  • This research was conducted to investigate the incidence of viral infection in Samcheok-shi and Jeongseon-gun, main garlic growing areas of Gangwon province in Korea. Four primers sets (GLV, LYSV, GCLV, OYDV) were used for RT-PCR to test of 100 samples collected from each location. Infection rates of GLV, LYSV, GCLV and OYDV of garlic samples from Samcheok-shi and Jeongseon-gun were 95, 95, 92 and 33%, respectively. All garlic samples tested in this research were infected at least one virus. Coinfection of 3 kinds of virus (GLV, LYSV and GCLV) and 4 kinds of virus (GLV, LYSV, GCLV and OYDV) were 60 and 25%, respectively. While infections of GLV, LYSV and GCLV were evenly found in both two regions, OYDV infection of garlic samples collected from Jeongseon-gun was higher than Samcheok-shi. Viral infections at garlic fields of whole country and Gangwon province are very serious now. Therefore, it is very necessary to develop technic for virus-free bulb and renew seed bulb of garlic.

  • The lily symptomless virus (LSV) is the most common virus in Korean native lilies and causes various types of damage to overall plant growth. This study was carried out to investigate the elimination rate of the LSV by the in vitro scale culture (scaling) method in Korean native lilies and to test reinfection rates of the LSV under several field culture conditions of bulb production. Four Korean native lilies (Lilium dauricum, L. distichum, L. lancifolium, and L. maximowitzii) were used and their scales were cultured in vitro for micro-scale formation. The micro-scales were subcultured repeatedly using MS culture medium supplemented with 30 or $90g{\cdot}L^{-1}$ sucrose. The culture conditions were $24{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD with 16 hour daylength using fluorescent lamps and maintained at $22{\pm}2^{\circ}C$. The virus-free bulblets were grown for one to three years in the greenhouse and transplanted to the field in October or March. Virus infection rates were investigated by direct tissue blotting immunobinding assays and measurement of chlorophyll and protein contents. Virus-free plants could be obtained from the 5th subculture of micro-scales in L. lancifolium and L. maximowitzii or from primary culture in L. dauricum and L. distichum. LSV-free plants were reinfected during bulb production in the field. Reinfection rates were higher at older bulb ages and under higher planting density. The plants planted in October and at inland Gyeongsan had higher infection rates than those planted in March and at coastal area Pohang. The reinfection rate of L. maximowitzii was higher than those of L. dauricum and L. lancifolium. The LSV-infected plants had lower chlorophyll contents and unchanged protein contents compared to virus-free plants.

  • Most orchids are propagated by tissue culture. To survey the viral infection of tissue cultured Orchids, total RNA was extracted from in vitro Cymbridium and Phalaenopsis spp. collected from companies producing tissue-cultured orchids, and RT-PCR analysis was conducted with primer pairs specific to Cymbidium mosaic virus (CymMV) and Odontoglossum ring spot virus(ORSV), which are infecting wide range of orchid genera. The bulb size of Cymbidium infected with CymMV and ORSV was compared with healthy one at 10 months after planting in vitro orchids in the glasshouse. The CymMV or ORSV infection in 97 Cymbidium and 55 Phalaenopsis plants was 84.5 and 89.1 %, respectively. Mixed infection was found in 52.6 and 47.3% of Cymbidium and Phalaenopsis tested, whereas virus-free orchids were 15.5 and 10.9%, respectively. The CymMV and ORSV reduced the bulb size by 2.7-50% depending on the cultivars of Cymbidium. The both viruses caused yellowing, mottle and mosaic with or without necrosis in 4 Cymbidium cultivars.

  • The experiment was conducted to culture callus tissue induced from foliage leaf of garlic bulb for the production of virus-free stocks and for the reduction of expenses for seeds, The following results were reached. 1. Linsmaier-Skoog basal medium containing 2,4-dichlorophenoxyacetic acid (2,4-D) $10^{-5}M$ and benzyladenine $10^{-5}M$ showed the most effective for the induction for the induction of garlic callus. 2. The growth rate of callus was the highest in Linsmaier-Skoog basal medium containing kinetin $10^{-6}M\;and\;2,4-D\;10^{-6}M$ 3. The results of periodical assay of virus concentration in callus tissues showed that virus was almost eliminated by repeated transfer of translucent and soft tissue for eight generations. 4. When virus-free garlic callus tissues were transfered to Murashige-Skoog basal medium containing kinetin $10^{-5}M$ and naphthaleneacetic acid $5\times10^{-6}M$, the tissues were redifferentiated and formed plantlet.

