Objectives : This study was conducted to evaluate the antioxidative and anticancer activity of the water and ethanol extracts from medicinal herbs. Methods : Three kinds of medicinal herbs(Corni fructus, Astragalus membranaceus and Glycyrrhiza uralensis) were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidative and anticancer activities. Results : The highest polyphenol contents of the water and ethanol extracts from medicinal herbs were 342.14 mg and 435.62 mg per 100 g of Cornus officinalis, respectively. The highest electron donating abilities (EDA) of the water and ethanol extracts from Glycyrrhiza uralensis were 88% and 91% at 1,000 ppm, respectively. The water and ethanol extracts from Astragalus membranaceus had the highest nitrite scavenging abilities (NSA) at 1,000 ppm. The highest anticancer activity of the extracts were from Glycyrrhiza uralensis against both of MDA and A549 cells. Conclusions : These results suggest that the medicinal herbs can be used as natural antioxidant to prevent oxidative damage in normal cells probably because of their antioxidant characteristics.
Objectives : This study was conducted to evaluate the antioxidative and anticancer activity of the water and ethanol extracts from pickled soybean products by manufacturing stage. Methods : Yakkong (YK) was pickled for 15 days to prepare Chokong (CK) and Chokong pills (CKP) were mixed Chokong (CK) powder with vinegar. Herbal Chokong pills (HCKP) were made by addition five kinds of medicinal herbs to Chokong. Pickled soybean products by each manufacturing process were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidative and anticancer activities. Results : The highest electron donating abilities (EDA) of the water and ethanol extracts were from HCKP among pickled soybean products and those were 81.2% and 91.5% at 1,000 $\mu$g/mL, respectively. The ethanol extracts of pickled soybean products had higher activities of EDA and superoxide dismutase (SOD) than water extracts. The highest anticancer activity was the water extracts of HCKP against both of MDA and A549 cells. Conclusions : These results suggest that herbal Chokong pills (HCKP) can be used as natural antioxidant to prevent oxidative damage in normal cells probably because of their antioxidative characteristics.
Objectives : This study was conducted to confirm the anti-inflammatory effects of Aster glehni Water extracts. Methods : In this study, MTT assay was performed to detect cell viability. To evaluate the anti-inflammatory effects of Aster glehni Water extracts, we examined NO production in LPS-induced macrophages. Expressions of iNOS, COX-2, ERK, p38, JNK were also investigated by using western blot assay. Results : Aster glehni Water extracts have no cytotoxicity at 15.625-1,000㎍/㎖ in RAW 264.7 cells. Aster glehni Extracts inhibited the NO production in a dose-dependent manner in RAW 264.7 cells treated with LPS. Pretreated 250, 500, 1,000㎍/㎖ of Aster glehni water extracts had significantly suppressed expression levels of iNOS, COX-2, p-ERK, p-p38, p-JNK. Conclusions : These results suggest that Aster glehni Water extracts have anti-inflammatory effects and can be used for various inflammatory skin diseases.
