Affinity for 57Co-Vitamin B12 by a Wide Histologic Variety of Tumor Types in Mice (생쥐 종양모델에서 57Co-Vitamin B12의 종양 친화성에 관한 연구)

  • The search for tumor-avid agents for use in nuclear medicine imaging is an ongoing field of importance. The purpose of this study was to determine the affinity for radio labeled vitamin $B_{12}$ by a wide histologic variety of tumor types in mice. Seventeen different types of tumor were grown subcutaneously in female Balb/C or Balb nu/nu(nude) mice. When the tumors reached about 1 cm in diameter, mice were injected intraperitoneally with $^{57}Co$-vitamin $B_{12}$. Twenty-four hours later, the mice were sacrificed. Organs and tissues were removed, weighed, and activity per mg determined by gamma counter. Values represented cpm/mg tissue that was normalized to 20 grams body weight for each mouse. A wide variety of tumor types showed significant uptake and concentration of $^{57}Co$-vitamin $B_{12}$, as evidenced by tumor:tissue activity ratios. For many tissues of great importance in terms of background(bone, muscle, blood), the tumor:tissue activity ratios of uptake were high. These data strongly suggest that further efforts to evaluate the utility of radio labeled adducts of vitamin $B_{12}$ for clinical use in oncologic imaging are warranted.

Production of Vitamin $B_{12}$ by Using Protoplast Fusion between Bacillus natto and Bacillus megaterium (Bacillus natto 및 Bacillus megaterium의 원형질체 융합에 의한 Vitamin $B_{12}$의 생산)

  • This study was conducted to breed a high vitamin $B_{12}$ producer by the fusion of protoplasts between Bacillus natto and Bacillus megaterium. Auxotrophic mutants of Bacillus natto SH-34 ($thr^-try^-rif^r$) and Bacillus megaterium BK-13 ($arg^-ade^-lys^-str^r$) which showed high protease activity and production of vitamin $B_{12}$, respectively, were isolated for the fusion experiment. Protoplasts were induced by incubating the cells with lysis solution containing $500{\mu}/ml$ lysozyme, and the ratio of protoplast and regeneration formation were ranged from 99% and 67%, respectively. Fusion frequencies of fusants between Bacillus natto SH-34 and Bacillus megaterium BK-13 were appeared in the ranges of $1.0{\times}10^{-5}$ under the treatment of 30% PEG 6000 containing 3% PVP. The fusant, MNF-72 showed the highest product yield of $7.85{\mu}g/g-cell\;vitamin\;B_{12}$ in production medium. For the improvement of productivity, the immobilization of fusants with sodium alginate was carried out. In batch and continuous fermentation systems, the productivity were determined to be $0.58{\mu}g/ml.hr\;and\;0.80{\mu}g/ml.hr\;vitamin\;B_{12}$ under optimum condition, respectivity.

Determination of Vitamin B12 (Cyanocobalamin) in Fortified Foods by HPLC

  • This study was conducted to develop an HPLC method for determining vitamin B$_{12}$ in fortified foods which has typically been determined by microbiological assays according to AOAC and Korean Food Code approved methods. Vitamin B$_{12}$ (cyanocobalamin) was determined by reversed-phase HPLC with a triple column and UV/VIS dectector (550 nm) using the column switching technique after extraction with 5 mM potassium phosphate solution by sonication without a clean-up procedure. The recovery of spiked samples and limit of detection (LOD) by HPLC were 78.6 ∼107.5 % and 2 ppb ($\mu\textrm{g}$/kg), respectively. The LOD of the microbiological assay (MBA) was much lower than that of HPLC. The concentrations of vitamin B$_{12}$ analyzed in all tested samples (n=12) confirmed compliance with declared label claims. The range of recovery ratio by the HPLC method when compared to the microbiological assay was 76.2 ∼140.0 %. There was not significant difference between the HPLC and MBA methods (p < 0.01) with r=0.9791 and linear regression y=0.9923x-0.04. The HPLC method for determining vitamin B$_{12}$ using the column-switching technique appears to be suitable for determining vitamin B$_{12}$ concentrations above 1 $\mu\textrm{g}$/100 g in fortified foods.ied foods.

  • Elevated plasma homocysteine is an independent risk factor for the development of cardiovascular disease. Exercise is generally believed to reduce the plasma homocysteine levels and therefore, being beneficial for cardiovascular disease (CVD). However, there is a possibility that athletes undergoing strenuous training and competition which increase oxidative stress may suffer from increased plasma homocysteine levels. The purpose of this study was to investigate the influence of endurance training on the plasma concentrations of B vitamins and homocysteine in 23 male adolescent field hockey players. Data collection and blood sampling was performed during the training period and non-training period. Following the training period, significant changes in energy and vitamin B6 intakes were observed in these subjects. Plasma vitamin B2, pyridoxal phosphate (PLP) and homocysteine levels were significantly higher during the training period than non-training period, whereas no difference was observed in plasma folate and vitamin B12 levels. Positive correlation was observed between plasma folate and folic acid intakes. When energy, B vitamin intakes were adjusted there was a significant negative correlation between plasma homocysteine levels and plasma riboflavin, folate and vitamin B12 levels. In conclusion, it is suggested that athletes with oxidative stress by strenuous exercise may need B vitamins since riboflavin, folic acid and vitamin Bl2 were shown to be negatively correlated with plasma homocysteine in athletes during the training period.

