics but for medicines or healthcare food.

The Antioxidant and Skin-whitening Effects of Saccharomyces cerevisiae FT4-4 Isolated from Berries Grown in Sunchang (화장품 소재로서 순창 베리류 유래 Sacchromyces cerevisiae FT4-4의 항산화 활성 및 미백 효과)

  • Saccharomyces lysate has the well-known function of soothing the skin in various ways: it is an anti-irritant and can treat skin care conditions, such as skin whitening and antioxidative activity. However, data on the safety for use of Saccharomyces lysate in cosmetics and skin care products are still limited. To design a new cosmetic material with antioxidant and skin-whitening effects, 80 yeast strains were isolated from berries grown in Sunchang. Among the isolates, the FT4-4 strain, which exhibited superior biological activities, was selected for further experiments. The FT4-4 strain was identified as Saccharomyces cerevisiae by 18S rRNA gene sequencing analysis. S. cerevisiae FT4-4 showed higher DPPH radical-scavenging (51.41%), superoxide dismutase (62.23%), and tyrosinase inhibition (64.75%) activities. The highest yield of biomass (3.16 g/l) and maximum growth rate of S. cerevisiae FT4-4 were observed within 16 h. Furthermore, the cytotoxicity potential of S. cerevisiae FT4-4 on B16F10 melanoma cells was measured by an MTT assay, and the results indicated that S. cerevisiae FT4-4 had a capacity to inhibit melanin up to 72.02% at an initial 10 mg/ml concentration. These results suggest that S. cerevisiae FT4-4 could be a promising candidate as a multi-functional material for application in the cosmetic industry, especially because of its antioxidant and skin-whitening effects.

Studies on the Antioxidant and Whitening Effects of Chamaecyparis obtusa Extract (편백나무(Chamaecyparis obtusa) 추출물의 항산화, 미백효과에 관한 연구)

  • This study was conducted as follows in order to find out the possibility of using (Chamaecyparis obtusa) by part as a cosmetic material. Chamaecyparis obtusa was separated leaves, bark, wood, and root, and extracted with 99.9% ethanol, and the antioxidant and whitening effects of the sample were confirmed. For this, the following studies were conducted. Antioxidant evaluation was confirmed by 1 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2 2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS+) radical scavenging activity. To evaluate the whitening effect, mushroom tyrosinase inhibitory activity evaluation and cytotoxicity evaluation of Chamaecyparis obtusa extract through MTT assay were conducted, and cellular tyrosinase inhibition rate was measured and melanin contents was confirmed. As a result of antioxidant activity, Bark extract among Chamaecyparis obtusa parts showed the best efficacy, and bark in B16F10 melanoma cell showed tyrosinase activity inhibition and melanin contents inhibitory effects. Therefore, it was confirmed that the ethanol extract of Chamaecyparis obtusa was developed as a natural material for safe and excellent whitening functional cosmetics.

Whitening Effect and Antioxidant Activity of Horseradish Subcritical Water Extracts (겨자무 아임계수 추출물의 항산화활성 및 미백효과)

  • Melanin is a natural pigment found in most plants and animals, and it is involved in determining the color of the skin and hair. Melanogenesis is a reactive occurrence in melanocytes aiming to protect the skin from external stimuli, such as ultraviolet rays. Tyrosine is involved in the biosynthesis of the substrate tyrosine into melanin. However, melanin overproduction can lead to skin diseases, such as melasma, blotching, hyperpigmentation, and skin cancer. Although many studies have been conducted on whitening substances, such as kojic acid and arbutin, some countries have banned or refrained from using them due to their side effects. Therefore, this study assessed the potential of horseradish (HR) as a new whitening agent in cosmetic products. For efficient extraction, subcritical water extraction was conducted. The results showed that the horseradish subcritical water 200℃ (SW 200) extract showed high DPPH radical scavenging ability, total phenolic contents (TPC), inhibiting tyrosinase activity and inhibiting melanin production of B16-F10 melanoma cell lines. To investigate its cytotoxicity to the B16-F10 melanoma cell lines, MTT reduction assay and morphological changes were observed. No cytotoxicity was found in horseradish methanol extract and SW 200. In conclusion, this research suggests the possibility of horseradish subcritical water may be useful as a natural whitening ingredient to be used in cosmetic products.

