  • Probiotics are live organisms that when administered in adequate amounts confer a health benefit on hosts. The administration of direct-fed microbials (DFM) such as lactobacilli and bacillus, may be a more direct approach to beneficially alter gastrointestinal microflora than altering dietary ingredients or supplementing with growth-promoting levels of antibiotics. It is apparent that microbes have an important influence on immune development and resistance to infection; that microbes are not static colonizers of our bodies, but are dynamic, symbiotic coresidents. And it can improve the surrounding environments; decrease the malodor caused by degrade the excrement. Recently, new paradigm such as environment protection and safe food have been settled. In domestic farm house, there is a great demand for probiotics as a substitute of antibiotics for the improvement of environmental quality and the production of a competitive goods. Probiotics circulated in a country have three categories: an animal medicine permitted by national veterinary research quarantine service (NVRQS), a support feed registered in city or country house, and not-registered goods. However, lots of unqualified goods were produced and circulated. And thus, it is in urgent need of evaluating the present situation and effect of probiotics. This study was conducted to evaluate the system of a probiotics as a livestock-environment improving agents for the alternation of antibiotics and quality control of it.

Effects of Potassium Ion and Caffeine on Contraction and Cytosolic Free $Ca^{2+}$ Levels in Vascular Smooth Muscle (혈관평할근 세포에서의 칼륨이온과 카페인의 영향: 수축과 세포내 칼슘이온 농도에 대하여)

  • Effects of high concentration of KC1 and caffeine on cytosolic $Ca^{2+}$ level $([Ca^{2+}]_{cyt})$, measured simultaneously with muscle tension using a fluorescent intracellular $Ca^{2+}$ indicator fura 2, were examined in isolated smooth muscle of rat aorta. High $K^+$ (72.7 mM) solution induced sustained increase in both $([Ca^{2+}]_{cyt})$ and tension. In contrast to this, caffeine (20 mM) induced a rapid increase in $([Ca^{2+}]_{cyt})$ followed by a decrease to a level which was higher than the resting level. However, muscle tension showed only a transient increase followed by a decrease below the resting level. In a $Ca^{2+}-free$ solution, high $K^+-induced$ neither $([Ca^{2+}]_{cyt})$ nor tension, whereas caffeine induced a transient increase in both $([Ca^{2+}]_{cyt})$ and muscle tension. These results suggest that high $K^+-induced$ contraction in vascular smooth muscle of rat aorta is due to $Ca^{2+}$ influx whereas caffeine-induced contraction is due to $Ca^{2+}$ release from cellular store. Further, caffeine seems to have an additional effect to decrease the sensitivity of the contractile elements to $Ca^{2+}$.

Feeding Territory of the Maiden Goby, Pterogobius virgo, and Invasion Feeding of the Multicolorfin Rainbowfish, Halichoeres poecilopterus (금줄망둑 Pterogobius virgo의 섭식세력권과 용치놀래기 Halichoeres poecilopterus의 침입 섭식)

  • Feeding of the maiden goby, Pterogobius virgo and multicolorfin rainbowfish, Halichoeres poecilopterus, was studied at Kurahashi Island in the Seto Inland Sea, Japan. P. virgo foraged on only benthic invertebrates in a pit on the bottom by spot-fixed fin digging. Territory size of P. virgo was very small, because they only defended the excavated pit. H. poecilopterus frequently intruded into the feeding territory of P. virgo and fed on benthic invertebrates flushed out by the fin digging of P. virgo. Feeding activity and vigilance time of P. virgo tended to decrease significantly more in such an association than when solitary. Frequency of aggression of P. virgo when H.poecilopterus intruded into the territory was higher the intruder came from the front of the territory than when the invasion was from the side and back, regardless of the size of the intruder. In small H.poecilopterus, feeding success rate was greater from the side and back of the territory than from the front. However, similar-sized H.poecilopterus were more successful in feeding from the front and side. Feeding association provides an antipredatory benefit to P. virgo because of less alert time against predators and earlier detection of predators, although their cost is great because of territory defense and decrease of feeding activity.

Effects of Hexestrol Administration on Serum Components in Hypophysectomized Female Rats (Hexestrol의 투여(投與)가 하수체척출(下垂體剔出) 암흰쥐의 혈청성분(血淸成分)에 미치는 영향(影響))

  • This study was made to investigate whether there would be any direct relationship between ovary and serum components without the control mechanism from the hypophysis. After the administration of hexestrol (6.0mg/head) to the hypophysectomized female rats, the serum components such as total lipid, cholesterol, total protein, non-protein nitrogen, Na, Cl, and K were measured for 8 weeks after treatments. The results were obtained as follows; 1. The contents of total lipid and cholesterol for the treatment groups were increased significantly as compared with the control group at the 2nd week and there on. However, there were no ditferences between the hypophysectomized and the hexestrol treated-hypophysectomized groups. 2. The contents of total protein and non-protein nitrogen for the hypophysectomized and the hexestrol treated-hypophysectomized groups were gradually increased as the time passed in comparison with the control group. The differences between the control group and the treatment groups were significant at the 4th week and there on, and the differences between the hypophysectomized and the hexestrol treated-hypophysectomized groups were insignificant, but the hypophysectomized group had slightly high value. 3. For the contents of Na, Cl, and K, there was not any unique change but only a slight flactuation in its level, and the differences were thus statistically insignificant in all cases. 4. In the hypophysectomized famale rats, administration of hexestrol had a litlle effect on the metabolism of protein, but on the other hand, no effect of those of lipid and minerals.

