사슴에서의 구제역 증상과 전파 가능성 (Clinical sign and transmission of foot-and-mouth disease in deer, Review)

  • Foot-and-mouth disease (FMD) commonly infects cloven-hoofed livestock animals such as cattle, pig, sheep, and goat and its clinical signs are well-known. Besides livestock, FMD can be transmitted among cloven-hoofed animals in the wild. FMD mostly affects livestock animals in farms, but, wild animals are likely to play a pivotal role in spreading the disease due to their way of free living. In the case of deer, the clinical signs of FMD vary widely from subclinical to severe infections. Thus, in some deer species, it may be hard to verify clinical signs of FMD. A deer may carry the virus up to 11 weeks after exposure, shedding the virus during the period. However, deer is not considered as a typical host for persistent infection like buffalo, cattle or sheep. In Korea, small-scale livestock farms which have less than 10 animals make up 63.6% of the entire livestock farms. Considering raising environment in deer farms, it is assumed that the risk of virus excretion and consequent transmission of FMD among deers is relatively lower than other cloven-hoofed animals. However, Sika deer and Elk which are typical deer species in Korea would manifest mild to subclinical symptoms upon FMD infection. Therefore, laboratory testing is necessary to confirm FMD in these animals because of difficulty in verifying clinical signs and the risk of virus shedding during inapparent infection.

Differentiation of Lymphocystis Disease Virus Genotype by Multiplex PCR

  • Lymphocystis disease virus (LCDV) is the causative agent of lymphocystis disease. The viruses have been divided into three genotypes (genotype I for LCDV-1, II for Japanese flounder isolates, and III for rockfish isolates) on the basis of major capsid protein (MCP) gene sequences. In this study, we developed a multiplex PCR primer set in order to distinguish these genotypes. We also analyzed the MCP gene of a new LCDV isolate from the sea bass (SB98Yosu). Comparison of sequence identities between SB98Yosu and eight Japanese flounder isolates, revealed identity of more than 90.1 % at nucleotide level and 96.5% at deduced amino acid level, respectively. Phylogenetic analyses based on the MCP gene showed that SB98Yosu belongs to genotype II, along with Japanese flounder isolates. Multiplex PCR based on the MCP gene allowed us to identify these genotypes in a simple and rapid manner, even in a sample that contained two genotypes, in this case genotypes II and III.

Implementing Expanded Rapid Human Immunodeficiency Virus Testing in Public Health Centers in Seoul, 2015

  • In 2015, rapid human immunodeficiency virus (HIV) testing was implemented in all 25 public health centers in Seoul. During March and December 2015, 20,987 rapid HIV tests were performed, of which 116 (0.5%) were positive. Compared to those of the period before application of the rapid HIV test in place of conventional enzyme immunoassay method, the number of HIV tests performed and the number of positive results increased by sevenfold and twofold, respectively. In conclusion, expansion of the provision of rapid HIV tests in public health centers increased the number of voluntary HIV tests.

Outbreak of Cucumber mosaic virus and Tomato spotted wilt virus on Bell Pepper Grown in Jeonnam Province in Korea

  • In August 2006, a severe disease incidence showing mosaic and/or necrotic symptoms on two bell pepper varieties including red-colored 'Special' and yellow-colored 'Fiesta' was observed in a greenhouse located in Gwangyang, Jeonnam province, Korea. To identify causal viruses, total RNAs were extracted from 11 fruit samples with and without symptoms. Specific oligonucleotide primers for Cucumber mosaic virus (CMV), Pepper mottle virus (PepMoV), Tomato spotted wilt virus (TSWV) and Pepper mild mottle virus (PMMoV) were designed based on the sequences available on GenBank. Database comparisons of the deduced amino acid sequences of each sequence produced 100% and 98% matches with nucleocapsid protein gene of TSWV (Acc. No. ABE11605) and coat protein gene of CMV (Acc. No. DQ018289), respectively, suggesting that the symptoms on bell pepper fruits might be caused by the infection of CMV and TSWV. To our knowledge this is the first report of necrotic as well as mosaic virus disease on bell pepper fruits by the infection of CMV and TSWV in Jeonnam province, Korea.

Production and characterization of monoclonal antibodies against an avian influenza virus (H9N2)

  • In this report, fifteen monoclonal antibodies (MAbs) against an avian influenza virus (H9N2 subtype) were newly produced and characterized. These MAbs proved to react to the epitopes of nucleocapsid protein (NP), hemagglutinin (HA), neuraminidase (NA) and non-structural protein 1 (NS1) of Korean H9N2 strain, respectively. Two HA-specific MAbs showed the ability to inhibit the hemagglutination activity of H9N2 subtype avian influenza virus when tested by hemagglutination inhibition (HI) assay. All MAbs did not cross-react with other avian-origin viruses (Newcastle disease virus, infectious bursal disease virus, infectious bronchitis virus and avian rotavirus) by immunofluorescence test or enzyme-linked immunosorbent assay. The MAbs produced in this study could be useful as the materials for diagnostics and therapeutics against Korean-lineage H9N2 virus infections.

