Localization of Single Chain Fv Antibodies (scFv) in Transgenic Tobacco Ptants Showing Resistance against Tomato Bushy Stunt Virus

  • To develop an effective protection strategy against tomato bushy stunt virus (TBSV), tobacco plants expressing single-chain Fv antibodies (scFv), were established. A previous had shown that the replication activity of viral replicase was inhibited by the selected scFvs. Moreover, no systemic symptom was found after virus inoculation on leaves of wt N. benthamiana infiltrated with an Agrobacterium suspension resulting i3l expression of the scFvs. However, control plants showed systemic symptoms. In this study the localization of the scFvs within two transgenic plant lines, (CP28H3, CP-P55) was demonstrated using immunogold labelling. The gold particles, indicating the presence of scFv, were mostly found In the cytoplasm of the plant cells including chloroplasts and in the cell walls. However, they were hardly found in the vacuole, nucleoplasm and intercellular spaces. Gold particles often accumulated in either the cytosol or chloroplasts showing a specific labeling, There was no difference in type of gold labeling between both transgenic lines. The localization of the scFv in the cytoplasm further conforms the inhibition of the RNA-dependent RNA polymerase (RdRp) by the selected scFv because it is known that the RdRp is localized to membraneous cytosolic structures.

  • The present study was done to demonstrate ADV antigens in frozen and paraffin sections from ADV-infected pigs and cell cultures by using of the IGS method. Tissue specimens from 3 young pigs infected with ADV-phylaxia strain and of 2 healthy pigs were used. Fibroblastic cells originated from pig brain and BHK cells were grown and confluent monolayers were infected with the virus. Two monoclonal antibodies and a specific hyperimmune serum to ADV were used as the source of primary antibodies for both the IGS and immunoperoxidase methods. Application of the IGS method yielded a black fine granular reaction in positive areas, and the results were superior to those obtained using the immunoperoxidase technique for all cases tested. The IGS method might be useful in the detection of various viral antigens in tissue sections.

Production of Monoclonal Antibodies Against Nervous Necrosis Virus (NNV, RGNNV genotype) (신경괴사증바이러스(nervous necrosis virus, RGNNV genotype)에 대한 단클론 항체 생산)

  • We developed and subsequently characterized mouse monoclonal antibodies (MAbs) against nervous necrosis virus (NNV, RGNNV genotype). We established six hybridoma clones secreting MAbs against NNV antigen: 2B1, 2B11, 2C12, 13C1-1, 13C1-2 and 14D11. All six MAbs belonged to the IgG2a isotype with a kappa light chain and their reactivity recognized against the 41 kDa coat protein of NNV by Western blot analysis. The affinity constants of the six MAbs were measured by enzyme-linked immunosorbent assay (ELISA). All six MAbs reacted with two NNV isolates (SgNag05 and Gemunodo06), while no reactivity was observed with five know fish viruses, namely marine birnavirus, infectious pancreatic necrosis virus, viral hemorrhagic septicemia virus, hirame rhabdovirus, and infectious hematopoietic necrosis virus. Moreover, high ELISA optical density (OD) values (0.87-1.42) were observed in the brain tissues of NNV-infected sevenband grouper, while low OD values (less than 0.12) were recorded in the brain tissues of uninfected fish. These results suggest that these six MAbs are highly competent and useful for the detection of NNV with the RGNNV genotype.

  • The most important progress in diagnostic sciences is the increased sensitivity and specificity in diagnostic procedures due to the development of newer micromethodologies and increasing availability of immunological and molecular biological reagents. The outcome of researches in this field has already provided DNA probes and antibodies which can be used for diagnosing various kinds of diseases including inherited ones. This development can be also applied to diagnose diseases in oral and maxillofacial regions. Technological advances have yielded highly sensitive test methodologies so that low analyte concentration and small sample volume are no longer limiting factors. Therefore, saliva can be useful test fluid for an array of analytes. Salivary constituents of diagnostic significance include steroid hormones, antibodies, drugs, and tumor markers. Of the proteins present in saliva, viral-specific immunoglobulins are of the greatest diagnostic interest. The development of conjugates and antigens by recombinant DNA technique and peptide synthesis is necessary for clinical application. Several kits developed for the purpose of blood testing should be modified to permit their application to saliva. The final practical outcome of researches in diagnostic sciences will be various diagnostic agents which can be used for detection of bacteria and viruses, screening and diagnosis of diseases, genetic screening for forensic individual identification. For these purposes, collaboration researches and development between institutions and companies are essential.