Verification of Whitening Activity of Inonotus obliquus Extracts in B16F10 Melanoma Cells (차가버섯(Inonotus obliquus) 추출물의 B16F10 멜라노마 세포에서의 미백활성 검증)

  • In this study, the various whitening activities of Inonotus obliquus were assessed for potential use as functional cosmetic materials. When measuring electron donating ability to confirm the antioxidant ability of I. obliquus extract, increased activity was observed as the concentration increased, and it was found an outstanding antioxidant capacity of 82.1% at a 1,000 ㎍/ml concentration. Also, the tyrosinase inhibitory effect, related to a whitening effect, was found to have inhibitory activity that increased in a concentration-dependent manner. The results of verifying the viability of melanoma cells (B16F10) using an MTT assay showed cell viability of more than 80% at concentrations below 100 ㎍/ml. Therefore, cell-related experiments were conducted at 25, 50, and 100 ㎍/ml concentrations. By measuring protein expression related to melanin synthesis via treating B16F10 cells with I. obliquus extract, it was confirmed that protein expression was inhibited in all factors, depending on the concentration. TRP-1 and MITF appeared by 40.1% and 64.2% amount of expression, respectively, at 100 ㎍/ml concentrations, and tyrosinase and TRP-2 were verified as having better protein expression inhibition than arbutin. In measuring mRNA expression related to melanin synthesis by treating B16F10 cells with I. obliquus extract, it was confirmed that mRNA expression was suppressed as the concentration increased. Accordingly, it was confirmed that I. obliquus extract has excellent whitening activity and could be used as a cosmetic material.

Antioxidant and Tyrosinase Inhibitory Activities of Dicaffeoylquinic Acid Derivatives Isolated from Gnaphalium Affine D. DON (떡쑥 추출물로부터 분리된 Dicaffeoylquinic Acid 유도체들의 항산화 및 타이로시네이즈 저해 활성)

  • In this study, three dicaffeoylquinic acids (DCQAs) isolated from Gnaphalium affine D. DON. extracts were structurally identified and evaluated for their antioxidant activities, cellular protective effects, and tyrosinase inhibitory activities. The ethyl acetate fraction of G. affine was chromatographed, which yielded 3 DCQA derivatives of 1-3 : 3,5-dicaffoylquinic acid (3,5-DCQA, 1), 4,5-dicaffeoylquinic acid (4,5-DCQA, 2), 1,5-dicaffoylquinic acid (1,5-DCQA, 3). The structure of each compounds was determined using $^1H$ NMR and MS analyses. Compounds of 1-3 showed strong free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities ($FSC_{50}=3.70$, 5.80, and $5.50{\mu}M$, respectively) compared to those of a commonly used lipophilic antioxidant, (+)-${\alpha}$-tocopherol ($21.90{\mu}M$). Cellular protective effects of 1-3 compounds on the $^1O_2$ sensitized photohemolysis of human erythrocytes were similar to (+)-${\alpha}$-tocopherol. 1-3 compounds also exhibited higher tyrosinase inhibitory effects ($IC_{50}=0.15$, 0.16, and 0.13 mM) compared to arbutin (0.33 mM), known as a skin-whitening agent. These results indicate that three DCQA derivatives may be applied as an antioxidant and a skin whitening agent in food or cosmetic industries.

Antioxidant Effect of Nelumbo nucifera G. Leaf Extract and Inhibition of MITF, TRP-1, TRP-2, and Tyrosinase Expression in a B16F10 Melanoma Cell Line (연잎 추출물의 항산화 활성 및 멜라노마 세포(B16F10)에서 MITF, TRP-1, TRP-2, tyrosinase의 발현 저해 효과)

  • The purpose of this study was to investigate the potential of Nelumbo nucifera G. leaf (NNL) extract as a cosmetic additive. The electron-donating ability of the NNL extract at a concentration of 1,000 μg/ml was 67.83%. In xanthine oxidase, the inhibition effect of the NNL extract was 92.7% at the same concentration. For whitening effects, tyrosinase inhibition effect of NNL extract was 42.7% at a 1,000 μg/ml concentration. The cell toxicity of the NNL extract was examined in melanoma cells (B16F10) using a 3-[4, 5–dimethyl–thiazol–2–yl]-2, 5-diphenyl-tetrazoliumbromide (MTT) assay. The cell toxicity assay revealed that the NNL extract had a toxicity of 81.61% at a concentration of 1,000 μg/ml The microphthalmia-associated transcription factor (MITF), tyrosinase related protein-1 (TRP-1), tyrosinase related protein-2 (TRP-2), and tyrosinase protein expression inhibitory effect by Western blot of NNL extract were measured by a Western blot at concentrations of 25, 50, and 100 μg/ml. At a 100 μg/ml concentration of the NNL extract, the expression of the MITF, TRP-1, TRP-2, and tyrosinase protein was decreased by 69.59%, 27.74%, 67.33%, and 67.78% respectively. The MITF, TRP-1, TRP-2 and tyrosinase mRNA expression inhibitory effect were measured by reverse transcription- polymerase chain reaction (PCR) at concentrations of 25, 50, and 100 μg/ml. GAPDH was used as a positive control. At a concentration of 100 μg/ml of the NNL extract, the expression of MITF, TRP-1, TRP-2, and tyrosinase mRNA was decreased by 67.51%, 71.36%, 85.74%, and 83.64%, respectively. These findings suggest that the NNL extract has antioxidant and whitening effects and that it has great potential as a cosmetic ingredient.

