effects of Vitamin E on the Antioxidative Defense System of Kidney in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨쥐 신장조직의 항산화계에 미치는 Vitamin E의 영향)

  • The purpose of this study was to investigate the effects of vitamin E on the antioxidative defense system of kidney in streptozotocin induced diabetic rats. Sprague Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal and three STZ induced diabetic groups, which were subdivided into vitamin E free diet(DM 0E group), 40mg vitamin E per kg diet(DM 40E group) and 400mg vitamin E per kg diet(DM 400E group). Vitamin E level of normal group was 40 mg per kg diet. Diabetes was experimentally induced by intravenous injection of 55mg/kg of body weight of streptozotocin(STZ) in citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Animals were sacrificed at the 6th day of diabetic states. There were no significant on body weights, food intakes, and food efficiency ratio before the diabetic occurrence. But after the injection of STZ, body weights and food efficiency ratios were significantly decreased and the food intakes was increased. Kidney weights were significantly increased in diabetic groups compared to normal group. However, there were no significant differences among the diabetic groups. Plasma insulin levels of diabetic groups were significantly decreased, whereas, blood sugar levels were increased compared to that of normal group. There were no significant differences among diabetic groups in plasma insulin and glucose levels. Activities of superoxide dismutase(SOD) in DM 0E and DM 40E groups were signi ficantly decreased by 33% and 27%, respectively, compared to normal group. But that of DM 400E group was increased by 35% compared to DM 0E group. Activity of glutathione peroxidase(GSHpx) in DM 0E group was decreased by 20% compared with normal group. GSHpx activity in DM 400E group was increased by 29% compared to normal group. The contents of vitamin E in kidney were 58% and 49% lower in DM 0E and DM 40E group, respectively, than normal group. There was no significant difference in renal vitamin E contents between DM-400E group and normal group. The contents of superoxide radical(O2 ) in kidney were 150% and 98%, respectively, higher in DM 0E and DM 40E groups than normalgroup. DM 400E and normal groups were similar levels in their superoxide radical contents of kidneys. These results indicate that vitamin E functioned as chain breaking antioxidant in kidney such as in other tissues.

  • This study was performed to investigate the effect of dietary vitamin E supplement and kinds of lipid on the vitamin E concentration and lipid composition in serum of the rats fed fro seven weeks. Supplemented vitamin E was 300mg/kg diet as DL-α-tocopherol acetate, and 15% of corn oil or beef tallow was added to standard diet. The results were as follows ; 1) Body weight and feed efficiency ratio were significantly increased by supplement with vitamin E, corn oil and beef tallow. 2) Serum vitamin E concentration increased significantly by supplement of vitamin E and beef tallow on the diets, while one of corn oil fed group decreased significantly . 3) The contents of triglyceride, total cholesterol, phospholipid in serum were significantly increased in the beef tallow group, and the vitamin E only -supplemented group showed significantly lower serum cholesterol in serum increased significantly in the vitamin E only -fed group and vitamin E plus corn oil-fed group. 4) The content of HDL-cholesterol in serum increased significantly in the vitamin E only -fed group and vitmain e plus corn oil -fed group. 5) The ratio of VLDL, LDL-cholesterol content to HDL-cholesterol one in serum was significantly low in the vitamin E only-supplemented group and corn oil group, and was significantly higher in the beef tallow group than in the corn oil group.

Effects of Vitamin E on the Microsomal Mixed Function Oxidase System of Kidney in Streptozotocin-induced Diabetic Rats (식이 Vitamin E가 Streptozotocin 유발 당뇨쥐 신장조직에서의 Microsomal Mixed Function Oxidase System에 미치는 영향)

  • The purpose of this study was to investigate the effects of vitamin E on microsomal mixed function oxidase system of kidney in streptozotocin(STZ) induced diabetic rats. Sprague-Dawley male rats weighing 140$\pm$10g were randomly assigned to one control and three STZ-diabetic groups which were subdivided into vitamin E free diet(DM-0E group) 40mg vitamin E per kg diet(DM-40E group) and 400mg vitamin E per kg diet(DM-400E group). Vitamin E level of normal group was 40 mg per kg diet. Diabetes was experimentally induced by intravenous administration of 55 mg/kg B.W of STZ in citrate buffer(pH4.3) after 4 weeks feeding of experimental diets. Animals were sacrificed at the 6th day of diabetic state. The contents of cytochrome P450 in kidney were increased by 82, 54, 41% in DM-0E, DM-40E and DM-400E groups respectively when compared with normal group. The contents of cytochrome b5 in kidney were increased by 28% in DM-0E when compared with normal group but those of DM-40E and DM-400E groups were similar to that of normal group. The activities of NADPH-cytochrome P450 reductase in kidney that were increased by 35% in DM-0E group. Levels of TBARS(thiobarbituric acid reactive substance) in kidney were increased by 207, 129% and 72% in DM-0E and DM-400E groups respectively when compared with normal group but those of DM-40E and DM-400E groups were 26,44% lower than that of DM-0E groups. It is know that the activities of MFO system and lipid peroxidation were inhibited in kidney of STZ-induced diabetic rat by administeration of high doses of vitamin E.(Korean J Nutrition 33(6) : 619~624, 2000)

