IHNV (Infectious Hematopoietic Necrosis Virus): 과거, 현재, 그리고 미래 (INHV (Infectious Hematopoietic Necrosis Virus): Past, Present and Future)

  • A global increase in fish consumption has led to a rapid expansion of aquaculture production, which has been linked to enhancing the spread of infectious diseases. Viral diseases can cause high mortality in many cultured fish species, posing a serious threat to the aquaculture industry. Infectious hematopoietic necrosis virus (IHNV) is one of the primary threats to aquacultured salmonid species, causing huge economic losses. Since the first report in cultured sockeye salmon Oncorhynchus nerka during the 1950s in North America, IHNV has spread to other regions, including Europe, Asia, South America, and Africa by transportation of infected fish and eggs, causing disease and increasing mortality in a wide variety of salmonid species. Here, we review existing information relevant to IHNV: its phylogenetic characteristics, origin, infection history, virulence determinants, susceptible hosts, vectors, and vaccine development. This review also addresses a possible cross-species transmission of IHNV to a new host, olive flounder Paralichthys olivaceus, a cultured fish of economic importance in East Asian countries.

Production of virus-like particles of nervous necrosis virus displaying partial VHSV's glycoprotein at surface and encapsulating DNA vaccine plasmids

  • In order to use nervous necrosis virus (NNV) virus-like particles (VLPs) as a delivery tool for heterologous antigens or plasmids, we attempted to produce red-spotted grouper nervous necrosis virus (RGNNV) VLPs displaying a partial region of viral hemorrhagic septicemia virus (VHSV) glycoprotein at the surface and VLPs that are harboring DNA vaccine plasmids within the VLP. A peptide encoding 105 amino acids of VHSV glycoprotein was genetically inserted in the loop region of NNV capsid gene, and VLPs expressing the partial part of VHSV glycoprotein were successfully produced. However, in the transmission electron microscope analysis, the shape and size of the partial VHSV glycoprotein-expressing NNV VLPs were irregular and variable, respectively, indicating that the normal assembly of capsid proteins was inhibited by the relatively long foreign peptide (105 aa) on the loop region. To encapsulate by simultaneous transformation with both NNV capsid gene expressing plasmids and DNA vaccine plasmids (having an eGFP expressing cassette under the CMV promoter), NNV VLPs containing plasmids were produced. The encapsulation of plasmids in the NNV VLPs was demonstrated by PCR and cells exposed to the VLPs encapsulating DNA vaccine plasmids showed fluorescence. These results suggest that the encapsulation of plasmids in NNV VLPs can be done with a simple one-step process, excluding the process of disassembly-reassembly of VLPs, and NNV VLPs can be used as a delivery tool for DNA vaccine vectors.

Surveillance of African swine fever infection in wildlife and environmental samples in Gangwon-do

  • African swine fever (ASF) is fatal to domestic pigs and wild boars (Sus scrofa) and affects the domestic pig industry. ASF is transmitted directly through the secretions of infected domestic pigs or wild boars, an essential source of infection in disease transmission. ASFV is also very stable in the environment. Thus, the virus is detected in the surrounding environment where ASF-infected carcasses are found. In this study, ASF infection monitoring was conducted on the swab and whole blood samples from wild animals, various hematopoietic arthropod samples that could access infected wild boar carcasses or habitats to cause maintenance and spread of disease, and soil samples of wild boar habitats. ASF viral DNA detection was confirmed negative in 317 wildlife and environmental samples through a real-time polymerase chain reaction. However, ASF occurs in the wild boars and spreads throughout the Korean peninsula. Therefore, it is necessary to trace the route of ASF virus infection by a continuous vector. Additional monitoring of various samples with potential ASF infection is needed to help the epidemiologic investigation and disease prevention.

