감과실의 성숙과 추숙중 조직의 변화 (Changes in the cell structure during maturation and postharvest of persimmon fruits)

  • 감과실이 성숙함에 따라 세포가 비대성장하고 세포간극이 발달하였으며 연시에서는 세포가 분리되어 독립적으로 존재하였다. 또 성숙중에는 원형질내에 small vesicle이 생기고, 완숙감과실에서 middle lamella의 분해현상을 관찰하였다. 연시에서는 세포벽과 middle lamella가 분리 용해되었다.

Electron Tomography and Synapse Study

  • Electron tomography (ET) is a useful tool to investigate three-dimensional details based on virtual slices of relative thick specimen, and it requires complicated procedures consisted of image acquisition steps and image processing steps with computer program. Although the complicated step, this technique allows us to overcome some limitations of conventional transmission electron microscopy: (1) overlapping of information in the ultrathin section covering from 30 nm to 90 nm when we observe very small structures, (2) fragmentation of the information when we study larger structures over 100 nm. There are remarkable biological findings with ET, especially in the field of neuroscience, although it is not popular yet. Understanding of behavior of synaptic vesicle, active zone, pooling and fusion in the presynaptic terminal have been enhanced thanks to ET. Some sophisticated models of postsynaptic density with ET and immune labeling are introduced recently. In this review, we introduce principles, practical steps of ET and some recent researches in synapse biology.

Cisplatin의 신장독성에 관한 연구 (Studies on the Cisplatin Nephrotoxicity)

  • Cisplatin is useful for various cancers including advanced testicular and ovarian cancers. However, clinical use of cisplatin has been limited due to its dose-related neplrotoxicity. Transport studies across the membrane vesicles were performed to study the cisplatin nephrotoxicity. In these experiments, after cisplatin was administered to adult male New Zealand White rabbits, basolateral membrane (BLM) vesicles were prepared from the renal cortex. Para-aminohippurate (PAH) uptakes through BLM vesicles were measured to examine the interactioln of cisplatin on the transports of the substrates. As results of the uptake experiments using the vesicle systems, cisplatin had little effects on PAH transport through BLM vesicle. In conclusion, cisplatin did not cause the damage of basolateral membranes.

Identification and Expression Patterns of kif3bz during the Zebrafish Embryonic Development

  • We are reporting the identification, expression patterns, and possible biological functions of zebrafish kif3b (kif3bz) encoding 475 amino acids. Kif3Bz contains the kinesin motor domain, catalytic domain, KISc domain, and one single coiled coil domain. Phylogenetic analysis indicates that kif3bz is a highly conserved gene among the tested vertebrates. First of all, both maternal and zygotic messages of kif3bz were evenly distributed in the blastomeres at 2-cell stage. Its ubiquitous expression throughout the blastomeres continued till 40% epiboly. However, kif3bz transcripts became restricted in Kupffer's vesicle at tailbud and 6-somite stages. At 13-somite stage, kif3bz expression pattern became specific to the telencephalon, diencephalon, trigeminal placode, and somites. Such expression patterns were further intensified in the telencephalon, diencephalons, hind brain, pronephric ducts, optic vesicles, and spinal cord neurons in the 23-somite stage embryos, and last till 24 hpf. We discussed possible functions of Kif3Bz related to the vertebrate embryonic development.

Aspergillus phoenicis의 生活史를 통한 Cellulase 및 Xylanase의 活性 변화 (The changes in the activities of Cellulase and Xylanase of Aspergillus phoenicis during the life cycle)

  • The changes in the activities of Cellulase and Xylanase of A. phoencis during the life cycle were surveyed by using synchronized culture technique. Avicelase activity of the fungus was reached at peak in the initial hyphal growth stage, but decreased gradually during the conidiophore formation, vesicle-phialide formation, and sporulation stages in decreasing order. CMCase, Salicinase, and Xylanase activities were raised very high in the initial hyphal growth stage, but decreased gradually in conidiophore formation and vesicle-phialide formation stages, and then increased, more or less, in the sporulation stage.

