Antioxidant and Antimicrobial Activity of Rosa multiflora Thunberg Fruits Extracts

  • In this study, we selected some material to have potential bioactivity from natural plants, confirmed as basic data for industrializing and tried to develope the food materials using them. DPPH, ABTS, antioxidant protection factor, TBARs and antimicrobial activity of extracts from Rosa multiflora Thunberg fruits were determined. The total phenolics extracted from Rosa multiflora were 12.08, 11.82, 11.1 and 12.6 mg/g when using water, 70% ethanol, 70% methanol and 70% acetone as the solvent, respectively. The optimum conditions for extracting the phenolic compounds were 70% ethanol over for 12 hrs(11.82 mg/g). The electron donating ability and inhibition rate on ABTS of the 70% ethanol extracts were 97% and 92.2%, respectively while the antioxidant protection factor(PF) of the water extracts and 70% ethanol extracts were 1.79 and 1.34 PF, respectively. The TBAR (thiobarbituric acid reactive substance) value were $1.3{\mu}M$ for the control and $0.15{\mu}M$ for the 70% ethanol extracts. The inhibitory activity against ${\alpha}$-amylase was 26% for the 70% ethanol extracts. The 70% ethanol extracts from Rosa multiflora Thunberg fruits exhibited antimicrobial activity against H. pylori, S. epidermidis, S. aureusand and E. coli with clear zone diameters of 14, 25, 14 and 13 mm, respectively when using $200{\mu}g/mL$ of the phenolic compounds. An HPLC analysis identified 6 major phenolic metabolites in the Rosa multiflora Thunberg fruits extracts: rosmarinic acid, caffeic acid, chlorogenic acid, courmaric acid, protocatechuic acid and quercetin. In particular, the content of rosmarinic acid was the highest in the 70% ethanol extracts. Therefore these results indicate that 70% ethanol extracts from Rosa multiflora Thunberg fruits can be useful as a natural antioxidant and in functional foods.

Anti-Inflammatory Activity of Pinus koraiensis Cone Bark Extracts Prepared by Micro-Wave Assisted Extraction

  • In this study, we compared the anti-inflammatory activity of Pinus koraiensis cone bark extracts prepared by conventional extraction and microwave-assisted extraction (MAE). Water extracts and 50% ethanol extracts prepared using MAE were applied to RAW 264.7 cell at 5, 10, 25, and $50{\mu}g/mL$ of concentrations, and tested for cytoxicity. The group treated with $50{\mu}g/mL$ of 50% ethanol extracts showed toxicity. In order to investigate the inhibition of nitric oxide (NO) production in RAW 264.7 cells, extracts of water and ethanol were treated with 5, 10, and $25{\mu}g/mL$ concentrations. The inhibitory activity of water and 50% ethanol extracts groups were determined as 40% and 60% at $25{\mu}g/mL$ concentration, respectively. We found concentration dependent decreases on inducible NO synthase. The inhibitory effect against forming inflammatory cytokines, prostaglandin $E_2$, tumor necrosis factor-${\alpha}$, interleukin (IL)-6, and IL-$1{\beta}$, was also superior in the $25{\mu}g/mL$ treated group than the control group. According to these results, the water extracts and 50% ethanol extracts both inhibited inflammatory mediators by reducing the inflammatory response. Therefore, The MAE extracts of P. koraiensis cone bark can be developed as a functional ingredient with anti-inflammatory activity.

Screening Biological Activities of Grape Seed and Skin Extracts of Campbell Early (Vitis labruscana B.)

