Genome editing by CRISPR/Cas9, a third-generation gene scissor in molecular breeding at the genome level, is attracting much attention as one of the breeding techniques of the future. In this study, genetic and phenotypic analysis was used to examine the responsiveness of the Bakokjam variety of the silkworm Bombyx mori to molecular breeding using CRISPR/Cas9 in editing the white egg 2 (w-2) gene. The nucleotide sequence of the w-2 gene was analyzed and three different guide RNAs (gRNA) were prepared. The synthesized gRNA was combined with Cas9 protein and then analyzed by T7 endonuclease I after introduction into the Bm-N silkworm cell line. To edit the silkworm gene, W1N and W2P gRNA and Cas9 complexes were microinjected into silkworm embryos. Based on the results of microinjection, the hatching rate was 16-24% and the incidence of mutation was 33-37%. The gene mutation was verified in the heterozygous F1 generation, but no phenotypic change was observed. In F2 homozygotes generated by F1 self-crosses, a mutant phenotype was observed. These results suggest that silkworm molecular breeding using the CRISPR/Cas9 system is possible and will be a very effective way to shorten the time required than the traditional breeding process.
The attractant (orange light) or repellent (white light) signal is transmitted from SRI (Sensory Rhodopsin I) via protein-protein interaction with its transducer Htrl (Halobacterial Transducer for Sensory Rhodopsin I) which in turn controls a cytoplasmic phospho-transfer pathway that modulates flagella motor switching in Halobacterium salinarum. Some mutations in both SRI and Htrl showed an unusual mutant phenotype called inverted signaling, in which the cell produces a repellent response to normally attractant light. Twelve mutations at the Glutamate 56 (E56) position in the second transmembrane helix of Htrl were introduced by site-specific random mutagenesis. Almost all E56 mutants showed orange-light inverted responses in pH and temperature-dependent manners except E56D and E56Y. Except for these two mutants, all mutants accelerated the $S_{373}$ decay compared to wild-type at $18^{\circ}C$. This supported that there is an interaction between SRI and the second transmembrane of Htrl. Also a structural model of Htrl based on the Tar crystal structure and the secondary structure prediction program proposed the E56 residue to be in the middle of the proton channel. The most important observation is that the E56 mutant provides the evidence that this residue is very sensitive for signal relay, which can be explained by the open and closed conformations of the channel (A and R conformations) in SRI, as was postulated by the unified conformational shuttling model for transport and signaling.
Objective: Considering the physiological and clinical importance of leptin receptor (LEPR) in regulating obesity and the fact that porcine LEPR expression is not known to be controlled by lncRNAs and miRNAs, we aim to characterize this gene as a potential target of SSC-miR-323 and the lncRNA TCONS_00010987. Methods: Bioinformatics analyses revealed that lncRNA TCONS_00010987 and LEPR have SSC-miR-323-binding sites and that LEPR might be a target of lncRNA TCONS_00010987 based on cis prediction. Wild-type and mutant TCONS_00010987-target sequence fragments and wild-type and mutant LEPR 3'-UTR fragments were generated and cloned into pmiRRB-REPORTTM-Control vectors to construct respective recombinant plasmids. HEK293T cells were co-transfected with the SSC-miR-323 mimics or a negative control with constructs harboring the corresponding binding sites and relative luciferase activities were determined. Tissue expression patterns of lncRNA TCONS_00010987, SSC-miR-323, and LEPR in Anqing six-end-white (AQ, the obese breed) and Large White (LW, the lean breed) pigs were detected by real-time quantitative polymerase chain reaction; backfat expression of LEPR protein was detected by western blotting. Results: Target gene fragments were successfully cloned, and the four recombinant vectors were constructed. Compared to the negative control, SSC-miR-323 mimics significantly inhibited luciferase activity from the wild-type TCONS_00010987-target sequence and wild-type LEPR-3'-UTR (p<0.01 for both) but not from the mutant TCONS_00010987-target sequence and mutant LEPR-3'-UTR (p>0.05 for both). Backfat expression levels of TCONS_00010987 and LEPR in AQ pigs were significantly higher than those in LW pigs (p<0.01), whereas levels of SSC-miR-323 in AQ pigs were significantly lower than those in LW pigs (p<0.05). LEPR protein levels in the backfat tissues of AQ pigs were markedly higher than those in LW pigs (p<0.01). Conclusion: LEPR is a potential target of SSC-miR-323, and TCONS_00010987 might act as a sponge for SSC-miR-323 to regulate LEPR expression.
