별불가사리 난자형성 중 난모세포(직경 7-l7O Um) 표면의 구조적 변화를 투과 전자현미경으로 관찰하였다. 직경 25-855 Um의 난모세포에서 'endocvtosis'와 관련된 구조인 coated pit, coated vesicle과 내포소낭이 존재하였으며. 내포소낭은 난황립과 융합하였다. 특히, 이러한 융합은 직경 130-155 Um의 난모세포에서 많이 관찰되었다 난자형성 초기에 난모세포의 미세융모는 세포막을 따라 무질서하게 분포하고. 인접한 세포와 맞닿아 있었다. 직경 15 Um에서 65 Um에 이르는 난모세포는 세포막 부위에 따라서 미세융모의 굵기와 길이, 그리고 형태가 다르게 나타났고, 직경 100 Um 이상의 난모세포에서는 잘 발달된 미세음모가 난뢍층에 존재하였다 여포세포로부터 나온 돌기는 직경 7 Um의 난모세포와 난자형성 후기의 난모세포에서 난황층을 윤고 세포막과 닿아 'junctional complex'를 형성하였다. 이와같은 난모세포 표면의 다양한 구조적 변화는 난모세포의 세포막을 통한 물질투과 기능의 분화와 관련이 있을 것으로 생각된다.
토끼의 골격근 homogenate에서 23,000$\times$G, 60 분간의 원심분리와 얻은 근 microsome의 ATPase 활성, 근수축에 대한 이완작용, 및 Ca 의 흡수작용을 여러 가지 조건에서 측정하였다. ATPase 활성은 Ca++ Mg++ 양 이온의 존재에 의하여 활성화되며 , 5 mM Mg++ 의 존재하에서는 Ca++ 의 최적농도는 0.1mM이다. Oxalate의 존재하에서는 1 mM 의 Ca++ 이 최적농도이므로 oxalate의 작용은 불용성 Ca-oxalate의 작용은 불용성 Ca-oxalate를 microsome vesicle so 및 medium 내에 침전시켜 유리 Ca++ 농도를 저하시키는 것이라고 생각된다. Microsome의 이완작용은 조제후 120 시간까지 시간에 따라 감소되어 가나, 그이 ATPase 활성은 거의 변화가 없는 것으로 보아 Ca++ + Mg++ -의존성 ATPase 는 이완작용에는 직접 관련이 없는 것으로 해석된다. Oxalatedmlwhswo는 microsome의 Ca++ 흡수량을 현저히 증대시키며 동시에 흡수포화에 도달하는 시간을 지연시킨다. Oxalate의 이러한 효과도 Ca-oxalate의 형성에 기인하는 것으로 해석된다. Microsome 내에 축적되는 Ca 의 량은 ATP 농도가 커질수록 많아진다. 그러나 축적된 Ca 의 량과 ATP 농도사이에 화학정량론적 관계는 없는 것같다.

Flip-flop rate constants were measured by dithionite assay of NBD-PE fluorescence in DMPC/POPC vesicles made of various DMPC/POPC ratios. The activation energy, enthalpy, entropy, and free energy were determined based on the transition state theory. We found that the activation energy, enthalpy, and entropy increased as the amount of POPC increased, but the activation free energy was almost constant. These experimental results and other similar studies allow us to propose that the POPC molecules included in DMPC vesicles affect the flip-flop motion of NBD-PE in DMPC/POPC vesicles via increasing the packing order of the ground state of the bilayer of the vesicles. The increase in the packing order in the ground state seems to be a result of the effect of the overall molecular shape of POPC with a monounsaturated tail group, rather than the effect of the longer tail group.
The tadpole larva of the ascidian Halocynthia roretzi has two sensory pigment cells in its brain vesicle. To elucidate the temporal requirement for FGF signaling in formation of the pigment cells, embryos were treated with an FGF receptor 1 inhibitor, SU5402, or an MEK inhibitor, U0126 during various embryonic stages. In the present study, it is shown that the embryos treated with SU5402 from the 16-cell stage to the early gastrula stage do not form pigment cells, whereas those treated after the early gastrula stage form pigment cells. In pigment cell formation, embryos suddenly exhibited the sensitivity to SU5402 only for 1 h at the neural plate stage(-4 h after the beginning of gastrulation). When U0126 treatment was carried out at various stages between the 8-cell and late neurula stages, the embryos scarcely formed pigment cells. Pigment cell formation occurred when the embryos were placed in U0126 at early tail bud stage. These results indicate that FGF signaling is involved in pigment cell formation at two separate processes during ascidian embryogenesis, whereas more prolonged period is required for MEK signaling.
