Separation of Egg White Using HPLC with Change of Mobile Phase and Temperature (HPLC에서 이동상 변화와 온도에 따른 난백의 분리)

  • Lysozyme in egg white functions as bacteriolysis agent and ovalbumin plays a role as antigen in immune system. Egg white analysis methods usually include electrophoresis, gel permeation chromatography and reversed-phase HPLC(RP-HPLC). Among them, RP-HPLC was selected for rapid analysis and C18 column(Agilent, USA) was used as HPLC column. Optimum conditions were searched by changing mobile phase and temperature. Capacity factor and resolution were calculated and compared for various elution conditions. In the isocratic elution, mobile phase volume ratio was changed from 30/70/0.1 to 60/40/0.1(Acetonitrile(ACN)/Distilled water(DW)/Trifluoroacetic acid(TFA)). ACN composition was increased by 10% and temperature was set as $20^{\circ}C$. In the gradient elution, ACN/DW ratio was changed from 10/90 to 60/40 during 20 minute and temperature was varied as 20, 30 and $40^{\circ}C$. In the isocratic elution, three peaks were separated at 50/50/0.1. Lysozyme and ovalbumin were confirmed as first and third peak in three peaks respectively. In the gradient elution, four peaks were separated at $30^{\circ}C$. Lysozyme and ovalbumin were confirmed as first peak and third peak in four peaks respectively.

Electro-rheological Measurements of Phase Inversion of Emulsions under Shear Flow (전단응력 하에서 에멀젼 상 변이의 측정을 위한 전기 유변학적 연구)

  • This study aims at measuring electrical and rheological properties of cosmetic emulsions on the skin under shear flow. The effects of volume ratio and surfactants on structural changes of emulsions were examined by determining the changes of electrical resistance, viscosity, and morphology. As the ratio of the internal phase increased, the phase inversion occurred more quickly. The viscosity change was found to increase with increasing of the variation of electrical resistance of the emulsions. This phenomenon may be caused by decreased resistant force against the shear flow because of the breakdown of the internal phase. Surfactants a]so played a key ro]e on phase transition of emulsions. It is likely that polymeric surfactants anchoring on the emulsion surface reinforced the interfacial mechanical strength. As the concentration of surfactants increased, the phase transition occurred more slowly. It has been demonstrated that the phase changes of emulsions under shear flow can be monitored on the real-time basis by using a JELLI$\^$TM/ chip system, a combination of conductiometry and rheometry. Our approach is expected to a useful experimental tool for predicting the phase transition of the cosmetic products during skin application.

Irregular Waves-Induced Seabed Dynamic Responses around Submerged Breakwater (불규칙파동장하 잠제 주변지반의 동적거동에 관한 수치해석)

  • In case of the seabed around and under gravity structures such as submerged breakwater is exposed to a large wave action long period, the excess pore pressure will be generated significantly due to pore volume change associated with rearrangement soil grains. This effect will lead a seabed liquefaction around and under structures as a result from decrease in the effective stress. Under the seabed liquefaction occurred and developed, the possibility of structure failure will be increased eventually. Lee et al.(2016) studied for regular waves, and this study considered for irregular waves with the same numerical analysis method used for regular waves. Under the condition of the irregular wave field, the time and spatial series of the deformation of submerged breakwater, the pore water pressure (oscillatory and residual components) and pore water pressure ratio in the seabed were estimated and their results were compared with those of the regular wave field to evaluate the liquefaction potential on the seabed quantitatively. Although present results are based on a limited number of numerical simulations, one of the study's most important findings is that a more safe design can be obtainable when analyzing case with a regular wave condition corresponding to a significant wave of irregular wave.

Analysis of Noticing Characteristics Presented in Elementary Pre-service Teachers' Self-reflection Journals on the Science Class (초등 예비교사의 과학수업 성찰지에 나타난 노티싱 특성 분석)

