Identification of the Capsid Protein-binding Region of the SL1(+) RNA Located at the 5' Region of the Potato virus X Genome

  • $Potato$ $virus$ $X$ (PVX) contains $cis$-acting elements including stem-loop 1 (SL1) RNA at the 5' region; SL1 is conserved among all potexviruses. The SL1 at the positive-sense RNA, SL1(+), is required for PVX RNA replication, cell-to-cell movement, and translation. Previous research demonstrated that SL1(+) RNA also serves as the origin of assembly for encapsidation of PVX RNA. To identify the essential sequences and/or regions for capsid protein (CP) subunit recognition within SL1(+) RNA, we used electrophoretic mobility shift assays (EMSA), UV cross-linking, and yeast three-hybrid analyses. The EMSA and UV cross-linking analyses with PVX CP subunits and RNA transcripts corresponding to the SL1(+) RNA showed that the SL1(+) RNA formed complexes with CP subunits. We also conducted EMSA and yeast three-hybrid analyses with RNAs containing various mutations of SL1(+) RNA elements. These analyses indicated that SL1(+) RNA is required for the interaction with PVX CP and that the RNA sequences located at the loop C and tetra loop of the SL1(+) are crucial for CP binding. These results indicate that, in addition to being important for RNA accumulation, the SL1(+) RNA from the 5' region of the PVX genome is also required for specific binding of PVX CP.

Phylogenetic Analysis of New Isolates of Cucumber mosaic virus from Iran on the Basis of Different Genomic Regions

  • Molecular characterization of Cucumber mosaic virus (CMV) was done by using samples from tomato and cucurbitaceous plants collected from different locations in the northwest region of Iran. After screening by enzyme-linked immunosorbent assay, 91 CMV-infected samples were identified. Biological properties of eight representative isolates were compared with each other revealing two distinct phenotypes on squash and tomato plants. Phylogenetic analyses based on nucleotide sequences of the coat protein (CP), movement protein (MP) and 2b of the new isolates, together with that of previously reported isolates, led to the placement of the Iranian isolates in subgroups IA and IB according to CP and MP genes, but in subgroup IA according to the 2b gene. These data suggest that reassortment may have been a major event in the evolution of CMV in Iran, and that the Iranian isolates are derived from a common recent ancestor that had passed through a bottleneck event.

  • 본 논문에서는 COVID-19와 같은 위급한 상황에서 바이러스 검사를 빠르게 진행하기 위한 K-d 트리 기반의 그래프 구축과 환자 이동 경로 시스템을 제안한다. 가상환경에서 활용되는 대표적인 길 찾기 알고리즘은 A*나 NavMesh 자료구조는 정해진 정적 이동 경로만을 안내하려는 방법이기 때문에 가상환경에서 NPC를 제어할 때는 효율적이지만, 실제 환경에 적용하여 문제를 풀기에는 충분하지 않다. 특히, 빠른 바이러스 검사를 받기 위해서는 짧은 거리만을 이용하는 게 아닌, 실제 도로 교통상황, 병원의 크기, 환자 이동 수, 환자 처리 시간 등 고려해야 할 상황들이 많다. 본 논문에서는 위에서 언급한 다양한 속성들과 이를 이용한 최적화 함수를 모델링하여, 실제 도시 맵에서 바이러스 검사를 빠르고 효율적으로 제어할 수 있는 프레임워크를 제안한다.

