몇가지 식물이 Agaricus campestris 균사의 생장에 미치는 영향(예보) (Effects of the special media on the mycelium growth in Agaricus campestris)

  • Effects of the special media on the mycelium growth in Agaricus campestris were studied. The results might be summarized as follows: 1. The mycelium growth fo Agaricus campestris were scarecely stimulated on the Peptone basal medium which was added 0.5gr. of Peptone and Dextrose basal medium which was added 1.5gr. of dextrose during the culture for 144 hours. 2. The mycelium of Agaricus campestris on the media which was added the several kinds of vegetable extracts showed a considerable growth for 144 hours. The order is as follows; Carrot-basal medium(4ml./100ml.)>Tomato-basal medium(2ml./100ml.)>Spinach-basal medium(3ml./100ml.). However, the spinach-basal medium among these three media were no significant difference.

Enhancement of Succinate Production by Organic Solvents, Detergents, and Vegetable Oils

  • Bioconversion of fumarate to succinate by Enterococcus sp. RKY1 was enhanced when Tween surfactant, organic solvent, and vegetable oil were added to the fermentation medium. The maximum amount of succinate produced was 80.4 g/l after a 24 h incubation when Tween 80 was added to the culture to a final concentration of 0.1 g/l. Triton X-l00 was observed to damage the enzymes and inhibit the formation of succinate. The addition of 10 ml/l acetone increased the production of succinate by 110%. Vegetable oils used were found to be effective for succinate production as well as for the cell growth. Similar productivity increases were obtained with corn oil and Tween 80 plus biotin with the total productivity being 3.6 g/l/h, and 3.5 g/l/h, respectively, which was approximately 25% greater than that of the control. Therefore, these results indicate that com oil can be considered the most appropriate agent for the production of succinate where succinic acid was primarily used in the production of food, medicine, and cosmetics.

지역아동센터 급식을 위한 적정 배식량 제안 (Recommendation of Serving Size of the Meal Service of Community Child Centers in Korea)

  • Objectives: This study was performed to set easily applicable portion sizes by sex and age for children at the Community Child Centers (CCC) in Korea. Methods: Considering the age and gender specific energy level at Target Patterns for children aged 6-18 years, which were suggested as a part of the 2010 Korean Food Guidance System (KFGS), we set three meal sizes. We reclassified the recommended daily servings of Grains, Meat fish eggs beans and Vegetables group at Target Patterns into three meal sizes, and then calculated the recommended serving per meal. Each proposed amount of food per meal was calculated based on serving size of foods commonly eaten at KFGS, which was then allocated to five meal components; rice, soup stew, protein and vegetable side-dishes and Kimchi. Each proposed amount of food per meal was applied to 173 menus' recipes from CANpro 3.0 as main ingredient's amounts. We cooked the 173 menus at the medium size and measured their weights after cooking. Results: Each recommended serving per meal was 0.75, 0.9 and 1.2 for Grains; 1.2, 1.6 and 2.4 for Meat fish eggs beans; 2, 2.4 and 2.8 for Vegetables by meal sizes. Among five meal components, the ratio of small and large to medium size was 1/5 less and 1/3 more for rice and 1/3 less and 1/3 more for soup stew, protein side-dish and Kimchi, respectively. We suggested the same amount for a vegetable side-dish to encourage vegetable intake. Proper portion sizes per meal of medium were rice 190 g, soup stew 210 g (solid ingredients 60 g), protein side-dish 100 g (meat eggs beans) and 70 g (fish), vegetable side-dish 80g and Kimchi 30 g. Conclusions: Proper portion size per meal suggested in this study may be useful at the CCC where dietitians are not available and the approach could be applicable to the other types of meal services.

일반 소비자의 입장에서 본 시장에서의 채소류 선택 방법 I - Chlorophyll과 Ascorbic Acid 관계- (A Method for Choosing Vegetables at the Market from a General Consumers Standpoint I -The Relationship between Chlorophyll and Ascorbic Acid-)

