To evaluate the protective effects of calcium antagonists, oxygen radical scavengers and excitatory amino acid (EAA) antagonist on the ischemic brain damage, we induced in vitro ischemic condition (namely, lack of oxygen and glucose) to rat hippocampal slices. And the degree of ischemic damage was determined by assaying changes in biochemical parameters such as ATP content and lactate ralease, MDA production in the presence or absence of the various drugs. During experimental ischemia for up to 60 min, ATP content was decreased and the amount of lactate release was markedly increased time-dependently. By changing the reaction medium which contained oxygen and glucose those biochemical parameters were recovered. But the recovery was not complete in this experimental condition. In the same ischemic conditions verapamil and vitamine E prevented the decrease of ATP content and the increase of lactate release from the slices. And verapamil and diltiazem decreased MDA release to the reaction medium. Superoxide dismutase (SOD) and MK-801 (as EAA receptor antagonist) protected the decrease of ATP content and reduced MDA release in 20 min ischemic condition, but glutathione affected ATP content and lactate release at the same condition. When oxygen and glucose were resupplied for 20 min after ischemic condition, verapamil showed the protective effect on the changes of ATP content and lactate release, and vitamine E decreased lactate release (at 20 min ischemia) and MDA release (at 60 min ischemia). These results showed that calcium antagonist and vitamine E protect the ischemic biochemical changes from rat hippocampal slices and calcium antagonist is more potent than vitamine E to protect the ischemical brain damege.
Flavonoids are class of polyphenolic compounds widely distributed in the plant kingdom, which display a variety of biological activities, including antiviral, antithrombotic, antiiflammatory, antihistaminic, antioxidant and free-radical scavenging abilities. To determined flavonoid, myricetin in the presence of other antioxidants - vitamin C and vitamin E - can exert antioxidative properties not only directly by modulating the AOE system but also scavenging free radical, we investigated cell viability, antioxidant enzyme activities and ROS level in B16F10 murine melanoma cell. B16F10 cells were exposed to medium containing myricetin in the presence or absence of vitamin C or vitamin E for a period of 24 hr. Cell viability was measured by MTT assay. In co-treating myricetin with other antioxidants, CAT activities were increased, compared with control, but SOD and GPx activities were decreased, compared with each antioxidant treated groups . In the group of myricetin or myricetin present with other antioxidants, ROS levels were decreased dose-dependently. Especially, myricetin present of other antioxidants were decreased compared with myricetin.
Electrical stimulation and vitamin $AD_3E$ administration have been shown to enhance the repair of biological tissues such as bone, ligament and tendon, The objective of this study were (a) to investgate the therapeutic effects of different levels of electrical stimulation and vitamin $AD_3E$ administration on fracture healing in a rat model and (b) to identify the most effective voltage level. Ninety Sprague-dawley rats were divided into electrical stimulation group and vitamin $AD_3E$ administration group. Electricla stimulation group was divided into four groups on the basis of the level of current delivered. The experimental groups received current (which varied by group), while the controls, with identical electrodes, received no current. In vitamin $AD_3E$ administration group, experimental group was injected 2,500 IU of vitamin A, 1,250 IU of vitamine $D_3$, 1mg of vitamine E intramuscularly. And in control group 0.1ml of saline was injected intramuscularly. After time periods (7-day, 14-day, 21-day for stimulation all rats were tested with combination of biochemical, roentgenologic and histomorphological methods. The results obtained were as follows ; In electrical stimulation groups, serum calcium and inorganic phosphorus level of experimental and control groups showed non specific change within normal physiological ranges. In vitamin $AD_3E$ administration group, serum calcium level of experimental and control groups showed non specific changes within normal physiological ranges, while experimental group showed slightly higher serum inorganic phosphorus level. According to roentgenologic and histomorphological examination, 2V 25Hz electrical stimulation group and vitamine $AD_3E$ administration group showed statistically significant improvements in bone density and ossification reaction until day 14. The terapeutic effect of stimulation on fracture healing was similiar to that of vitamin $AD_3E$ administration. In this study stimulation of 2V 25Hz was the most effective level of electrical stimulation for the healing of fracture of rats.
Hemofilter was optimally designed and manufactured using polysulfone hollow fiber mem- brane with surface area of 0.6mE Molecular weight cut-off of the hemofilter was measured with polyethylene glycol and dextran aqueous solutions of various molecular weights and it was ranged from 9,500 to 38,900. Ultrafiltration rates were measured with pure water in a static system and flowing system. The clearance of urea, creatinlne, and vitamine BIB were measured using aqueous solutions.
The considerable researches have been done on the change,1 associated with age in the blood picture of domestic animals, but a survey has not been of the blood picture in Thoroughbred fears in Korea. Therefore, the purpose of this study is to outline the range of values in clinically not\\\\`- mal Thoroughbred foals. Some blood chemical parameters from 3 days to 8 weeks of age were exam-ined to obtain normal values in Thoroughbred foal7. Blood samples were obtained from the jugular vein in 11 Thoroughbred foals and collected at 3 days, and at 1,2,4 and 8 weeks of age, respectively. The results obtained ill this study were summarized as follows: The packed cell volume (PCV) at 4 weeks of age was significantly lower than that at 3 days of age (P<0.01) and that al 2 and 8 weeks of age were straightly lower than that at 3 days of age (P<0.05). The total protein (TP) at 8 weeks of age was higher than that at 3 days, 1 and 2 weeks of age (P<0.05). The activity of aspartate ami-notransferase (AST) at 2,4 and 8 weeks of age were significantly higher than that at 3 days of age (P<0.01). The total bilirnbin showed a tendency to decrease gradually according to the weeks and that at 2,4 and 8 weeks of age were significantly lower than that at 3 days of age (P<0.01). The BUN at 1 and 8 weeks of ago were lower than that at 3 days of age (P<0.05). The concentration of Ca at 1 week of alee was higher than that at 3 days of age (P<0.05). The vitamin A and vitamin E showed a tendency to decrease gradually from 3 days to 4 weeks of age and increased slightly at 8 weeks of age.
