Transition-State Variation in the Solvolysis of Benzoyl Chlorides$^*$

  • Solvolysis reactions of some substituted benzoyl chlorides were studied in ethanol-water, ethanol-trifluoroethanol and methanol-acetonitrile mixtures. Results showed that the reaction proceeds via an $S_N2$ process in which bond formation is more advanced than bond cleavage. Comparison of the two models for predicting transition state variation indicated superior nature of the quantum mechanical model relative to the potential energy surface model.

Kinetics of Base Hydrolysis of Some Chromen-2-one Indicator Dyes in Different Solvents at Different Temperatures (여러 온도 및 용매 하에서 수행된 chromen-2-one 지시약 염료들의 염기성 가수분해 반응에 대한 속도론적 연구)

  • Base hydrolysis of 7-hydroxy-2H-chromen-2-one (HC) and 7-hydroxy-2H-chromen-2-one-4-acetic acid (HCA) in aqueous-methanol and aqueous-acetone mixtures were studied kinetically at temperature range from 283 to 313 K. The activation parameters of the reactions were evaluated and discussed. Moreover, the change in the activation energy barrier of the investigated compounds from water to water-methanol and water-acetone mixtures was estimated from the kinetic data. It is observed that the change in activation barriers is more or less the same for the hydrolysis of HC and HCA. Base hydrolysis of HC and HCA follows a rate law with $k_{obs}=k_2[OH^-]$. The decrease in the rate constants of HC and HCA as the proportion of methanol or acetone increases is due to the destabilization of $OH^-$ ion. The high negative values of entropy of activation support the proposal mechanism, i.e. the investigated reaction takes place via the formation of an intermediate complex. Moreover, these values refer to the rigidity and stability of the intermediate complex. Thus, the ring opening of the intermediate complex would be the rate controlling step.

  • A simple and sensitive HPLC method was developed for the simultaneous determination of ten kinds of active ingredients formulated in commercial cough-cold mixtures. A group of Pseudoephedrine HCl, dl-Methylephedrine HCl, Noscapine, Chlorophenylamine maleate, Dextromethorphan HBr and Phenylpropanolamine HCl were determined at 254nm using a Novapak $C_{18}$ column with mobile phase consisting of a mixture of methanol-acetonitrile-1, 4dioxane-tetrahydrofuran-water(12 : 20 : 20 : 5 : 43, pH4.7) containing 0.013M-dioctyl sodium sulfosuccinate. The another group of Acetoaminophen, Caffeine, Guaifenesin and Ethenzamide were also determined at 254nm using a Novapak $C_{18}$ column as the stationary phase, and a mixture of methanol-1% aqueous acetic acid (3 : 7). The results indicate that these methods are accurate and precise with relative standard deviation of not more than 1% (n=5) for the above active ingredients.

  • A rapid and simple quantitative assay method for aminoglycoside-3'- phosphotransferase (APH(3')) derived from E. coli ATCC 21990 was developed using the thin layer chromatographic densitometry, 3'-phosphorylated kanamycin B (3'-PKMB), product of APH (3') reaction, was separated from reaction mixtures by developing on the silica gel TLC plate with chloroform-methanol-ammonia water (3:4:3). The quantity of the 3'-PKMB was measured by densitometry after color development by ninhydrin method. Densitometric TLC assay for APH (3') was showed a good quantitative result and reproducibility. Sensitivity of this assay was 1.56 nmol of 3'-PKMB and could be analyzed many samples at same time. This method may be applicable for the analysis of inactivating enzymes of aminoglycoside antibiotics.

Dispersion Polymerization of Acrylamide in Methanol/Water Media

  • Dispersion polymerization of acrylamide was carried out in the media of methyl alcohol/$H_2O$ mixtures using hydroxypropyl cellulose and ammonium persulfate as steric stabilizer and initiator, respectively. The effects of concentrations of initiator and steric stabilizer, amount of monomer, polymerization temperature, methyl alcohol/$H_2O$ ratio, and purification of monomer and nitrogen purge on the particle size of the latices and molecular weight of the polymers were investigated. The average particle diameter increased with increasing concentration of initiator, water content in methyl alcohol/$H_2O$ media, and polymerization temperature, but decreased with monomer and stabilizer concentrations. The viscosity average molecular weight increased with increasing concentrations of monomer, steric stabilizer, and water content in dispersion media, but decreased with initiator concentration and polymerization temperature. The PAM polymers prepared with the purified monomer and the nitrogen purging before the reaction showed the highest molecular weight.

