The purpose of this study was to investigate the nitrite scavenging activity of the ethanol and water extracts obtained from Hizikia fusiforme and Sargassum fulvelum using sodium nitrite under different pH conditions. The nitrite scavenging activity was appeared in all the extracts examined and it showed the highest value at pH 1.2 The ethanol extract of Hizikia fusiforme provided higher activity than the water extract of that. Ohterwise, the nitrite scavenging activity of Sargassum fulvelum ethanol extract was similar to that Sargassum fulvelum water extract. There was significant difference among concentration of extracts Hizikia fusiforme and Sargassum fulvelum. Also, nitrite scavenging activity of the extracts Hizikia fusiforme and Sargassum fulvelum decreased with higher pH condition. In addition, the activity was increaed with higher concentration of extracts Hizikia fusiforme and Sargassum fulvelum in the course of reaction time 30 minute to 3 hour.
The present study was to investigate the antioxidant activity in ethanol and water extracts of root and stem of Sargassum coreanum. Antioxidant activities were evaluated by total polyphenol contents, DPPH radical scavenging activity, chelating effect, reducing power, and rancimat method. Total polyphenol contents of ethanol and water extracts were 32.79 mg/g and 15.55 mg/g, respectively. Ethanol extract showed higher DPPH radical scavenging activity than water extract and similar activity to BHT. Reducing power of extracts was increased in a concentration-dependent manner and ethanol extract had more reducing power than water extract. Ethanol and water extracts have little chelating effect at all concentrations. Antioxidant index (AI) of ethanol extract measured by Rancimat was higher than that of water extract, but their AI was lower than that of BHT. These results indicate that ethanol extract of S. coreanum root and stem has more potent antioxidant activity than water extract through DPPH radical scavenging and reducing power, and could potentially be used as a good source of natural antioxidants.
본 연구는 미나리의 최적 추출 조건을 확립하기 위해 다양한 에탄올추출 농도가 미나리의 영양학적 성분 변화에 미치는 정도를 관찰함으로써 진행되었다. 본 실험에서는 열수추출물, 에탄올추출물(50%, 80%, 95%)로 추출 농도를 다르게 하여 미나리를 추출하였다. 수율을 측정한 결과 50% 에탄올 추출물에서 44.67%로 가장 높게 나타났다. 일반성분 분석 결과, 50% 에탄올추출물에서 단백질 함량이 6.76%, 탄수화물 함량이 19.60%로 가장 높게 나타났다. 지방 함량은 열수추출물과 비교하여 에탄올추출물에서 유의적으로 증가하였으며(P<0.05), 에탄올추출 농도에 따라 유의차가 있었다 (P<0.05). 식이섬유를 측정한 결과, 50% 에탄올추출물을 제외한 에탄올추출물(80%, 95%)에서 열수추출물과 유의차가 나타났으며(P<0.05), 두 실험군 간에도 유의적 차이가 있었다(P<0.05). 총당 함량을 측정한 결과, 열수추출물에 대해 에탄올추출물은 통계적 유의차가 있었고(P<0.05), 에탄올 농도에 따라서도 유의차를 나타냈으며(P<0.05), 95% 에탄올추출 시 5.35%로 가장 낮게 측정되었다. 비타민 A 함량은 열수추출물에 대해 에탄올추출물이 유의하게 증가하였으며(P<0.05), 에탄올추출 농도에 따라 유의차가 있었다(P<0.05). 무기성분 측정 결과, 나트륨, 칼슘, 철의 경우 열수추출물에 대해 에탄올추출물이 유의하게 감소하였고(P<0.05), 에탄올추출 농도가 증가함에 따라 유의적으로 감소하였다(P<0.05). 따라서 에탄올추출 방법을 사용 목적에 따라 적용함으로써 미나리를 이용한 우수한 가공식품 개발 가능성과 이용률 향상에 도움이 될 것으로 판단된다.