Effects of Vitamin E and Dehydroepiandrosterone on the Formation of Preneoplastic Lesions in Rat Hepatocellular Carcinogenesis (비타민 E와 Dehydroepiandrosterone이 화학적 발암원으로 유도한 쥐간의 전암성 병변에 미치는 영향)

  • This study is designed to examine the effects of dietary supplementation with vitamin E and dehydroepiandrosterone (DHEA) on the formation of preneoplastic lesions in diethylnitrosamine (DEN) induced rat hepatocarcinogenesis. All Weaning male Sprague-Dawley rats were initiated by a single dose of DEN (200mg/kg body weight), subjected to two­thirds partial hepatectomy 3 weeks later and were sacrificed 8 weeks after DEN initiation. Two weeks after initiation, rats were fed Purina purified rodent diet 5053 (Ralston Purina Rat chow, USA) with $1.5\%$ (15,000 IU/kg diet) vitamin E, $0.5\%$ DHEA and both of those supplemented diet for 6 weeks. Placental glutathione S-transferase (GST-P) positive foci, the activities of catalase, total-glutathione peroxidase (GPx) , glutathione reductase (GR), glutathione S-transferase (GST) and thiobarbituric acid reactive substances (TBARS) contents were decreased significantly by vitaimin E supplement. On the other hand GST-P positive foci number, Cu/Zn-superoxide dismutase (SOD) and glucose 6-phosphatase (G6Pase) activities weren't changed by vitamin E supplement. It might suggest that protective effect of vitamin E against hepatocarcinogens is not involved in the formation of the GST-P positive foci but related to the expansion of that. It seemed that vitamin E supplement helped endogenous defense system in carcinogenesis by decreasing TBARS contents, $H_2O_2$, organic peroxides. Therefore, vitamin E seemed to protect cell from free radical damage in carcinogenesis. By DHEA supplement liver weight and liver/body ratio were increased, the area and number of GST-P positive foci, the activities of catalase, GR, total GPx, GST and the TBARS contents were decreased significantly. On the other hand Cu/Zn-SOD and G6Pase activities weren't changed by DHEA supplement. In hepatocarcinogenesis the activities of antioxidant enzymes weren't increased by DHEA supplement. DHEA did not increase the oxidative stress, while DHEA seems to have anticarcinogenic effect in rats hepatocarcinogenesis.

  • Semi-micro-HPLC using a column-switching technique was developed for simultaneous determination of vitamin A and E contents in infant formula. Vitamin A and E were extracted by PDA - HPLC with reversed phase column using organic solvent and their contents in Certified Reference Material (CRM) and infant formula were determined and compared with hydrolysis method and rapid extraction. Developed method has many advantages of simple and rapid sample preparation and simultaneous determination of vitamin A and E by micro-HPLC using reversed phase column.

  • Biochemical studies such as growth rate, blood cholesterol and ascorbic acid contents in various organs of albino male rats were studied in the Garlic, Vitamin E, high and low protein diet fed groups. The results of this study were summarized as follows : 1) Either single 10% or 30% protein diet fed rats were not shown growth rate properly. In supplementation of Vitamin E or Garlic on 30% protein growth rate was a more increased than that of the 10% protein diet group. 2) The rate of food consumption of rats fed a 10% or 30% protein diet supplemented with Vitamin E and Garlic was more increased than that of the control Group receving 10% or 30% protein diet alone. 3) No essential difference was observed between the cholesterol level in blood of rats fed 10% protein diet supplemented with Vitamin E and Garlic and that of rats Receiving 10% protein diet alone. But the cholesterol level in blood of rats fed 30% protein diet supplemented with Vitamin E and Garlic was lower than that of control receiving 30% protein diet alone. 4) The Vitamin C contents in various organs of rats fed the diet supplemented with Vitamin E and Garlic seems to be a little higher than that of the control group fed the protein diet alone.

Effect of dietary vitamin E on the lipid peroxidation by dietary iron injected to male rats (Vitamin E가 철분 투여로 인한 rat의 지질 과산화에 미치는 영향)

  • This study was designed to investigate the effect of dietary vitamin E on the lipid peroxidation by dietary iron-injected to male rats. Sprague-Dawely strain male rats were divided into three experimental groups, namely control, iron injected and iron-vitamin E injected groups. The control group was fed with normal diet; the iron injected group was given normal diet and while injected intraperitoneally 30mg of ferric hydroxide/100g of body weight 20 times every 3 days. The iron-vitamin E injected group was intraperitoneally administered 30mg of ferric hydroxide/100g of body weight 20 times every 3 days and vitamin E every day with the dose of 5IU(5mg)/100g body weight. All experimental groups were maintained for 60 days with feeding on the respective ratio. The results obtained from this experiment were summarized as following: 1. The net weight gain was significantly decreased by the iron injection, but much increased by the vitamin E injection. 2. The contents of unsaturated fatty acid in phospholipid in liver, kidney, muscle and serum were decreased by the iron injection, but increased by the vitamin E injection. 3. The increment of malondialdehyde contents was induced by the iron overloading, but significantly decreased by the vitamin E injection. Therefore, it is suggested that dietary iron administration to male rats facilitates the lipid peroxidation in vivo and vitamin E has the inhibiting effect on lipip peroxidation process by iron.

