근적외분광분석법을 이용한 감귤잎의 수분 측정 (Determination of the water content in citrus leaves by portable near infrared (NIR) system)

  • 감귤의 생육단계별 토양 수분의 양은 감귤의 비대와 당도에 영향을 미치며, 토양 건조 처리를 통해 수분스트레스를 부여하면 과실의 당도를 높일 수 있다. 감귤잎 수분 함유율의 측정은 감귤 성장 시기에 따른 관수 시점, 관수량 조절의 지표가 될 수 있다. 본 연구는 근적외분광분석법(NIRS)을 이용하여 비파괴적으로 감귤잎의 수분을 측정하기 위하여 수행되었다. 온주밀감 (Citrus unshiu Marc.)의 잎 ('Okitsu' Satusuma mandarin leaves)은 건조감량시험법에 따라 건조시켜 수분 함유율 20.80-69.98% 범위로 하여 사용하였다. 감귤잎의 흡수 스펙트럼은 광섬유 반사 프로브를 이용하여 측정하였고, 1450 nm에서 수분 함유율 변화에 따른 OH band의 변화를 관찰할 수 있었다. 감귤잎 수분 측정을 위한 모델은 1100-1700 nm 파장 범위의 스펙트럼을 사용하여, 부분최소제곱법 (PLSR, Partial least squres regression)을 실시하여 개발되었다. 그 결과 SEP (Standard errors of prediction)는 0.97%였다. 개발된 모델을 검증하기 위하여 다른 감귤잎에 적용시킨 결과, 건조감량 측정 결과에 대하여 SEP 0.81%로 좋은 상관성을 보여주고 있다. 본 연구를 통해서 신속하고 비파괴적인 감귤잎의 수분 함유율 측정이 근적외분광분석법을 이용하여 성공적으로 수행되었다.

LC를 이용한 물과 토양 중 Cyhalofop-butyl과 대사물질의 분석 (Determination of Cyhalofop-butyl and its Metabolite in Water and Soil by Liquid Chromatography)

  • 실험실 내 조건에서, 물과 토양 중 cyhalofop-butyl과 그 대사물질인 cyhalofop acid를 위한 잔류분석법이 고감도에서도 간단하고 매우 효과적으로 개발되었다. 물과 토양 중 cyhalofop-butyl과 cyhalofop acid를 분석하기 위하여 액액분별추출과 silica gel chromatographic 정제를 수행하였으며 HPLC-UV를 이용하여 정성/정량하였다. cyhalofop-butyl의 회수율은 2 가지 농도에서 3 반복 수행하여 각각 82.5-100.0%와 66.7-97.9%이었고, 검출한계와 최소검출량은 두 시료에서 모두 0.02 ppm과 10 ng이었다. Cyhalofop acid의 회수율은 물과 토양에서 각각 80.7-104.8%와 76.9-98.1%이 었으며, 검출한계는 각각 0.005 ppm과 0.01 ppm이었고 최소검출량은 두 시료에서 모두 2 ng이었다. Cyhalofop-butyl의 반감기는 물과 토양에서 각각 4.14와 6.6 day였다. 개발되어진 본 시험법은 cyhalofop-butyl의 30% 유탁제를 처리한 물과 토양에서 그 잔류량을 분석하기 위하여 성공적으로 적용되었다.

DOF 공정에 의한 정수처리 효과 (Effect of Drinking Water Treatment by DOF(Dissolved Ozone Flotation) System)

  • 본 연구에서는 용존오존부상법(Dissolved Ozone Flotation; DOF)을 이용한 정수처리 효과를 파악하기 위하여 적정 오존 주입 농도와 오존 주입 농도별 처리 특성에 대해서 연구하였으며, 또한 용존공기부상법(Dissolved Air Flotation; DAF) 및 중력식 침전법(Conventional Gravity Sedimentation; CGS)과 DOF의 처리 성능을 비교하였다. 최적 오존 주입 농도를 산정하기 위한 실험에서 최적 오존 주입농도는 2.7 mg/L로 나타났으며, 오존 주입 농도가 증가할수록 탁도 및 KMnO$_4$ 소모량, UV$_{254}$ 흡수도, TOC 등과 같은 제거효율이 낮아지는 것으로 나타났다. DOF와 DAF, CGS의 처리성능을 비교한 결과 오존 주입 농도 2.7 mg/L로 운전한 DOF시스템의 탁도 제거효율은 88.9%, 유기물질 항목의 경우 KMnO$_4$ 소모량은 62.9%, TOC 47%, 그리고 UV$_{254}$ 흡수도 77.3% 그리고 THMFP 제거율의 경우 51.6%로 다른 두 공정보다 높거나 비슷하게 나타났다. 따라서 응집 부상 공정과 오존 산화 공정이 하나의 반응기에서 일어나는 DOF 시스템은 정수처리시설의 소유부지 감소와 기존 CGS 시스템에서 처리가 어려운 조류 및 용존성 유기물질의 제거를 통해 소독부산물의 생성을 감소시킴으로 향후 고도정수처리시스템으로 적용 가능성이 충분한 것으로 판단된다.