Solute Carrier SLC41A1 'A MINI REVIEW'

  • The human solute carrier, SLC41Al, is a $Mg^{2}+$ transporter that is regulated by extracellular magnesium. Although intracellular magnesium plays a fundamental role in cellular metabolism, little is known about how $Mg^{2}+$ is taken up and controlled by cells. Magnesium plays a fundamental role in cellular metabolism so that its control within the body is critical. Magnesium homeostasis is principally a balance between intestinal absorption of dietary magnesium and renal excretion of urinary magnesium. The kidney, mainly the distal convoluted tubule, controls magnesium reabsorption. Although renal reabsorption is under the influence of many hormones, selective regulation of magnesium transport is due to intrinsic control involving transcriptional processes and synthesis of transport proteins. Using microarray analysis, identification of the genetic elements involved with this transcriptional control has been begun. SLC41A1(GenBank Accession No. AJ514402), comprises 10 putative transmembrane domains, two of which are highly homologous to the integral membrane part of the prokaryote transports $Mg^{2}+$ and other divalent cations $Sr^2+,\;Zn^2+,\;Cu^2+,\;Fe^2+,\;Co^2+,\;Ba^2+,\;and\;Cd^2+,\;but\;not\;Ca^2+,\;Mn^2+,\;and\;Ni^2+.$ Transport of $Mg^{2}+$ by SLC41Al is rheogenic, voltage dependent, and not coupled to Na or Cl. Expressed SLC41Al transports a range of other divalent cations: $Mg^{2+},\;Sr^{2+},\;Zn^{2+},\;Cu^{2+},\;Fe^{2+},\;Co^{2+},\;Ba^{2+},\;and\;Cd^{2+}$. The divalent cations $Ca^{2+},\;Mn^{2+},\;and\;Ni^{2+}$and the trivalent ion $Gd^{3+}$ did not induce currents nor did they inhibit $Mg^{2+}$ transport. The nonselective cation $La^{3+}$ abolishes $Mg^{2+}$ uptake. Computer analysis of the SLC41Al protein structure reveals that it belongs to MgtE protein family & suggested that the human solute carrier, SLC41Al, might be a eukaryotic $Mg^{2+}$ transporter closely related $(60-70\%)$ protein encoded by SLC41A2 is a $Mg^{2}+$ transporter that might be involved in magnesium homeostasis in epithelial cells also transports a range of other divalent cations: $Ba^2,\;Ni^2,\;CO^2,\;Fe^2,\;or\;Mn^2,\;but\;not\;Ca^2,\;Zn^2,\;or\;Cu^{2+}$ that may have related functional properties.

Effects of Nitrite and Nitrate Contents of Chinese Cabbage on Formation of N-Nitrosodimethylamine during Storage of Kimchi (김치 저장 중 N-Nitrosodimethylamine 생성에 배추 내 아질산염과 질산염 함량의 영향)

  • Nitrite and dimethylamine (DMA) are the immediate precursors of carcinogenic N-nitrosodimethylamine (NDMA). This study investigated the effects of nitrite and nitrate contents in Chinese cabbage on changes in NDMA, nitrite, nitrate, and DMA contents during storage of kimchi. Contents of nitrite in Chinese cabbage 1 (HNC), Chinese cabbage 2 (MNC), and Chinese cabbage 3 (LNC) were $47.54{\pm}1.07$, $10.12{\pm}0.31$, and $6.10{\pm}0.09mg/kg$, respectively. Kimchi were assigned to one of the following three groups: kimchi used HNC (HNK), kimchi used MNC (MNK), and kimchi used LNC (LNK). HNK had higher levels of nitrite and nitrate than those in MNK and LNK after storage for 10 days and 20 days. DMA and NDMA contents decreased in all kimchi during the storage period. HNK had higher NDMA content than LNK until 20 days of storage. These results suggest that the contents of nitrite and nitrate in Chinese cabbage have significant effects on the formation of NDMA in kimchi.