명월초에서 분리한 오이모자이크바이러스의 감염 첫 보고 (First Report of Cucumber mosaic virus Isolated from Sambungai (Gynura procumbens))

  • 2016년 3월, 모자이크와 황화 병징을 보이는 명월초 잎에서 CMV를 분리하여 CMV-Gyp로 명명하였다. 기주식물의 생물적 반응과 RT-PCR, PCR-RFLP, 이동단백질 및 외피단백질 유전자의 염기서열 분석을 통해 CMV를 동정하였다. 가지과(담배, 고추, 꽈리)와 비름과(흰명아주, 붉은명아주)에서는 전형적인 CMV의 병징이 나타났으나 박과(쥬키니호박, 오이)에서는 병징이 발현되지 않는 특성을 보였다. 3a 및 CP 유전자의 계통학적 분석 결과, CMV-Gyp가 CMV Subgroup II에 속하는 것으로 나타냈다. CMV-Gyp의 이동단백질과 외피단백질유전자의 서열은 아미노산 수준에서 CMV-Hnt와 99.3% 및 100% 일치하였다. 이 논문은 명월초에서 CMV 감염에 대한 첫 보고이다.

오돈토글로썸 윤문 바이러스 Cy계통 게놈 RNA의 cDNA 구축 및 유전자 크로닝 (Construction of Complementary DNA Library and cDNA Cloning for Cy Strain of Odontoglossum Ringspot Virus Genomic RNA)

  • Genomic RNA was extracted from Cy strain of odontoglossum ringspot tobamovirus (ORSV-Cy) isolated from infected leaves of tobacco cv. Samsun. Size of the genomic RNA was about 6.6 kb in length. The genomic RNA was fractionated using Sephadex G-50 column chromatography into 2 fractions. They were polyadenylated at their 3'-end using E. coli poly(A) polymerase. Polyadenylated viral RNA was recovered by oligo (dT) primer adapter containing NotI restriction site and Moloney murine leukemia virus SuperScript reverse transcriptase (RNase H-). Second-strand cDNA was synthesized by using E. coli DNA ligase, E. coli DNA polymerase I and E. coli RNase H. Recombinant plasmids containing cDNAs for ORSV-Cy RNA ranged from about 800 bp to 3,000 bp. Among the selected 238 recombinants, pORCY-124 clone was the largest one covering 3'-terminal half of the viral RNA. This clone contained two restriction sites for EcoRI and XbaI and one site for AccI, AvaI, BglII, BstXI, HindIII, PstI, and TthIII 1. respectively. The clone contained partial viral replicase, a full-length movement protein and a complete coat protein genes followed by a 3' untranslated region of 414 nucleotides based on restriction mapping and nucleotide sequencing analyses. Clones pORCY-028, -068, -072, -187 and -224 were overlapped with the pORCY-124. Clones pORCY-014 and -095 covered 5' half upstream from the middle region of the viral RNA, which was estimated based on restriction mapping and partial sequence analysis. Constructed cDNA library covered more than 90% of the viral genome.

국내 분리 오제스키병 바이러스 접종자돈의 병리학적 관찰 (Pathological observation on the piglets experimentally infected with Aujeszky′s disease virus isolated in Korea)

  • Pathological studies were performed on the five piglets experimentally infected with Aujeszky's disease virus(pseudorabies), NYJ isolate, isolated from the naturally infected pigs in Korea: two piglets were inoculated intramuscularly, two piglets intranasally, and one piglet subcutaneously at the dose of 1$m\ell$ per animal with the 105.5 $TCID_50$/0.1ml titer. Clinical signs included dyspnea, high fever(>$41^{\circ}C$), anorexia, vomiting, diarrhea or constipation, ataxia, circling movement, posterior paralysis, intermittent convulsion, and coma followed by death although some variations by age and inoculated routes were observed. Gross features included multiple necrotic foci in the liver, congestion and hemorrhage in the lymph nodes and spleen, petechial hemorrhage in the kidney, hemorrhagic pneumonia, marked meningeal congestion, severe sub meningeal hemorrhage in the spinal cord, excessive cerebrospinal fluid retention, and muscular necrosis at the inoculated area. Microscopically, non suppurative meningoencephalitis with gliosis and perivascular cuffing in CNS, ganglioneuritis, necrohemorrhagic splenitis, necrotic hepatitis, tonsillitis and rhinitis, hemorrhagic or interstitial pneumonia, and non-suppurative myositis in the injected area were observed. Eosinophilic intranuclear inclusion bodies were found in a variety of tissues the including the liver, kidney, adrenal gland, spleen, lymph nodes, tonsil, and lung. Ultrastructurally, virus particles were confirmed in nucleus and cytoplasms of pneumocytes around the necrotic areas.