경기도 광주 여름재배 시설토마토의 가루이 매개 바이러스 TYLCV, ToCV 발생현황 (Patterns of the Occurrence of TYLCV and ToCV with Whitefly on Summer-Cultivated Tomato in Greenhouse in Gwangju, Gyeonggi Province)

  • 경기 광주시 완숙토마토 시설재배단지에서 가루이가 매개하는 바이러스 2종 tomato yellow leaf curl virus (TYLCV)와 tomato chlorosis virus (ToCV)의 발생양상을 조사하였다. 2020년 여름재배 작형에서 TYLCV와 ToCV의 감염률은 수확기에 급격히 증가하였고, 단독 감염률은 각각 30.9%, 5.0%였고, 복합감염률은 52.2%로 TYLCV 단독감염과 2종 바이러스 복합감염이 83.1%로 대부분을 차지하였다. 2종 바이러스를 매개하는 가루이의 발생분포 및 보독률을 조사한 결과, 작물 재배기간 내내 담배가루이가 높은 비율로 발생하였고, 조사한 담배가루이는 모두 Mediterranean (MED)으로 확인되었다. 담배가루이 MED는 TYLCV를 단독으로 보독하거나 ToCV와 복합 보독하는 경우가 48.5%로 많았으며, ToCV의 보독률은 29.1%로 온실가루이(7.2%)에 비해 높았다. 담배가루이 MED의 2종 바이러스 보독률이 토마토 정식 전부터 47.2%로 높았고, 이후 계속 증가하여 수확기에는 73.8%로 가장 높았다. 수확기 토마토 재배포장별 2종 바이러스의 감염률에 따라 수확량을 비교 분석해본 결과, TYLCV와 ToCV의 복합감염률 증가가 수확량을 감소시키는 것을 확인하였고, 복합감염률이 10% 증가함에 따라 405.4 kg/10a 수량감소를 보였다.

광고 글 필터링 모델 적용 및 성능 향상 방안 (Application of Advertisement Filtering Model and Method for its Performance Improvement)

  • 최근 기하급수적인 인터넷 데이터의 증가로 딥러닝 등의 많은 분야가 발전하였지만 바이럴 마케팅(viral marketing)과 같은 상업적 목적의 광고가 발견되면서 정보증가의 부작용이 발생하고 있다. 이는 양질의 정보를 공유하고자 하는 인터넷의 본질을 훼손하고 있을 뿐만 아니라 사용자는 양질의 정보를 습득하기 위해 검색시간이 증가하는 문제가 야기된다. 이에 본 연구에서는 광고(Ad: Advertisement, 이하 Ad) 글을 정보 전달의 본질을 흐리는 내용의 글이라 정의하였으며 본 정의에 부합하는 정보로 필터링하는 모델을 제안하였다. 제안하는 모델은 광고 필터링 경로와 광고 필터링 성능 개선경로로 구성되었으며 지속적으로 성능이 개선되도록 설계하였다. 광고 글 필터링을 위해 데이터를 수집하고 KorBERT를 사용하여 문서분류를 학습하였다. 본 모델의 성능을 검증하기 위해 실험을 진행하였으며 5개의 주제를 통합한 데이터에 대한 정확도(Accuracy), 정밀도(Precision)는 각각 89.2%, 84.3%의 결과를 나타냈고 광고의 비정형적 특성을 고려하더라도 높은 성능이 보임을 확인하였다. 본 모델을 통해 바이럴 마케팅으로 구성된 문서에서 광고 문단을 판단하고 필터링하여 사용자에게 양질의 정보를 효과적으로 전달하며 검색하는 과정에서 낭비되는 시간과 피로가 감소할 것으로 기대된다.

