Induction of Apoptotic Cell Death in Human Jurkat T Cells by a Chlorophyll Derivative (Cp-D) Isolated from Actinidia arguta Planchon

  • The chloroform and methanol (2;1, v/v) extract from an edible plant, Actinidia arguta Planchon, appeared to possess antitumor activity against human leukemias Jurkat T and U937 cells through inducing apoptosis. The substance in the solvent extract was purified by silica gel column chromatography, preparative TLC, and Sephadex LH-20 column chromatography. Characteristics of the substance analyzed by UV scanning analysis, $^1H$ and $^{13}C$ NMR spectra suggested that the substance belongs to the chlorophyll derivatives-like group. The $IC_{50}$ value of the chlorophyll derivative (Cp-D) determined by MTT assay was $15\mu\textrm{g}/ml$ for Jurkat, $10\mu\textrm{g}/ml$ for U937, and $11.4\mu\textrm{g}/ml$ for HL-60m and was more toxic to these leukemias than to solid tumors or normal fibroblast. In order to elucidate cellular mechanisms underlying the cytotoxicity, the effect of the Cp-D on Jurkat T cells was investigated. When cells were treated with the Cp-D at a concentration of $15\mu\textrm{g}/ml$, [3H]thymidine incorporation declined rapidly and wa undetectable in 1h. However, no significant changes were made in the cell cycle distribution of the cells by 24h. The sub-Gl peak representing apoptotic cells began to be detectable in 36h, at which time apoptotic DNA fragmentation was also detected on agarose gel electrophoresis, demonstrating that the cytotoxic effect of the Cp-D is attributable to the induced apoptosis. Under the same conditions, although the protein level of cyclin-dependent kinases such as cdc4, csk6, cdk2, and cdc2 was not significantly changed until 24h, the kinase activity of all c안 rapidly declined and reached a minimum level within 1-6h and then recovered to the initial level by 12h and sustained until 24h. These results suggest that inactivation of cdks at an inappropriate time during the cell cycle progression in jurkat T cells following a treatment with the Cp-D leads to induction of apoptotic cell death.

Behavioral Characteristics on Season and Group Size of Hanwoo Steer (한우 거세우의 계절 및 군 두수별 행동 특성)

  • This study was carried out to determine the behavioral characteristics on season and group size of Hanwoo steer. Forty eight Hanwoo steers were video-recorded at 15, 17 and 19 months of age for behavioral measurement. Space allowance was kept consistence with either group. The results showed that eating behavior of Hanwoo steers was strongly influenced (p<0.05) during seasonal transition, being the lowest in summer and the highest in autumn. With seasonal transition, standing time would linearly increase (p<0.05). In summer, steers spent more (p<0.05) time tying down but relatively less (p<0.05) time walking and fighting, meanwhile they much less (p<0.05) scratching and rubbing. In autumn, steers showed more grooming behavior (p<0.05) contrasted to the other seasons. Steers spent more (p<0.05) walking time when raised in large group and increased (p<0.05) grooming frequency when raised in small group.

Sequence Analysis and Expression of the VP7 Gene of G1 Rotavirus Isolated from an Infant in Korean (한국인 영아에서 분리된 G1 로타바이러스의 VP7 단백 유전자 염기서열 및 발현)

  • To determine the sequence and expression of the VP7 gene of Korean isolates (CAU-9), viral RNA was purified and used for cDNA amplification by RT-PCR. The VP7 cDNA was cloned, sequenced, and expressed using baculovirus expression system. The result showed that the sequence homologies CAU-9 compared with foreign isolated strains Wa, 417, TMC-II, 95B and SA11 were ranged from 74.0% to 95.1 % of nucleotide sequence and 35% to 43% of amino acid sequence, respectively. High homology of CAU-9 was observed in Japanease isolates 417 (nucleotide sequence homology was 95.1% and amino acid sequence homology was 43%). To express VP7 gene, the VP7 cDNA was cloned into pCR-Bac vector and inserted into the genome of baculovirus adjacent to the polyhedrin promoter by cotransfection of Spodoptera frugiperda (Sf9) insect cells with wild type baculovirus DNA. In antigenic analysis of Sf9 cells inoculated with the recombinant VP7, immunofluorescence assay revealed positive for viral antigens. In metabolic labeling of Sf9 cell lysates infected with recombinant baculoviruses, it was revealed that the protein of 34 kDa was expressed. The limited study of expressed VP7 protein inoculated with guinea pigs failed to elicit neutalizing antibody. As a results, the sequence analysis and expression of VP7 protein of rotavirus CAU-9 isolated from an infant in Korea could permit the conformation and development of virus like particles which may be useful in designing vaccine strategy.