In our previous study, pyrrolnitrin produced in Pseudomonas chlororaphis G05 plays more critical role in suppression of mycelial growth of some fungal pathogens that cause plant diseases in agriculture. Although some regulators for pyrrolnitrin biosynthesis were identified, the pyrrolnitrin regulation pathway was not fully constructed. During our screening novel regulator candidates, we obtained a white conjugant G05W02 while transposon mutagenesis was carried out between a fusion mutant $G05{\Delta}phz{\Delta}prn::lacZ$ and E. coli S17-1 (pUT/mini-Tn5Kan). By cloning and sequencing of the transposon-flanking DNA fragment, we found that a vfr gene in the conjugant G05W02 was disrupted with mini-Tn5Kan. In one other previous study on P. fluorescens, however, it was reported that the deletion of the vfr caused increased production of pyrrolnitrin and other antifungal metabolites. To confirm its regulatory function, we constructed the vfr-knockout mutant $G05{\Delta}vfr$ and $G05{\Delta}phz{\Delta}prn::lacZ{\Delta}vfr$. By quantifying ${\beta}-galactosidase$ activities, we found that deletion of the vfr decreased the prn operon expression dramatically. Meanwhile, by quantifying pyrrolnitrin production in the mutant $G05{\Delta}vfr$, we found that deficiency of the Vfr caused decreased pyrrolnitrin production. However, production of phenazine-1-carboxylic acid was same to that in the wild-type strain G05. Taken together, Vfr is required for pyrrolnitrin but not for phenazine-1-carboxylic acid biosynthesis in P. chlororaphis G05.
KGD1 유전자는 비허용온도에서 세포벽에 결함을 보이는 Saccharomyces cerevisiae LP0353 균주의 베타-1,3-글루칸 합성 효소의 활성을 회복시키는 유전자로 분리되었다. $\alpha$-ketoglutarate dehydrogenase를 암호화하는 KGD1 유전자의 효모의 세포벽 합성과 연관된 기능을 분석하기 위하여 유전자 파괴를 시도하였다. KGD1돌연변이는 생장속도가 감소하고, 키틴 합성 효소들의 활성이 증가하였으며, 세포벽 구성 당류의 함량에 변화를 보였다. 또한 Calcofluor white과 Nikkomycin Z 등과 같은 세포벽 합성 저해물질에 대해 감수성 변화를 나타냈다. 이러한 결과들은 KGD1이 효모의 세포벽 특히 베타-1,6-글루칸과 키틴의 생합성에 영향을 주고 있음을 시사한다.
Intensive taxonomical studies of the Aspergilli have long been made. Altogether 132 species and 18 varieties are recognized in the book "The Genus Aspergillus" written by Raper and Fennell (1965), in contrast to 77 species, 8 varieties and 4 mutations in " A Manual of Aspergilli" written 20 years earlier by Thom and Raper (1945). Classification of the Asperilli by Thom and Raper (1945) and by Raper and Fenell (1965). Classification of the Aspergilli by Thom and Raper (1945) and by Rapher and Fenell (1965) have been based mainly upon morphological and cultural detail both physiological and biochemical activities. In Korean there are many kinds of foods fermented natrually without the employment of selected microorganisms, and there are, of course, many different microorganisms serving in the fermentation fermented foods than other countries, the distribution and biological properties of the Asperigilli, in Korea are more variable. Taxonominical studies with 36 strains of Asperilli were based upon the examinations of morphological, cultural, and physiological characteristics. nineteen strains indigenous to Korea were selected from a lot of strains which had been isolated from meju and kokja and one strain from soil. They were identified according to the group key of Raper and Fennell. Ten strains were donated by Dr.Hesseltine of the Northern Utilization Research and Development Division in the U.S.A. From the Asp. japonicus supplied by Dr.Hesseltine, a white mutant was isolated and also studied. Two strains were donated by Dr. Murakami of the Research Institute of Brewing in Japan, and four strains came from Korean industrial companies.ndustrial companies.