In order to select the optimum drying method for the production of cinnamon extract, water extract and 70% ethanol extract of cinnamon were prepared. And then several drying method of oven drying, vacuum evaporation, spray drying and freeze drying were performed. Content of cinnamic acid, cinnamic aldehyde, eugenol, tannin and free sugar, and antioxidant activity, degree of browning, pH, color value, turbity and solubility were compared. In water extract, contents of cinnamic acid, cinnamic aldehyde, eugenol were 29.45mg/100g, 94.86mg/100g, 120.75mg/100g and decreased to 4.76%∼44.21%, 5.30%∼48.05%, 3.66%∼21.83% by oven dyring, vaccum drying, spray drying respectively, but freeze drying showed a little decrease of those components. In 70% ethanol extract, effectual components decreased to 76.05%∼88.38% and 26.86%∼78.76% by freeze drying and vacuum evaporation respectively. Antioxidant activity decreased by drying and decreasing rate in 70% ethanol extract was lower than water extract. Degree of browning increased as the drying temperature increased. Tannin and free sugars were little affected by drying temperature. Solubility decreased in oven drying and 70% ethanol extract. Overall data suggested that optimum drying methods of cinnamon extract were freeze drying in case of water extract and freeze drying and vaccum drying in case of 70% ethanol extract.
Skin anti-wrinkle activities of the stems and leaves of Abeliophyllum distichum Nakai were evaluated by the extracts obtained from various extraction processes such as using hot water at $100^{\circ}C$, 70% ethanol at $85^{\circ}C$, and 70% ethanol with ultrasonication at $60^{\circ}C$ The ultrasonicated extract showed 95.62% of the highest cell viability in addition of $0.3mg/m{\ell}$ of the extracts into the normal human fibroblast cell, CCD-986sk. For antioxidant activities, the extracts using ultrasonicated extract showed the highest DPPH free radical scavenging as 80.27%, followed by 75.88% and 62.44% for the extracts using ethanol extract and water extract. The ultrasonicated extract also showed the highest elastase inhibition activity as 25.32%, compared to ethanol extract and water extract based method at 22.01% and 12.88%, respectively. MMP-1 production was most effectively decreased down to $2908.1pg/m{\ell}$ with ultrasonicated extract while $6640.8pg/m{\ell}$ with water extract and $3609.3pg/m{\ell}$ with ethanol extract, in addition of $0.3mg/m{\ell}$. Collagen production was increased up to $154.7ng/m{\ell}$ in addition of ultrasonicated extract, and followed by $121.4ng/m{\ell}$ and $31.2ng/m{\ell}$ for ethanol extract and water extract, respectively. These results indicate that the ethanol extract should have skin anti-wrinkling activities and can be improved by the ultrasonication process that high energy input elute more amounts of bioactive substances eluting more amounts of bioactive substances from the high energy input of ultrasonication.
Alzheimer's disease (AD), most common form of dementia is characterized that memory deficit and loss of cognitive function. This study was evaluated cognitive enhancing effect of water and ethanol extracts of Codonopsis lanceolata and compared using Morris water maze and passive avoidance test. The water and 70% ethanol extracts (100, 300 and 500 mg/kg) were administered to mice. The neuroprotective effect on glutamate-induced cell death in HT22 cells was additionally investigated using MTT assay. Results showed 70% ethanol extract of Codonopsis lanceolata enhanced cognitive function than water extract, as shown by decrease in escape latency time in Morris water maze test. In passive avoidance test, 70% ethanol extract also increased the latency time compared to the water extract. Furthermore, 70% ethanol extract significantly protected neuronal cell against glutamate cytotoxicity and showed higher than neuroprotective effect of water extract. These results indicate that 70% ethanol extract more improve spatial cognitive ability and protected neuronal cells than water extract.
The effects of dried leaf powders and water and ethanol extracts of persimmon and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity were investigated in 12-month-old rats. Forty-nine male Sprague-Dawley rats weighing 520$\pm$19g were blocked into seven groups according to body weight. Rats were raised for four weeks with control(no tea leaf powder or extracts) and experimental diets containing either 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) or green tea(Camellia sinensis O. Ktze), or water or ethanol extract from equal amounts of each dried tea powder. Food intakes of all tea diet groups were higher than that of control. Weight gains and food efficiency ratios of all tea diet groups were not significantly different from those of control. All tea diets decreased plasma triglyceride level, especially, green tea powder and persimmon ethanol diets were more effective than other diet. All the tea diet groups showed decrease in liver triglyceride level, and persimmon powder and ethanol extract increased fecal triglyceride excretion. Plasma cholesterol levels of all the tea diet groups were not significantly different from the control, but control. Fecal cholesterliver cholesterol concegroups were significantlntrations of all tea y lower than