Comparative Analyses of Tomato yellow leaf curl virus C4 Protein-Interacting Host Proteins in Healthy and Infected Tomato Tissues

  • Tomato yellow leaf curl virus (TYLCV), a member of the genus Begomovirus, is one of the most important viruses of cultivated tomatoes worldwide, mainly causing yellowing and curling of leaves with stunting in plants. TYLCV causes severe problems in sub-tropical and tropical countries, as well as in Korea. However, the mechanism of TYLCV infection remains unclear, although the function of each viral component has been identified. TYLCV C4 codes for a small protein involved in various cellular functions, including symptom determination, gene silencing, viral movement, and induction of the plant defense response. In this study, through yeast-two hybrid screenings, we identified TYLCV C4-interacting host proteins from both healthy and symptom-exhibiting tomato tissues, to determine the role of TYLCV C4 proteins in the infection processes. Comparative analyses of 28 proteins from healthy tissues and 36 from infected tissues showing interactions with TYLCV C4 indicated that TYLCV C4 mainly interacts with host proteins involved in translation, ubiquitination, and plant defense, and most interacting proteins differed between the two tissues but belong to similar molecular functional categories. Four proteins-two ribosomal proteins, S-adenosyl-L-homocysteine hydrolase, and 14-3-3 family protein-were detected in both tissues. Furthermore, the identified proteins in symptom-exhibiting tissues showed greater involvement in plant defenses. Some are key regulators, such as receptor-like kinases and pathogenesis-related proteins, of plant defenses. Thus, TYLCV C4 may contribute to the suppression of host defense during TYLCV infection and be involved in ubiquitination for viral infection.

New Normal and Business Sustainability in the Age of Global pandemic

  • Purpose - This study examines the awareness regarding the symptoms of COVID-19 exposures and safety distances strategies whether they were useful to resile the businesses a mid of the pandemic. Besides exploring the awareness and safety distances, the effectiveness of offering free use of protective equipment (mask, hand sanitizer, frequent hand washing, etc.) to the customers for visiting the business centers was also examined. Design/methodology/approach - This study collected 264 survey data in Bangladesh which is one of the most densely populated country and very vulnerable for COVID-19 due to its socio-economic condition. The multiple regression analysis is used to analyze the data. Findings - The findings of the study indicate that the awareness about the symptoms of virus exposures (cough, fever, diarrhea, and weakness) has significant affirmative effects to enhance the public movement for business purposes with the lower possibility to be affected by the virus. The study also indicates that safety distances and protective equipment can mediate the significant positive relationship between the awareness of the disease and the businesses' resilient capacity. Research implications or Originality - COVID-19, as an apprehensive health issue in the current world, has sharpened the uncertainty of the businesses. One essential technique as lockdown, has been followed by almost every country to protect the transmission of the virus even though the scholars criticized it due to the substantial adverse effects on the country's economy. Under this circumstances, this study provides implications to the relevant businesses by assessing the nexus between the safety distances and the proper uses of protective equipment with the business resilient.

칸나에서 분리한 Cucumber mosaic virus의 특성 (Characterization of an Isolate of Cucumber mosaic virus Isolated from Canna generalis Bailey)