  • This study was conducted to determine biological activities, such as lipid peroxidation inhibition, cytotoxicity, sun blocker, inhibition of tyrosinase, and antioxidative effect, of ethanol extracts, and of solvent fractionated ethanol extracts obtained from grape seeds and skins. The strongest lipid oxidative inhibition of 66.9% and 67.6% was observed respectively, in the presence of 20 $\mu\textrm{g}$/$m\ell$ of both ethanol extract and water fraction of grape seeds. Overall, the ethanol extracts and their fractions of grape seeds exhibited stronger lipid oxidative inhibition than that of skin extracts. On the other hand, the ethanol extracts of grape skins showed stronger cytotoxicity than that of seeds on MCF-7, Hep3B, and A549 cancer cell lines. However, the water fraction of seed ethanol extracts showed the strongest cytotoxic effect of 76.52% and 67.01% on MCF-7 and Hep3B, respectively among their fractions. Ethanol seed extracts obtained at 3$0^{\circ}C$ had the strongest absorbance both at UVA region (350 nm) and UVB region (308 nm) and the chloroform fraction showed the strongest absorbance at W region and butanol fraction at UVA region among their tractions, respectively. In the meantime, the ethanol extracts obtained at 3$0^{\circ}C$ and butanol fraction showed the strongest tyrosinase inhibitory effect of 39.4% and 37.6%, respectively. This study shows that ethanol extracts and their fractions of grape seeds and skins could be potential good materials for functional food and cosmetic products.

정향 열수 및 에탄올 추출물의 항산화 및 항염 효과 (Antioxidant and Anti-Inflammatory Activities of Extracts from Eugenia caryophyllata Thunb.)

  • The objective of this study was to investigate the antioxidant and anti-inflammatory activities of Eugenia caryophyllata Thunb. water and 70% ethanol extracts. The content of total polyphenol was significantly higher in water extract than in 70% ethanol extract. The DPPH radical scavenging activity of water extract was similar to that of Vit. C at a concentration of $1,000{\mu}g/mL$. The ABTS radical scavenging activities of water and 70% ethanol extract were similar to that of Vit. C at a concentration of $1,000{\mu}g/mL$. SOD-like activity of water extract was higher than that of 70% ethanol extract at a concentration of $1,000{\mu}g/mL$ but lower than that of Vit. C. The DPPH radical scavenging activity, ABTS radical scavenging activity, and SOD-like activity increased as concentrations of water and 70% ethanol extracts increased. Cell cytotoxicity was not observed at all concentrations except at $100{\mu}g/mL$ concentration of water extract. Inhibitory activity on NO production effect of water extract was significantly higher than that of 70% ethanol extract. These results show that E. caryophyllata Thunb. has potent biological activities, and their activities were different depending on extraction solvent.

강황(Curcuma longa L.)으로부터 초임계 유체 추출한 curcumin의 생리활성 (Physiological Effects of Curcumin Extracted by Supercritical Fluid from Turmeric (Curcuma longa L.))

  • Curcumin의 추출효율은 에탄을 추출방법이 가장 높았고, 항산화효과는 에탄올, 초임계, 열수 추출물 순으로 나타났다. Ames의 mutagenicity test를 실시한 결과 각각의 추출물은 돌연변이원이 없는 것으로 확인되었고, TA98에 직접변이원 2-NF 처리시 초임계 추출물의 저해율이 20.1%로 에탄올과 열수 추출물에 비하여 가장 높은 항돌연변이 효과를 나타냈다. 또한 간접 변이원 2-acetamidofluorene($10\;{\mu}L/plate$; Sigma Co.; 2-AF)에 대한 항돌연변이 효과를 알아본 결과 2-AF로 유도된 TA98은 초임계 추출물을 0.1 mL/plate로 처리시 항돌연변이의 효과는 12.2%였으며, 알코올 추출물과 열수 추출물의 경우는 항돌연변이 효과가 나타나지 않았다. Nitrite의 분해능의 경우에 있어서도 에탄올 및 열수 추출물에 비하여 초임계 추출물이 높은 분해율을 나타냈으며 pH 1.2에서 가장 높은 분해율을 보였다. 이와 같이 초임계를 이용해 추출한 curcumin은 항산화효과, nitrite 분해능, 항돌연변이효과 등에 대한 생리활성 기능을 보였으며, 기존의 에탄올이나 열수 추출법에 비하여 유사하거나 더 높은 생리활성 효과가 나타나기도 했다.