A Dog Park Planning Model for Promoting Animal Welfare Culture in Regional Community, Korea (지역사회의 동물복지문화 증진을 위한 도그파크 계획 모형)

  • Korea's pet-related society has been fast growing without the base and preparations of socio-cultural animal welfare. For the most part, Korea's current public parks, in particular, have a space structure in which a lot of friction between the general public who are not fond of dogs and dog owners in public places has been happening unavoidably. A lot of countries that are taking a lead role in animal welfare have built dog parte that the general public and dogs can enjoy together. This research starts with the realization that the concept of 'dog park' needs to be introduced in Korea as more and more Koreans own dogs and other pets. This research aims to provide guidance for dog park planning by drawing a dog park planning model and to promote basics for co-existence of dogs and people in a park and set in place animal welfare early and raise the level of awareness. Total 7 types of dog put 56 planning items including dog zone, dog playground, and agility, and a selective matrix by which it is possible to select essential recommendatory optional items by each type of dog park were drawn in this research. Overall there were the highest needs in separated area for dogs in park. A dog park planning model is designed as follows. 1) To select a dog park type that was introduced. 2) To set up facilities applicable in common and planning items desired in advance by each type. 3) To zone dog park areas by the priority order of planning items that were introduced and to position necessary facilities. 4) To plan program that should be were introduced. A dog park was expected to take some positive roles such as providing opportunities to socialize dogs, activating social communities, more effective park management by participating local community members and so on.

The Novel Approach of Gene Detection by Single-neuronal Cell Manipulation (단일 도파민뉴런을 이용한 새로운 유전자발현 검출기법)

  • RT-PCR is an useful method to investigate the expression of target gene as detection tools. Although RT-PCR is the powerful detection method for tissues, it was difficult to amplify the target gene product using the single cell. To clarify the expression level of the genes related to Parkinson's disease (PD), I performed the laser dissection of single cell from Substantia nigra. I examined the mRNA expression level in the dopaminergic neuron isolated from the PD patients by the single cell RT-PCR method. It is known that tyrosine hydroxylase (TH), DOPA decarboxylase (DDC) are involved in biosynthesis of the catecholamine such as dopamine. Little has been known about the gene expression features of these enzymes in single dopaminergic neuron. I could detect the specific gene products in single cell level. The different expression was observed in PD-related gene products from the single neuron of PD patients. Interestingly, TH gene expression was significantly decreased with comparing the ratio of decrease in other PD-related genes. Hence, I represented data that indicate the RT-PCR method described in this report is an effective method in detecting a specific single-cell mRNA level related with diseases.

Effects of Estradiol-$17{\beta}$ and 2,4-D on Vitellogenin Synthesis in the Hepatocytes Primary Culture of the Olive Flounder, Paralichthys olivaceus (넙치 Paralichthys olivaceus 초대 배양 간세포의 난황 전구물질 합성에 미치는 estradiol-$17{\beta}$와 2,4-D의 영향)

  • Effects of Estradiol-$17\beta(E_2)$ and 2, 4-dichlorophenoxy acetic acid (2, 4-D) on vitellogenin(VTG) production were investigated in primary hepatocyte culture of olive flounder, Paralichthys olivaceus. Highest survival rate of hepatocyte were observed at $27^{\circ}C$, which markedly declined equal to 50% of those of $15^{\circ}C$. Vitellogenin production peaked at the concentration of $10^{-6}M\;E_2$. No effect was observed on VTG production at various concentrations of 2, 4-D. However, a low concentration of 2, 4-D (ie, $10^{-8}M$) only appeared increased VTG production. $E_2$ or $10^{-8}M$ 2, 4-D-primed VTG production was markedly inhibited by the addition of $10^{-6}M$ tamoxifen to the culture medium(P<0.01). Inhibition was not affected by combinational treatment with $10^{-6}M$ $E_2$ and $10^{-6}M$ 2, 4-D. These results from the current investigation suggest that 2, 4-D mimics $E_2$, but the mechanism of reaction in inducing the $E_2$ receptor are different in VTG production in oliver flounder hepatocytes.

The influence of root surface distance to alveolar bone and periodontal ligament on periodontal wound healing

  • Purpose: The purpose of this animal study was to perform a 3-dimensional micro-computed tomography (micro-CT) analysis in order to investigate the influence of root surface distance to the alveolar bone and the periodontal ligament on periodontal wound healing after a guided tissue regeneration (GTR) procedure. Methods: Three adult Sus scrofa domesticus specimens were used. The study sample included 6 teeth, corresponding to 2 third mandibular incisors from each animal. After coronectomy, a circumferential bone defect was created in each tooth by means of calibrated piezoelectric inserts. The experimental defects had depths of 3 mm, 5 mm, 7 mm, 9 mm, and 11 mm, with a constant width of 2 mm. One tooth with no defect was used as a control. The defects were covered with a bioresorbable membrane and protected with a flap. After 6 months, the animals were euthanised and tissue blocks were harvested and preserved for micro-CT analysis. Results: New alveolar bone was consistently present in all experimental defects. Signs of root resorption were observed in all samples, with the extent of resorption directly correlated to the vertical extent of the defect; the medial third of the root was the most commonly affected area. Signs of ankylosis were recorded in the defects that were 3 mm and 7 mm in depth. Density and other indicators of bone quality decreased with increasing defect depth. Conclusions: After a GTR procedure, the periodontal ligament and the alveolar bone appeared to compete in periodontal wound healing. Moreover, the observed decrease in bone quality indicators suggests that intrabony defects beyond a critical size cannot be regenerated. This finding may be relevant for the clinical application of periodontal regeneration, since it implies that GTR has a dimensional limit.
