새로운 천연염료로서 수련 잎 추출색소의 염색성과 기능성(2): 견·모섬유를 중심으로 (The Dyeing Properties and Functionality of Water Lily(Nymphaea tetragona) Leaves Extract as a New Natural Dye Resource(2): Dyeing of Silk and Wool Fibers)

  • This study was to investigate the practicality and functionality of water lily(Nymphaea tetragona) leaves as a natural dye resource while searching for various dyeing methods to utilize them. Effect of dyeing condition including methanol ratio of dyebath, dyeing temperature and time, mordanting method, etc were investigated. Colorants were prepared by extraction in methanol and followed drying process. When composing 30% of methanol in the dyebath, better dyeuptake and uniform dyeing were resulted. Pre-mordanting method gave better results in terms of dye uptake than post-mordanting method on the wool fabrics. Fe and Ti were effective for increasing the dye uptake on the silk fabrics. Depending on mordant type and mordanting method, the dyed fabrics got various color showing green, khaki, brownish yellow, dark brown, dark gray and so on. As for color fastness, the silk fabrics dyed with water lily extract showed relatively high rating in light fastness(3~4, 4~5 rating), washing fastness(4~5, 5 rating), and rubbing fastness(4, 4~5 rating). The silk and wool fabrics dyed with water lily leaves extract showed excellent antimicrobial activity over 98% of bacterial reduction rate against Staphylococcus aureus and Klebsiella pneumoniae. It was confirmed that water lily leaves can be used as a natural dye resource for dyeing wool and silk fabrics because its colorants showed excellent affinity and antimicrobial functionality as well as good colorfastness.

Betula Platyphylla var. Japonica Extract Prevent Ultraviolet C Light-induced Cell Damage in Chinese Hamster Fibroblast (V79-4) Cells

  • The present study reports the protective properties of a total methanol extract of B. platyphylla var. japonica against ultraviolet (UV)-C irradiation. Pretreatment of Chinese hamster fibroblast (V79-4) cells with a total methanol extract significantly increased cell survival following $300\;J/m^2$ of UV-C irradiation. The total methanol extract was further fractionated into 5 fractions: n-hexane, dichloromethane, ethylacetate, n-butanol and water fractions. Among these fractions, B. platyphylla var. japonica ethylacetate, butanol and water fractions showed significant protective effects against the cellular damage induced by UV-C irradiation. In order to elucidate the mechanism underlying this protective effect, DPPH (Editor note: abbreviations should be spelled out at first use.) radical scavenging and lipid peroxidation inhibitory activity were measured. Significant radical scavenging and lipid peroxidation inhibitory activities were observed for the ethylacetate fraction. In summary, the present data demonstrate that an extract of B. platyphylla var. japonica has a significant protective effect against UV-C irradiation. The underlying mechanism of this protective effect may involve radical scavenging and inhibition of lipid peroxidation by the B. platyphylla var. japonica extract.

산초 (Zanthoxylum schnifolium)열매 추출물의 항산화 및 $\alpha$-Glucosidase 저해 활성 (Antioxidant and $\alpha$-glucosidase Inhibition Activity from Different Extracts of Zanthoxylum schnifolium Fruits)

  • This study was performed to the activity of antioxidant and anti-diabetic from Zanthoxylum schnifolium fruit. The Zanthoxylum schnifolium fruits were extracted with water, 100% methanol, 100% ethanol, 70% methanol and 70% ethanol. The activities of each extracts were measured by antioxidant tests, such as total phenolic contents, total flavonoid contents, hydroxyl radical activity, DPPH radical scavenging activity, reducing power and $\alpha$-glucosidase inhibition activity assay. The water extract represented the highest activity in the antioxidants properties in vitro, including hydroxyl radical activity, DPPH radical scavenging activity, reducing power. Furthermore, water extract also showed significantly high total phenolic contents but the 100% methanol extract showed high total flavonoid contents estimated as $196.48{\mu}g/mg$. The $IC_{50}$ values for $\alpha$-glucosidase inhibition activity of the 100% ethanol and 100% methanol extracts were $275.66{\mu}g/mg$ and $261.44{\mu}g/mg$. Our results indicated that the Zanthoxylum schnifolium fruit expected to be useful as antioxidants.

Assessment of Antioxidant and Phenolic Compound Concentrations as well as Xanthine Oxidase and Tyrosinase Inhibitory Properties of Different Extracts of Pleurotus citrinopileatus Fruiting Bodies