쑥 추출물의 사람 Low Density Lipoprotein에 대한 항산화능 (Antioxidative Activity of Mugwort extracts(Artemisia Princeps Var. Orientalis) on Human Low Density Lipoprotein)

  • The antioxidative effect of mugwort extracts was measured by DPPH and LDL with four different solutions (70% acetone, ethanol, hot water, cold water). Mugwort extracts contained 3.2% of polyphenol, 380 RE/l00 g of vitamin A, 16.2 mg/100 g of vitamin C, and 5.1 ${\alpha}$-TE/100 g of vitamin E. DPPH revealed the effect in the order of 70% acetone, hot water, ethanol, and cold water. In particular, 70% acetone showed outstandingly stronger activity than the control group. Also, when 10 ${\mu}\ell$ was added, the effect was well noticed. But the antioxidative activity was hardly seen at 15 ${\mu}\ell$. LDL exhibited the same order of strength in proportion to mugwort's concentration. Against the control group, the activity of 70% acetone was 7 times, hot water and ethanol 6 times, and cold water 2 times. This result is attributable to the antioxidative increase of polyphenol and antioxidative vitamins.

Antioxidant Activities of Various Solvent Extracts from Ginseng (Panax ginseng C.A. Meyer) Leaves

  • Water, methanol and ethanol extracts of ginseng leaves were assayed for total phenolics and flavonoids, ascorbic acid, cupric and ferrous ion chelating activities, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, ferric reducing antioxidant power (FRAP) assay and ABTS radical cation decolourization (TEAC) assay for their antioxidant properties. The ethanol extract of ginseng leaves contained significantly (p<0.05) higher amounts of total phenolics and flavonoids (600.57 and 1701 mg/100 g) than methanol (374.43 and 1512.64 mg/100 g) and water extracts (248.30 and 680.05 mg/100 g). Among solvent extracts of ginseng leaves, the ethanol extract showed the most powerful antioxidant activities. However, the ferrous ion chelating activity of ginseng leaf extracts were lower than the cupric ion chelating ability. These differences in concentrations of key antioxidants among various solvent extracts seemed to be responsible for their differences in antioxidant activities. These results suggest that ethanol extract of ginseng leaves has the most effective antioxidant capacity compared to the methanol and water extracts tested in the present study. Thus, it can be applied for the effective extraction of functional material from ginseng leaves for the usage of pharmaceutical and/or food industries.

Yam Extracts Increase Cell Proliferation and Bone Matrix Protein Collagen Synthesis of Murine Osteoblastic MC3T3-E1 Cells

  • Yam extracts (Dioscorea batatas) have been reported to possess a variety of functions. However, studies on its osteogenic properties are limited. In this study, we investigated the effect of ethanol and water extracts on osteoblast proliferation and bone matrix protein synthesis, type I collagen and alkaline phosphatase (ALP), using osteoblastic MC3T3-E1 cell model. MC3T3-E1 cells were cultured with yam ethanol and water extracts (0~30 mg/L) within 39 days of osteoblast differentiation period. Cell proliferation was measured by MTT assay. Bone matrix proteins were assessed by the accumulation of type I collagen and ALP activity by staining the cell layers for matrix staining. Also, the secreted (media) matrix protein concentration (type I collagen) and enzyme activity (ALP) were measured colorimetrically. Yam ethanol and water extracts stimulated cell proliferation within the range of 15~30 mg/L at 15 day treatment. The accumulation of type I collagen in the extracellular matrix, as well as secreted collagen in the media, increased with increasing doses of yam ethanol (3~15 mg/L) and water (3~30 mg/L) extracts. ALP activity was not affected by yam ethanol extracts. Our results demonstrated that yam extracts stimulated osteoblast proliferation and enhanced the accumulation of the collagenous bone matrix protein type I collagen in the extracellular matrix. These results suggest that yam extracts may be a potential activator for bone formation by increasing osteoblast proliferation and increasing bone matrix protein type I collagen. Before confirming the osteogenic action of yam, further studies for clarifying how and whereby yam extracts can stimulate this ostegenesis action are required.

