A Study on the Effects of Rhodiola rosea Root on the Cancers (홍경천(紅景天)(Rhodiola rosea Root)의 항암(抗癌)효과에 대한 연구(硏究))

  • Objectives : Effects of Rhodiola rosea root on cancers on stomach, breast, lung, and liver were examined. Methods : Water extracts and methanol extracts of Rhodiola rosea root were treated on cancer cells, and its effects on cancers were examined. Results : 1. Water extracts and methanol extracts of Rhodiola rosea root was less harmful in its lowest density 0.25 mg/mL (9.l%와 10.5%), but became more harmful as its density increased. 2. As for human stomach adenocarcinoma cells AGS, breast adenocarcinoma cells MCF-7, and lung carcinoma cells A549, methanol extracts showed 70-77% inhibition of cancer cells in high density(1 mg/mL), and water extracts showed 60-70% inhibition rate and its selective death rate was less than 2.5. Conclusion : Rhodiola rosea root can be used to treat cancers and to increase immunity.

Effects of Samul-tang on Nitric Oxide Induced-cytotoxicity in C6 Glial Cell (Nitric Oxide에 의해 유발된 C6 glial 세포독성(細胞毒性)에 대한 사물탕(四物湯)의 방어효과(防禦效果))

  • The water extracts of Samul-tang(SMT) has been used for treatment of ischemic brain damage in Oriental traditional medicine. However, little is known about the mechanism by which the water extracts of SMT rescues brain cells from ischemic damages. To elucidate the protective mechanism on ischemic induced cytotoxicity, I investigate the regulation of LPS and PMA induced iNOS expression in C6 glial cells. LPS and PMA treatment for 72 h in C6 glial cells markedly induce nitric oxide(NO), but treatment of the cells with the water extracts of SMT decrease. dose dependently nitrite formation. In addition, LPS and PMA treatment for 72 h induce severe cell death and LDH release in C6 glial cells. However treatment of the cells with the water extracts of SMT dose not induce significant changes compare to control cells. Furthermore, the protective effects of the water extracts of SMT is mimicked by treatment of $N^{G}MMA$, a specific inhibitor of NOS. LPS and PMA induced iNOS activation in C6 glial cells cause chromosomal condensation and fragmentation of nuclei by caspase activation. The treatment of the cells with the water extracts of SMT may suppress apoptosis via caspase inhibition by regulation of iNOS expression. Taken together, I suggest that the protective effects of the water extracts of SMT against ischemic brain damages may be mediated by regulation of iNOS during ischemic condition.

Antiasthmatic effects on Pinellia ternate Breitenbach(PTB) water extracts against airway inflammation and hyperresponsiveness induced by diesel exhaust particles with ovalbumin sensitization in BALB/c mice (생쥐에서 ovalbumin과 디젤배기가스 입자로 유도된 기도염증과 기도 과민성에 대한 반하 물 추출물의 항천식 효과)

  • Objectives : Asthma is a chronic, complex respiratory disease, caused by airway obstruction, airway eosinophilic inflammation(AEI), and airway hyperresponsiveness(AHR). This study was conducted to determine whether oral administration of crude water extracts of Pinellia ternate Breitenbach(PTB) has an antiasthmatic potential in the treatment of asthma in mice. Methods : Asthmatic AEI and AHR were induced by systemic sensitization to ovalbumin(OVA) by intratracheal instillation with 0.1 mg/mL suspension of diesel exhaust particles(DEP) once a week for 10 weeks in BALB/c mice. Crude PTB water extracts(50 mg/kg and 200 mg/kg) were orally administered 5 times a week for 10 weeks. Cyclosporin(10 mg/kg) was administrered the same manner as a positive control. Results : Long-term treatment with crude PTB water extracts suppressed the infiltration of inflammatory cells, including eosinophils, into airways from blood. It also reduced asthmatic AEI and AHR by attenuating the increase in the levels of cytokines such as interleukin(IL)-4, IL-5 and IL-13 in bronchoalveolar lavage fluid(BALF), as well as the levels of histamine and OVA-specific IgE in blood. However, the effect of crude PTB water extracts(200 mg/kg) was not likely to be stronger than that of cyclosporin(10 mg/kg). Conclusion : These results suggest that crude PTB water extracts have inhibitory effects on AEI and AHR in a mouse model of asthma and may act as a potential Th2 cytokine antagonist, and have a therapeutic effect on allergic asthma.

In vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and inhibition of nitric oxide production activities of methanol and hot water extracts of Russula rosacea mushroom

  • Russula rosacea, a mycorrhizal fungus, has been used for edible and medicinal purposes. This study was conducted to evaluate the in vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and nitric oxide inhibitory effects of the fruiting bodies from R. rosacea extracted with methanol, and hot water. The 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activities of the methanol and hot water extracts (2.0 mg/ml) of R. rosacea were comparable with BHT, the positive control. The chelating effects of the mushroom and hot water extracts were significantly higher than that of BHT. The reducing power of methanol and hot water extract (6 mg/ml) were significantly lower than that of BHT. Seven phenolic compounds were detected from acetonitrile and hydrochloric acid solvent extract of the mushroom. alpha-amylase and alpha-glucosidase inhibitory activities of methanol and hot water extracts were lower than that of acarbose, the positive control. The acetylcholinesterase and butyrylcholinesterase inhibitory effects were moderate compared with galanthamine, the standard drug. Nitric oxide (NO) production in lipopolysaccharide (LPS) induced RAW 264.7 cells were inhibited significantly by the mushroom extracts in a concentration dependent manner. Therefore, we demonstrated that fruiting bodies of R. rosacea possess in vitro antioxidant, anti-hyperglycemic, anti-cholinesterase, and NO production inhibitory activities. The experimental results suggest that the fruiting bodies of R. rosacea are good natural antioxidant, anti-hyperglycemic, anti-cholinesterase, and anti-inflammatory sources.

Toxicity Monitoring of River Sediments in the Geum River Basin using Daphnia magna and Moina macrocopa (Daphnia magna와 Moina macrocopa를 이용한 금강수계 하천퇴적물 생태독성 모니터링)

  • In this study, toxicity monitoring of sediments collected from 25 stations in the Geum river basin was conducted using Daphnia magna and Moina macrocopa. According to the results of acute toxicity tests (immobilization and mortality) of organic extracts of semdiments, Miho stream showed much less toxicity than Gap and Nonsan streams. In particular, significant toxicity was observed in both species for St.15 and St.16 sediment samples that passed through Deajeon city as a branch of Gap stream. For Nonsan stream, St.23 sediment showed high toxicity toward M. macrocopa. This site seemed to be affected by upper agricultural industrial complex. Additionally, M. macrocopa showed a higher sensitivity than D. magna for organic extracts of sediments. In the case of toxicity tests using sediment pore water and aqueous extracts, only pore water of St.27 sediment was toxic against D. magna. Toxicity identification evaluation showed that hydrogen sulfide was likely a major toxicant in the pore water.

Effect of Hot-water Extracts from Laminaria japonicus on Melanin Production in B16 Melanoma Cells (Bl6 Melanoma 세포에서 다시마 열수추출물이 멜라닌 생성에 미치는 영향)

  • To investigate the whitening activity of Laminaria japonicus, we measured the effect of hot-water extracts on melanin production in B16 melanoma cells. Melanocyte stimulating h ormone (MSH, $1{\mu}M$)-stimulated melanin production in B16 melanoma cells was significantly inhibited by fucoidan and hot-water extracts, but not by alginate. The purified tyrosinase activity was not affected by hot-water extracts, alginate and fucoidan. However, tyrosinase expression was significantly inhibited by fucoidan and hot-water extract in Western blot. These results suggest that inhibitory mechanism of hot-water extracts on melanin production may be due to the inhibition of tyrosinase expression but not to direct inhibition of tyrosinase, such an effect of which may be dependent on fucoidan.

Antioxidant activity of water and alcohol extracts of Thuja orientalis leaves

  • Water and alcohol extracts were prepared from dried and powdered leaves of Thuja orientalis (T. orientalis). The reducing power, total phenolic content, the 1,1-diphenyl-2-picrylhydrazyl scavenging activity, inhibitory effect on Fe (II)-EDTA-$H_{2}O_{2}$ (Fenton reaction system) induced DNA damage and inhibitory effect on human red blood cell (RBC) hemolysis were evaluated in the present study. At a concentration of 200 mg, water and alcohol extracts of T. orientalis inhibited the hydrolysis of DNA by 72.859% and 65.312%, respectively. Water and alcohol extracts of T. orientalis also inhibited 2,2'-Azobis(2-amidinopropane) dihydrochloride induced RBC hemolysis to the extent of 69.30% and 54.55%, respectively. The reducing power and antioxidative activity of water extract was found to be more than that of alcohol extract. This is attributable to the presence of higher amount of phenolic compounds in water extract. The present results indicate that the T. orientalis extracts are rich sources of natural antioxidants and can protect DNA and human red blood cells against free radical induced oxidative damage.