Antiobesity effects of the water-soluble fraction of the ethanol extract of Smilax china L. leaf in 3T3-L1 adipocytes

  • BACKGROUND/OBJECTIVES: Several medicinal properties of Smilax china L. have been studied including antioxidant, anti-inflammatory, and anti-cancer effects. However, the antiobesity activity and mechanism by which the water-soluble fraction of this plant mediates its effects are not clear. In the present study, we investigated the lipolytic actions of the water-soluble fraction of Smilax china L. leaf ethanol extract (wsSCLE) in 3T3-L1 adipocytes. MATERIALS/METHODS: The wsSCLE was identified by measuring the total polyphenol and flavonoid content. The wsSCLE was evaluated for its effects on cell viability, lipid accumulation, glycerol, and cyclic adenosine monophosphate (cAMP) contents. In addition, western blot analysis was used to evaluate the effects on protein kinase A (PKA), PKA substrates (PKAs), and hormone-sensitive lipase (HSL). For the lipid accumulation assay, 3T3-L1 adipocytes were treated with different doses of wsSCLE for 9 days starting 2 days post-confluence. In other cell experiments, mature 3T3-L1 adipocytes were treated for 24 h with wsSCLE. RESULTS: Results showed that treatment with wsSCLE at 0.05, 0.1, and 0.25 mg/mL had no effect on cell morphology and viability. Without evidence of toxicity, wsSCLE treatment decreased lipid accumulation compared with the untreated adipocyte controls as shown by the lower absorbance of Oil Red O stain. The wsSCLE significantly induced glycerol release and cAMP production in mature 3T3-L1 cells. Furthermore, protein levels of phosphorylated PKA, PKAs, and HSL significantly increased following wsSCLE treatment. CONCLUSION: These results demonstrate that the potential antiobesity activity of wsSCLE is at least in part due to the stimulation of cAMP-PKA-HSL signaling. In addition, the wsSCLE-stimulated lipolysis induced by the signaling is mediated via activation of the ${\beta}$-adrenergic receptor.

정수공정별 천연유기물질의 특성 변화 (Variation of Natural Organic Matter Characteristics through Water Treatment Processes)

  • 천연유기물질은 지표수와 지하수내 탁도를 유발하고 humic (humic acid와 fulvic acid)과 nonhumic 성분으로 구성된다. 일반적으로 NOM은 소독부산물(DBP) 전구물질로써 수처리시 관심의 대상이 된다. 원수의 특성은 정수공정의 선택과 적용에 중요한 정보를 제공한다. 수원에 따른 NOM의 상당한 차이가 다양한 원수들 사이에서 관찰되었고, 문제시되는 NOM을 제거하고 전환시키기 위한 목적으로 정수공정을 적용한다. 본 연구의 결과, 산화처리에 의해 전체 DOC 농도의 변화는 거의 없었지만, 고분자의 소수성 물질이 저분자의 친수성 물질로 전환되었고, 염소보다 오존의 경우 그 산화력의 차이 때문에 SUVA 값의 감소가 더 크게 나타났다. 응집처리에 의해 hydrophilic 성분보다 분자량이 더 큰 hydrophobic 성분이 응집 침전되어 훨씬 잘 제거되었다. THMFP는 50% 정도까지 제거되었는데, 이것은 상대적으로 THMFP를 잘 유발시킨다는 humic acid 성분의 제거가 원활하게 일어났기 때문이다.

PVdF 나노섬유 나권형 정밀여과와 입상 활성탄의 혼성 수처리 공정에서 pH의 영향 (Effect of pH in Hybrid Water Treatment Process of PVdF Nanofibers Spiral Wound Microfiltration and Granular Activated Carbon)

  • 강도가 강하고 내약품성, 무독성, 내연소성의 장점을 가지고 있는 PVdF (polyvinylidene fluoride) 나노섬유로 기공이 $0.4{\mu}m$ 평막을 제조한 후, 그 평막으로 부직포를 첨가하여 나권형 모듈을 제작하였다. 카올린과 휴믹산으로 조제한 모사용액과 순수를 대상으로 나권형 모듈의 투과선속과 처리율을 비교하여 pH의 영향을 알아보았고, 여과실험 후 물 역세척을 하여 회복률과 여과저항을 계산하였다. 또한, 나권형 모듈을 통과한 처리수를 입상 활성탄(GAC, granular activated carbon)으로 채워진 컬럼에 통과시킨 후, 탁도와 $UV_{254}$ 흡광도를 측정하여 GAC의 흡착 효과를 고찰하였다.