복분자(Rubus coreanum F.) 열수 및 에탄올추출물의 생리활성비교 (Comparison of Physiological Activities between Hot-Water and Ethanol Extracts of Bokbunja (Rubus coreanum F.))

  • 복분자의 열수추출물과 에탄올 농도별 추출물의 항산화 및 생리 활성효과를 비교하였다. 각 추출물의 총 페놀 함량은 열수추출물에서 41.4 mg/g으로 가장 높았으며, 에탄올 추출물은 $60\%$에서 41.3 mg/g으로 높았다. 복분자 추출물의 항산화 효과는 ABTS가 $60\%$ 에탄을 추출물에서 99.8$\%$, 열수추출물에서 $99.3\%$로 나타났으며, PF는 1.3, 1.2, DPPH가 $93.3\%$, $91.1\%$, TBARS가$ 0.03(\times\;100\mu M),\;0.04(\times\;100\mu M)$로 나타났다 복분자 추출물의 생리활성효과는 XOase 활성억제 효과와 pancreatin $\alpha$-amylase 활성억제효과가 열수추출물이 $60\%$ 에탄을 추출물보다 높았으며, ACE 활성억제효과는 열수추출물과과 $60\%$ 에탄올 추출물이 같은 활성억제효과를 나타내었다. 복분자 추출물로부터 생리활성효과에 영향을 미치는 페놀물질을 알아보기 위해 4가지 페놀화합물인 protocatecuic acid, caffeic acid, courmaric acid, rosemarinic acid를 선정하여 HPLC로 분석해 본 결과 $60\%$ 에탄을 추출물과 열수추출물 모두 protocatecuic acid가 12.5 mg/g, 10 mg/g으로 4가지 페놀화합물 가운데 가장 많이 검출되었다. 이상의 결과로 복분자 추출물의 생리활성효과는 천연항산화제 및 기능성 식품으로 이용 가능할 것으로 판단되었다

새삼, 실새삼 및 갯실새삼 추출물이 Clone M-3 세포주의 Melanin 생합성 및 Tyrosinase 활성에 미치는 영향과 세포독성 및 항산화효과 (Inhibitory Effects on Melanin Biosynthesis and Tyrosinase Activity; Cytotoxicity in Clone M-3 and Antioxidant Activity by Cuscuta japonica, C. australis, and C, chinensis Extracts)

  • Water extracts, ethanol extracts, and juice of Cusuta japonica, C, australis, and C. chinensis were prepared, and their cytotoxicity, antioxidant activity and inhibitory effects on tyrosinase activitiy and melanin biosynthesis were estimated by using melanoma Clone M-3. From this study; the following conclusions were attained. Extracts of Cuscuta japonica, C. australis, and C. chinensis showed noticeable cytotoxicity except ethanol extracts from the stem of C. australis. A maximual cytotoxicity was observed with tile ethanol extract from the seed of C, australis (87.39%). While the ethanol extract from the seed of C. japonica (91.88%) showed the most pronounced inhibitory effect on melanin biosynthesis, the water extract from the stem of C. japonica (1.05%) possessed very little inhibitory effect. The most inhibitory effect on tyrosinase activity was observed with the water extract from the stem of C. australis (76.67%). Howeverr the water extract from the stem of C. japonica showed a very poor effect on the inhibition of tyrosinase activity All the preparations, except extracts from the seed of C. australis were able to remove reactive oxygen species (ROS) in a dose-dependent manner. The juice of C. japonica demonstrated the strongest activity (59.02%).
