Biological Activities of Water and Ethanolic Extracts from Allium victorialis L. Mature Leaves

  • Allium victorialis L. (A. victorialis) is a very popular vegetable in Korea. The most commonly used parts of this vegetable are the bulbs and young leaves. To determine if the mature leaves have any beneficial properties, we investigated antioxidant, anti-${\alpha}$-glucosidase, anti-inflammatory, and anticancer activities of water and ethanol extracts from A. victorialis. Antioxidant activity was evaluated by measuring total phenolic content, DPPH and superoxide radicals scavenging activities. The water extract from A. victorialis ($W{\cdot}A$. victorialis) exhibited higher antioxidant ability than the ethanol extract ($E{\cdot}A$. victorialis). Moreover, the water extract showed strong inhibitory effect on ${\alpha}$-glucosidase. On the other hand, the ethanol extract had greater anti-inflammatory activity on murine macrophage cells (RAW 264.7) and greater anticancer activities against human colon cancer cells (HT-29). These results suggest that mature leaves from E·A. victorialis may have health-enhancing effects.

고지방식이 비만 마우스에 대한 야채 조성물의 항비만 효과 (Anti-obesity Effect of Vegetable Formula on Obese Mice in High Fat Diet)

  • This study was conducted to investigate the cholesterol and Triglyceride (TG) lowering and anti-obesity effects of water extract of vegetable formula (onion 25%, carrot 20%, Cabbage 20%, Sweet Pumpkins 20%, Chinese plum 5%, turmeric 5%, morus leaves 5%, SLS) in mice fed high fat-diet. ICR mice were divided into 3 groups; a normal diet group (ND), a high-fat diet group (HFD), a high-fat diet and SLS with 300 mg/kg treated group (HFD+SLS). Body fat gain was increased by high-fat diet and HFD+SLS group showed a definite weight loss. The total cholesterol level in the HFD+SLS group was 328.4 mg/dl which was 27.4% lower than that in the HFD group and the cholesterol-lowering effect of SLS was confirmed. The HFD + SLS group showed 118.1 mg/dl and the triglyceride level in the serum was decreased by 88.7% compared to the HFD group and SLS significantly decreased blood triglyceride levels. RT-PCR showed that the expression of PPAR-${\gamma}$ and the target gene SCD-1 was inhibited in a dose-dependent manner in the SLS-treated group. These results suggest that the SLS water extract may have a cholesterol and triglyceride-lowering effect and inhibit the expression of PPAR-${\gamma}$ and SCD-1 to have an anti-obesity effect.

  • Sixteen vegetable food extracts were screened for the presence of saponin with the methods of foaming test, hemolytic test and Liebermann-Burchard reaction. Six samples were found to contain saponin. The ethanal extract of six positive samples and its hydrolysates, sapogenin parts and sugar parts were applied t.l.c to detect saponin moiety.

Development of Fruit and Vegetable Peels Extracts for Memory Improvement of Prevention and Treatment of Cognitive Impairment

  • This study relates to a composition for improvement of memory or prevention and treatment of cognitive impairment using waste resources rich in beneficial substances. This study makes good effects to inhibit the activity of acetylcholinesterase in brain tissue and to improve the cognitive functions in a simulation model of cognitive impairment induced by scopolamine, so it can be available in the promotion of memory and the prevention and treatment of cognitive impairment. The composition uses the extract of fruit peels, which have long been used without causing toxicity in a wide range of food applications; therefore, it can be used safely without a risk of side effects, even in the case of a long-term administration for the preventive purpose. Furthermore, this research is a very beneficial invention in the environment-friendly aspect in association with the recycling of resources, as it is based on the novel efficacies of fruit peels, which have been conventionally disposed as a refuse of fruits due to their poor sensory qualities despite the content of beneficial substances.

Phenolic constituents and biological activities of leaf extracts of traditional medicinal plant Plectranthus amboinicus Benth (Lamiaceae)

  • Plectranthus amboinicus Benth (Lamiaceae) is a medicinal plant native to India, and its leaves are widely used in several traditional medicinal preparations. The purpose of this study was to detect and quantify phenolics present in ethyl acetate and acetone extracts of P. amboinicus leaves, and evaluate their antioxidant, antibacterial, antimutagenic and anticancer activities. The HPLC chromatograms of crude leaf extracts indicated the presence of phenolics like caffeic acid, coumaric acid, rutin, quercetin and gallic acid, which were present in the range of 0.01 - 1.41 mg/g in ethyl acetate and 0.03 - 1.93 mg/g in the acetone extract. The acetone extract showed statistically (p < 0.05) higher antioxidant activity ($IC_{50}$, 99.59 ${\mu}g/ml$) than ethyl acetate extract ($IC_{50}$, 149.96 ${\mu}g/ml$). Statistically (p < 0.05) higher antimutagenicity was shown by acetone extract (46.16%) as compare to ethyl acetate extract (12.16%) at 500 ${\mu}g/plate$ concentration. The acetone extract showed higher antibacterial activity than ethyl acetate extract, and both the extracts showed highest activity against B. cereus (375 and 625 ${\mu}g/ml$, respectively) and lowest activity against Y. enterocolitica (1000 and 1125 ${\mu}g/ml$, respectively). Both the extracts also showed inhibitory effect on cancer cell lines HCT-15 and MCF-7. These results suggest that the leaves of P. amboinicus possess various biological activities, and validate the traditional use of the leaves of P. amboinicus against cold, infection and ulceration.

