Antifungal Activity of Fistulosides, Steroidal Saponins, from Allium fistulosum L (대파(Allium fistulosum L)로부터 fistulosides의 분리와 분리 물질의 항진균 활성)

  • Allium fistulosum L. (Welsh onion) is a perennial herb that is widely cultivated throughout the world, especially in China, Japan and Korea. Although, various activities were reported, little is known about antimicrobial activity of A. fistulosum L.. In this study, strong antimicrobial substances, fistuloside A, B, and C were isolated from the edible parts of A. fistulosum L. and their antimicrobial activity was evaluated with pathogenic- or food-spoilage microorganism based on disk-diffusion assay, minimal inhibitory concentration (MIC) and minimal fungicidal concentration (MFC) determination. Fistuloside A and fistuloside C showed strong antifungal activity and anti-Proteus activity, while fistuloside B is effective to only fungi. The fistuloside C showed a prominent antifungal activity with $3.1{\sim}6.2{\mu}g/ml$ of MIC and MFC. Our results indicated that fistuloside C has a prominent antifungal activity and support the use of A. fistulosum to treat microbial infection.

Changes in Quality of Welsh Onion (Allium fistulosum L.) during the Freezing Storage Period under Different Freezing Conditions (냉동조건에 따른 대파의 냉동저장 중 품질변화)

  • Purpose: This study was conducted to find the optimal freezing method and storage conditions for welsh onion. Methods: Cut welsh onions (0.3 cm) were packed in nylon/linear low density polyethylene (LLDPE) film bags, and frozen utilizing still-air freezing at -$20^{\circ}C$ (SAF20) and -$40^{\circ}C$ (SAF40), and immersed-liquid freezing at -$40^{\circ}C$ (ILF40); they were then stored at -$20^{\circ}C$ for 7 months. During storage, quality characteristics were measured monthly. Results: Drip loss was the lowest in the ILF40 packaging. Color difference in the stem (white part) did not differ significantly according to freezing conditions and storage time. Color difference in the leaf (green part) and stem was the lowest in SAF20. pH remained unchanged, while total aerobic bacterial count, pyruvic acid and moisture content decreased during storage. Pyruvic acid content of ILF40 was the highest among the freezing treatments. Fructose and glucose contents increased gradually during storage. Citric acid, malic acid, succinic acid and fumaric acid contents were unaffected, regardless of the freezing conditions. Conclusion: The optimal freezing method for welsh onions with the least quality changes was determined to be immersed liquid freezing, following by preservation up to 7 months by freeze-storing.

AlLTPs from Allium species represent a novel class of lipid transfer proteins that are localized in endomembrane compartments

  • Lipid transfer proteins (LTPs) are widely distributed in the plant kingdom, but their functions remain elusive. The proteins AlLTP2-4 were isolated from three related Allium plants: garlic (A. sativum L.), Welsh onion (A. fistulosum L.), and Nanking shallot (A. ascalonicum L.). These novel proteins comprise a new class of LTPs associated with the Ace-AMP1 from onion (A. cepa L.). The AlLTP genes encode proteins harboring 132 common amino acids and also share a high level of sequence identity. Protein characteristics and phylogenetic analysis suggest that LTPs could be classified into five distinct groups. The AlLTPs were clustered into the most distantly related plant LTP subfamily and appeared to be restricted to the Allium species. In particular, the number of amino acids existing between the fourth and fifth Cys residue was suggested as a conserved motif facilitating the categorization of all the LTP-related proteins in the family. Unlike other LTPs, AlLTPs harboring both the putative C-terminal propeptide and N-terminal signal peptide were predicted to be localized to cytoplasmic vacuoles. When a chimeric GFP protein fused with both N-terminal and C-terminal AlLTP2 signal peptides was expressed in rice cells, the fluorescence signal was detected in the endomembrane compartments, thereby confirming that AlLTPs are an unprecedented intracellular type of LTP. Collectively, our present data demonstrate that AlLTPs are a novel type of LTP associated with the Allium species.