  • Kinetic studies on the solvolysis of para-methyl benzyl nitrate and benzyl nitrate were carried out in aqueous methanol, ethanol, acetonitrile, acetone, tetrahydrofuran and dioxane mixtures at 60$^{\circ}$C. The rates were faster in protic solvent mixtures than in aprotic solvent mixtures. This was considered in the light of transition state stabilization by hydrogen bonding solvation of protic solvent mixtures. Grunwald-Winstein equation, extended Grunwald-Winstein equation and correlation between E$_T$(30) and rate constant were applied in order to discuss the transition state variations caused by changing benzyl substituents and solvents. The results showed that strong electrophilic assistance of solvent is operative in the the water-rich solvents.

  • By the 505 chromotest which utilized Escherichia bolt PQ 37, Korean medicinal plants had been screened to Investigate the antimutagenic effect to aflatoxin B1(AFBl). Ikmocho(IMC, Leonurus sibiricus L.) was extracted with hot water. The extract was not found to be mutagenic in the Salmonella mutation test with or without metabolic activation, and the extract was showed to possess the antimutagenic properties towards AFB1-induced metation. The mutagenicity of AFB1 was inhibited by methanol soluble fracstion (IMC-MS) in dose-dependent. However, water-soluble fraction exhibited comutagenic activity. The greatest inhibitory effect of IMC-MS on AFB1 mutagenicity occurred when IMC-MS was first incubated, AFB1 followed by a second incubation with the cells and 59 mixture. Also lower inhibition was occurred when S9 mixtures were first incubated, with IMC-MS followed by a second incubation with AFBI. The results of the sequential incubation study support the probability that one mechanism of inhibition could involve the formation of chemical complex between IMC-MS and AFB1 rather than deactivation of S9 enzyme.

  • In order to prepare polyion complex membranes useful for the separation of water-alcohol by pervaporation, k-carrageenan containing artionic sulfate groups in the backbone and good hydrophilicity was selected for the polyanion membrane material and poly{1,3-bis[4-butyl pyridinium] propane. bromide}, one of the polycations synthesized in our lab and containing cationic pyridinium groups., was used. The polyion complex membranes were prepared by the ion complex formation between kcarrageenan films and poly{1,3-bis[4-butyl pyridinium] propane. bromide}. On the formation process of polyion complex membranes, the way of potyion complex formation was carefully studied. In order to study the effect of the morphology on the permeation properties of the polyion complex membranes, which is one of the important factors affecting on the permeation properties of membranes but rarely studied, the microstructure behaviors of the polyion complex mem6ranes in methanol-water mixtures with different compositions Were also studied with x-ray diffractometry and polarizing microscopy.

Vapor Permeation Characteristics of TiO2 Composite Membranes Prepared on Porous Stainless Steel Support by Sol-Gel Method

  • Composite membranes with a titania layer were prepared by soaking-rolling method with the titania sol of nanoparticles formed in the sol-gel process and investigated regarding the vapor permeation of various organic mixtures. The support modification was conducted by pressing $SiO_2$ xerogel of 500 nm in particle size under 10 MPa on the surface of a porous stainless steel (SUS) substrate and designed the multi-layered structure by coating the intermediate layer of ${\gamma}-Al_2O_3$. Microstructure of titania membrane was affected by heat-treatment and synthesis conditions of precursor sol, and titania formed at calcination temperature of 300$^{\circ}C$ with sol of [$H^+$]/[TIP]=0.3 possessed surface area of 210 $m^2$/g, average pore size of 1.25 nm. The titania composite membrane showed high $H_2/N_2$ selectivity and water/ethanol selectivity as 25-30 and 50-100, respectively. As a result of vapor permeation for water-alcohol and alcohol-alcohol mixture, titania composite membrane showed water-permselective and molecular-sieve permeation behavior. However, water/methanol selectivity of the membrane was very low because of chemical affinity of permeants for the membrane by similar physicochemical properties of water and methanol.