that of ol excretions of persimmon powder, green tea ethanol extract, persommon ethanol extract and green tea ethanol extract groups were significantly higher than that of control. Plasma and liver thiobarbituric acid reactive substance(TBARS) concentrations of all the tea diet groups were lower than that of control. Especially, plasma TBARS concentrations of green tea powder and persimmon ethanol extract groups were sinificantly low. Red blood cell(RBC) superoxide dismutase(SOD) activities of persimmon ethanol extract and green tea water extract groups were increased, and RBC catalase activities of all experimental groups were not significantly different. RBC glutathione peroxidase(GSH-px) activities of persimmon ethanol extract, persimmon water extract and green tea powder groups were increased. Liver SOD activities of all the tea diet groups except green tea ethanol extract group were higher than that of control. Liver catalase activities of all experimental groups were not significantly different, and liver GSH-px activity of green tea powder group was significantly higher than that of control. In conclusion, dried leaf powders, and water and ethanol extracts of persimmon and green tea were effective in lowering lipid level, inhibiting lipid peroxidation and increasing antioxdative enzyme activities in 12-month-old rat. Green tea leaf powder with high contents of flavonoids and water soluble dietar fiber was most effective in lowering plasma triglyceride, cholesterol and TBARS level. This study was designed to investigate the effect of medicinal herb extract on the antioxidant and antimicrobial activities against Helicobacter pylori, which is known as a ulcerogenic pathogen. The concentration of total phenolic compound of Scutellaria baicalensis(13.14%) was highest among water extracts and that of Ulmus parvifolia(15.12%) was highest among the ethanol extracts. The antioxidant activity of the water extract of Scutellaria baicalensis and of the ethanol extract of Ulmus parvifolia were 91.00% and 65.03%, respectively, in DPPH assay. The antioxidant activity of the water extract of Scutellaria baicalensis and of the ethanol extract of Ulmus parvifolia were 32.90% and 27.70%, respectively, in SOD assay. The antioxidant activity of the water extract of Ulmus parvifolia and of the ethanol extract of Glycyrrhiza uralensis Fischer was 2.15 and 2.17, respectively, in TBARS assay. In disc method, Scutellaria baicalensis showed the highest anti-microbial activity against H. pylori, followed by Glycyrrhiza uralensis Fischer among the water extracts and Glycyrrhiza uralensis Fischer showed the highest anti-microbial activity followed by Radix puerariae among ethanol extract.
Objectives : This study aimed to evaluate the effect of Wongisaengmaek-san (extracted with water or 50% ethanol) on antioxidative activity and recovery of fatigue induced by weight-loaded forced swimming exercise. Methods : Antioxidant activity of Wongisaengmaek-san was evaluated in terms of total amount of polyphenol, 1.1-diphenyl-2-picrylhvdroxyl (DPPH) radical scavenging activity and xanthine oxidase inhibition. The anti-fatigue effect of Wongisaengmaek-san was investigated using an acute weight-loaded forced swimming test by monitoring swimming test times and blood biochemical parameters creatinine. BUN, lactic acid dehydrogenase (LDH), free fatty acid (FFA) and lactic acid (LA). Results : 1. 50% EE (ethanol extract) had 2.3 times higher amount of total polyphenol compared to water extract. 2. It was identified that 50% ethanol extract showed enhancement of xanthine oxidase inhibition and DPPH scavenging effect in vitro. 3. According to anti-fatigue effect in the scale of 5% weight loaded mouse swimming test, both water and 50% ethanol extract showed significant improvement of swimming time elongation respectively, and 50% ethanol extract induced a more positive result. 4. After swimming for 1% weight-loaded mouse. water extract and 50% ethanol showed significant anti-fatigue effect with manifestation of blood sampling among both of the intervention: 50% ethanol extract showed a greater result. Conclusions : 50% ethanol extract of Wongisaengmaek-san has more effective antioxidant activity and anti-fatigue than water extract.
In Korea and China, cherry elaeagnus (Elaeagnus multiflora Thunb.) has been used traditionally to treat cough, diarrhea, itching, and foul sores. Therefore, in this study, the ethanol and water extracts of cherry elaeagnus leaves were examined for their antioxidant activities. The ethanol extract of the cherry elaeagnus leaves contained more phenolics than the water extract. All the cherry elaeagnus leaf extracts had higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability than ascorbic acid at concentrations of $250-1,000\;{\mu}g/mL$. The ethanol extract also showed higher superoxide dismutase (SOD)-like activity compared to the water extract. Furthermore, the SOD-like activity of the ethanol extract amounted to 89% of that of ascorbic acid at a concentration of $500\;{\mu}g/mL$. The nitrite scavenging ability and xanthine oxidase inhibitory (XOI) activity of the ethanol extract were higher than those of the water extract. In particular, the ethanol extract had higher XOI activity than ascorbic acid at a concentration of $1,000\;{\mu}g/mL$.