  • For teachers, noticing refers to paying attention to something, indicating they interpret it and how they are willing to react to it in the context of their own instruction. Analysis of noticing features enables us to understand the overall characteristics of the teacher's lesson design, practice, and reflection, which are core agents in the educational design and implementation. This can also be taken to be the basis of education design for competency reinforcement for teachers. Therefore, in this study, the characteristics of noticing shown in teachers' reflections after class design and demonstration were identified. For this purpose, the self-reflection journals of 106 elementary pre-service teachers enrolled in the College of Education in Gangwon-do were analyzed. In particular, the journals were gathered that were written after the demonstration dealing with the change of gas volume by temperature in science class. After designing a noticing analysis frame consisting of the five dimensions 'agent', 'stage', 'topic', 'focus', and 'stance', the frequency and ratio of noticing by each dimension's components were derived. The frequency and ratio of noticing for the dimension of 'focus' were analyzed for the dimensions of 'stage' and 'topic'. The results of the study were as follows. For the dimension of 'agent', the frequency of teacher and student was the highest, and for the dimension of 'stage', inquiry activity was the highest. For the 'topic' dimension, class design according to the teaching strategy appeared most frequently, and in the 'focus' dimension, the cases that did not specify the goal of the class and the competencies to be achieved by the students appeared most frequently. In the 'stance' dimension, description showed the highest frequency. From the analysis of how the 'focus' changes according to the 'stage' and 'topic', it was found that a characteristic focus appeared for each component of the dimension. From these results, the implications of the noticing characteristics of pre-service teachers for the design and implementation of teacher education were discussed.

Improving Diagnostic Performance of MRI for Temporal Lobe Epilepsy With Deep Learning-Based Image Reconstruction in Patients With Suspected Focal Epilepsy

  • Objective: To evaluate the diagnostic performance and image quality of 1.5-mm slice thickness MRI with deep learningbased image reconstruction (1.5-mm MRI + DLR) compared to routine 3-mm slice thickness MRI (routine MRI) and 1.5-mm slice thickness MRI without DLR (1.5-mm MRI without DLR) for evaluating temporal lobe epilepsy (TLE). Materials and Methods: This retrospective study included 117 MR image sets comprising 1.5-mm MRI + DLR, 1.5-mm MRI without DLR, and routine MRI from 117 consecutive patients (mean age, 41 years; 61 female; 34 patients with TLE and 83 without TLE). Two neuroradiologists evaluated the presence of hippocampal or temporal lobe lesions, volume loss, signal abnormalities, loss of internal structure of the hippocampus, and lesion conspicuity in the temporal lobe. Reference standards for TLE were independently constructed by neurologists using clinical and radiological findings. Subjective image quality, signal-to-noise ratio (SNR), and contrast-to-noise ratio (CNR) were analyzed. Performance in diagnosing TLE, lesion findings, and image quality were compared among the three protocols. Results: The pooled sensitivity of 1.5-mm MRI + DLR (91.2%) for diagnosing TLE was higher than that of routine MRI (72.1%, P < 0.001). In the subgroup analysis, 1.5-mm MRI + DLR showed higher sensitivity for hippocampal lesions than routine MRI (92.7% vs. 75.0%, P = 0.001), with improved depiction of hippocampal T2 high signal intensity change (P = 0.016) and loss of internal structure (P < 0.001). However, the pooled specificity of 1.5-mm MRI + DLR (76.5%) was lower than that of routine MRI (89.2%, P = 0.004). Compared with 1.5-mm MRI without DLR, 1.5-mm MRI + DLR resulted in significantly improved pooled accuracy (91.2% vs. 73.1%, P = 0.010), image quality, SNR, and CNR (all, P < 0.001). Conclusion: The use of 1.5-mm MRI + DLR enhanced the performance of MRI in diagnosing TLE, particularly in hippocampal evaluation, because of improved depiction of hippocampal abnormalities and enhanced image quality.

Effect of the Pretreatment by Thermal Hydrolysis on Biochemical Methane Potential of Piggery Sludge (열가수분해 전처리가 양돈 슬러지의 메탄생산퍼텐셜에 미치는 영향)