Investigation of Optimal ionic Concentration of Nutrient Solution for the Water Culture of Young Welsh Onion (실파의 수경재배에 적합한 양액농도 구명)

  • The purpose of this experiment was to investigate optimal ionic concentration of nutrient solution for water culture of young welsh onion (Allium fistulosum). For the purpose of clarification of optimal nutrient concentration to maximize growth of young welsh onion, different nutrient concentrations of Yamazaki's solution for welsh onion seedling $(NO_3^--N\;9.0,\;NH_4^+-N\;3.0,\;PO_4^{3-}-P\;6.0,\;K^+7.0,\;Ca^{2+}\;2.0,\;Mg^{2+}\;2.0,\;and\;SO_4^{2-}-S\;4.4me{\cdot}L^{-1})$ which selected by prior experiment were treated as 0.6, 1.2, 1.8, and $2.4dS{\cdot}m^{-1}$. Increments of fresh weight, dry weight and top length were the highest in 1.2 and, in the next, were placed by the order of 1.8, 2.4, and $0.6dS{\cdot}m^{-1}$ The regression coefficients for the maximal growth of fresh weight of cv. 'Geurnjanguedaepa' and 'Tokyokuro' were $y=-42.091x^2+171.79x+11.047 (R^2=0.8946,\; R=0.9458^*)\;and\;y=-50.069x2+157.58x+15.414(R^2=0.9343,\;R=0.9692^{**})$, respectively, and optimal EC levels according to regression coefficients were 1.68 and $1.57dS{\cdot}m^{-1}$. As the conclusions, optimal nutrient levels far young welsh onion were $1.2dS{\cdot}m^{-1}$ EC in the early growth stage and $1.6\~l .7dS{\cdot}m^{-1}$ in the later growth stage.

Selection of Binder and Solid Materials for Pelleting Welsh Onion(Allium fistulosum L.) Seeds (파 펠렛종자 생산을 위한 접착제 및 피복재료 선발)

  • This study was carried out to select new pelleting binder and material for Welsh onion seeds. The optimum treatments of the various plant growth regulators to improve seed germination of the Welsh onion was also estimated. There were no significant effects of growth regulators on the germination percentage, but germination was faster according to the number of days to 50% of the final germination ($T_{50}$) and the mean number of days to germination (MDG) than those of the control. Germinability was increased when the seeds were soaked in gibberellic acid ($GA_3$) solution for 24hrs, even though there was no synergy effect on the germinability when $GA_3$ was mixed with 6-benzylaminopurine (BAP). The optimum treatment for improving germination of Welsh onion was observed when the tested seeds was soaked in 500 $\mu$M of $GA_3$ at $20^{\circ}C$ for 24hrs. Also, when the seeds soaked in the aforementioned treatment, the rate of germination was increased at lower temperature than at $20^{\circ}C$, the optimal temperature. The percentage and the speed of seed germination depended on the kinds of pelleting binder and their concentration. It showed that the higher the concentration of the binder for seeds pelleting, the lower the percentage of seed germination. Among the pelleting binder, polyvinyl alcohol (PVA) and polyvinyl pyrrolidone (PVP) were the best binders for seed pelleting, because seed germination using those binder did not affect on the concentration of binder. On the other hand, Carboxymethy cellulose (CMC) and methyl cellulose (MC) severely inhibited the seed germination The germinability was also different arcording to the pelleting materials. Among the different 58 pelleting materials, kaoline alone, the mixture of bentonite and kaoline, the mixture of bentonite, calcium carbonate and diatomaceous earth #300 were found as the best pelleting materials for welsh onion seeds.

Control Thresholds for the Management of Beet Army Worm, Spodoptera exigua (Lepidoptera : Noctuidae) on Welsh Onion (Allium fistulosum L.) (파에서 파밤나방 요방제 수준 설정)

  • Control thresholds for the management of beet army worm, Spodoptera exigua (Lepidoptera : Noctuidae) were evaluated on welsh onion of two different planting time. Two inoculation times were tested for each planting and the number of inoculated larva was 5, 10, 20, 40 per twenty plants. The injured rate of leaves was 33.5% to 10.9% at 7th September at 40 to 5 larva inoculation plot on the five days planting. That of leaves was 20.5% at 40 larva on the twenty days planting. The yields of welsh onion on non-inoculation plot were 4,395 kg per 10a. The rate of reduced yields was 49.2%(1,774.2 kg) and 36.5%(2,220.5 kg) at 40 larva inoculation on the 5 and 20 days planting, respectively. The linear relationships between population density and yield reduction were as following; it was Y=-43.474X + 3419.6 ($R^2\;=\;0.9698$) for 5 days and Y=-32.977X + 3518.9 ($R^2\;=\;0.9928$) for 20 days. Based on these results the control threshold level was estimated to be 1.8 larva for 5 days and 4.6 larva for 20 days per 20 plant. The more damaged leaves by S. exigua larva, the more yield reduction on welsh onion.

