Depositions of Egg Capsules by Female Shell Heights and Comparisons of Sizes at 50% of Group Sexual Maturities of the Female Rapa Whelk Rapana venosa in Three Different Salinity Concentration Regions

  • We investigated fecundities in egg capsules and sizes at 50% of group sexual maturities in female Rapana venosa in three different salinity concentration regions (S-1, S-2, and S-3). In three different salinities, egg capsule heights, the number of egg capsules and the number of eggs and embryos were remarkably increased with the increase of female shell heights (or ages) and also increased with the increase of salinity concentrations. Heights of egg capsules, the number of egg capsules and fecundities (the number of eggs and embryos) were the maximum at S-1 (Gwangyang Bay (average 31.5 psu)) and the minimum at S-3 (the upper reaches of Seomjin River (average 15.5 psu)). Total numbers of fecundities of R. venosa $individual^{-1}year^{-1}$ were about $1{\times}10^6$ at S-1 region, about $8{\times}10^5$ at S-2 region, and about $2{\times}10^5$ at S-3 region. Rates (50%) of individuals reaching first sexual maturities in three different salinity regions (S-1, S-2 and S-3) were over 50% in females measuring 7.1-8.0 cm in shell height (considered to be two years old), and 100% in those > 10.1 cm (considered to be five years old). Biological minimum sizes ($RM_{50}$) in females in three different regions are 72.0 mm SH at S-1 region, 70.9 mm SH at S-2 region, and 74.6 mm SH at S-3 region, respectively. Exceptionally, smaller individuals (considered to be one year old) were participated in reproduction.

Mesh selectivity of gill net for swimming crab Potunus trituberculatus in the western coastal waters of Korea (서해 꽃게 자망의 망목 선택성 연구)

  • A series of fishing experiments were carried out in the western coastal waters of Korea from May, 2006 to September, 2007, using gill nets of different mesh sizes (m=101, 121.8, 152.5, 176.8, and 191 mm) to determine the mesh selectivity of gill net for swimming crab, Potunus trituberculatus. The catch species was composed of swimming crab (87%), Thomas's rapa whelk Rapana venosa (5%), arthritic neptune Neptunea cumingi (2%), Japanese swimming crab Charybdis japonica (2%), marbled sole Pleuronectes yokohamae (1%), pen shell Atrina pectinata, ocellate spot skate Okamejei kenojei, and so on. The carapace length (CL) of each swimming crab caught in the experimental nets was measured. Sponge crab (egg-bearing female crab) was appeared from the end of June to August in the coastal waters. The master selection curve was estimated by applying the extended Kitahara's method. The selection curve showed that the gill nets of larger mesh size allowed more crabs of small carapace size to escape. The optimum value (CL/m) was 0.645 and the CL/m was estimated to be 0.301, 0.354, 0.391, 0.422, and 0.450 when the retention probabilities were 0.1, 0.2, 0.3, 0.4 and 0.5, respectively.

  • Nowadays, mankind is suffering from serious pollutions. In proportion to industrial modernization, polltional problems have been increased and arised new one endlessly. Todays, we are cope with a newkind pollution materials threatening the life of mankind with root out. It is Environmental Hormon, that is, Endocrine Disrupting Chemical(EDs). EDs was made by dioxin, PCB, DDT, cadmium, lead, ether and even diethylstillbestrol(DES) that used to contraceptive pill etc. These EDs makes changing from normal human hormon to abnormal one and it kill off or fade out spermatozoa. Then the spermatozoa was infected with EDs makes man imposex, sexual malformations and infertility also. It is the more critical phenomenons that EDs invade into the field of wild-life; birds, invertebrate animals, fishes, mammalia especially. England TV(BBC) journalist Cadury, D. introduced a lot of case studies that many kind of male animals was changed feminization by EDs in her book (The Feminization of Nature;1997). Otherwise, Gibbs, P.E. and Bryan, G. W. reported about dog-whelk snail (a sort of sea shell) was changed gender from male to female by EDs in their study. Briefly speaking, Environmental Hormon (EDs) stem from polluted materials maloperate human and animal informational codes of hormon. It is a significant event to an individual ontogeny and ecological system. To overcome these problem WHO, OECD, UNEP held many work-shops, international seminars and researches but it is not enough yet. On May of 1997, the International Meeting of Environmental Administrators makes a $\ulcorner$Declaration on EDs Problems$\lrcorner$ for solving it fortunately and NHK(japan) reported Super Critical Fluid as a new materials for decrease toxicity of dioxin in recent. Protein Consumption Market Trends in Korea: Focusing on Meat, Fishery, and Plant-based Protein Foods (국내 단백질 소비시장 동향: 축산물, 수산물, 식물성 단백질 식품을 중심으로)