고전하밀도 양이온성폴리머와 Sodium dodecyl sulfate가 만드는 콤플렉스에 대한 비이온계면활성제의 영향 (The transformation of the complex of high charge density cationic polymer with sodium dodecyl sulfate into vesicles by nonionic surfactant)

  • The transformation of the liquid crystal complex made by binding of anionic surfactant, sodium dodecyl sulfate (SDS), into high charge density cationic polymer, the homopolymer of diallyldimethylammonium chloride (PDADMAC) was induced by adding of nonionic surfactants and investigated by means of microscopy and FE.SEM. Among nonionic surfactants in this experiments polyethylene glycol (3 mol) ether of lauryl alcohol (laureth-3) made variation in the complex. The laureth-3 transformed the complex into spherulite vesicle with the size of ca.$100{\mu}m$. This change increased the viscosity and the turbidity of the solution phase separated originally. Microscope showed that they are spherulite particles and polarized microscope suggested they are multi.lamellar liquid crystals. FE-SEM also proved that explicitly.

Catalytic Properties of Phospholipase D using Phosphatidic Acid as an Activator

  • The effects of phosphatidic acid(PA) on the activity of phospholipase D were examined in detail. The enzyme activity was examined in the liposome system containing phosphatidylcholine and PA, which was suspended in a desired buffer solution by ultrasonication. The substrate of large unilamella vesicle (LUV) state by ultrasonication was more effective on the enzyme activity than that of multilamella vesicle(MLV) by water-bath type sonication. The most effective molar ratio of PC-PA liposome for enzyme activity was found to be 1:0.7. The other optimum conditions were found 5 mM $Ca^{2+}$ ion, pH 6.6, and incubation temperature of $27^{\circ}C. K_m \;and \;V_{max}$ values were estimated to be 1.43 mM and 0.8 $nmole/min/{\mu}g$ protein respectively. These properties in a PC-PA liposome system were compared with those in a PC-SDS mixed micelle system. The effects of other phospholipids and organic phosphates on the enzyme activity were also examined.

키토산 올리고당 유도체의 베지클 형성에 관한 연구 (A Study on the Formation of Vesicle by Chitosan Oligosaccharide Derivative)

  • 게 껍질에서 얻어진 키토산에 아질산나트륨을 가한 후 수산화붕소산 나트륨으로 환원시켜 중합도 2∼3인 2,5-anhydro-D-mannitol의 말단기를 갖는 키토산 올리고당을 합성하였다 염화팔미틴산을 DUP 촉매하에서 키토산 올리고당과 반응시켜 키토산 올리고당에 세 개의 아실기가 결합된 키토산 올리고당 유도체를 얻었으며, 이를 다시 수산화칼륨으로 가수분해하여 N-팔미토일 키토산 올리고당을 합성하였다. 최종적으로 수용액에 분산시킨 후 초음파를 가한 후 형성된 베지클을 TEM 을 이용하여 확인할 수 있었다.

LRRK2 and membrane trafficking: nexus of Parkinson's disease

  • Recent evidence from genetics, animal model systems and biochemical studies suggests that defects in membrane trafficking play an important part in the pathophysiology of Parkinson's disease (PD). Mutations in leucine-rich repeat kinase 2 (LRRK2) constitute the most frequent genetic cause of both familial and sporadic PD, and LRRK2 has been suggested as a druggable target for PD. Although the precise physiological function of LRRK2 remains largely unknown, mounting evidence suggests that LRRK2 controls membrane trafficking by interacting with key regulators of the endosomal-lysosomal pathway and synaptic recycling. In this review, we discuss the genetic, biochemical and functional links between LRRK2 and membrane trafficking. Understanding the mechanism by which LRRK2 influences such processes may contribute to the development of disease-modifying therapies for PD.

Role of Type 1 Inositol 1,4,5-triphosphate Receptors in Mammalian Oocytes

  • The ability of oocytes to undergo normal fertilization and embryo development is acquired during oocyte maturation which is transition from the germinal vesicle stage (GV), germinal vesicle breakdown (GVBD) to metaphase of meiosis II (MII). Part of this process includes redistribution of inositol 1, 4, 5-triphosphate receptor (IP3R), a predominant $Ca^{2+}$ channel on the endoplasmic reticulum membrane. Type 1 IP3R (IP3R1) is expressed in mouse oocytes dominantly. At GV stage, IP3R1 are arranged as a network throughout the cytoplasm with minute accumulation around the nucleus. At MII stage, IP3R1 diffuses to the entire cytoplasm in a more reticular manner, and obvious clusters of IP3R1 are observed at the cortex of the egg. This structural reorganization provides acquisition of $[Ca^{2+}]_i$ oscillatory activity during fertilization. In this review, general properties of IP3R1 in somatic cells and mammalian oocyte are introduced.