치즈 유청의 오이모자이크바이러스와 고추모틀바이러스 감염 억제 효과 (Inhibitory Effect of Cheese Whey on Cucumber Mosaic Virus and Pepper Mottle Virus in Capsicum annuum)

  • 치즈 제조공정에서 나오는 부산물인 유청이 진딧물에 의해 전염되는 오이모자이크바이러스(cucumber mosaic virus, CMV)와 고추모틀바이러스(pepper mottle virus, PepMoV)의 감염 억제효과가 있는지를 고추 '청양' 품종을 대상으로 조사하였다. 온실에서 유청 원액 및 원액을 물로 2-20배로 희석하여 고추 '청양' 품종에 분무한 후 복숭아혹진딧물을 이용하여 CMV-Vch를 접종한 결과, 원액을 처리한 실험 구의 감염률이 6.6%로, 대조로 물을 처리한 실험 구의 감염률 23.3%에 비해 저하되었다. 노지에서 재배하는 고추 '청양' 품종에 CMV는 인위적으로 접종하지 않고 자연 감염되도록 둔 상태에서 정식일부터 6월 말일까지 1주일 간격으로 유청 원액을 분무 처리한 결과, 6월5일 이전에 자연적으로 CMV에 감염된 식물체의 비율이 무처리와 살충제 처리에 비해 각각 18.9%와 16.7% 감소하였다. 유청의 식물 바이러스 감염억제에 대한 작용 기작을 알기 위하여 온실에서 PepMoV-kr에 감염된 식물체 즙액과 유청을 1:1로 혼합하여 고추에 인공접종하여 시간경과에 따른 외피단백질 농도 변화를 웨스턴 블로팅 방법으로 분석한 결과, 접종 6일 후 대조에 비하여 적었으나, 접종 9일 후에는 대조와 동일하게 증가하였다. 본 연구 결과는 유청이 고추에 발생하는 서로 다른 속에 속하는 두 종류의 바이러스인 CMV와 PepMoV에 의한 감염 억제에 효과가 있다는 것을 보여주었다.

Molecular Identification and Sequence Analysis of Coat Protein Gene of Ornithogalum mosaic virus Isolated from Iris Plant

  • A potyvirus was isolated from cultivated Iris plants showing leaf streak mosaic symptom. Reverse transcription and polymerase chain reaction (RT-PCR) product of 1 kb long which encoded partial nuclear inclusion B and N-terminal region of viral coat protein (CP) genes for potyviruses was successfully amplified with a set of potyvirus-specific degenerate primers with viral RNA samples from the infected leaves: The RT-PCR product was cloned into the plasmid vector and its nucleotide sequences were determined. The nucleotide sequence of a CDNA clone revealed that the virus was an isolate of Ornithogalum moseic virus (OrMV) based on BLAST search analysis and was denoted as OrMV Korean isolate (OrMV-Ky). To further characterize the CP gene of the virus, a pair of OrMV-specific primers was designed and used for amplification of the entire CP gene of OrMV-Kr, The virus was easily and reliably detected from virus-infected Iris leaves by using the RT-PCR with the set of virus-specific primers. The RT-PCR product of the CP gene of the virus was cloned and its sequences were determined from selected recombinant CDNA clones. Sequence analysis revealed that the CP of OrMV-Kr consisted of 762 nucleotides, which encoded 253 amino acid residues. The CP of OrMV-Ky has 94.1-98.0% amino acid sequence identities (20 amino acid alterations) with that of other three isolates of OrMV, Two NT rich potential N-glycosylation motif sequences, NCTS and NWTM, and a DAC triple box responsible for aphid transmission were conserved in CPs of all the strains of OrMV. The virus has 58.5-86.2% amino acid sequence identities with that of other 16 potyviruses, indicating OrMV to be a distinct species of the genus. OrMV-Ky was the most related with Pterostylia virus Yin the phylogenetic tree analysis of CP at the amino acid level. This is the first report on the occurrence of OrMV in Iris plants in Korea. Data in this study indicate that OrMV is found in cultivated Iris plants, and may have mixed infection of OrMV and Iris severe mosaic virus in Korea.