  • Antigenicity of recombinant human interferon $\beta$(LB00013), a newly developed drug for anti-cancer and anti-viral therapeutic use, was investigated in mice and guinea pigs. The following results were obtained: 1. Mice showed no production of antibodies against LB00013 sensitized with aluminum hydroxide gel (Alum) as an adjuvant, when judged by the heterologous passive cutaneous anaphylaxis (PCA) test in rats. Meanwhile, antibodies against ovalbumin (OVA) sensitized with Alum were clearly detected. 2. In guinea pigs, the sensitization of neither LB00013 only nor LB00013 with complete Freund's adjuvant (CFA) produced positive reactions in the homologous active systemic anaphylaxis (ASA) and the PCA tests. Meanwhile, the sensitization of OVA with CFA produced positive reactions in both PCA and ASA. 3. A LB00013-specific reaction was not observed in an indirect hemagglutination(IHA) assay using sera isolated from LB00013 sensitized mice. The present results suggested that LB00013 may have no antigenic potential in mice and guinea pigs.

  • The neutralization epitopes of the outer capsid protein VP4 of a porcine rotavirus, Gottfried strain, were studied using neutralizing monocolonal antibodies(N-MAbs). Eight N-MAbs which are specific for the VP4 of Gottfried strain were used for analyzing the antigenic sites of VP4. Three different approaches were used for this analysis; i)testing the serological reactivity of each N-MAb against different G and P types of human and animal rotavirusese ii) analyzing N-MAb-resistant viral escape mutants and iii) performing nucleotide sequence analysis of the VP4 gene of each N-MAb-resistant viral escape mutant. From experimental results, at least four antigenic sites(I, II, III, and IV) were identified. Antigenic site I recognized by N-MAbs 24B9, 23G10, and 26A2 was separated from antigenic site II recognized by N-MAbs 30H5, 32B3, and 29B3. However, these antigenic sites were overlapped with antigenic site III recognized by N-MAb 21A1. The other antigenic site IV recognized by N-MAb 16D2 was separated from antigenic sites I, II, and III.

Improved Immune Response to Recombinant Influenza Nucleoprotein Formulated with ISCOMATRIX

  • Current influenza vaccines elicit antibodies effective against homologous strains, but new strategies are urgently needed for protection against emerging epidemic or pandemic strains. Although influenza vaccine candidates based on the viral nucleoprotein (NP) or matrix protein do not elicit sterilizing immunity, they have the advantage of inducing immunity that may cover a larger number of viral strains. In this study, recombinant NP produced in Escherichia coli was purified and formulated in combination with the adjuvant ISCOMATRIX. This formulation increased a NP-specific immunity in mice, with a Th1 profile, and may constitute a promising low-cost influenza vaccine candidate, with ability to stimulate humoral and cellular immune responses.

Immunohistochemical study of causative agents for the porcine viral diarrhea disease on newborn piglets in Cheju (제주지역 양돈장에서 발생한 자돈 설사형 바이러스의 면역조직화학적 확인)

  • In an outbreak of acute porcine diarrhea in newborn piglets, an etiological study was carried out using piglets submitted in Cheju Province Institute for Livestock Promotion(Cheju Veterinary Service for the disease diagnosis). Sixteen piglets(2-7 days old) were collected from 4 farms during outbreaks of diarrhea disease(from January to April 2000). Specimens were taken after necropsy and examined by immunohistochemistry using of monoclonal antibodies for porcine epidemic diarrhea(PED) virus, transmissible gastroenteritis(TGE) virus, and porcine rotavirus. Immunohistochemistry showed that PED virus antigens, but both TGE virus and rota virus antigens not, were localized in the some epithelial cells of the intestines of 14 animals among 16 piglets examined. PEB virus antigens were mainly detected in the cytoplasm of enterocytes. Infected cells, which were most abundant in the villous epithelial cells of the jejunum and ileum, were uncommon in the crypt, epithelial cells, the lamina propria and Peyer's patches of piglets examined. The results suggest that PED virus is one of the most prevailing agents in an outbreak of fatal diarrhea in newborn piglets on Cheju island and PED virus was need to further study to prevent this disease.