  • Several studies have reported a possible link between anti-aquaporin-4 antibody and vitamin B12 deficiency in neuromyelitis optica spectrum disorder (NMOSD). Bilaterally symmetric hyperintense signals on magnetic resonance imaging (MRI) of the posterior columns, called the inverted V sign, are a characteristic feature of subacute combined degeneration associated with vitamin B12 deficiency. We report a patient with anti-aquaporin-4 antibody-positive NMOSD and an inverted V sign on MRI of the spinal cord and address the association between anti-aquaporin-4 antibody and functional vitamin B12 deficiency.

  • Simultaneous deficiency of Vitamin $B_{12}$ and iron induces that the bone marrow erythroid megaloblastosis and peripheral blood macroovalocytosis are masked because of countervailing the tendency of iron deficiency to produce hypochromic microcytic erythrocytes. We report two cases of Vitamin $B_{12}$ deficiency anemia with low mean corpuscular volume (MCV) due to combined iron deficiency anemia with review of literature.

  • The purpose of this study was to investigate the effect of 4 week folic acid and vitamin B12 intake on hematological, immune and aerobic anaerobic exercise performance in female basketball players. The subjects were divided into nine groups: ingestion and control group. The results were as follows; 4 weeks of folic acid and vitamin B12 before and after ingestion. There was a significant effect on intake of aerobic and anaerobic capacity, and blood factor analysis showed a significant effect on intake of RBC and Hct. In conclusion, considering the results of the above study, it is expected that folic acid and vitamin B12 intake for improvement of anaerobic and anaerobic exercise ability of female basketball player can improve the performance.

Stuy on the Composition of Pleurotus ferulae Fruit Body (아위버섯의 성분에 관한 연구)

  • The composition of Pleurotus ferulae fruit body (PFF) has been investigated to determine the possible use for medicinal and edible purposes. The nutritional compositions of PFF were as follows; carbohydrate 54.3 g%, crude protein 20.2 g%, moisture 12.5 g%, crude fat 8.0 g% and ash 5.0 g% (w/w). Potassium concentration in PFF was high up to 2,337.0 mg% (w/w) and Mg, Na, Zn, Ca and Fe were followed. There were seventeen amino acids in PFF. The glutamic acid content was high up to 13.65% mol and aspartic acid, glysine, alanine, leucine, arginine, valine and lysine were followed. Since PFF contains major free amino acids such as alanine, phenylalanine, tyrosine and leucine, it will be a favorable food stuff, Vitamin contents of PFF were vitamin A 0.12, vitamin B$_1$ 0.31, vitamin B$_2$ 0.68, vitamin C 7.99, vitamin E 316.88 and vitamin D$_3$ 0.29 mg%. PFF will have a good anti-aging effect due to the high content of vitamin C and E.

  • The agar diffustion method using Escherichia coli was investigated for determination of Vitamin $B_{l2}$ instead of turbidimetric method using Lactobacillus leichannii (USP XXII method). The turbidimetric method is difficult to control the test organism and it has complicated procedure. From this study, it was found that the agar diffusion method on the determination of Vitamin $B_{12}$ in pharamaceutical preparation is simple and convenient as compared with turbidimetric method. Also we found that the coefficient of variation in reproducibility and the standard error in recovery were 2.18% and 1.83%, respectively.

The Effects of Environmental Conditions on the Reduction Rate of TNT by $Fe^0$ (환경요인이 $Fe^0$에 의한 TNT의 환원 반응속도에 미치는 영향)

  • The effects of environmental conditions, initial dissolved oxygen concentrations, pH, and the presence of electron carrier vitamin $B_{12}$, on the reduction rate of Tn by $Fe^0$ was quantitatively analyzed using a batch reactor In all experiments, TNT reduction was best described with a first order reaction and the reduction rate decreased with the increase in the initial DO concentration. However, the specific reaction rate did not decrease linearly with the increase in the initial DO concentration. In the presence of HEPES buffer 0.2 and 2.0 mM(pH 5.7$\pm$0.2), the specific reaction rate increased more than 5.8 times, which showed reduction rate is rather significantly influenced by the pH of the solution. To test the possibility of reaction rate enhancement, well-known electron carrier(or mediator) , vitamin $B_{12}$, has augmented besides $Fe^0$. In the presence of 8.0 $mu\textrm{g}$/L of vitamin $B_{12}$, the specific reaction rate increased as much as 14.6 times. The results indicate that the addition of trace amount of vitamin $B_{12}$ can be a promising rate controlling option for the removal of organics using a $Fe^0$ filled permeable reactive barrier.