Recently many researches on plaque removal effect and therapeutic effect of toothpaste containing natural medicines are being studied in early periodontal diseases. The purpose of this study is to examine the clinical and microbiological effect of toothpaste containing natural medicines such as camomile, rhatany, myrrh, sage oil, glycyrrhetinic acid and vitamin E. Sixty three subjects with gingivitis were divided into an experimental group which performed normal oral hygiene procedure with toothpaste containing natural medicines and vitamine E and a control group which also performed normal oral hygiene procedure with Syrinmed? toothpaste without containing herbal extracts and vitamine E. At the baseline, 2 weeks, and 4 weeks, subjects were analyzed for clinical study and microbiological study. After 2 weeks and 4 weeks use of their respective toothpastes, statistically significant decreases of gingival index, plaque index, and bleeding index were shown in both the control and the experimental group. The degree of decrease was more significant in the experimental group than the control group. A statistically significant decrease of pocket depth, and gingival crevicular fluid were shown in both the control and lie experimental group. A statistically significant increase of cocci was shown in both the control and the experimental group, the degree of increase was more significant in the experimental group than control group. A statistically significant decrease ofnon-motile rods, and motile rods were shown in both the control and the experimental group, the degree of decrease was more significant in the experimental group than the control group. Spirochetes increased weakly in both the control and the experimental group but a statistic significance was not shown. A statistically significant decrease of anaerobic bacteria, aerobic bacteria, and black pigmented Bacteroides were shown in both the control and the experimental group. These results indicate that the use of toothpaste containing natural medicines is effective in the prevention and the treatment of periodontal diseases.
Background : Acute pulmonary injury by paraquat are caused by multiple mechanisms including direct injury with oxygen free radicals and several mediators released from inflammatory cells. In order to clarify whether vitamin E could reduce tissue damages induced by intraperitoneal administration of paraquat and to investigate the pathogenetic mechanisms of paraquat-induced pulmonary injury, vitamin E as a free radical scavenger was administered. Method : Rats were divided into three groups (group 1 : control, group 2 : paraquat treated group, group 3 : paraquat and vitamin E treated group). Animals were sacrificed on day 1, day 2, day 3, and day 8 after the administration of saline, paraquat, or paraquat/vitamin E. Results : Treatment with vitamin E decreased the death rate of rats treated with paraquat. Comparing with control group ($1.37{\times}10^6/ml$), mean total cell counts recovered from the lavage fluid from animals treated with paraquat($1.65{\times}10^6/ml$) were increased(p=0.06). Magnitudes of increment of the total cell counts on the Day 8 in the vitamin E treated group were smaller than those of the animals treated with paraquat alone. The neutrophils began to appear in significant amounts in the lavage fluid on Day 8 after the administration of paraquat(37.0+12.7%). A significant decreasing neutrophil concentration at Day 8 was observed in the paraquat/vitamin E treated group(20.6+13.4%). Histologically the degree of pulmonary fibrosis was most prominent in the paraquat treated group while diffuse alveolar damage was continuously observed in the paraquat/vitamin E treated group and extensive interstitial lymphocytic infiltration was seen in the paraquat/vitamin E treated group. The paraquat/vitamin E treated group showed the less histologic changes. Conclusion : In this study vitamin E acting as a scavenger of neutrophil-derived free radicals and suppressant of lipid peroxidation, seemed to be the effective antioxidant in the inhibition of paraquat-induced pulmonary injury.
Fruits of Arctium lappa L. (Compositae), which has been used as the antiinflammatory, detoxifying and diuretic agents in the folk remedies, was examined on the in vitro oxidation of human low density lipoprotein (LDL). Several lines of evidence indicate that oxidized LDL (Ox-LDL) may promote atherogenesis. Hence. The role of antioxidants in the prevention of LDL oxidation needs to be determined. The activity of fractions of Arctii Fructus treated with oxidized LDL which was incubated with $16\;{\mu}M$ of $Cu^{2+}$ for metal catalyzed oxidation was investigated. The BuOH fraction (4\;ppm) inhibited the oxidative modification of LDL as evidenced by a decrease in the lipid peroxide content (thiobarbituric acid reacting substances activity), the negative charge of LDL (electrophoretic mobility) and increase of the vitamine E content.

The purpose of this study was to evaluated the antioxidative constituents and their activities of the 80% methanolic extracts from fruit of Sorbaria sorbifolia var. stellipila MAX. The isolation of active compound was performed in three steps: solvent partition, open column chromatography, and high-performance liquid chromatography (HPLC). The solvent fractions were tested for their antioxidant activities by oxygen radical absorbance capacity (ORAC). The antioxidant activity of 80% methanolic extracts by various solvent partitions was in the order of 80% MeOH (1.68 ${\pm}$ 0.027), n-hexane (1.02 ${\pm}$ 0.036), $CH_2Cl_2$ (0.95 ${\pm}$ 0.025), EtOAc (1.98 ${\pm}$ 0.065), n-BuOH (1.94 ${\pm}$ 0.054) and Water (1.28 ${\pm}$ 0.032). Therefore, the results indicated that the potential antioxidant activities and functional values were observed significantly at EtOAc fraction from fruit of S. sorbifolia, flavonoid compound isolated.