  • This study examined the relationship between chlorophyll(Chl) and ascorbic acid(ASA) as an index of freshness for vegetables, when the general consumers purchase several kinds of vegetables at the market. The leaves and fruits of several types of vegetables were divided into top, middle, and basal sections. Chl was determined by a spectrophotometer, and ASA was analyzed by high-performance liquid chromatography. For the vegetable sections, we found that Chl and ASA levels were highest in the top sections, followed by the middle, and were lowest in the basal portions. For the relationship between Chl and ASA in vegetables, there were seven classifications including high Chl(above 80mg/100g of fresh weight) and high ASA(above 80 mg/100 g of fresh weight) for mallow, and kail; the second group had high Chl and low ASA(below 80 mg), such as in spinach, burdock, and perilla leaves; the third group contained medium Chl($40{\sim}80mg$) and medium ASA($40{\sim}80mg$) levels, such as for gonddalbee, angelica, pumpkin leaf and coriander; the fourth group had medium Chl and low ASA levels, as in crown daisy and lettuce; the fifth group contained low Chl(below 40 mg) and high ASA, such as in hot peppers and parsley; the sixth group had low Chl and medium ASA levels, and included broccoli, pak choi, and sweet peppers; finally the seventh group contained low Chl and low ASA levels, as in dropwort, young radishes, shallots, cucumbers and swiss chard.

High cell density cultivation of Bacillus sp.

  • 본 연구는 Bacillus sp.의 고농도 생산을 위한 최적배지 조건의 확립과 배양중 발생하는 foam control에 대해 알아보았다. PBD와 “one factor at a time", BBD로 이여지는 배지 최적화 과정을 통해 최초 생산 배지에서의 균체량보다 100% 이상의 향상된 균사체량을 볼 수 있었다. 또한 scale-up 과정에서 발생하는 foam control를 위해 각기 다른 type의 antifoam을 사용하였는데, 적당한 농도에서 좋은 생장을 보이고 있었고, vegetable oil은 foam control과 값비싼 탄소원인 soluble starch을 대신할 수 있는 좋은 기질임을 알 수 있었다. 또한 배양액 내의 산소를 잡고 있으면서 좋은 전달자의 역학을 하여 급격한 DO의 감소를 막아주었다.

Comet Assay를 이용한 케일, 명일엽, 당근, 돌미나리 녹즙의 Chinese Hamster Lung 세포 DNA 손상 보호 효과 (Protective Effect of Yellow-Green Vegetable Juices on DNA Damage in Chinese Hamster Lung Cell Using Comet Assay)

  • The present study was attempted to investigate the antioxidant capacity of popular yellow-green vegetable juices (kale, Angelica keishei, carrot, small water dropwort) and to investigate the effect of vegetable juices on protecting oxidative damage to DNA in cultured Chinese hamster lung (CHL) cells. Antioxidant capacity was analyzed by TRAP assay (Total radical-trapping antioxidant potential). Cellular DNA dmamage was measured by SCGE (single-cell gel electrophoresis, also known as comet assay. Cells incubated in medium with PBS (negative control) or with various concentration of the freeze dried green juices (25, 50, 100, 250 $\mu\textrm{g}$/$m\ell$) resuspended in PBS were treated with $H_2O_2$ (200 ${\mu}{\textrm}{m}$) as an oxidative stimulus for 5 min at 4$^{\circ}C$. The physiological function of each vegetable juice on oxidative DNA damage was analyzed and expressed as tail moment (tail length X percentage migrated DNA in tail) . Kale juice had the highest TRAP value suggesting that kale has the highest antioxidant capacity followed by Angelica keishei, small water dropwort and carrot. Cells treated with $H_2O_2$ had extensive DNA damage compared with cells treated with PBS or pre-treated with vegetable juice extracts. All green juices inhibited $H_2O_2$-induced DNA damage with kale being the most effective juice among the tested juices. These results indicate that green juice supplementation to CHL cells followed by oxidative stimulus inhibited damage to cellular DNA, supporting a protective effect against oxidative damage induced by reactive oxygen species. 부모의 어린이급식관리지원센터 인지도와 유아의 채소 섭취 관련 식생활 및 보육기관의 식단 메뉴에 관한 만족도 (Parents' Recognition of Center for Children's Food Service Management and Preschoolers' Satisfaction with Menu Provided by Childcare Centers and Food Life Regarding Vegetable Intake)

