Distribution and Isolation of Soil borne Wheat Mosaic Virus in Korea

  • This study was conducted to investigate the occurrence of Soil borne wheat mosaic virus(SbWMV) in barley fields in Korea and to examine the host pathogenicity of SbWMV. By using the ELISA test, SbWMV was detected in the six regions : Suwon, Milyang, Jinju, Youngkwang, Iksan, and Chonju. SbWMV was isolated from the two strains, Albori strain from Jinju and Eunpamil strain from Milyang. SbWMV was collected from leaves showing mosaic, yellowing and necrosis stripes. SbWMV was inoculated mechanically on 1∼1.5 leaf stages with leaf-rubbing to identify the host pathogenicity of 36 Korean barley cultivars, a wheat cultivar, two rye cultivars, three Japanese barley cultivars and Chenopodium amaranticola. Viral sympoms of inoculated leaves appeared on moulted loaves about 4 to 6 weeks of inoculation. Baegdong and Tapgolbori, infected from Albori strain and Eunpamil strain infected from Samdobori showed much higher susceptibility than C. amaranticola and C. quinoa which showed ring spots and chlorotic spots respectively. Virus particles were observed by the electron microscope. They were rod-shapes, which are bipartite, of 142 nm or 281 nm in length with 20 nm diameter on infected leaves. Specific detection and identification of SbWMV was set up using the RT-PCR. PCR fragments of SbWMV(0.5kb) were obtained by using the designed primers for SbWMV RNA 2.

Flour Quality Characteristics of Korean Waxy Wheat Lines

  • Flour physicochemical properties of six Korean waxy wheat lines and their parental plants, including Kanto 107 and BaiHuo, which have partially null in GBSS (granule bound starch synthase), were evaluated in this study. The very low amylose content (3.20%) of Korean waxy wheat lines, which had been influenced by the null in all three GBSS isoforms encoded by three Wx loci, could result in the higher starch swelling power (25.15%), lower starch and flour pasting temperature (61.37$^{\circ}C$; 65.85$^{\circ}C$), and higher starch pasting peak viscosity and breakdown (246.60 RVU; 161.50 RVU) than those of their parental plants. In addition to high swelling and pasting properties, Korean waxy wheat lines had the higher protein content (12.80%), alkaline water retention capacity (97.39%), SDS sedimentation volume (80.33 $m\ell$) and damaged starch content (4.35 %) than those of their parental plants.

ISP, CSP의 대체제로서 Wheat Fiber 첨가한 유화형 소시지의 저장성에 미치는 영향

  • 본 실험은 ISP, CSP의 대체제로서의 wheat fiber 첨가가 유화형 소시지의 저장성에 미치는 영향을 조사하였다. pH를 비교한 결과는 저장 4주차에서만 wheat fiber 첨가한 처리구가 유의적으로 낮았으며, 가열수율은 control과 비교하여 모든 처리구가 유의적으로 높게 나타났다. 저장감량은 저장 초기에는 wheat fiber를 첨가한 처리구가 유의적으로 낮은 감량을 보였으나, 저장 4주째에서는 유의적인 차이를 보이지 않았다. 물성은 hardness, gumminess, chewiness의 경우 모든 처리구에서 높게 나타났으며, 대체적으로 저장기간이 증가함에 따라서 증가하는 것으로 나타났다. TBA수치와 VBN수치는 저장기간이 경과함에 따라서 수치가 증가하는 것으로 나타났으며, 관능검사 결과도 모든 항목에서 유의차가 나타나지 않았다. 이상의 결과 ISP, CSP의 대체제로 wheat fiber를 첨가한다면 유화형 소시지의 저장성에 영향을 주지 않으며, 경제적인 측면에서 제품의 단가를 절감할 수 있을 것으로 생각된다.