In the neuron, SNARE (soluble N-ethylmaleimide-sensitive factor attachment protein receptors) assembly plays a central role in driving membrane fusion, a required process for neurotransmitter release. In the cytoplasm, vesicular SNARE VAMP2 (vesicle-associated membrane protein 2) engages with two plasma membrane SNAREs syntaxin 1A and SNAP-25 (synaptosome-associated protein of 25 kDa) to form the core complex that bridges two membranes. While various factors regulate SNARE assembly, the membrane also plays the regulatory role by trapping VAMP2 in the membrane. The fluorescence and EPR analyses revealed that the insertion of seven C-terminal core-forming residues into the membrane controls complex formation of the entire core region, even though preceding 54 core-forming residues are fully exposed and freely moving. When two interfacial Trp residues in this region were replaced with hydrophilic serine residues, the mutation supported rapid complex formation.
An efficient method for labeling individual proteins in live cells is required for investigations into biological mechanisms and cellular processes. Here we describe the preparation of small quantum dots (QDs) that target membrane surface proteins bearing a hexahistidine-tag ($His_6$-tag) via specific binding to an nitrilotriacetic acid complex of nickel(II) ($NTA{\cdot}Ni^{2+}$) on the QD surfaces. We showed that the $NTA{\cdot}Ni^{2+}$-QDs bound to His-tag functionalized beads as a cellular mimic with high specificity and that QDs successfully targeted $His_6$-tagged vesicle-associated membrane proteins (VMAP) on cell surfaces. This strategy provides an efficient approach to monitoring synaptic protein dynamics in spatially restricted and confined biological environments.
A method of preparation of magnetite nanoparticles in ordered systems, as in vesicle and microemulsion, consisting of soybean lecithin and water has been introduced. The size of magnetite grain was controlled by the content of soybean lecithin and size of liposomes in the systems. It was found by experiment that magnetite nanoparticles were formed inside the double layer vesicles. The magnetite nanoparticles were separated by magnetic separation and centrifugation and the dispersion of the magnetite nanoparticles prepared at 10% (w/w) soybean lecithin was particularly stable. The formation of pure magnetite nanoparticles was confirmed by analyses of XRD and electron diffraction pattern.
Synapse is the basic structural and functional component for neural communication in the brain. The presynaptic terminal is the structural and functionally essential area that initiates communication and maintains the continuous functional neural information flow. It contains synaptic vesicles (SV) filled with neurotransmitters, an active zone for release, and numerous proteins for SV fusion and retrieval. The structural and functional synaptic plasticity is a representative characteristic; however, it is highly vulnerable to various pathological conditions. In fact, synaptic alteration is thought to be central to neural disease processes. In particular, the alteration of the structural and functional phenotype of the presynaptic terminal is a highly significant evidence for neural diseases. In this review, we specifically describe structural and functional alteration of nerve terminals in several neurodegenerative diseases, including Alzheimer's disease (AD), Parkinson's disease (PD), Amyotrophic lateral sclerosis (ALS), and Huntington's disease (HD).
북방산개구리, 참개구리 및 옴개구리를 사용하여 성숙된 난자의 세포질에서 활성을 띠는 성숙촉진요인( maturation promoting factor, MPF)을 미세주입 법으로 확인하고 이들의 성질을 조사하였다. 핵붕괴(germinal vesicle breakdown, GVBD)된 난자의 세포질을 미성숙 난자(GV난자)에 주입하고(75-100 nl) 이들을 15-24시간 배양했을 때 대부분의 GV 난자들이 핵붕괴를 일으켰으나(약 80%) 미성숙 난자(GV 난자)의 세포질을 주입했을 때에는 약 200nl의 난자들만이 핵붕괴를 일으켰다. 핵붕괴된 난자들의 crude extract를 주입했을 때에도 역시 성숙유도 효과가 있었으며 이종간에도 효과가 있었다. 난자의 성숙을 잘 일으키지 않는 옴개구리의 난자를 사용하여 MPF를 가진 세포질을 계대주입(serial transfer)하였을 때에도 MPF가 계속 활성을 띠는 것을 확인할 수 있었다. 아울러 개구리 난자의 MPF생성과 증폭과정에 CAMP의 증가나 단백질 합성의 저해가 미치는 영향을 조사한 결과. MPF의 작용이 유의하게 이들에 의해 억제되는 것을 알 수 있었다.