  • The objective of this study was to investigate the organic solubilization (SCOD) and improvement of methane production for pig slurry by thermal hydrolysis. A sludge cake was pretreated by thermal hydrolysis at different reaction temperatures (200, 220, 250, $270^{\circ}C$). Ultimate methane potential (Bu) was determined at several substrate and inoculum (S/I) ratios (1:9, 3:7, 5:5, 7:3 in volume ratio) by biochemical methane potential (BMP) assay for 73 days. Pig slurry SCOD were obtained with 98.4~98.9% at the reaction temperature of $200{\sim}270^{\circ}C$. Theoretical methane potentials ($B_{th}$) of thermal hydrolysates at the reaction temperature of $200^{\circ}C$, $220^{\circ}C$, $250^{\circ}C$, $270^{\circ}C$ were 0.631, 0.634, 0.705, $0.748Nm^3\;kg^{-1}-VS_{added}$, respectively. $B_u$ of $200^{\circ}C$ thermal hydrolysate were decreased from $0.197Nm^3\;kg^{-1}-VS_{added}$ to $0.111Nm^3\;kg^{-1}-VS_{added}$ with the changes of S/I ratio from 1:9 to 7:3, and also $B_u$ of different thermal hydrolysates ($220^{\circ}C$, $250^{\circ}C$, $270^{\circ}C$) showed same tendency to $B_u$ of $200^{\circ}C$ thermal hydrolysate according to the changes of S/I ratio. Anaerobic biodegradability ($B_u/B_{th}$) of $200^{\circ}C$ thermal hydrolysate at different S/I ratios was decreased from 32.2% for S/I ratio of 1:9 to 17.6% for S/I ratio of 7:3. $B_u/B_{th}$ of $220^{\circ}C$, $250^{\circ}C$, and $270^{\circ}C$ thermal hydrolysat were decreased from 36.4% to 9.6%, from 31.3% to 0.8%, and from 26.6% to 0.8%, respectively, with the S/I ratio change, respectively. In this study, the rise of thermal reaction temperature caused the decrease of anaerobic digestibility and methane production while organic materials of pig slurry were more solubilized.

  • Background: The opitmal ventilator setting during partial liquid ventilation(PLV) is controversial. This study investigated the effects of various gas exchange parameters during PLV in normal rabbit lungs in order to aid in the development of an optimal ventilator setting during PLV. Methods: Seven New-Zealand white rabbits were ventilated in pressure-controlled mode with the following settings; tidal volume($V_T$) 8 mL/kg, positive end-expiratory pressure(PEEP) 4 $cmH_2O$, inspiratory-to-expiratory ratio(I:E ratio) 1:2, fraction of inspired oxygen($F_TO_2$) 1.0. The respiration rate(RR) was adjusted to keep $PaCO_2$ between 35~45 mmHg. The ventilator settings were changed every 30 min in the following sequence : (1) Baseline, as the basal ventilator setting, (2) Inverse ratio, I:E ratio 2:1, (3) high PEEP, adjust PEEP to achieve the same mean inspiratory pressure (MIP) as in the inverse ratio, (4) High $V_T$, $V_T$ 15 mL/kg, (5) high RR, the same minute ventilation (MV) as in the High $V_T$. Subsequently, the same protocol was repeated after instilling 18 mL/kg of perfluorodecalin for PLV. The parameters of gas exchange, lung mechanics, and hemodynamics were examined. Results: (1) The gas ventilation(GV) group showed no significant changes in the $PaO_2$ at all phases. The $PaCO_2$ was lower and the pH was higher at the high $V_T$ and high RR phases(p<0.05). No significant changes in the lung mechanics and hemodynamics parameters were observed. (2) The baseline $PaO_2$ for the PLV was $312{\pm}$ mmHg. This was significantly lower when decreased compared to the baseline $PaO_2$ for GV which was $504{\pm}81$ mmHg(p=0.001). During PLV, the $PaO_2$, was significantly higher at the high PEEP($452{\pm}38$ mmHg) and high $V_T$ ($461{\pm}53$ mmHg) phases compared with the baseline phase. However, it did not change significantly during the inverse I:E ratio or the high RR phases. (3) The $PaCO_2$ was significantly lower at high $V_T$ and RR phases for both the GV and PLV. During the PLV, $PaCO_2$ were significantly higher compared to the GV (p<0.05). (4) There were no important or significant changes in of baseline and high RR phases lung mechanics and hemodynamics parameters during the PLV. Conclusion: During PLV in the normal lung, adequate $V_T$ and PEEP are important for optimal oxygenation.