  • Cellular damage caused by reactive oxygen species has been implicated in several diseases, thus establishing a significant role for antioxidants in maintaining human health. Acetone, methanol, and hot water extracts of Pleurotus citrinopileatus were evaluated for their antioxidant activities against ${\beta}$-carotene-linoleic acid and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, reducing power, ferrous ion-chelating abilities, and xanthine oxidase inhibitory activities. In addition, the tyrosinase inhibitory effects and phenolic compound contents of the extracts were also analyzed. Methanol and acetone extracts of P. citrinopileatus showed stronger inhibition of ${\beta}$-carotene-linoleic acid compared to the hot water extract. Methanol extract (8 mg/mL) showed a significantly high reducing power of 2.92 compared to the other extracts. The hot water extract was more effective than the acetone and methanole extracts for scavenging DPPH radicals. The strongest chelating effect (92.72%) was obtained with 1.0 mg/mL of acetone extract. High performance liquid chromatography analysis detected eight phenolic compounds, including gallic acid, protocatechuic acid, chlorogenic acid, ferulic acid, naringenin, hesperetin, formononetin, and biochanin-A, in an acetonitrile and hydrochloric acid (5 : 1) solvent extract. Xanthine oxidase and tyrosinase inhibitory activities of the acetone, methanol, and hot water extracts increased with increasing concentration. This study suggests that fruiting bodies of P. citrinopileatus can potentially be used as a readily accessible source of natural antioxidants.

추출용매에 따른 다릅나무 수피 추출물의 항산화 활성 (Stem bark of Maackia amurensis Extract according to extraction Solvent)

  • Objectives : The objective of this research was to investigate the antioxidant activities of stem bark of Maackia amurensis extract.Methods : Stem bark of Maackia amurensis extract were prepared using 70% methanol. Methanol extracts were fractionated to hexane, chloroform, ethyl acetate, butyl alcohol, water fractions and investigated. The antioxidant activities of fractions was evaluated by four different assays as total polyphenol contents, total flavonoid contents, DPPH(1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity and ABTS(2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) scavenging ability.Results : The yield of methanolic extracts from stem bark of Maackia amurensis was 10.16%, whereas those of its solvent fractions (hexane, chloroform, ethyl acetate, butyl alcohol, and water) were 5.45, 11.39, 13.88, 26.07, and 40.80%, respectively. The total polyphenol contents and electron donating ability of 70% methanol extracts from stem bark of Maackia amurensis were 15.44 mg/g and 194.15 μg/mL of its IC50, respectively. The 70% methanol extracts showed the highest antioxidant activity. The total polyphenol content and total flavonoid content of chloroform fractions were higher in each of 201.98 mg/g and 13.55 mg/g. The chloroform fraction showed the lowest levels of DPPH(IC50, 183.95 μg/mL) and ABTS scavenging activity(IC50, 10.0 μg/mL). The antioxidant activity was detected in methanol extract, chloroform fractions.Conclusions : These results indicate that 70% methanol extract and its fractions of stem bark of Maackia amurensis, especially chloroform fraction, have the properties of anti-oxidant suggesting stem bark of Maackia amurensis may be a candidate for natural and functional materials.

Benzo(a)pyrene에 의해 유도된 간기능 장해에 미치는 쑥의 효과 (The Effect of Mugwort Extracts on the Benzo(a)pyrene-induced Hepatotoxicity in Rats)

  • Mugwort has been used as a Korean folk medicine in treating liver diseases acting as an analgesics, sedative, diuresis, choleretics. This study was perfomed to evaluate the effect of mugwort extracts on the changes of enzyme activities, lipid accumulation of the serum and liver, when hepatotoxicity was induced by benzo(a)pyrene. The results are as follows: 1. Mugwort water extract administration prevented the increase of serum and liver AST, ALT, LDH, ${\gamma}$-GTP, liver ALP activities and bilirubin content caused by B(a)P injection. 2. The increase of serum and liver ALT, LDH, ${\gamma}$-GTP, serum AST activities and liver bilirubin contents in B(a)P treated group were decreased by mugwort methanol extract treatment. 3. Serum and liver total cholesterol, phospholipid, triglyceride level and serum HDL-cholesterol level were increased by B(a)P treatment. After combined treatment of mugwort water and methanul extracts, these lipid content were significantly decreased. 4. The hepatotropic effect of mugwort water extract and after-treatment against B(a)P induced hepatotoxicity was superior to that of methanol extract and pretreatment.

보리(Hordeum vulgare) 추출물의 항변이원성 (Studies on the Antimutagenicity of extract from Barley (Hordeum vulgare))

  • This study was performed to determine the effects of antimutagenicity from Barley (Hordeum vulgare). In Salmonella typhimurium reversion assay (In vitro test), the extract of Barley (Hordeum vulgare) inhibited mutagenic activity of 4-NQO and Trp-p-1 with 59 mix. in Salmonella typhimurium TA98 and TA100. In Micronucleus test (In vivo test), the methanol extract of Barley (Hordeum vulgare) inhibited micronucleus formation in bone marrow by cyclophosphamide. The $\beta$-glucan of Barley (Hordeum vulgare) showed inhibitory effects of 59-77% in mutagenic activity of 4-NQO by Salmonella typhimurium TA100. The mutagenicity of Trp-p-1 with S9 mix. by Salmonella typhimurium TA98 showed inhibitory effects of 24-56%. The methanl extract (M) was fractionated with ether (MI), ethylacetate (M2), buthanol (M3) and water (M4). The Antimutagenicity of Trp-p-1 with 59 mix. by Salmonella typhimurium TA98 in Barley fraction showed the following: methanol extract (99.58%)>ether fraction (98.05%)>buthanol fraction (56.90%)>water fraction (56.72%)>ethyl acetate fraction (28.72%). Among them, ether fraction in TA 98 showed strong antimutagenicity effects (85.56%, 98.05%) against mutation induced by 4-NQO and Trp-p-1. As concentration of the methanol extract increased (1.25~5 g/kg/10 cc), micronucleus formation in bone marrow by chemical mutagen (CP) showed inhibitory effects of 50% (p< 0.05).