Studies on Transfer of Dietary Vitamins A and E to Chicken Eggs (사료 비타민 A와 E의 계란내 이행에 관한 연구)

  • Vitarnin A levels of egg yolk from hens fed diets supplemented with vitamin A increased slowly up to Week 4, while the values rapidly increased thereafter. The highest value(360% of initial value) was observed at Week 8 in birds fed 64,000 IU of vitamin A/kg diet. On the other hand, the contents of vitamin E in egg yolk rapidly increased in the first 2 weeks of the experiment and then stabilized up to Week 8. However, the efficiency of egg transfer decreased(P<0.05) as dietary vitamins A and E levels increased. It took at least 4 weeks for the increased levels of vitamin A and E in eggs from hens fed diets supplemented with each vitamin to recover the normal levels. Vitamin E levels of egg yolk in hens fed the vitamin A supplemented diets decreased as dietary vitamin A level increased (P<0.05). On the other hand, a slightly reduced levels of vitamin A levels of egg yolks from hens fed the vitamin E supplemented diet for 4 weeks of experiment was recovered back to control level at Week 8(P<0.05). Supplemental vitamin A markedly decreased the yolk deposition of vitamin F when two vitamins were supplemented simultaneously. The yolk deposition of vitamin A decreased as high level of vitamin B was supplemented, suggesting an antagonistic interactions between the two vitamins.

Optimization of Supercritical Fluid Extraction of Tocotrienol from Grape Seed (초임계유체 추출을 이용한 포도씨 tocotrienol 추출조건 최적화)

  • In this study, supercritical carbon dioxide extraction (SFE) was utilized for the extraction of tocotrienol from grape seeds. The optimal conditions for vitamin E and tocotrienol extraction were determined via response surface methodology (RSM). Central composite design was utilized to assess the effects of oven temperature (30-$50^{\circ}C$, X1), operating pressure (17-25 MPa, X2), and extraction time (1-5 hr, X3) of supercritical fluid extraction. Vitamin E and tocotrienol contents were 8.65 mg/100 g and 7.88 mg/100 g at $40^{\circ}C$, 20MPa and 5 hr, respectively. The predicted extraction condition was validated via actual experimentation. The predicted extraction conditions were $40^{\circ}C$, 3.8 hr, and 20.7MPa. The vitamin E and tocotrienol contents under these conditions were 8.20 mg/100 g and 7.42 mg/100 g, respectively. The vitamin E and tocotrienol contents of solvent extraction with hexane were 8.18 mg/100 g and 7.24 mg/100 g, respectively.

High Vitamin E Supplement is Needed to Have an Anticarcinogenic Effect of Fish Oil (어유와 비타민 E 보강 수준이 쥐간의 전암성 병변에 미치는 영향)

  • The influences of fish oil and different levels of vitamin I supplement on hepatocellular chemical carcinogenesis have been studied. Male Sprague-Dawley rats received diethylnitrosamine (DEN)(200mg/kg body weight) and were subjected to two-thirds partial hepatectomy to induce murine chemical hepatocarcinogenic procedure. Placental glutathione S-transferase(GST-P) positive foci area, antioxidant enzymes(Cu/Zn-superoxide dismutase(SOD), catalase, glutathione reductase (GR), total- glutathione peroxidase (TGPx), glutathione S -transferase (GST)), glucose 6-phosphatase (G6Pase) activities, and lipid peroxidation of microsomes(thiobarbituric acid reactive substances (TBARS)) were measured. Experimental animals were fed 15% corn or fish oil with 0, 40, 1,000, 10,000IU vitamin E /kg diet for 8 weeks. Vitamin E supplements decreased the area of GST-P positive foci in both groups. The higher the vitamin E levels, the smaller the area of GST-P positive foci were noticed. Compared to 0 IU vitamin E, 40 IU in corn oil and 1,000 IU in fish oil groups were effective in decreasing G57-P positive foci area. Fish oil groups tended to have smaller area of GST-P positive foci. fish oil groups showed lower body weight, lower activities of Cu/Zn-SOD and TGPx, higher TBARS contents, higher activities of GST, catalase, G6Pase, GR and higher liver/body ratio than corn oil groups. As the level of vitamin I increased, GST-P positive foci count, catalase activities, and TBARS tended to decrease. G6Pase activities tended to increase in both groups. At higher vitamin E levels, GST activities tended to decrease in fish oil groups. These results suggest that vitamin I has suppressive offects on hepatocellular chemical carcinogenesis probably through antioxidant eH:cts decreasing TBARS contents, $H_2O$$_2$, and organic peroxides. fish oil tended to have greated suppressive offects than corn oil on hepatocellular carcinogenesis.