Objectives : The purpose of this study is to investigate the effects of Chowiseungcheng-tang extracts on the preadipocytes proliferation in 3T3-L1 cell line, lipolysis of adipocytes in rat's epididymal adipocytes and localized fat accumulation of porcine by extraction methods(alcohol and water). Methods : Diminish preadipocytes proliferation and promote lipolysis of adipocytes do primary role to reduce obesity. So, we used 3T3-L1 mouse embryo fibroblasts(preadipocytes) and rat epididymal adipocytes from Sprague-Dawley rats to investigate the effects of Chowiseungcheng-tang extracts on the preadipocytes proliferation, lipolysis of adipocytes. They were treated with 0.01, 0.1, $1.0mg/m{\ell}$ Chowiseungcheng-tang alcohol and water extracts. And for the purpose of investigating the effects of Chowiseungcheng-tang alcohol and water extracts on the localized fat accumulation, we injected 0.1, 1.0, $10.0mg/m{\ell}$ Chowiseungcheng-tang extracts to porcine fat tissues and observed histological changes of them. Results : Following results were obtained from the preadipocytes proliferation and lipolysis of adipocytes and histological investigation of fat tissues. 1. Chowiseungcheng-tang extracts suppressed preadipocytes proliferation on the high dosage(especially $1.0mg/m{\ell}$), and especially alcohol extracts had better effects. 2. The alcohol extracts of Chowiseungcheng-tang decreased the activity of glycerol-3-phosphate dehydrogenase (GPDH) on the concentrations of 0.1, $1.0mg/m{\ell}$. Alcohol extracts had better effects than water extracts. 3. Chowiseungcheng-tang extracts increased lipolysis of adipocytes on the concentrations of 0.1, $1.0mg/m{\ell}$, and especially on the concentration of $1.0mg/m{\ell}$ alcohol extract of Chowiseungcheng-tang had better effect. 4. The water extract of Chowiseungcheng-tang had significant activity to the destruction of porcine fat cell membranes only on the concentration of $10.0mg/m{\ell}$, but alcohol extracts of Chowiseungcheng-tang had it on all concentrations. Conclusions : The alcohol extracts of Chowiseungcheng-tang had much better effects on the preadipoeytes proliferaton, lipolysis of adipocytes and localized fat accumulation than water extracts.
우리 나라에서 쉽게 구할 수 있고, 값이 저렴하여 대중적으로 널리 이용될 수 있는 감잎으로부터의 열탕 추출물과 함께 탄닌을 분리하여 xanthine oxidase 활성을 측정함으로써 감잎의 통풍 예방효과를 살펴보았다. 감잎의 열탕 추출물을 농도별로 살펴본 결과 열탕 추출물의 농도가 증가할수록 저해효과가 증가하여 2.0mg 첨가시 82.4%의 높은 저해효과를 나타냈으며, 감잎탄닌도 감잎의 열탕 추출물과 마찬가지로 농도에 비례해서 santhine oxidase 저해효과가 증가하는 것으로 나타났다. 한편 감잎 열탕 추출물 및 감잎 탄닌은 xanthine oxidase에 대해 경쟁적 저해를 하는 것으로 나타났다.
We investigated the effects of bean sprouts (Glycine max), dropwort (Oenanthe javanica), and radish (Raphanus sativus var. hortensis for. acanthiformis) extracts on alcohol dehydrogenase (ADH). The extracts from three kinds of vegetables were prepared by extracting with boiling water, distilling water, and ethyl alcohol. Among extracts, boiling water extract showed the highest activating effect on ADH, respectively and distilled water extract had a greater effect on ADH activation than that of alcohol extract. The ADH facilitating effect of bean sprout extract by distilled water was significantly higher than dropwort or radish, hut the effect of the bean sprout extract by ethyl alcohol was lower than others. The facilitating effect on ADH of mixture extracts of bean sprout and dropwort were mixed at 1 : 1 mixture of boiled-water extract showed the highest effectiveness. And bean sprout extract separated below 3000 molecular weight (MW) range of extract fraction had greater ADH activity than large MW parts.
The aim of this study was to investigate the quality characteristics of makgeolli that was produced by using different percentages (control, 3%, 5%, 7%, 9% by rice weight) of Angelica gigas Nakai water extracts during fermentation at $25^{\circ}C$ for 7 days. Yeast cell count in makgeolli steadily increased and showed the highest value at day 4 of fermentation, since then, the count decreased consistently. The final alcohol contents were higher than 15% in all samples. Soluble contents and antioxidation activity were significantly increased in percentage as Angelica water extracts increased (p<0.05). As the amount of Angelica water extracts increased, total polyphenol contents increased as well. Soluble contents and total polyphenol contents were increased according to the fermentation period. Total acidity increased up to day 3, but slowly decreased afterwards. However, the pH rapidly decreased by day 3, but then increased steadily. Reducing sugar amount was increased sharply by day 2 of fermentation and then, diminished gradually. Hunter's color value was not significantly different among samples however, they were significantly different between day 2 samples of fermentation and day 3 or more samples (p<0.05). In sensory evaluations, the sample produced using 5% Angelica water extracts received the highest score in herbal flavor, herbal taste, sourness, sweetness, and overall acceptance. Makgeolli using Angelica gigas Nakai water extracts showed excellent characteristics in antioxidative activity and higher total polyphenol content. Makgeolli produced using 5% Angelica water extracts received the highest score in sensory evaluation. Therefore, makgeolli produced using 5% Angelica water extracts was considered to be the most suitable for manufacturing makgeolli.