  • 전형적인 줄무늬 모자이크 증상을 나타낸 칸나로부터 Cucumber mosaic virus(CMV)의 한 계통(Can-CMV)을 분리하고, 특성을 조사하였다. Can-CMV는 대부분의 전신감염 기주에서 병징이 발현되지 않았고, 이들 기주의 접종엽 및 상엽에서 RT-PCR로 바이러스의 감염여부를 조사한 결과, N. benthamiana와 N. glutinosa에서만 바이러스가 검출되었으며, 다른 기주로부터는 확인되지 않았다. 한편 Chenopodium amaranticolor의 접종엽에 발현된 국부 괴사병반은 대조로 접종한 Fny-CMV나 LS-CMV에 의해서 형성된 병반보다 매우 작은 크기의 반점을 형성하는 특성을 보였다. 또한 Vigna unguiculata의 접종엽에는 Fny-CMV나 LS-CMV가 접종 2-3일 후에 뚜렷한 괴사병반을 형성하는데 반해서, Can-CMV는 접종 4-5일 후에 윤곽이 확실치 않은 퇴록병반을 형성하였다. Can-CMV에 감염된 N. benthamiana로부터 추출한 dsRNA는 4종의 밴드가 검출되었으며, 이들 분자의 크기 및 종수는 Fny-CMV나 LS-CMV와 차이를 나타내지 않았다. Can-CMV의 항원은 Fny-CMV의 항혈청에 대해서 한 종의 뚜렷한 침강선을 형성하였으며, Fny-CMV의 항원에 의해서 형성된 침강선과 융합하였고, LS-CMV의 침강선과는 분지선을 형성함으로서, 혈청학적으로 서브그룹 I에 속하는 계통으로 판단되었다. 또한 Can-CMV에 감염된 N. benthamiana로부터 RNA를 추출하여 CMV-specific 프라이머를 이용한 CMV-RNA3의 외피단백질 유전자를 포함하는 3'영역에 대해서 RT-PCR을 실시하였다. Can-CMV는 Fny-CMV나 LS-CMV와 마찬가지로 예상되었던 약 950bp크기의 cDNA가 증폭되었으며, 이 cDNA를 EcoRI으로 처리하였을 때에는 절단되지 않았고, MspI에서는 595bp 및 350bp의 절편으로 절단되었다. 이와 같은 결과는 Fny-CMV의 패턴과 일치하였으며, 이러한 결과로부터 Can-CMV가 서브그룹 IA에 속하는 계통으로 확인되었다. 이상과 같은 결과들로부터, Can-CMV는 앞으로 바이러스와 기주의 다양한 상호관계를 이해하는데 있어서 중요한 병원학적 성질을 가지고 있는 것으로 생각되었다.

Tomato spotted wilt virus Isolates Giving Different Infection in Commercial Capsicum annuum Cultivars

  • $Tomato$ $spotted$ $wilt$ $virus$ (TSWV)-infected $Capsicum$ $annuum$ plants were collected from open fields during June to July in 2010. The TSWV isolates were designated as Gneung, Ghang-Kjj, Gchang-Njc, Ghae, and Pap. The nucleotide sequence of the nucleocapsid protein (N) and movement protein (NSm) of the five isolates was determined. The pathogenicity of the five isolates was determined on 14 $C.$ $annuum$ cultivars two times by using mechanical inoculation. The five isolates induced different response: Both Gneung and Gchang-Kjj did not infect any of the cultivars in the 2nd trial, while Gchang-Njc, Ghae and Pap infected 11, 6 and 13 of 14 cultivars, respectively. The five isolates also were tested on $Solanum$ $lycopersicum$ breeding line TGC09-71 and three $Nicotiana$ species. $S.$ $lycopersicum$ showed a similar response to the five isolates as did $C.$ $annuum$. Both Gchang-Njc and Ghae infected systemically all three $Nicotiana$ species tested. While both Pap and Gneung did not infect any of the $Nicotiana$ species tested. In conclusion, five TSWV isolates induced different infection spectra in $C.$ $annuum$ cultivars, $Nicotiana$ species and an $S.$ $lycopersicum$ breeding line. Amino acid sequence analysis of the NSm gene could not support or explain the different infection spectra of the five isolates. This study indicated that various isolates must be used as virus inocula for evaluation of $C.$ $annuum$ and $S.$ $lycopersicum$ cultivars in breeding programs for TSWV resistance.