  • This study aims to investigate protein consumption market trends in Korea. Protein consumption was divided according to the protein source into meat, fishery, and plant-based protein. To accomplish the goal of this study, food purchase data from 525 households panels collected by the Rural Development Administration over the last 10 years were used. The results of the study showed an increase or decrease in protein consumption by protein type over the last 10 years, and a reason to explain this change has been suggested. Specifically, this study found a dramatic increase in the consumption of several proteins, including beef sirloin, beef tenderloin, seasoned beef & steak, pork belly, pork shoulder, pork neck, seasoned pork, pork cutlet, sweet and sour pork, canned ham, chicken drumstick, chicken breast, dak gangjeong, Chinese fried chili chicken, salmon, eel, abalone, squid, octopus, webfoot octopus, octopus minor, canned whelk, tofu, cold bean soup,and plant-based milk. Some items showed no increase in consumption (such as beef jerky, pork rib, sausage, bacon, whole raw chicken, cutlass fish, oyster, fish cake, crab stick, surimi sausage,and canned fishery), whereas a few items showed decreased consumption (e.g., mackerel, pollack, cod,and canned tuna)

Pathogenicity and PCR detection of Vibrio tapetis in Manila clams, Ruditapes philippinarum (양식 바지락, Ruditapes philippinarum에 대한 Vibrio tapetis의 병원성과 PCR법에 의한 진단)

  • Pathogenicity of Vibrio tapetis, the causative bacterium of 'brown ring disease (BRD)' was evaluated in Manila clams (Ruditapes philippinarumi by artificially 0.1 $m\ell$ infection of $1.0\times10^5$cells and $1.0\times10^8$ cells at 20 $^{\circ}C$. A PCR assay based on 16S rRNA to detect the bacteria in clam tissues was established. Accumulative mortality of clams infected with $1.0\times10^7$cells and $1.0\times10^4$ cells per an individual of the bacteria was 67.5% and 7.5%, respectively. However, the deposit of brown pigment in the inner shells by accumulation of chonchiolin was not found. The bacteria were not be able to re-isolate from the infected clams by the conventional agar plate method but were easily detected by PCR assay established in this experiment. In clams artificially infected with 10 species of Vibrio, a 414bp for V. tapetis was detected in PCR assay. The specific band in the clams infected with $1.0\times10^4$cells per an individual of V. tapetis was detected only in gills one day after the infection but never be found in any tissues including gills three days after the infection. In the case of clams infected with $1.0\times10^8$cells per an individual of V. tapetis the specific band was detected in gills and intestine one day after the infection, in all tissues three days after the infection, and then in gills and adductor muscle nine days after the infection. The PCR assay was applied to detect V. tapetis in manila clam, surf clam (Mactra veneriformis), oyster (Crassostrea gigas) and Thomas' rapa whelk (Rapana venosa) taken from Taean and Gochang from April to July 2004. The infection rates were detected to 23.1% and 9.4% in the oyster and surf clam, while manila clam and Thomas' rapa whelk were not found.

  • Ultrstructural studies of germ cell differentiation and vitellogenesis in the oocytes of the female Rapana venosa in the brackish water area of Seomjin River were investigated by transmission electron microscope observations. In the early vitellogenic oocytes, the Golgi complex and mitochondria were involved in the formation of glycogen particle, lipid droplets, and yolk granules. In the late vitellogenic oocytes, the rough endoplasmic reticulum and multivesicular bodies were involved in the formation of proteid yolk granules in the cytoplasm. However, heterosynthetic vitellogenesis in this species were not observed in vitellogenic oocytes during oogenesis. A mature yolk granule was composed of three components: crystalline core, electron lucent cortex and the limiting membrane. As shown in some large gastropods, vitellogenesis in R. venosa occurred by way of endogenous autosynthesis without heterosythetic vitellogenesis (exogeneous endocytosis), which are found in the oocytes in bivalves. The mating period and spawning activity were related with the increases of seawater temperatures and salinities.

Prevalence and Detection of Perkinsus sp. infection in the Manila clams, Ruditapes philippinarum (양식 바지락, Ruditapes philippinarum의 Perkinsus sp. 검출현황과 현장조사)

  • Hemacolor stain, histopathological observation and FTM incubation assay were applied to detect Perkinsus sp. infection in Manila clams (Ruditapes philippinarumi taken from culture beds at Tean and Gochang from March 2002 to August 2003. The prevalency was 100% in the clams from Gochang and 20-70% from Tean. Of the three methods, histopathological observation was the most effective to detect the infection. And the parasites was most abundant in gills. When PCR assay was applied to detect Perkinsus sp. for four species of Mollusc such as manila clam, surf clam (Mactra veneriformis), oyster (Crassostrea gigas) and Thomas' rapa whelk (Rapana venosa) taken from Tean and Gochang from April to July 2004, the parasites were detected from all the species at the infection rates of 95%, 62%, 46.9% and 10% in that order. The infection rate was much higher in the species burrowing in the bottom sediments than those attaching on the tidal rock. The results suggest that Perkinsus sp. may affect almost all the molluscs inhabiting western coastal areas of Korea.

