Purification and Characterization of Catalase-2 from Deinococcus radiophilus

  • A bifunctional catalase-peroxidase, designated catalase-2, of a UV resistant Deinococcus radiophilus was purified to electrophoretic homogeneity by both chromatographic and electrophoretic methods. Its molecular weight was 310 kDa and composed of a tetramer of 80 kDa subunits. The catalase-2 exerted its optimal activity at $30^{\circ}C$ and around pH 9. Its $K_m$ value for $H_{2}0_{2} $ was about 10 mM. It showed the typical ferric heme spectrum with maximum absorption at 403 nm which shifted to 419 nm in the presence of cyanide. The ratio of A40i' A2S0 was 0.48. Fifty percent inhibition of the enzyme activity was observed at $4.6{\times}10^{-6}$, $7.7{\times}10^{-6}$, and $3.0{\times}10^{-6}$ M of NaCN, $NaN_3$, and $NH_{2}OH$, respectively. The enzyme was thermostable and not sensitive to 3-amino-1,2,4-triazole. Treatment of the enzyme with ethanol-chloroform caused a partial loss (30%) of its activity. The catalase-2 was distinct from the Deinococcal bifunctional catalase-3 in a number of properties, particularly in its molecular structure and substrate affinity.

  • The objective of this study is to find the long-age strength property and the compressive strength of age which is used as the specified concrete strength. The W/W ratio (45%, 50%, 55%, 60%) fine aggregate of useful river sand or blended sand(river sane : sea sand=1:1) were chosen as the experimental parameters. the experimental results show that pH(it means the material segregation resistance) & suspension were increased larger, so W/C become larger, and slump flow was increased as W/C increased (except W/C=60%), air-contents were decreased as W/C became increase and all of this results are satisfied with the under of 40%. The compressive strength ( a case use only river sand as fine aggregate) is showed less than the case of blended asnd. Because the unit weight of the blended sand is more heavy than the unit weigh of the river sand. The results of the case which haven been used only river sand, and the case have been used blended sand), both case have considered W/C. So it's possible to use the compressive strength of age 28 day like the case of plain concrete.

  • We studied the maturation and spawning of female Spanish mackerel (Scomberomorus niphonius) based on 445 specimens collected monthly from January to December 2004 in the coastal waters off Busan. The fork length (FL) of S. niphonius ranged from 26.1 to 105.4 cm, the gonadosomatic index (GSI) of the females was highest in May 2004, and the spawning season take place from April to July. A significant difference was detected in the sex ratio between females and males ($X^2$ test, p<0.01). The percentage of sexually mature females exceeded 50% in the 50-60 cm (FL) size group and reached 100% in the over 70 cm (FL) size group. Fecundity (F) varied between 201,156 and 836,426 egg per female. The relationship between F and FL of the fish was expressed as $F=5.8756FL^{3.8465}$. The relationship F and body weight (BW) was expressed as F=581.421 n (BW)-4108.5. The first spawning length was 41.8 cm (FL.)

Effects of Dietary Resveratrol on Growth Performance, Blood Biochemical Parameter, Immunoglobulin, and Blood Antioxidant Activity in Broiler Chicks (Resveratrol의 첨가가 육계의 생산성, 혈액 생화학 특성, 혈액 내 면역글로불린과 혈액 내 항산화 인자에 미치는 영향)

  • This study was conducted to investigate the effects of dietary resveratrol on growth performance, blood biochemical parameters, immunoglobulin, and blood antioxidant activity in broiler chicks. Three hundred twenty one-day old broiler chicks were divided 8 treatments (C(-), basal diet; C(+), basal diet with antibiotics; DL-${\alpha}$-tocopherol 20 IU; DL-${\alpha}$-tocopherol 200 IU; resveratrol 20 ppm; resveratrol 200 ppm; methylated resveratrol 20 ppm; methylated resveratrol 200 ppm) with 4 replicates and 10 birds per replicate. Birds were reared for 35 days, and, at the age of 35 days, eight birds of average weight from each replicate were selected for blood samples collection. There were no significant differences on feed intake and feed conversion ratio. But final body weight and weight gain in antibiotics, resveratrol and methylated resveratrol treatments were significantly higher than no-antibiotics and ${\alpha}$-tocopherol treatments (P<0.05). There were no significant differences on carcass rate and relative organ weights among treatments, however, weights of liver and bursa of februcius in antibiotics, resveratrol and methylated resveratrol treatment were lower than other treatments. Weight of pancreas was high in resveratrol and methylated treatment. On the cecal microflora (total microbes, Coliform bacteria, Salmonella spp., and lactic acid bacteria), these in resveratrol and methylated resveratrol treatments didn't show the differences compared with those in no-antibiotics, antibiotics, and ${\alpha}$-tocopherol treatments. In the serum, there were no significant differences on creatinine, blood urea nitrogen (BUN), total protein, albumin, globulin, alanine aminotransferase (ALT) and aspartate aminotransferase (AST) among treatments, though globulin contents of reseveratrol 200 ppm and methylated resveratrol 20 ppm treatments decreased compared to those of other treatments. Immunoglobulin (IgA, IgG and IgM) were significantly decreased in antibiotics and resveratrol treatments compared to that of no-antibiotics and ${\alpha}$-tocopherol treatments (P<0.05). Superoxide dismutase (SOD) like activity tended to increase in resveratrol groups (P<0.05), however, there was no significant difference on malondiakdehyde (MDA) content among treatments. In conclusion, these results showed that resveratrol derived from mulberry can be used as alternative of antibiotics through improvement of broiler's performance and maintain of health.