  • This study was conducted to determine parents' recognition of the Center for children's foodservice management (CCFSM) and to compare preschoolers' satisfaction for meals served by childcare centers and some aspects regarding the vegetable intake according to the parents' recognition of CCFSM. The subjects were 255 parents, whose children were 2~5 year old and attended a childcare center, were grouped according to the recognition of CCFSM (high recognition, HR, 27.5%; medium recognition, MR, 47.4%; low recognition, LR, 25.1%). Information was obtained by a self-administered questionnaire and data were analyzed by SPSS 25.0. Only 58.6% of HR and 10.7% of MR answered the they had participated education/event held by the CCFSM. More parents in the HR group (88.6%) acknowledged the helpfulness of CCFSM on the children's food habits compared to those in the MR group (63.6%) (P<0.001). Compared to the MR and LR groups, more parents in the HR group answered not only that they were 'satisfied'/'very satisfied' with the meals served by childcare centers (P<0.05), but also they tended to think that their children were also satisfied (P=0.061). Up to 31.2% of parents in the LR group answered that there was no need for education to increase the vegetable intake of their child compared to 14.3% and 17.4% in the HR and MR groups, respectively (P<0.05). Moreover, up to 26.6% of parents answered that school cook planned menus compared to 5.7% and 13.2% in the HR and MR group, respectively (P<0.001). In conclusion, the results provided the association between parents' high recognition of CCFSM and preschoolers' satisfaction for meals from childcare centers as well as a better chance for a desirable food life regarding vegetable intake.

박 자엽조직을 이용한 효율적인 식물체 분화와 Agrobacterium에 의한 형질전환 (Efficient shoot regeneration using cotyledon explants and Agrobacterium tumefaciens - mediated transformation of bottle gourd (Lagenaria sicraria Standl.))

  • 본 연구는 대목용으로 이용되는 박에 있어서 형질전환에 이용 가능한 품종을 선발하고 재분화 효율 및 형질전환 효율을 높이기 위하여 수행되었다. 3 mg/L BA가 첨가된 MS배지에서 덩굴마름병 저항성 순계계통의 평균 신초형성 수는 0 ~ 7.3개였으며 3종류의 상용 품종의 신초형성 수는 2.0 ~ 6.5개였다. 덩굴마름병 저항성 순계 계통의 재분화율은 품종 간 편차가 상용품종에 비하여 컸다. 형질전환 효율을 검정하기 위해 GFP와 gus발현을 조사한 결과 공동배양 과정 중에 GFP가 67%와 100% 로 높은 일시적인 발현을 보였다. 식물체 재분화는 BA와 2.4-D를 조합한 배지조합에서 자엽의 절단부위에서 신초가 발생하였으며, 4주 후 BA를 단독으로 1 mg/L 처리한 배지에서 한 개의 절편체에서 발생하는 신초의 개수 및 신초의 신장도 좋았다. 캘러스의 발생은 1 mg/L BA와 2,4-D를 0.1 mg 첨가한 배지에서 가장 활발하게 분화하였다.

팽이버섯 균사체 생장에 미치는 Natural Oil의 영향 (Effect of Natural Oil on the Mycelial Growth of Flammulina velutipes)

  • Vegetable oils supplemented to the basal medicum stimulated mycelial growth of Flammulina velutipes. The mycelial yield was increased 3.5 folds by addition of 3% (v/v) ricebran oil. Maximum mycelial yield (18.2mg/ml) was obtained by addition of 3.0% ricebran oil with 1.0% $CaCl_2$ to the basal medium. There was no significant difference between the liquid and solid spawn in the yield of sporophores.

Effects of Temperature and Moisture on the Survival of Colletotrichum acutatum, the Causal Agent of Pepper Anthracnose in Soil and Pepper Fruit Debris

  • The survival of Colletotrichum acutatum was investigated in soil, infected fruits, and infected fruit debris incorporated into soil at several temperatures with different soil moisture levels. Samples were examined at 2-week intervals for 18 weeks to determine the survival of the pathogen based on the number of colony forming unit (CFU) of C. acutatum recovered on a semi-selective medium. C. acutatum conidia survived in both sterile and non-sterile soil at 4 and $10^{\circ}C$ for 18 weeks. If infected pepper fruits were completely dried, C. acutatum survived for 18 weeks at temperature from 4 to $20^{\circ}C$. Soil temperature and moisture affected the survival of C. acutatum in infected fruit debris incorporated into soil after air-drying. The effect of soil moisture on survival was weaker at low temperatures than at high temperatures. For up to 16 weeks, conidia were recovered from fruit debris in soil that had been kept at 4 to $20^{\circ}C$ and below 6% soil moisture. Conidia were recovered from fields until approximately 6 months after pepper fruits were harvested. Using PCR with species-specific primers and a pathogenicity test, we identified conidia recovered from soil and infected fruit from both the laboratory and field as C. acutatum and as the primary inoculum causing pepper anthracnose.