Antimutagenic and Cytotoxic Activity of Ethanol and Water Extracts from Rubus coreanum (복분자 딸기(Rubus coreanum) 에탄올 및 열수추출물의 항돌연변이 활성과 암세포 성장 억제 효과)

  • The antimutagenic and cytotoxic activities of ethanol and water extracts from Rubus coreanum were investigated in this study. Their antimutagenic activities were measured by the Ames test and their cytotoxic activities were evaluated by the growth inhibition of cancer cells via the 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) assay and the sulforhodamine B (SRB) assay. In the results, the inhibition rates of the ethanol and water extracts toward mutagenicity induced by 4-NQO were 95.0% and 93.6% at 5 mg/plate, respectively, while their inhibition rates against mutagenicity induced by sodium azide were 27.2% and 40.8%, respectively. According to MTT assay, the cytotoxicity values of the ethanol extract against Hep3B and HeLa cells were 67.2% and 68.5%, respectively, and the values for the water extract were 65.8% and 66.4%, respectively. In the SRB assay, the ethanol and water extracts inhibited over 60% of cancer cell growth. In conclusion, both the ethanol and water extracts of Rubus coreanum offer potentially good antimutagenic and anticancer effects.

Anti-diabetic Effect of Dried-silkworm Dongchunghacho in Streptozotocin-induced Diabetic Rats (Streptozotocin 유발 당뇨모델을 이용한 건조누에 동충하초의 항당뇨 효과)

  • The anti-diabetic effect of dried-silkworm Dongchunghacho water extracts was investigated in streptozotocin (STZ)-induced diabetic rats. Seven-week-old SD rats were randomly assigned to six groups: normal control (NC) group, diabetic control (DC) group, water extracts of Paecilomyces japonica grown on a dried-silkworm (PJ-DS, 50 mg/kg) group, PJ-DS (250 mg/kg) group, water extracts of Cordyceps militaris grown on a dried-silkworm (CM-DS, 250 mg/kg) group, and water extracts of dried-silkworm (DS, 250 mg/kg) group. These groups were orally provided with sample dissolved in water respectively for four weeks after an injection of STZ (60 mg/kg, ip) followed by identification of diabetic control (DC) group. After four weeks, body weight of all diabetic groups was significantly lower than that of the normal control (NC) group, but among diabetic groups there were no significant differences. Blood glucose levels of the PJ-DS (50 mg/kg), PJ-DS (250 mg/kg), CM-DS (250 mg/kg) and DS (250 mg/kg) groups were reduced compared to the DC group by 3.0 percent, 18.5 percent, 6.9 percent and 13.1 percent, respectively. Concentration of total cholesterol and triglyceride in blood plasma were to some degree decreased in PJ-DS (250 mg/kg) and DS (250 mg/kg) groups compared to the DC group. According to these results dried-silkworm Dongchunghacho water extracts (PJ-DS) boost protection against STZ-induced toxicity.

Effects of Water Extracts in fruits of Omija (Schizandra chinensis Baillon) on Alcohol Metabolism (오미자 열매의 물추출물이 알콜대사에 미치는 효과)

  • To assess the effects of water extracts in fruits of Omija (Schizandra chinensis Baillon) on alcohol metabolism, rats were orally administrated with alcohol (25% alcohol, 0.75g/200g B.W., 40% alcohol, 0.8g/200g B.W.). The level of metabolites and enzyme activities of the serum and liver were unchanged by the 25% ethanol or 40% ethanol treatment with acute orally administration. Blood alcohol level was markdely decreased by the treatment with water extracts in fruits of Omija. The serum level of Urea nitrogen, Free fatty acid, GPT and LDH were tended to decreased, level of GOT was unchanged. Contents of hepatic microsomal protein, glycogen, pyruvate in the liver were increased by water extracts in fruits of Omija. In conclusion, the present study clearly demonstrates that water extract in fruits of Omija promotes the overall metabolism and detoxication of alcohol.