느타리 버섯류의 새로운 품종을 개발하기 위하여 백색느타리 신품종을 육성하였다. 2005년부터 2007년까지 백색느타리 유전자원을 수집하여 특성검정을 하였다. 2007년에 육종모본 수한의 백색변이체 ASI2842와 원형의 백색변이체 MGL0205를 선발, 교잡하여 84개의 교잡주를 육성하였다. 이중에서 우수한 Po2007-63 ($2842-7{\times}0205-7$) 을 선발하여 특성검정, 확대재배를 실시하여 농작물 직무육성 신품종 선정심의회에서 '고니 '로 명명되었다. 주요특성으로 균사 생장 적온이 $25{\sim}30^{\circ}C$이며 버섯 원기 형성 및 발생 온도는 $10{\sim}16^{\circ}C$이였다. 자실체의 갓 색깔은 백색으로 자연 상태에서 봄, 가을에 재배가 알맞은 특성을 가지고 있다. 균사체 배양기간은 25~30일이며 균 긁기 후 초발이소요일수는 3~5일로 온도가 높을수록 단축된다. 자실체 형태는 얕은 깔때기형이다. 유효경수는 병당 $8{\pm}2$개, 대굵기는 $14{\pm}1mm$, 대길이는 $67.6{\pm}8mm$로 다른 느타리 종에 비해 자실체 대가 굵고 길며, 갓두께는 $4.7{\pm}0mm$로 갓이 작고 부스러짐에 강하여 품질이 우수하다. 자실체 수량은 병당 ($850m{\ell}$) $91{\pm}13g$로 대조군 미소의 수량지수를 100으로 보았을 때 97이였다. 품질을 높게 하려면 재배온도를 $11^{\circ}C$ 정도로 다소 낮은 생육온도에서 관리하는 것이 좋다. 가변특성으로는 감자배지와 버섯완전배지에서 균사를 배양한 결과 $20-25^{\circ}C$에서는 버섯완전배지에서 생장이 빠르다. 그러나 $30^{\circ}C$에서는 감자배지에서 생장이 양호하였다. 이러한 현상은 대조구인 미소에서도 동일한 경향이었다. 또한 2종류의 primer를 이용하여 새로운 품종 '고니'와 모균주에 대한 DNA profile을 분석한 결과 primer URP 1에서는 양친주의 주요 밴드를 가지며 대조구인 미소와는 뚜렷하게 구분되었고, primer URP 2에서는 양친주 주요 밴드는 가지나 대조구와도 유사한 밴드를 나타내었다. 신품종 백색느타리 '고니'는 백색이 갖는 깨끗하고 신선한 이미지로 웰빙식품이 각광을 받고 있는 시대에 맞게 다양하고 우수한 버섯을 요구하는 소비자를 만족시키는데 기여 할 것으로 기대된다.
갓과 대의 색택이 백색인 백색느타리 신품종 "백선"의 주요특성은 다음과 같다. 균사생장적온은 $28{\sim}31^{\circ}C$이고 버섯발생온도는 $22^{\circ}C$, 버섯생육온도 $20^{\circ}C$로 '미소'보다 균사생장적온이 높으며 버섯발생 및 생육온도가 유사하며, 발생형은 다발형태를 나타내었다. 병재배시 배양기간은 30일, 초발이 소요일수는 4일, 생육일수는 4일로 총재배기간은 38일이 소요되었다. 형태적 특성에 있어 갓직경은 32.5 mm, 대직경 9.1 mm, 대길이 91.4 mm로 '미소'에 비하여 가늘고 긴 형태를 나타내었으며, 갓색도(L)는 84.2, 대색도(L)은 83.4로 '미소'에 비하여 밝은 백색을 나타내었다. 수량은 생산력 검정시 1100 ml병에서 185 g을 나타내었으며, 농가실증재배시 A(평택) 184g/1100 ml, B(여주) 178 g/850 ml으로 대조품종 대비 40%이상 증수 되었다. 대의 물리성은 탄력성, 응집성, 씹음성, 깨짐성이 각각 80%, 57%, 720 g, 57 kg을 나타내었다. DNA다형성을 비교 분석한 결과 UFPF1, UFPF3, UFPF4의 primer에서 교배모본인 'GMPO20410'와 'MGL2205'의 DNA의 밴드가 혼합되어 있었으며 품종 간, 균주 간의 밴드 차이가 있었다. 저장기간에 따른 신선도는 $4^{\circ}C$에서 28일 저온저장시 8점으로 신선한 상태였으며, $4^{\circ}C$저장 후 상온보관 시 5점으로 식용 가능한 상태로 대조품종인 '미소' 보다 저온 및 상온보관 시 신선도가 우수하였다.
