Hot Water Extract of Triticum aestivum L. (Common Wheat) Ameliorates Renal Injury by Inhibiting Apoptosis in a Rat Model of Ischemia/Reperfusion

  • Objectives : Interruption and subsequent restoration of blood flow into the kidney result in renal injury. As an approach to preventing the renal injury, we determined the optimal conditions and the underlying mechanisms by which supernatant of hot water extract of ground Triticum aestivum L. (extract) attenuated ischemia/reperfusion (I/R) injury. Methods : One hour after administration of the extract (400 mg/kg) by intraperitoneal injection, renal I/R injury was generated by clamping the left renal artery in rats after surgical removal of the right kidney, followed by reperfusion. The maximal difference between the vehicle-treated and the extract-treated group under ketamine/xylazine or enflurane anesthetization was assessed at varying periods of ischemia (30-45 min) and reperfusion (3-48 hr), based on the renal function assessed with serum creatinine levels, tissue injury with hematoxylin/eosin staining, and apoptosis with terminal deoxynucleotidyltransferase-mediated dUTP nick-end labeling staining. Results : Enflurane anesthetization with 40 min of ischemia and 24 hr of reperfusion was identified to be the optimal condition, under which condition serum creatinine levels and tubular damage in the extract-treated group were significantly reduced compared with those in the vehicle-treated group ($1.3{\pm}0.2$ versus $2.7{\pm}0.3$ mg/dL, P < 0.01, and average score $1.8{\pm}0.1$ versus $3.5{\pm}0.3$, P < 0.01, respectively). These beneficial effects were mediated by inhibition of apoptotic cascades through attenuation of renal tissue malondialdehyde levels, Bax/Bcl-2 ratio and caspase-3 levels. Conclusions : The extract conferred renal protection against ischemia/reperfusion injury in rats by scavenging reactive oxygen species and consequently blocking apoptotic cascades, plausibly augmented by enflurane protection.

온도 및 일장조건이 소맥의 생육 및 수량에 미치는 영향 (Effects of Temperature and Daylength on Growth and Grain Yield in Wheat (T. aestivum))

  • To observe the effects of temperature and photoperiod on the growth and yield of winter wheat (Triticum aestivum, L) eight varieties including Chang Kwang were tested at various treatments like longday-high temperature (24hrs-20\circ), shortdayhigh temperature (12hrs-20\circ), longday-low temperature (24hrs-14\circ) and shortday-low temperature (12hrs-14\circ). Among the traits measured, days to heading, plant height, spike length, number of spikes per hill and grain yield per hill were generally decreased at high temperature and long day treatment and increased at low temperature and shortday condition. Number of grains per spike was decreased at low temperature and short day condition while increased at longday and high temperature conditions. Grain weight was decreased significantly at high temperature and shortday while increased at low temperature and longday treatment.

Proteomics Approach on Puroindoline Gene of Pre-harvest Sprouting Wheat

  • Wheat (Triticum aestivum L.) grain texture is an important determinant of milling properties and end product use. Two linked genes, puroindoline a (PINA) and puroindoline b (PINB), control most of the genetic variation in wheat grain texture. Wheat seed proteins were examined to identify PINA and PINB gene using two pre-harvest sprouting wheat cultivars; Jinpum (resistant) and Keumgang (susceptible).Wheat seed proteins were separated by two-dimensional electrophoresis with IEF gels over pH ranges: pH 3-10. A total of 73 spots were digested with trypsin resulting peptide fragmentation were analyzed by matrix assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF/MS). Mass spectra were automatically processed and searched through NCBInr, SWISS-PORT and MSDB database with mono isotopic masses and complete gene sequence were found by UniProt database. Puroindoline a and puroindoline b that is responsible for grain texture related with baking performance and roughness. Two spots were found Pin b (16.7 kDa) and Pin a (16.3 kDa) in Jinpum compare to seven spots were identified Pin a (16.1 kDa, 16.3 kDa) and Pin b (16.7 kDa, 9.5 kDa and 14.4 kDa) in Keumgang. Some selected spots were identified puroindoline like grain softness protein (16.9 kDa, 17 kDa and 18.1 kDa) in Keumgang. Moreover, to gain a better inferring the identification of puroindoline related proteins using proteomics, we accomplished a complete gene sequence of PINA and PINB gene in pre-harvesting sprouting wheat seeds between resistant (Jinpum) and susceptible (Keumgang).

밀 Frucdtan Exohydrolase의 분리 및 특성 (Purification and Properties of Wheat Fructan Exohydrolase)

  • 밀의 영양체 저장탄수화물인 fructan의 대사에 관련된 fructan exohydrolase(FEH) 효소의 특성을 구명하기 위해 FEH를 밀의 줄기와 엽초에서 정제, 실험한 결과를 요약하면 다음과 같다. 1. FEH의 분자량은 63.7kD이었으며, pH 5.5와 3$0^{\circ}C$에서 최고의 활성을 보였다. 2. FEH의 $K_{m}$$V_{max}$는 fructan의 크기에 따라 각각 10~37mM, 5g~179mM로 변이를 보였으며, tetrasaccharide가 본 실험에서 사용된 기질 중에서 가장 낮은 $K_{m}$$V_{max}$를 보였다. 3. 밀의 FEH는 $eta$(2 $\longrightarrow$1)로 연결된 fructan에 대하여 기질 특이성을 보였으며 $\beta$(2 $\longrightarrow$6) fructan과 sucrose는 분해할 수 없었다.

  • 연질밀(Triticum aestivum L. em Thell.)로서 soft white spring(SWS) 밀과 soft white winter(SWW) 각각 10품종(또는 계통)을 공시재료로하여 종실의 이화학적 품질특성, 제분특 성, 호화특성, mixograph특성 및 sugar-snap cookie의 제조특성을 측정하고 이들 특성간의 상관관계를 구명하고자 일연의 실험을 실시한 바 그 결과를 보고한다. Single Kernel Characterization System(SKCS) kernel hard-ness와 용적중, near-infrared reflectance(NIR) hardness와는 각각 $r=0.947^{*},\;r=0.495^{*}$의 정의 상관이 있었으며 SKCS weight와 SKCS size와는 $r=0.942^{**}$의 고도의 정의 상관이 있었다. 용적중과 straight-grade flour yield, break flour yield, 밀가루 회분 함량 및 제분평점과는 각각 $r=0.720^{**},\;r=0.490^{*},\;r=-0.781^{**}$$r=0.838^{**}$ 의 고도의 정 또는 부의 상관이 있었다. 종실의 단백질 함량과 straight-grade flour yield, break flour yield 밀가루 회분 함량 및 제분평점은 각 각 $r=-0.786^{**},\;r=-0.765^{**},\;r=-0.889^{**}$$r=-0.938^{**}$로 서 고도의 정 또는 부의 상관이 있었으며 이와 같은 현상은 밀가루 단백질 함량에서 동일한 결과를 보였다. Mixograph 흡수을과 제분특성과는 고도의 정 또는 부의 상관이 있었으며, 특히 종실의 단백질 함량과는 $r=0.938^{**}$, 밀가루의 단백질 함량과는 $r=0.901^{**}$의 고도의 정의 상관이 있었다. Cookie 직경과 cookie top grain score는 $r=0.946^{**}$의 고도의 정의 상관이 있었다. Cookie특성과 제분특성, 단백질 함량 및 mixograph 흡수율과는 고도의 정 또는 부의 상관이 있었으나 전분특성과는 단지 swelling volume과 cookie top grain score 만이 $r=0.447^{*}$의 정의 상관이 있었다.