  • In vitro culture of shoot tip of garlic (Allium sativium L. cv. Seosan) was carried out to find medium condition of the induction of multiple shoots and bulbing for muliproduction of virus-free seed bulbs. For this work, tank culture was introduced. In shoot tip culture on MS solid medium the induction of multiple shoots and bulbing were better by adding 3% sucrose than 8%. Supplementation with 2mg/L 2ip and 0.2 mg/L IAA in this medium was effective. Three point three shoots including 2.7 bulbs were formed from a shoot tip after cultivation for 30 days on this medium. Bulbing of garlic in liquid culture with plastic water tank of 20L supplied air at the side of the lower part was better by adding 3% sucrose than 8% by subculture for 45 days with shoots obtained from shoot tip culture for 30 days on soid MS medium. Shoot growth was vigorous at 3% sucrose however bulb growth was more effective on the medium of 8% sucrose. Because of the effectiveness on solid medium added 3% sucrose, 2 mg/L 2ip and 0.2 mg/L IAA for initial production of multi-shoot in stem tip culture and the effectiveness in liquid culture with water tank for growth of bulbs, the method of two-step culture could be introduced for the multiple production of seed bulb of high quality. It was more desirable by supply of 0.2 mg/L BA and 0.02 mg/L NAA at tank culture time. But growth of the bulbs became poor by increasing concentration of NAA of the medium.

  • This study was conducted to investigate the effect of bulblet size on growth and bulb enlargement of Lilium Oriental hybrids, such as 'Sorbonne', 'Siberia', and 'Casa Blanca', grown in areas of high altitude (780 m), in 2009. The bulblets were oriented from tissue culture and planted to produce virus-free bulblets. All the three cultivars showed high vitality, with more than 93% producing shoots, even from small bulblets with a bulb circumference (BC) of less than 6 cm. Bulblets with BC 9-10 cm showed an increased height and number of leaves rapidly to induce the phase conversation from vegetative growth to reproductive growth. The bulblets of BC 9-10 cm in 'Sorbonne', 'Siberia', and 'Casa Blanca' had 2.0, 2.9, and 2.8 flower bud sets per plant, respectively. However, the flowers from those bulblets were not adequate as standard bulbs to produce cut-flowers for the domestic market. The larger bulblets showed more prosperous characteristics in growth and development of Lilium Oriental hybrids. Small bulblets had a high occurrence of viruses and leaf blight symptoms during cultivation, indicating the aggravated disease symptoms in the previously infected bulbs. 'Sorbonne' cultivars showed a high rate of enlargement of bulbs, and small bulblets under BC 6 cm produced more than 23% of the standard bulbs of BC 14-16 cm, which could produce cut-flowers for export. However, 'Casa Blanca' bulblets with BC 6-9 cm produced low bulbs of BC 14-16 cm at 21% level. 'Casa Blanca' cultivars had more bulb roots, longer root lengths, and higher weights than 'Sorbonne' and 'Siberia' cultivars. 'Casa Blanca' cultivars with BC over 9-10 cm grew rapidly and produced a high quantity of bulbs. In these results, 'Sorbonne' bulblets of BC 9-10, 'Siberia' bulblets of BC 10-12 cm, and 'Casa Blanca' bulblets of BC 10-12 cm produced 89%, 86%, and 93% of the standard bulbs of BC 14-16 cm, respectively. It is recommended that bulblets larger than the above sizes be used to produce cut-flowers for the export market. Experiment results suggest that production of bulbs larger than BC 18 cm requires bulblets that are larger than BC 12-14 cm of the three cultivars for the highest quality cut-flowers.

  • Since garlics (Allium sativum L.) are propagated through cloves, infection by virus or other pathogens may become severe problem if not using high quality seed bulbs every year resulting in the reduction of yield and bulb quality, In order to solve this problem, the establishment of virus-free bulb production and its supply system have been required because no chemicals were found to eliminate viruses from seed bulbs. This experiment was conducted to develop an effective production technique of high quality seed bulbs using shoot-tip culture. Over 90% of shoot-tips explanted on January L 1990 were survived at constant temperature of either 20, 24 or 28$^{\circ}C$, wheres 88% at alternate temperature (28/20$^{\circ}C$). The growth of shoot and root was most vigorous at constant 24$^{\circ}C$, and least at alternate temperature (28/20$^{\circ}C$) condition. When shoot-tips were explanted June 21 to August 1,1991, survival and growth of shoot-tips was most vigorous on MS medium supplymented with 0.1 mg/L NAA and 2 mg/L kinetin and least 1 mg/L Gh$_3$. The shoot-tips taken from the seed bulbs stored at 4$^{\circ}C$ for 15 to 60 days were placed on MS medium, shoot growth and in vitro bulblet formation increased slightly as affected by the increase of told treatment period at 4$^{\circ}C$.