본 연구는 화청벼와 IR24의 돌연변이 계통인 분상질 및 심백 변이체의 작물학적 및 이화학적특성과 제병 및 알콜발효특성을 조사하여 그 가공적 이용성을 검토하고 분상질배유 유전자의 유전분리와 표지인자들과의 연관 관계를 밝히고자 수행하였으며 그 결과는 다음과 같다. 1. 돌연변이체의 종실은 원품종에 비해 작았으며, 천입중과 1$\ell$중도 전반적으로 가벼운 것으로 나타났다. 2. 변이체들의 출수기는 원품종과 같았으며, 정조수량은 원품종에 비해 낮았다. 3. 분상질 배유의 전분립은 느슨한 결정구조를 갖고 있었고, 전분립의 크기는 원품종과 변이체들간에 동일경향이 아니었으며, 원품종에 비해 변이체들은 밀도와 경도가 낮았다. 4. 변이체들의 아밀로스 함량은 16.9~28.5%로 변이복이 크게 나타났다. 현미의 조단백 함량에서 화청벼의 분상질변이체들은 원품종과 같았으나 IR24의 심백변이체인 47106은 원품종(8.30%)에 비해 11.32%로 높게 나타났다. 5. Gel consistency는 원품종들이 soft의 특성을 보였으나 분질 및 심백의 변이체에서는 모도 medium의 특성을 보였고, 백미의 알칼리 붕괴도는 원품종에 비해 분수질 변이체들에서 높게 나타났다. 6. Amylogram에 의한 호화개시 온도는 IR24의 분상질변이체(47160)를 제외하고 모도 원품종과 비슷하였으나, 최고점도와 최저점도 및 굳음성은 원품종 화청벼와 IR24에 비해 변이체에서 월등히 낮게 나타났다. 7. Instron으로 측정된 증편의 경도와 gumminess로 볼 때 화청벼 분상질 및 심백변이체로 조제된 증편은 다소 부드러운 특성을 나타냈으나, IR24의 경우는 일정 경향이 없었다. 분상질미와 심백미의 증편부피는 원품종에 비해 대체로 크게 나타났다. 8. 알콜발효 과정 중 당도 저하는 변이체들 특히 분상질에서 빨리 일어났고, 발효 10일 후의 알콜생성량도 원품종에 비해 변이체들에서 높게 나타나서 분상질미는 양조용으로서의 이용성이 크게 기대되었다. 9. 화청벼와 IR24에서 유래한 분상질 배유유전자들의 대립성 검정결과 서로 다른 3개의 유전자좌를 탐색하였으며, 그 유전자들을 각각 flo-a, flo-b, flo-c로 표시하였다. 10. flo-a 유전자는 IV번 연관군의 lg(liguleless ; 무엽설) 유전자와 5.76$\pm$1.72%(상반)로 연관되어 있었다.
패랭이꽃속의 분포와 개발을 위한 기초자료로 강원도에서 자생 2종과 수집가능한 7 외래종에 대한 기내배양체의 특성을 조사하였다. 캘러스의 유도를 위해 조직부위별 다양한 절편체들을 2,4-D. NAA, BA가 농도별로 처리된 MS 배지에 치상한 후, 27$^{\circ}C$ 광조건과 2.0mg/L 2,4-D와 0.5 mg/L BA의 조합배지에서 배양했을 때 3주 후 절단면에서 callus가 형성되기 시작하였으며, 패랭이꽃 (Dianthus chinensis)의 잎절편체에서 가장 양호하였다. Organogenic 캘러스의 선발은 기내에서 증식된 adventitious shoot의 절편체 유래 캘러스로부터 가능했으며 이들 캘러스를 1.0 mg/L BA와 0.1 mg/L NAA가 조합된 배지에 치상하였을 때 약 30% 정도의 shoot 형성률을 보였고 multiple shoot의 분화를 관찰할 수 있었다. In vitro에서 증식된 adventitious shoot 절편체를 1.0 mg/L BA와 0.1 mg/L NAA가 포함된 N6 배지에 치상했을 때, 캘러스 단계를 거치지 않고 직접 shoot이 형성되었고 D. gratianopol 유래 adventitious shoot 절편체는 52%의 높은 shoot 형성률을 보였다. 이들 multiple shoot로부터 재분화된 소식물체는 0.1 mg/L NAA가 포함된 MS 배지에서 뿌리가 유도되고 vermiculite가 담긴 pot에 이식 후 85% 습도하에서 활착되어 정상적인 개화된 개체를 획득할 수 있었다. 0.03 M EMS 처리된 패랭이꽃의 adventitious, shoot 절편체 유래 캘러스로부터 힌꽃의 패랭이꽃의 도연변체식물 (M23)이 분화되었으며 종자 형성이 가능하였다.

제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