Comparative Study of Protocorm-like body and Multiple Shoots from Dendrobium Candidum on Biological Activities (철피석곡의 원괴체상구체 및 다신초 추출물의 생리활성에 관한 비교 연구)

  • Dendrobium candidum is one of the well-known orchid on traditional and rare medicinal herb in China. We artificially cultured protocorm-like body and multiple shoots using the bioreactor culture system from Dendrobium candidum and experimented an efficacy as a cosmetic ingredient for skin care. Several studies previously have reported anti-tumor, wound healing and immunological function of Dendrobium candidum but other efficacies were not well known. In the present study, we investigated the cosmetic efficacy to know applicable value of protocorm-like body and multiple shoots cultured from Dendrobium candidum as a cosmetic ingredient. The biological activities of extracts from protocorm-like body, multiple shoots, aerial part and underground part of Dendrobium candidum were investigated. Results were found that extracts of protocorm-like body are superior to other extracts (underground part, aerial part and multiple shoots extracts) on anti-oxidant effect. Also, protocorm-like body extract contained the phenolic and flavonoid compounds more than aerial part, underground part and multiple shoots extracts. In addition, we investigated skin whitening effect related to whitening of skin. In tyrosinase activity and melanin synthesis assay, multiple shoots extract is superior to other extracts (aerial part, underground part and protocorm-like body), on inhibitory effect of tyrosinase activity and melanin synthesis respectively. These results indicate that the protocorm-like body and multiple shoots extracts cultured from Dendrobium candidum presents a new possibility of being applicable to skin improvement as a cosmetic ingredient.

The Study on Tissue-Cultured Echinacea purpurea Adventitious Roots Extract for Application as a Cosmetic Ingredient (조직 배양한 에키네시아 추출물에 관한 효능 연구)

  • Echinacea purpurea, an indian traditional plant medicine has been widely used as herbal remedy for the treatment of disease such as colds or other infections. However, Echinacea purpurea extracts recently have been applied as a cosmetic ingredient for skin care. We artificially cultured Echinacea purpurea by using the bioreactor culture system for this study. We induced callus from Echinacea purpurea and separated adventitious roots, harvested and extracted after cultured in bioreactors. Previously, several studies have been reported on anti-oxidant and immuno-enhancing effects of Echinacea purpurea extract but other efficacies were not well known. In this study, we investigated the whitening, anti-wrinkle and anti-oxidant effects to know applicable value of tissue-cultured Echinacea purpurea adventitious roots extract(TCEPARE) as a cosmetic ingredient. TCEPARE did not show cytotoxicity until a concentration of 2% and showed the anti-oxidative effect in DPPH and NBT tests. Also, the extract decreased tyrosinase expression in a dose-dependent manner and inhibited melanin synthesis in B16 melanoma cells. TCEPARE reduced protein levels of MMP-1, 2 secreted in culture medium or in cell lysates. From these results we suggest that TCEPARE has potential benefits applicable as to cosmetic ingredient for skin care products.

Study for Whitening Activity of Mixture of Arbutin and Oil Soluble Licorice Extract (알부틴과 유용성감초 추출물 혼합물에 의한 미백활성 연구)

  • The aim of this study was to investigate whitening effect of combination between arbutine and oil soluble licorice extract. Inhibitory effects of arbutin and oil soluble licorice extract against tyrosinase activity and melanogenesis in B16 melanoma cells were assessed in vitro to determine whitening effect. MTT assay with B16 melanoma cells showed that mixture (arbutin and oil soluble) was not each concentration. Both oil soluble licorice extract and arbutin induced dose-dependent inhibition of mushroom tyrosinase activity. Various concentrations (oil soluble extract : arbutin = 1:1. 1:1.5, 1:2, 1:2.5, 1:5) of mixtures also significantly inhibited tyrosinase activity in B16 melanoma cells by 40-51%. In addition, the mixtures reduced the melanin contents of B16 melanoma cells by more than 50% at each concentration. These results suggest that mixtures of arbutin and oil soluble licorice extract are very effective whitening ingredients.