침출 조건에 따른 보리잎차와 녹차의 항산화능 비교 (A Comparison of the Antioxidant Activity of Barley Leaf Tea and Green Tea according to Leaching Conditions in Distilled Water)

  • This study was conducted to investigate the antioxidant amounts and properties of barley leaf tea and green tea at under various leaching conditions. The leaching temperatures and times of the distilled water were 50, 70 and $90^{\circ}$C for 1, 3 and 5 min, respectively. The levels of crude fat and ash in the barley leaf tea were higher than those in green tea. Brown color intensity and flavonoid absorbance increased with leaching temperature. The highest vitamin C levels and water-soluble phenol resulted at $90^{\circ}$C for 3 min. The DPPH radical scavenging activities of the barley leaf tea and of green tea were 11.06 and 50.56%, respectively, compared to 2.9% for L-ascorbic acid (150 ppm). The nitrite scavenging activities of barley leaf tea and green tea were 95.11 and 74.88%, respectively. The SOD-like activities of barley leaf tea, green tea, and L-ascorbic acid (150 ppm) were 12.99, 8.33, and 12.75%, respectively. The antioxidant effect of green tea was higher than that of barley leaf tea and lastly, the SOD-like activity of barley leaf tea was as high as that of green tea.

Investigation of Filamentous Fungi Producing Safe, Functional Water-Soluble Pigments

  • The production of water-soluble pigments by fungal strains indigenous to South Korea was investigated to find those that are highly productive in submerged culture. Among 113 candidates, 34 strains that colored the inoculated potato dextrose agar medium were selected. They were cultured in potato dextrose broth and extracted with ethanol. The productivity, functionality (radical-scavenging activities), and color information (CIELAB values) of the pigment extracts were measured. Five species produced intense yellowish pigments, and two produced intense reddish pigments that ranked the highest in terms of absorbance units produced per day. The pigment extracts of Penicillium miczynskii, Sanghuangporus baumii, Trichoderma sp. 1, and Trichoderma afroharzianum exhibited high radical-scavenging activity. However, the S. baumii extract showed moderate toxicity in the acute toxicity test, which limits the industrial application of this pigment. In conclusion, P. miczynskii KUC1721, Trichoderma sp. 1 KUC1716, and T. afroharzianum KUC21213 were the best fungal candidates to be industrial producers of safe, functional water-soluble pigments.

Anti-Oxidative and Anti-Inflammation Activities of Pork Extracts

  • This study was conducted to evaluate the antioxidative and anti-inflammatory effects of boiled pork powder (BPP) and hot water extract powder (HWEP) from 4 cuts of meat from Landrace × Yorkshire × Duroc (LYD). The highest DPPH radical scavenging activities determined were from BPP of Boston butt (13.65 M TE) and HWEP of loin (19.40 M TE) and ham (21.45 M TE). The 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activities of BPP from shoulder ham (39.28 M TE) and ham (39.43 M TE) were higher than those of other meat cuts, while HWEP of ham exhibited the highest ABTS radical scavenging activity. A higher oxygen radical absorbance capacity was determined for BPP from ham (198.35 M TE) and in HWEP from loin (204.07 M TE), Boston butt (192.85 M TE), and ham (201.36 M TE). Carnosine content of BPP and HWEP from loin and were determined to be 106.68 and 117.77 mg/g on a dry basis, respectively. The anserine content of BPP (5.26 mg/g, dry basis) and HWEP (6.79 mg/g, dry basis) of shoulder ham exhibited the highest value as compared to the extracts from the other meat cuts. The viability of RAW 264.7 cells was increased with increasing HWEP from loin and ham treatment. In addition, the expression of IL-6 and TNF-α was significantly reduced by HWEP from loin and ham, in a dose dependent manner. These results suggested that boiled pork and hot water extract of pork have antioxidative and cytokine inhibitory effects.

Enhanced Hydrogen Production from Methanol/Water Photo-Splitting in TiO2 Including Pd Component

  • The future use of hydrogen as an energy source is expected to increase on account of its environmentally friendliness. In order to enhance the production of hydrogen, Pd ions (0.01, 0.05, 0.1, and 0.5 mol%) were incorporated $TiO_2$ (Pd-$TiO_2$) and used as a photocatalyst. The UV-visible absorbance decreased with increasing level of palladium incorporation without a wavelength shift. Although all the absorption plots showed excitation characteristics, there was an asymmetric tail observed towards a higher wavelength caused by scattering. However, the intensity of the photoluminescence (PL) curves of Pd-$TiO_2$ was smaller, with the smallest case being observed at 0.1 and 0.5 mol% Pd-$TiO_2$, which was attributedto recombination between the excited electrons and holes. Based on these optical characteristics, the evolution of $H_2$ from methanol/water (1:1) photo-splitting over Pd-$TiO_2$ in the liquid system was enhanced, compared with that over pure $TiO_2$. In particular, 2.4 mL of $H_2$ gas was produced after 8 h when 0.5 g of a 1.0 mol% Pd-$TiO_2$ catalyst was used. $H_2$ was stably evolved even after 28 h without catalytic deactivation, and the amount of $H_2$ produced reached 14.5 mL after 28 h. This is in contrast to the case of the Pd 0.1 mol% impregnated $TiO_2$ of $H_2$ evolution of 17.5 mL due to the more decreasedelectron-hole recombination.