Protective Effects of Welsh Onion (Allium fistulosum L.) on Drug-induced Hepatotoxicity in Rats (랫드의 약물성 간장 장해에 대한 대파의 보호 효과)

  • The aim of this study was to evaluate the effects of Welsh onion administration on $CCl_4$-induced hepatic injury in rats. The increasing rate of the body and organ weight was not significantly different by the ad-ministration of the Welsh onion. Welsh onion (2, 4, $10\%$ w/w) was given for 4 weeks with the injections of $CCl_4$. Total serum lipid was significantly decreased in all treatment groups compared to $CCl_4$ only treatment group (p<0.05). The Welsh onion from winter season was more protective effect by lowering the serum levels of transminase (SGOT and SGPT) compared with others, the levels of transminase - phosphatase (ALP) in serum. The Welsh onion from winter at a dose of $4\%,\;10\%$ (w/w) showed significantly hepatoprotective activity which was comparable to that $CCl_4$-induced hepatic damage. Histological evaluation showed that Welsh onion partially prevented $CCl_4$-induced inflammation, necrosis and vacuolation. Pretreatment of Welsh onion reduced extent of the necrosis found 24 hr after the intraperitoneal administration of $CCl_4$. The present study shows the liver protective action of the Welsh onion against experimentally induced liver damage in rats. This suggests that the Welsh onion may be used as an effective hepatoprotective agent.

Effects of Nutrient Solution Temperatures on the Growth and Quality of Welsh Onion (Allium fistulosum L.) in Winter Season (겨울철 배양액 온도가 파의 생육 및 품질에 미치는 영향)

  • This experiment was carried out to investigate the effect of different nutrient solution temperatures on the growth and quality of welsh onion in winter season. The fresh weight increased with the increase of temperature within the range of 13-23$^{\circ}C$. The top length was longer 18 and 23$^{\circ}C$ than at 13$^{\circ}C$, the dry weight was the highest at 18$^{\circ}C$. The potassium content in plant was the highest at 18$^{\circ}C$, calcium and magnesium contents were higher at 13 and 18$^{\circ}C$ than at 23$^{\circ}C$. The vitamin C content decreased with the increase of nutrient solution temperature, but there was no statistical significance within treatments. The pyruvic acid was higher at 13 and 18$^{\circ}C$ than at 23$^{\circ}C$, while nitrate content was lower at 13 and 18$^{\circ}C$ than 23$^{\circ}C$. Therefore, for growth and quality of welsh onion, it was concluded that 18$^{\circ}C$ may be suitable nutrient solution temperature in winter season.

The Acceleration of Germination in Welsh Onion Seed Irradiated with the Low Dose ${\gamma}-ray$ Radiation (저선량 감마선 조사가 파종자의 발아에 비치는 영향)

  • To investigate the hormetic effects of the low dose ${\gamma}-ray$ radiation on the germination rate, Welsh onion (Allium fistulosum L. cv. Eunchun and cv. Sukchangwoidae) seeds were irradiated at the dose of $0.5\;{\sim}24.0$ Gy with the ${\gamma}-ray$ radiation (Co-60). The germination rate of 'Eunchun' cultivar increased about 10% in the low dose ${\gamma}-ray$ irradiation group compared with that of the control. In the 'Sukchangwoidae' cultivar, the germination rate of the 4 Gy irradiation group increased 40% more than that of the control. Broadly, it seemed that the hormetic effects of the low-dose ${\gamma}-ray$ radiation were taken more promisingly in the uncultivated soil than in the fertile soil. The germination rate from the paper towel and filter paper based cultivation increased 10% and 16% more, respectively, in the 1 Gy irradiation group than that in the control group. And the electric conductivities of the above groups supposed to be taken hormetic effects of the ${\gamma}-ray$ radiation were lower than those of the control group. From the above results, it is suggested that the low dose ${\gamma}-ray$ radiation ranged from 1 Gy to 10 Gy could have the hormetic effects on the germination rate related characters in Welsh onion seeds.