ADV 국내 분리주를 접종한 실험 동물의 조직 병리 소견과 항체가 연구 (Histopathoiogical Findings and Antibody Titers of the Experimental Animals Infected with Aujeszky′s Disease Virus(Korea Isolates))

  • We surveyed the antibody titers in the slaughtered pigs by enzyme immunodiffusion method for the epidemiological distribution of Aujeszky's disease in Korea. And also we observed the clinical symptoms, histopathological findings and antibody titers by serologic neutralization test in experimental animals infected with Aujeszky's disease virus(ADV) isolated from Korea. The results of the experiments were summarized as follows. 1. We detected 2 pigs infected with ADV of 1000 in 1990 and 1 of 600 in 1991 by enzyme immunodiffusion method. 2. In histopathological findings of experimental animals inoculated ADV, the typical characteristics of Aujeszky's disease were not observed in pig, but edema and degeneration in the epidermis were observed in rats, vasculoendotheriosis, perivascular cuffing and cellular infiltration in the cerebrum were appeared in rabbits and perivascular cuffing and forcal infiltration of glial cells in the cerebrum were appeared in mice. 3. The increasing antibody titers(1 : 64) in the serological neutralization test were ascertained in 2 carrier pigs inoculated ADV. 4. Rabbits, mice and rats died all about 5 days after inoculation, but pigs and chickens didn't die. 5. In 1 cat and 2 rabbits inoculated with the Aujeszky's disease virus(ADV), the typical clinical findings of Aujeszky's disease were observed in rabbit, but not in cat and so we slaughtered without any signs of Aujeszky's disease.

Pathological findings of the mixed infection with canine distemper virus and Streptococcus canis on farmed badger

  • Herein, we report a case of badgers showing high morbidity and mortality rate due to the mixed infection of canine distemper virus (CDV) and Streptococcus canis (S. canis) in a farm where wild animal, badger, is being reared for herbal medicine. During the period of about one month, 120 out of 320 badgers showed severe respiratory symptoms and died, and 3 bodies were submitted to the Animal and Plant Quarantine Agency for disease diagnosis. The lung with the most severe necropsy findings failed to collapse and showed dark reddening and had yellowish nodules on the cut surface. The characteristic and common histopathologic findings include multifocal necrosis with hemorrhage of the lung, severe lymphoid depletion of the spleen and intracytoplasmic or intranuclear inclusion bodies in almost all organs. Finally, CDV and S. canis were identified by immunohistochemistry and bacterial isolation, respectively. This is the first mixed infection case of CDV and S. canis in badgers being raised on the farm.

Lumpy skin disease as an emerging infectious disease

  • Lumpy skin disease (LSD) is one of the most important emerging transboundary diseases. Recently, LSD has emerged in many countries in the northern hemisphere. The LSD virus has a huge genome and is highly resistant to environmental conditions. The virus is also host-specific and large ruminants, such as cattle and domestic water buffalo, are particularly susceptible. In addition, wild ruminants can serve as potential reservoirs for spreading the LSD virus. The emergence might be related to climate change in various regions because LSD is an arthropod-borne infectious disease. This disease causes enormous economic losses, such as leather damage, decreased milk production, abortion, and death in infected ruminants. The economic importance of LSD in the bovine industry has forced countries to develop and implement control strategies against the disease. With the recent global spread and the economic impact, LSD will be discussed intensively. In addition, effective preventive measures are suggested based on the presence or absence of LSD outbreaks.

Molecular Identification and Genetic Diversity Analysis of Papaya Leaf Curl China Virus Infecting Ageratum conyzoides

  • Papaya leaf curl China virus (PaLCuCNV) is a damaging plant pathogen causing substantial losses to crop. The complete genomes of three PaLCuCNV isolates from Ageratum conyzoides were obtained and combined with the 68 reference isolates in GenBank for comprehensive genetic diversity analyses using specialized computational tools. Sequence alignment revealed nucleotide sequence similarity ranging from 85.3% to 99.9% among 71 PaLCuCNV isolates. Employing phylogenetic analysis, 71 PaLCuCNV sequences were clustered into five groups, with no significant correlation observed between genetic differentiation and either host species or geographical origin. Additionally, 13 recombination events across all PaLCuCNV isolates were identified. Genetic diversity analysis indicated the ongoing expansion and evolution of PaLCuCNV populations, supported by a neutral model. Moreover, significant genetic differentiation was observed among distinct viral populations, primarily attributed to genetic drift. Overall, our findings provide valuable insights into the detection, genetic variation, and evolutionary dynamics of PaLCuCNV.

Production of Newcastle Disease Virus by Vero Cell Culture

  • Newcastle disease virus (NDV) vaccines were produced from Vero cells by using lively attenuated virus strain. The MOI of 0.1.' serum concentration of 2%. initial pH of 8.0. and infection time of 3 days were found to be optimum conditions for vaccine production. The treatment of polycation enhanced the virus production. When ascorbic acid was added as an antioxidant, NDV production was also enhanced. Utilization of $CaCl_2$ showed an inhibitory effect on the propagation of NDV. It was also found the ammonium ion concentration higher than 4mM inhibited virus production. Thus ammonium ion removal system was tried for the efficient production of NDV vaccine.