Five Newly Collected Turnip Mosaic Virus (TuMV) Isolates from Jeju Island, Korea are Closely Related to Previously Reported Korean TuMV Isolates but Show Distinctive Symptom Development

  • For several years, temperatures in the Korean peninsula have gradually increased due to climate change, resulting in a changing environment for growth of crops and vegetables. An associated consequence is that emerging species of insect vector have caused increased viral transmission. In Jeju Island, Korea, occurrences of viral disease have increased. Here, we report characterization of five newly collected turnip mosaic virus (TuMV) isolates named KBJ1, KBJ2, KBJ3, KBJ4 and KBJ5 from a survey on Jeju Island in 2017. Full-length cDNAs of each isolate were cloned into the pJY vector downstream of cauliflower mosaic virus 35S and bacteriophage T7 RNA polymerase promoters. Their fulllength sequences share 98.9-99.9% nucleotide sequence identity and were most closely related to previously reported Korean TuMV isolates. All isolates belonged to the BR group and infected both Chinese cabbage and radish. Four isolates induced very mild symptoms in Nicotiana benthamiana but KBJ5 induced a hypersensitive response. Symptom differences may result from three amino acid differences uniquely present in KBJ5; Gly(382)Asp, Ile(891)Val, and Lys(2522)Glu in P1, P3, and NIb, respectively.

Molecular Characterization of Apple stem grooving virus Isolated from Talaromyces flavus

  • Talaromyces flavus mediates the transmission of Apple stem grooving virus (ASGV) to several host plants. The ASGV-F carried by T.flavus was partially purified from the fungus. Based on sequence analysis and homology searches, this is closely related to other ASGV strains isolated from host plants. The partially purified viral coat protein (CP) was separated on a 12% SDS-polyacrylamide gel and analyzed by Western blotting with an ASGV anti-serum. A single band at 28 kDa reacted with the ASGV anti-serum. The deduced amino acid sequence of the ORF-l showed conserved domains, including an NTP-binding helicase motif, GFAGSGKT. The amino acid sequences of the helicase and CP showed strong homology to other ASGV strains (98%). All ASGV isolated from plants and fungi had salt bridges composed of the CP and the GFAGSGKT motif of the helicase, which are commonly conserved in plant viruses. These results suggest that ASGV-F is one of ASGV strains isolated from T.flavus based on sequence similarity as well as the serological analysis of CP.

Late Respiratory Infection after Lung Transplantation

  • Background: Aiming to improve outcome of lung transplantation (LTx) patients, we reviewed risk factors and treatment practices for the LTx recipients who experienced respiratory infection in the late post-LTx period (>1 month after LTx). Methods: We analyzed the clinical data of 48 recipients and donors from 61 LTx, who experienced late respiratory infections. Late respiratory infections were classified according to the etiology, time of occurrence, and frequency of donor-to-host transmission or colonization of the recipient prior to transplantation. Results: During the period of observation, 42 episodes of respiratory infections occurred. The organisms most frequently involved were gram (-) bacteria: Acinetobacter baumannii (n=13, 31.0%), Pseudomonas aeruginosa (n=7, 16.7%), and Klebsiella pneumoniae (n=4, 10.0%). Among the 42 episodes recorded, 14 occurred in the late post-LTx period. These were bacterial (n=6, 42.9%), fungal (n=2, 14.3%), viral (n=4, 28.5%), and mycobacterial (n=2, 14.3%) infections. Of 6 bacterial infections, 2 were from multidrug-resistant (MDR) A. baumannii and one from each of MDR P. aeruginosa, extended spectrum ${\beta}$-lactamase (+) K. pneumoniae, methicillin-resistant Staphylococcus aureus and Streptococcus pneumoniae. Infection-related death occurred in 6 of the 14 episodes (43%). Conclusion: Although the frequency of respiratory infection decreased sharply in the late post-LTx period, respiratory infection was still a major cause of mortality. Gram (-) MDR bacteria were the agents most commonly identified in these infections.