PRODUCTION RESPONSES OF CROSSBRED HOLSTEIN MILKING COWS FED UREA-TREATED RICE STRAW AT THREE DIFFERENT FIBER LEVELS

  • The experiment was conducted to determine the effect of different fiber levels on milk production of crossbred Holstein milking cows fed urea-treated rice straw (UTS) as a roughage. Eight cows were allotted into 2 squares of 4 cows each with 4 treatments by a balanced design. The treatments were 17%, 22%, and 24% crude fiber (CF) diets and Thai feeding system (free choice of roughage and 1 kg of concentrates/2 kg of milk) as a control. Body weight change was not significantly different among the treatments during the experiment. Milk production (4% FCM) and milk protein content wee not different among the treatments, but milk fat content was low in the 17% CF group and high in the control group. Cows fed the 17% CF diet consumed less UTS and more concentrates than the others, and consequently total DM intake was not different among the treatments. The feed conversion ratio was significantly higher in the control. Feed cost per kg milk was lowest in the control and highest in the 17% CF diet. The fiber content of the diet would be more than 17%, preferably 22-24% for normally producing Thai crossbred Holstein cows when the UTS was fed as a main roughage source.

Effects of Nuruk or Wheat Bran Supplemented Diet on the Serum Levels of Cholesterol and Activities of Hepatic Oxygen Free Radical Metabolizing Enzymes in Rats (누룩 또는 밀기울 첨가식이로 성장시킨 흰쥐의 혈중 Cholesterol 및 간조직 유해산소 대사효소 활성 변동)

  • The effects of nuruk and wheat bran on cholesterol level in serum and activities of free radical metabolizing enzymes were investigated in rats. The rats were fed a diet containing nuruk or wheat bran for one month. Body weight and food intake were measured. Animals were sacrificed after one month. The increased food efficiency ratio throughout whole growth period was observed in the rats fed with either nuruk containing Aspergillus terreus or wheat bran compared with control group on normal diet. In the rats fed with nuruk, hepatic GSH content, glutathione S transferase activity, hepatic cytochrome P 450 content, and aniline hydroxylase activities were generally increased. In the rats fed with nuruk containing other fungi except Aspergillus terreus, xanthine oxidase activity was decreased. The decreased cholesterol level in serum was observed in rats fed with nuruk prepared from Aspergillus terreus and wheat bran. LDL cholesterol level was decreased in rats fed with nuruk prepared with other fungi such as Penicillium sp. and Rhizopus sp. But HDL cholesterol level was increased in all groups fed with nuruk from any fungi and wheat bran. These results suggested that nuruk or wheat bran supplemented diet might exert their effect by decreasing cholesterol level in serum and amount of oxygen free radical level.

The Relation between Nutrient Intakes and Blood Parameters of Cardiovascular Function of Female College Students in Chungnam (일부 여대생의 영양소섭취와 심혈관기능 혈액지표와의 관련성에 관한 연구)

  • The purpose of this study was to investigate the relationship between nutrient intakes and blood parameters of cardiovascular function in 40 female college students on self-selected diet in Chungnam. Anthropometric measurements, analysis of dietary intakes and blood composition were conducted. Serum levels of triglyceride, total cholesterol, HDL-cholesterol, LDL-cholesterol, CK-MB, and LDH were measured by biochemical analyzer. The results were summarized as follows. The mean age of the subjects was 22.5$\pm$1.2 years. The weight, height and BMI were 52.4$\pm$7.2kg, 161.3$\pm$5.7cm and 20.2$\pm$2.4, respectively. Daily energy and lipid intakes were 1634.2$\pm$ 437.7kcal and 46.8$\pm$17.0g. And the ratio of carbohydrate, protein, and lipid to energy intake was 59.2 : 14.5 : 26.3. Serum concentrations appeared to be 61.1$\pm$ 24.2mg/dl(triglyceride), 141.2$\pm$330.mg/dl(total cholesterol), 64.8$\pm$ 20.1mg/dl(HDL-cholesterol), 64.1$\pm$ 25.4mg/dl(LDL-cholesterol), 7.2$\pm$3.2IU/l (CK-MB), 107.9$\pm$23.4IU/l (LDH). In the relation between nutrient intakes and serum parameters, vitamin $B_1$ intake showed positive correlation with triglyceride, HDL-cholesterol, and CK-MB, respectively(p<0.001, p<0.05, p<0.05). Vitamin $B_2$intake had positive correlation with triglyceride(p<0.05). And vitamin C intake had negative correlation with HDL-cholesterol(p<0.05). From these results, it is suggested that the intakes of micro nutrients such as vitamin $B_1$, vitamin $B_2$, vitamin C have relation with cardiovascular function in female college students. However, further systematic research is needed to investigate the functions of micro nutrients in healthy persons for prevention of chronic diseases.