Changes in the serum immunoglobulin levels and viral antibody titers of colostrum-conferred Korean native calves during the first 12 weeks postpartum (초유를 섭취한 한우 송아지의 출생후 12주 동안의 혈청 면역글로불린과 각종 바이러스 항체가의 변화)

  • The changes in serum total protein and immunoglobulin levels, and BVD, IBR and PI-3 viral neutralizing antibody titers in colostrum-conferred Korean native calves during the first 12 weeks postpartum were studied, and the results obtained were summerized as follows: The Mean concentration of total protein, total immunoglobulin, IgG, IgM and IgA in sera of 9 calves at birth were $3.8{\pm}0.5g/dl$, $0.27{\pm}0.15mg/ml$, $0.06{\pm}0.08mg/ml$, $0.21{\pm}0.11mg/ml$, and extremely low concentration, respectively. Serum total protein level reached a maximum at 20 hours after birth, total immunoglobulin, IgG, and IgM levels at 24 hours, and IgA level at 28 hours, respectively. Serum IgA level reached a minimum at 4 weeks old, IgM level at 5 weeks, total immunoglobulin level at 8 weeks, and IgG level at 10 weeks, respectively. After then those levels had begun to increase, but total protein level was still decreasing at 12 weeks old. The half-lives of IgG, IgM, and IgA were 21.1 days, 4.0 days, and 2.6 days-respectively. In 10 Korean native cows immediately after parturition, serum neutralizing antibody titers specific to BVD, IBR and PI-3 virus were $8.7{\pm}1.5{\log}_2$, $5.7{\pm}1.2{\log}_2$, and $6.8{\pm}1.01{\log}_2$, respectively. And colostral neutralizing antibody titers against BVD, IBR, and PI-3 virus were $10.1{\pm}1.4{\log}_2$, $6.8{\pm}1.3{\log}_2$ and $7.8{\pm}1.7{\log}_2$, respectively. Before suckling the colostrum, SN antibody titers against BVD, IBR, and PI-3 virus were undetectable from all of 9 Korean native calves. Nevertheless SN antibody titer against BVD virus reached a maximum level ($9.2{\pm}0.6{\log}_2$) at 24 hours after birth, that against IBR virus ($6.1{\pm}1.0{\log}_2$) at 20 hours after birth, and that against PI-3 virus ($6.8{\pm}0.9{\log}_2$) at 32 hours after birth, respectively. In 12 weeks old calves, the SN antibodies against BVD and IBR virus were still decreasing, but that against PI-3 virus reached a minimum at 10 weeks, and increased after 12 weeks of age. The half-lives of SN antibodies against BVD, PI-3 and IBR, virus were 16.0 days, 22.6 days, and 25.5 days, respectively.

Development of Test System for Detection of Antibody to Human Immunodeficiency Virus Type 1 Subtype O (HIV-1 O형 항체 진단시료의 개발)

  • In Korea, all domestic made test systems for detecting antibodies in HIV-1 contain the antigens from human immunodeficiency type 1 (HIV-1) subtype B. However, because HIV-1 subtype O is significantly different in amino acid sequences from all other subtypes of HIV-1, there has been a need for developing a test for detecting antibodies in subtype O. For this purpose, the entire nucleotide sequence corresponding to the extracellular domain of the transmembrane glycoprotein of HIV-1 subtype O was synthesized with consideration of Escherichia coli condon usage. Various regions of the extracellular domain were cloned into E. coli expression vectors and tested for levels of protein production. The nucleotide sequence, named ECTM, that can encode a 129 amino acid-long peptide, was found to be expressed at a high level in E. coli. The protein of approximately 17 kDa specifically reacted with sera from individuals infected with HIV-1 subtype O. The ECTM protein was purified to near homogeneity by the CM-T gel chromatography, using concentrated, denatured inclusion bodies. In Western blot analysis, the purified viral antigen reacted with sera from individuals infected with subtype O more efficiently than subtype B. The enzyme linked immunoabsorbent assay (ELISA) system was developed using the subtype O viral protein and compared with the commercially available kit lacking the antigens from subtype O. The ELISA kit containing the subtype O antigen ECTM alone efficiently reacted with sera from individuals infected with subtype O. The subtype O antigen-containing kit produced a positive absorbence even when sera were diluted 512-fold, suggesting a high sensitivity. The commercially available kit also reacted with subtype O sera, but produced a negative result at a dilution of 8-fold. Our results suggest that the currently available kit may not be able to efficiently detect subtype O sera and that the viral protein developed in this study may be added to the current system to maximize the detection of sera from individuals infected with subtype O.