Records on Ginseng and Medical Book during the Goryeo Dynasty (고려 시대 인삼과 의약서에 대한 기록)

  • Korean ginseng, a special product of Korea, has been one of the most important exports since the era of the Three Kingdoms. However, not many records were kept about ginseng in Korea until the Goryeo Dynasty. This paper summarizes the records relating to international diplomatic relations and trade of ginseng in the Goryeo Dynasty and the medicinal books known to have been published at the time. During the Goryeo Dynasty, ginseng was actively transported to the neighboring countries of Bohai, Song, Wa, Later Jin, and Yuan as a diplomatic gift or as a trade item. Ginseng was mainly exported from Goryeo to these countries, but it was also received as a diplomatic gift from Bohai and Khitan. Arabian merchants came to Byeokran Port, a representative international trading port of Goryeo, and traded ginseng. After the Mongol invasion, the demand for ginseng in the Yuan Dynasty was excessive, which became a big social problem. During the Goryeo Dynasty, several medicinal books were published, including Jejungiphyobang, Eouuichwalyobang, Hyangyakgobang, Samhwajahyangyakbang, Hyangyak Hyemin Gyeongheombang, Hyangyak Gugeupbang, and Biyebaekyobang. Hyangyak Gugeupbang, which was reprinted during the Joseon Dynasty, has been handed down to the present time, although this has not been the case for the originals of the remaining books. Recently, some of the latter books have been restored through the study of the references in various medicinal books of later generations. While the medicinal books used in the royal court showed that a high proportion of the prescriptions containing ginseng, not a single prescription for ginseng has been found in theHyangyak Gugeupbang, which was mainly used for commoners. This is thought to be because ginseng was very rare and expensive at the time, so it was difficult for commoners to access it.

  • Through specific cases, this article examines the causes and measures to overcome cultural divergences most frequently encountered by Koreans living in Vietnam and Korean-contacting Vietnamese in terms of mode of communication, mode of living and mode of doing business. Historical reality proves that Vietnam and Korea have a lot in common in history, culture and society, due to the influence from the Chinese culture on both countries. However, each country has its own acculturation, hence, there exist, apart from the normal dissimilarities, differences between the two countries especially in natural environment andpolitical systems. As a result, in Vietnam, Koreans and Vietnamese have met with quite a lot of divergences in communication, business and living activities. In order to overcome these cultural divergences, in addition to the ceaseless efforts made by Koreans in Vietnam and Korean-contacting Vietnamese, there should be concern, assistance as well as cooperation between the people and the authorities of the two countries. We should draw lessons from the groundless or trivial affairs which, springing from the Chinese people's dislike for Koreans, have been blown up and disseminated on the internet; where by we are to reconsider the relationship between Koreans and Vietnamese. What is more, Korea still feels historically ashamed to have sent nearly 350,000 soldiers to the wa rin Vietnam for economic purposes in the past. As our predecessors brought about that historic shame to get bread for our fatherland in exchange, our generation today should approach Vietnam out of a sense of responsibility towards history. Having experienced a period of economic difficulties, Vietnam innovated and started its open-door policy in 1987. However, this event should not be regarded as the reason for us to approach Vietnam with only one rigid economic principle.

Hypoglycemic and Hypolipidemic Effects of Orostachys japonicus with Medicinal Herbs in Streptozotocin-Induced Diabetic Rats (생약재와 와송 추출물의 복합조성물이 Streptozotocin 유발 당뇨성 흰쥐의 혈당강하 및 혈중 지질성분에 미치는 영향)

  • The hypoglycemic and hypolipidemic effects from Wa-song (Orostachys japonicus) extracts with mixtures of medicinal herbs (such as Baekbokyung, Changchul and Sa-in) were evaluated in the streptozotocin (STZ)-induced diabetic rats. Thirty-five Sprague-Dawley rats were assigned to five groups; normal, diabetic control (D-control), a group fed a medicinal herbs mixture (D-MH), and a group fed composites of O. japonicus with mixtures of medicinal herbs (1:1, D-MHOA and 3:1, D-MHOB). All were fed on their diet for 4 weeks. After 4 weeks, the body weight of all experimental groups increased compared to the diabetic control group, with the D-MHOB group having a significantly higher increase. Fasting glucose levels in all experimental groups (compared to the D-control group) significantly decreased after 2 weeks, but between 2~3 weeks, the levels in the D-MH group were similar to the D-MHOA group. After 4 weeks, the fasting glucose level of the D-MHOB group was the lowest compared to the other groups. In a test of oral glucose tolerance, blood glucose levels were highest 60 minutes after feeding glucose; however this level improved significantly in the groups fed an experimental diet compared to the D-control group. Glycosylated hemoglobin levels were 1.9 times higher in the D-control group than the normal (3.9%), but levels in the experimental groups were significantly decreased in D-MHOA and D-MHOB groups compared to the D-MH group. In the high amounts of O. japonicus to medicinal herbs mixture, total lipids and cholesterol significantly decreased in the serum, while HDL-C levels increased. GPT activity was significantly lower in the D-MHOB group compared to the other groups. Lipid peroxide levels decreased in the D-MHOA and D-MHOB groups compared to the D-MH group. Antioxidant activity was higher depending on the dose of O. japonicus. Overall, O. japonicus exhibited effective hypoglycemic and hypolipidemic actions enhanced by a combination of medicinal herbs.