Quality and Antioxidant Properties of Cookies Supplemented with Cabbage Powder (양배추 분말을 첨가한 쿠키의 품질 및 산화방지 활성)

  • The feasibility of incorporating cabbage powder (CP) as a value-added food ingredient into convenient food products was investigated using cookie as a model system. CP was incorporated into cookies at amounts of 0, 2, 4, 6, and 8% (w/w) based on total weight of wheat flour. pH level and moisture content of cookie dough decreased and increased significantly, respectively, with increasing levels of CP (p<0.05), whereas density was not directly affected by levels of CP incorporation. The spread ratio of cookies and their hardness increased significantly while the loss rate decreased significantly with increasing levels of CP (p<0.05). In terms of color, lightness and yellowness decreased while redness increased significantly (p<0.05) with increasing levels of CP. 2,2-Diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) radical scavenging activities were significantly elevated by CP addition, and they increased significantly as CP concentration increased in the formulation (p<0.05). Finally, consumer acceptance test indicated that the higher than 4% of CP incorporation had an adverse effect on general consumer preferences. In contrast, cookies with moderate levels of CP (2%) were recommended based on overall scores to take advantage of the antioxidant properties of CP without sacrificing consumer acceptability.

Effect of Dietary Yeast (Saccharomyces exiguus) on Growth Performance, Cecal Microflora and Fecal Ammonia Gas in Broiler Chickens (효모(Saccharomyces exiguus)의 급여가 육계 생산성, 맹장내 미생물 및 분내 암모니아 가스 발생에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary yeast (Saccharomyces exguus) supplementation on growth performance, cecal microflora and fecal ammonia gas in broiler chicks. A total of two hundred seventy, 1-d-old male broiler chicks (Ross strain) were randomly allotted to nine pens (replicates), 30 birds per pen. There were three dietary treatments with three replicates. The treatments were control (virginiamycin 0.05%+salinomycin 0.03%), Saccharomyces exguus 0.5 and 1.0%. Total body weight gain were significantly higher in Saccharomyces exguus1 1.0% treatment than the control (P<0.05). Although not significant, the yeast supplementation tended to improve the feed conversion ratio. No significant differences were observed on the numbers of cecal E. coli, Salmonella and Lactobacillus in yeast treatments compared to those of control. The production of fecal ammonia gas was significantly lower in yeast treatments than the control (P<0.05). The concentrations of fecal short chain fatty acids and volatile organic compounds were not different among the groups. These results suggest the possibility that yeast (Saccharomyces exguus) could be used as the alternative of antibiotic growth promoters by improving the performance of broiler chicks. In addition, dietary yeast could improve the environment of broiler houses by reducing fecal ammonia production.

The Relation between Net Rate of Endogenous Noncarbonic Acid Production from Diet Potassium and Protein Intakes and Bone Mineral Density in Korean Women (일부 한국 여성에 있어 칼륨과 단백질의 섭취량에 의한 NEAP(net rate of endogenous noncarbonic acid production)와 골밀도와의 관련성)

  • The purpose of this study was to investigate the relation between NEAP (net rate of endogenous noncarbonic acid production) from dietary potassium and protein intakes, and bone mineral density and urinary deoxypyridinoline (DPD) in Korean women. The subjects were 276 women aged $18\sim81$, and they were asked about general characteristics and nutrient intake using 24-hr recall method. Also, their bone mineral density of spine and femoral neck were measured using DEXA. Urinary DPD analysis was done in 106 postmenopausal women. The average energy, protein and potassium intake were 1503.0 kcal, 57.7 g and 60.8 mEq, respectively. Mean NEAP [54.5$\times${protein intake (g)/potassium intake (mEq)}-10.2] was 44.6. NEAP showed a significantly negative correlation with spine bone mineral density of the subjects after controlling for confounding factors (age, height and weight) (p<0.05). The lowest quartile of protein intake had the highest spine bone mineral density after controlling for confounding factors (p<0.05). There was no significant correlation between urinary DPD and calculated NEAP. These results provided positive relation between a ratio of lower protein to higher potassium dietary intake and bone mineral density.