  • Germ cell development and cyclic changes in the epithelial cells of the seminal vesicle of the male rapa whelk, Rapana venosa, were investigated by cytological and histological observations. The process of germ cell development can be classified into five stages: (1) spermatogonial, (2) primary spermatocyte, (3) secodary spermatocyte, (4) spermatid, and (5) spermatozoon. In particular, four atypical cells (Type IA, IB, IIA and IIB cells) occur among normal germ cells in the acini during spermatogenesis. Presumably, the atypical cells, which have lysosome-like vacuoles or lysosome-like bodies in the cells, are involved in breakdown and absorption themselves in the acini. However, atypical cells were not found in the epithelial cells of the inner layer of the seminal vesicle. A considerable amount of spermatozoa are transported from the testis towards the the seminal vesicles until late July. The main coupulation period is between June and July. The process of the cyclical changes of the seminal vesicles can be classified into three phases: (1) resting, (2) accumulating, and (3) spent. Yellow granular bodies are involved in resorption or digestion of residual spermatozoa.

Effect of Preparation method and Fermentation Conditions on Microbiological Characteristics of Sikhae (어류를 이용한 식해의 제조 방법에 따른 미생물의 특성변화)

  • This study investigated microbial populations and the sensory quality of sikhae including globefish (GLS), flounder (FLS), gastropods (GAS) and whelks (WHS) during storage at 4C for 16 d following fermentation at 10C for 4 d. General bacterial numbers increased to 102 in GLS and FLS, and to 1045 in WHS and GAS after the 20 d fermentation/storage period. Lactic acid bacteria increased to 108 log cycle in GLS and FLS after 10 d ripening time, and reached this level in GAS and WHS after 15 d and 20 d, respectively. After 20 days the number of lactic acid bacteria in each of the four samples was 108. There were 104105 yeast cells/g in each of the four samples after 20 d. The number of Leuconostoc increased to over 108 log cycle after 10 d in GLS and FLS, and 15 days in GAS for WHS the increase was to 107 log cycle. The pH values of GLS, FLS, GAS and WHS 4.42, 4.56, 4.31 and pH 4.26, respectively. The Sikhae acidity for all four samples ranged from 1.551.85%. From the sensory evaluation the overall acceptability was in the order of FLS > GLS > GAS > WHS.

Study on the Contents of Trace Elements in Foods (on the Trace Element Contents of Shellfish in Korean coastal Water) (식품중의 미량금속에 관한 연구조사 (연안 견류중의 중금속 함유량에 관하여))

  • In 1987, the level of heavy metals were determined ina total of 200 samples of 9 species of shellfish of Korea. The samples were collected at the fish. markets by 10 Public Institute of Health. The samples were whelk (Buccinum striatiBBimum), oyster (Crassostrea gigas), ark shell(Tegillarca granesa), shartnecked clam (Venerupis semidecussta), hard clam (Meretrix lusoria), top shell (Turbo cornutus), abalone (Haliotis gigantea), ark shell (Scapharea broughtonii), sea-mussel (Mytilus conuscus gould), respectively. The levels of total mercury, lead, cadmium, arsenic, copper, zinc and manganese were determined. The total mercury levels were determined by mercury analyzer using the combustion gold amalgamation method. The arsenic level were determined by spectrophotometry using colorimetric sil ver diethyldithiocarbamate method after dry ash dige8tion of the samples with magnesium oxide and magnesium nitrate. The levels of other metals were determined by inductively coupled pluma spectrophotometry after wet digestion of the samples with nitric acid and su1furic acid. The results were summerized as follows; 1. The overallranges and mean(ppm) were; Hg, ND-O.221 (0.036); Pb, 0.05-1.51 (0.37); Cd, 0.02-1.86 (0.61); As, 0.5-3.97 (1.22); Cu, 0.14-54.16 (4.93); Zn, 7.40-207.17 (30.09); Mn, 0.13-s.72 (3.40). 2. The levels of all 6 metals were found to be below the maximum permissible Iimits set by the Japan lor mercury, the Netherland for lead the Hong Kong for cadmium. The Finland for Arsenic no statutory Iimits for Zn and Mn in shellfish in any countries. 3. The results show that all the 9 species of shellfish studied, none have accumulated levels dangerous enough to pose a health problem.roblem.