  • 본 연구에서는 참외의 부위별 물 및 알코올 추출물의 항산화효과, ${\alpha}$-Glucosidase 저해활성 및 항균효과를 조사하였다. 총 phenol 함량을 측정한 결과 물 추출물에서는 껍질이 151.64 ${\mu}g/g$, 에탄올 추출물에서는 껍질이 224.77 ${\mu}g/g$로 가장 높은 페놀함량을 나타내었다. 총 flavonoid 함량은 껍질이 물 추출물의 경우 45.53 ${\mu}g/g$, 에탄올 추출은 338.37 67.16 ${\mu}g/g$로 가장 높게 나타내었다. 참외의 항산화 효과는 물 추출물의 경우 1%에서 껍질이 25%로 가장 높았으며, 에탄을 추출은 1%에서 껍질이 83.3로 가장 높은 전자공여능을 나타내었으며, 과육과 태좌는 20% 미만의 낮은 활성을 나타내었다. ABTS는 물 추출물은 1%에서 껍질이 79.2%로 가장 높았으며, 과육이 57.6%, 태좌가 74.0%로 50%이상의 활성을 나타내었고, 에탄올추출물은 99.9%로 껍질이 가장 높게 나타났고, 과육과 태좌는 52.1%, 41.2%의 활성을 보였으며, Xanthine oxidase의 저해활성의 경우 1%에서 껍질이 물 추출물이 29.3%, 에탄올추출물이 30.5%의 가장 높은 활성을 보였지만, 비교적 낮은 저해율을 나타내었다. ${\alpha}$-Glucosidase 저해활성을 측정한 결과 1%에서 열수 추출물은 25.9%, 에탄올 추출물은 37.5%의 활성을 보인 껍질이 가장 높은 활성을 보였다. 또한 Streptococcus agalactiae, Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Candida albicans, Staphylococcus epidermidis 및 Salmonella typimimum에 대한 항균활성을 평가한 결과 참외의 에탄올 추출물에서 Streptococcus agalactia에서만 껍질, 과육, 태좌에서 각각 15, 13 및 12mm의 clear zone를 나타내었다.

Sanghuangporus sanghuang extract inhibits the proliferation and invasion of lung cancer cells in vitro and in vivo

  • BACKGROUND/OBJECTIVES: Sanghuangporus sanghuang (SS) has various medicinal effects, including anti-inflammation and anticancer activities. Despite the extensive research on SS, its molecular mechanisms of action on lung cancer are unclear. This study examined the impact of an SS alcohol extract (SAE) on lung cancer using in vitro and in vivo models. MATERIALS/METHODS: Different concentrations of SAE were used to culture lung cancer cells (A549 and H1650). A cell counting kit-8 assay was used to detect the survival ability of A549 and H1650 cells. A scratch assay and transwell cell invasion assay were used to detect the migration rate and invasive ability of SAE. Western blot analysis was used to detect the expression of B-cell lymphoma-2 (Bcl-2), Bcl2-associated X (Bax), cyclin D1, cyclin-dependent kinases 4 (CDK4), signal transducer and activator of transcription 3 (STAT3), and phosphorylated STAT3 (p-STAT3). Lung cancer xenograft mice were used to detect the inhibiting ability of SAE in vivo. Hematoxylin and eosin staining and immunohistochemistry were used to detect the effect of SAE on the structural changes to the tumor and the expression of Bcl-2, Bax, cyclin D1, CDK4, STAT3, and p-STAT3 in lung cancer xenograft mice. RESULTS: SAE could inhibit lung cancer proliferation significantly in vitro and in vivo without cytotoxicity. SAE suppressed the viability, migration, and invasion of lung cancer cells in a dose and time-dependent manner. The SAE treatment significantly decreased the proapoptotic Bcl-2/Bax ratio and the expression of pro-proliferative proteins Cyclin D1 and CDK4 in vitro and in vivo. Furthermore, SAE also inhibited STAT3 expression. CONCLUSIONS: SAE reduced the cell viability and suppressed cell migration and invasion in human lung cancer cells. Moreover, SAE also exhibited anti-proliferation effects in vivo. Therefore, SAE may have benefits in cancer therapy.

스트렙토조토신으로 유도된 흰쥐에서 곤달비 추출물의 항당뇨 효과 (Anti-diabetic Effect of the Methanolic Extract of Ligularia stenocephala Leaves in the Streptozotocin-induced Rat)

  • 곤달비 메탄올 추출물은 100mg/kg 및 200mg/kg의 경구투여로 STZ로 유도된 당뇨병의 제반 증상을 완화하였다. 즉, STZ의 투여는 흰쥐의 체중, 수분 섭취, 식이섭취를 증가시켰으며, 혈당량, 혈청 triglyceride 량, 동맥경화 위험지수를 증가시켰으나, 곤달비 추출물의 투여는 이러한 당뇨병에 관련된 증상을 개선하였다. 이와 더불어, 위의 당뇨병 모델 쥐에서 당내성 실험에서도 곤달비 추출물이 현저한 효과를 나타내었으므로 혈당의 세포내 이행을 촉진시키는 것으로 확인되었다. 이상과 같은 결과로부터 곤달비 추출물은 당뇨병 및 그에 따르는 고지혈증에 유효하였으므로 당뇨병 질환의 완화를 위하여 사용될 수 있는 산채 식품인 것으로 나타났다. 이러한 결과를 바탕으로 곤달비의 활성 물질의 입증과 그 이용에 관한 계속적인 연구가 필요한 것으로 생각된다.