Annual Analysis of the Agronomic Traits of Global Wheat Germplasms in the Korean Environment (국내환경에서 밀 유전자원의 연차간 농업특성 분석)

  • Securing a range of wheat resources is of particular importance with respect to wheat breeding, as it provides a broad genetic foundation. Although wheat breeders have used different wheat germplasms as material resources in current breeding systems, the traits of most germplasms collected from foreign countries differ from the unique traits that have evolved in the Korean environment. In this study, conducted over a 2-year period (2018 and 2019), we therefore evaluated the agricultural traits 1,967 wheat germplasms collected not only in Korea but also worldwide. During the period from sowing to February, the average temperature in 2019 was greater than 1℃, whereas from March to June, the average temperature was approximately 0.9℃ higher in 2018. Compared with the growth recorded in 2018, the stem length in 2019 increased by approximately 20 cm in 2019, and there were notable differences heading date and maturation between 2018 and 2019. In 2019, the heading dates of 973 and 713 wheat resources were earlier and later than those in 2018, respectively. Moreover, stem length was found to be highly correlated with the heading date and maturation. In Korea, where the rainy season and tine of rice transplantation overlap with the time of wheat harvest, early flowering time with high grain yield has been the most important selection target with respect to wheat breeding. We anticipate that the findings of this study will provide would use a foundation for the selection of elite materials and the development of resource core-sets for Korean wheat breeding programs.

  • Production of spring wheat, the major crop in Mongolia, accounts for 98% of the cultivated area. Understanding genetic variability in existing gene bank accessions is critical for collection, conservation and use of wheat germplasms. To determine genetic diversity and population structure among a representative collection of Mongolian local wheat cultivars and lines, 200 wheat accessions were analyzed with 15 SSR markers distributed throughout the wheat genome. A total of 85 alleles were detected, with three to five alleles per locus and a mean genetic richness of 5.66. Average genetic diversity index was 0.69, with values ranging from 0.37-0.80. The 200 Mongolian wheat accessions were mainly divided into two subgroups based on structure and phylogenetic analyses, and some phenotypes were divergent by the subgroups. Results from this study will provide valuable information for conservation and sustainable use of Mongolian wheat genetic resources.

Protein and Arabinoxylan Contents of Whole Grains from Wheat Genetic Resources Cultivated in Korea (국내에서 재배된 밀 유전자원의 통밀에서 단백질 및 아라비노자일란 함량 분석)

  • Recently, phytochemicals in whole grains have received increasing attention because the increased consumption of whole grains and whole grain products has been associated with a reduced risk of chronic diseases. Among the phytochemicals in whole wheat, arabinoxylan influences various physiological activities and can aid the treatment of numerous health conditions, either directly or indirectly. In this study, 614 wheat germplasms, collected from the world, were cultivated in 2018 and harvested in 2019 in South Korea. The qualities of these whole grains, including seed protein content, were evaluated using the NIR spectrophotometric method, and arabinoxylan content was determined using enzymatic methods. The ash content was 0.36 to 2.80% and protein content was 7.66 to 20. The SDS-sedimentation for gluten complex ability ranged from 26.85 to 80.63 mL, and protein content showed a high correlation with SDS-sedimentation in the germplasms. Verify the result of the enzymatic method of arabinoxylan, the reliability of the experimental method was determined through repeatability, reproducibility, and recovery. The average value of the 614 resources was 51.64 mg/g of whole grain, and when classified according to the country of origin, the South Korean origin resources tended to have a higher content compared to the genetic resources of other countries. These results are expected to be used as basic data for setting standards for whole grain quality in wheat breeding systems.

Differences in Productivity among Wheat, Barley and Rye for Forage

  • TO select the most suitable crop and variety of forage for productivity at the southern part of Korea, The crops of wheat, barley and rye were grown from Oct. 1999 to June 2000. Paldanghomil variety of rye crop was shown to have the highest productivity in comparison to other varieties of crops used in this experiment. It showed relatively high plant height, number of tiller, forage yield and dry matter yield. There fore, it was concluded that Paldanghomil of rye crop was the most suitable crop with high yield in the southern part of Korea. The heritabilities of all characters were estimated to be high.