Antioxidant Activities and Tyrosinase Inhibitory Effects of Different Extracts from Pleurotus ostreatus Fruiting Bodies

  • We evaluated the antioxidant activity and tyrosinase inhibitory effects of Pleurotus ostreatus fruiting bodies extracted with acetone, methanol, and hot water. The antioxidant activities were tested against $\beta$-carotene-linoleic acid, reducing power, 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity, and ferrous chelating ability. Furthermore, phenolic acid and flavonoid contents were also analyzed. The methanol extract showed the strongest $\beta$-carotene-linoleic acid inhibition as compared to the other exracts. The acetone extract (8 mg/mL) showed a significantly high reducing power of 1.54 than the other extracts. The acetone extract was more effective than other extracts for scavenging on 1,1-diphenyl-2-picrylhydrazyl radicals. The strongest chelating effect (85.66%) was obtained from the acetone extract at 1.0 mg/mL. The antioxidant activities of the extracts from the P. ostreatus fruiting bodies increased with increasing concentration. A high performance liquid chromatography analysis detected seven phenolic compounds, including gallic acid, protocatechuic acid, chlorogenic acid, naringenin, hesperetin, formononetin, and biochanin-A in an acetonitrile and 0.1 N hydrochloric acid (5 : 1) solvent extract. The total phenolic compound concentration was $188{\mu}g$/g. Tyrosinase inhibition of the acetone, methanol, and hot water P. ostreatus extracts increased with increasing concentration. The results revealed that the methanol extract had good tyrosinase inhibitory ability, whereas the acetone and hot water extracts showed moderate activity at the concentrations tested. The results suggested that P. ostreatus may have potential as a natural antioxidant.

수비전(壽脾煎) 추출물의 항우울 효과에 관한 연구 (Antidepressant effect of the extracts of Subi-jeon, a Korean medicinal prescription)

  • Objective : The Korean famous medicinal prescription of Subi-jeon was investigated for their antidepressant effects by tail suspension test, hot plate test, reserpine-induced hypothermia test. In addition, the monoamine oxidase activity was determined in vivo. Methods : The methanol extract reduced dose-dependently the duration of immobility in the tail suspension test, by 31.4 and 34%(p<0.05) at doses of 500mg/kg and 1g/kg, respectively, compared with control group. In comparison with this, the effect of the water extract was very weak. Results : 1. In the hot plate test, the methanol extract potently increased the jump latency time(p<0.05) compared to the control group, exhibiting the inhibition rate of 197% and 256% at doses of 500mg/kg and 1g/kg(per os), respectively, which is more effective than the water extract. 2. Both extracts suppressed the fall of body temperature induced by reserpine(reserpine-induced hypothermia) in a dose-dependent manner, showing the less effect at lower doses and better effect at higher doses compared to the water extract. 3. Both extracts inhibited the brain monoamine oxidase activity in an in vivo assay compared to the control group, the activity of water extract was better than that of the methanol extract. Conclusion : The prescription of Subi-jeon can be useful for the prevention and treatment of depression.

소리쟁이(Rumex crispus) 뿌리로부터 가용성 고형분의 추출특성 (Extraction Characteristics of Soluble Solid from Rumex crispus(Curled Dock) Roots)

  • In this study, the extraction characteristics of soluble solid from Rumex crispus(Curled dock) was studied from the investigation of the effects of experimental conditions on extraction rate; extraction ratio, composition of extractants, extraction time and pH of extractant, etc. The proximate composition of Rumex crispus was 2.58% crude lipid, 5.59% crude protein, 7.39% crude ash, 6.13% moisture and 78.31% carbohydrate, respectively. Turbidity of extract by distilled water was higher and increased with extraction time and extraction temperature, where as the turbidity didn't increase by ethanol and methanol in 20 folds of extraction ratio. Turbidity was inversely proportional to the extraction ratio for the three extractants at 25$^{\circ}C$ and 1 hour extraction. But turbidity of extract was highest by composition of 50% methanol-water extractant than any other compositions of extractants. Eighteen and fifteen free aminoacids were detected in extracts with distilled water, methanol and ethanol extractant, respectively, and it's contents were order of glutamic acid>proline>aminobutyric acid>alanine. The extraction rate of soluble solid from Rumex crispus was order of distilled water>methanol>ethanol within experimental extraction ratio. In extraction with distilled water, the contents of soluble solid was inversely proportional to the pH of extractant.