The antioxidative activities of two varieties of egg plant (Chunyang No 2: Dangaji, Jinju Janggaji: Janggaji) extracts were investigated. The total polyphenol contents of Dangaji peel hot water extract and the Janggaji flesh and fruit hot water extracts were higher than those of the other samples. However, the DPPH radical scavenging activities(electron donating activity) of Dangaji flesh-ethanol, peel-cold water, and fruit- ethanol extracts, as well as the Janggaji peel cold water extract, were higher than those of the other samples. Furthermore, the in vitro inhibitory effects of the Dangaji peel cold water and hot water extracts on rat hepatic xanthine oxidase were highest among the samples, and were exhibited in a dose dependant manners. Although there were marked changes in the xanthine oxidase Km values for xanthine as a substrate, the Vmax value changes by the addition of the Dangaji water extracts were minor compared with the control. This result suggests that Dangaji water extracts may regulate the activity of xanthine oxidase-via the inhibition of binding affinity between the enzyme and substrate. The present study provides experimental evidence that constituents of egg plant extracts may ameliorate reactive oxygen species(ROS)-induced oxidative stress via hepatic hepatic xanthine oxidase activity, but further studies to identify the active antioxidants and compounds and inhibitors of xanthine oxidase are required.
Hamcho (Salicornia herbacea) extracts were evaluated for total polyphenol content, antioxidant and pro-oxidant activities. The total polyphenol content was 1.81 g and 0.72 g per 100 g of dried sample in water and ethanol extracts respectively. Both water and ethanol extracts of Hamcho significantly exhibited antioxidant activity. The scavenging activity of hydroxyl radical was 13.8~26.2% and 14.2~16.0% in water and ethanol extracts respectively. The inhibitory effect of conjugated diene formation was 24.6~39.1% and 28.4~39.6% in water and ethanol extracts respectively. However, pro-oxidative effect was also observed in the Hamcho extracts. The Hamcho water extract showed the pro-oxidant effect by enhancing the formation of hydroxyl radical and conjugated diene. The ethanol extract of Hamcho induced conjugated diene formation at 0.5 mg/mL but not at 1 mg/mL. The hydroxyl radical formation was not induced by the Hamcho ethanol extract. Taken together, these results show that Hamcho extracts can act as pro-oxidants by generating hydroxyl radical or conjugated diene in addition to their antioxidant properties. Therefore, this study suggests that the physiological properties of Hamcho and its use as food materials should be considered with caution because antioxidant dietary plants such as Hamcho possess possible adverse effects induced by pro-oxidant activity.
In this study the effects of water extracts of endocarps and seeds of omija(Schizandra chinensis Baillon)on alcohol metabolism in rats were investigated. 25% alcoholic solution(v/v,0.75g/200g B.W.)and 40% alcoholic solution(v/v,0.80g/200g B.W.)were orally administered to rats for 2 hours. The levels of metabolites and enzyme activities both on serum and liver were not changed by acute oral adminiatration of 25% and 40% alcoholic solution. Blood alcohol levels were significantly lowered by treatment of water extracts of endocarps and seeds of omija in rats administered with 25% and 40% alcohol. In 25% alcohol treated group, serum GPT level was decreased but hepatic G-6-P DH and pyruvate levels were increased. In 40% alcohol treated group, water extracts of endocarps and seeds of omija resulted in a significant decreased in serum levels of FFA, GPT but increased in serum glucose level. Hepatic levels of cytosolic protein, glycogen and pyruvate were increased by water extracts of omija parts in 40% alcohol treated group.