Artificial Multiplication of the Black Soldier Fly(BSF), Hermetia illucens(Diptera: Stratmyidae) Using the Livestock Feces (가축분뇨를 이용한 동애등에 증식기술 개발)

  • This study was conducted to investigated the distribution, ecological character and life cycle of Black Soldier Fly(BSF), Hermetia illucens, to determine effects of BSF on composing ability to the livestock feces. The distribution of BSF was defined in all parts of the country in Korea. Its main habitat was found to be areas near cattle sheds, feces sheds, living waste dump grounds, and food waste dump grounds. Excellent species of the bio-conversion using the livestock feces were selected out of two kinds of solider flies(Hermetia illucens and Ptecticus tenebrifer). And effects of BSF on composing ability to the livestock feces were pig feces 0.5 g, cattle feces 0.43 g and chicken feces 0.3 g per a larvae of BSF. In the examination of the ability of BSF to decompose food waste, volume of the livestock feces decreased by weight pig feces 77, cattle feces 83.62 and chicken feces 80%. Observed characteristics of BSF larvae and pupae stage were by developmental stage may be summarized as follows: size and length of larvae and pupae of BSF were higher than in pig feces and mixed sawdust treatments. In order to develop artificial indoor rearing techniques, The pupation and emergence rate were not visible the difference but important factor of the mating and egg-laying was a sun light. And it was visible the difference in mating ratio. Mating rate of H. illucens by seasonal change was the highest from May to July. Mating mostly occurred between 10 : 00 and 12 : 00 during which light intensity is high. As the egg-laying medium for artificial egg collection, calf feed and food waste were most effective. The ratio of egg-laying medium is waste food : sawdust(1 : 1) in volume. For egg-laying materials, flower foam and wood with holes were used; holes sized 3 ~ 5 mm in diameter, 7 ~ 10 mm in depth were most preferred for egg-laying.

Solvothermal Preparation of Nanocrystalline TiO2 Using Alcohol-water Mixed Solvent (알코올-물 혼합용액을 이용하는 Solvothermal 법에 의한 나노크기의 TiO2 제조)

  • In this study, a solvothermal reaction to prepare nanocrystalline titania was carried out using $TiCl_4$ and mixed solvents of alcohol and water. The effects of the type and the composition of alcohol on the crystal structure and agglomeration of final $TiO_2$ products were investigated. The products were characterized by X-ray diffraction (XRD), transmission electron microscopy (TEM) as well as scanning electron microscopy (SEM). In the solvothermal reaction using the n-butanol solutions with different volume ratios of n-butanol/water (100/0, 75/25, 50/50, 25/75, 0/100), the extent of agglomeration of obtained rutile $TiO_2$ was found to change with the volume ratio of n-butanol/water, and the n-butanol/water ratio of 75/25 revealed the best result for the preparation of well-dispersed nanocrystalline $TiO_2$ powders. The crystal phase of $TiO_2$ prepared through the solvothermal reaction changed with the type of alcohol in solvent (alcohol/water = 75/25). $TiO_2$ products obtained with the aqueous solutions of methanol, ethanol and isopropanol have an anatase phase, while that with n-butanol has a rutile phase. The results showed that, in the solvothermal reaction using both $TiCl_4$ as a starting material and the alcohol-water mixed solvents without any other additive, the enhancement of dispersion and control of crystal structure of $TiO_2$ products can be feasible by simply varying the composition and type of alcohol in the mixed solvents.

Color revelation characteristics of color mortar using iron oxide and carbon black (산화철과 카본블랙을 사용한 컬러 모르터르의 색상발현 특성)

  • Recently, as there has been growing demand for aesthetic quality in structural materials, the development of excellent color cement concrete having good coloration properties has become a requirement. This study is designed to analyze the basic physical properties of mortar and the properties of keeping the coloration under an ultraviolet ray accelerated weathering test environment according to changes in the mixing ratio between carbon black and iron oxide used as colorants. The test results show that the use of carbon black and iron oxide reduces the initial flow by 6.3~17.2 % and the air content by 3.5~31.5 % but increases the unit volume weight by 3.4~5.5 %, compared to ordinary Portland cement (OPC). In addition, the study shows that the addition of iron oxide increases the self drying shrinkage. So, caution needs to be taken on the workability of the concrete. The brightness value L represented by black showed the most excellent black colour when carbon black 3 % and iron oxide 5 % are added. According to UV accelerated weatherproof test, the brightness value L was found to increase in all experiment specimens by 4.28~11.97 %, and the color change by UV was found to be higher for the case where carbon black colorant was not used. Therefore, in terms of color revelation characteristics, the case using carbon black 3 % and iron oxide 5 % was found to show the best black color.
