The coat protein (CP) gene of cucumber mosaic virus-As (CMV-As) strain was engineered for expression in the plant by using the cauliflower mosaic virus 35S transcript regulatory sequences. The CP gene was cloned into an Agrobacterium-derived binary vector. A chimeric gene was constructed by the cDNA of CMV-As CP and plant expression vector pBI121. The clone, pCMAS66, was first introduced into the phagemid vector pSPORT1 for situating sense orientation for translation and making restriction sites in order to re-introduce plant expression vector, pHI121. The resulting subclone pCASCP02 and plant expression vector pBI121 were treated with BamHI-SacI for excising the target gene and removing GUS gene, respectively. After Agrobacterium transformation by freeze-thaw technique, the clone, pCMASCP121-123 which contains sense orientation of the target gene, was selected and confirmed by restriction endonuclease analysis. The CMV-As CP gene was introduced into A. tumefaciens. The results on tobacco plant transformation with the vector system revealed that the system could be successfully introduced and showed high frequency of selection to putative transformations.
An advanced direct torque control (DTC) with pulse width modulation (PWM) method is presented in this paper. The duty ratio calculation of the selected voltage vector is based on the voltage functions of the selected voltage vector according to the sector angle. The proposed DTC uses a conventional DTC scheme with six sector divisions and switching rules. However, the winding voltages are supplied by the PWM approach. Furthermore, the duty ratio of the switching voltage vector is determined by the flux, torque error, and motor speed. The base voltage that shall determine the duty ratio can be calculated by approximate voltage functions according to the voltage angle. For the calculation of base voltages, second-order quadratic functions are used to express the output voltage of the selected voltage vector according to voltage angle. The coefficients for the second-order quadratic functions are selected by the voltage vector, which is determined by the switching rules of the DTC. In addition, the voltage functions are calculated by the coefficients and voltage angle between the voltage vector and rotor position. The switching voltages from the calculated duty ratio can supply the proper torque and flux to reduce the ripple and error. The proposed control scheme is verified through practical experimental comparisons.
본 연구는 최근 들어 DDoS 공격이 단순히 서비스를 방해하는 것에서 벗어나, 다양한 공격 기법을 혼합한 멀티벡터(Multi-Vector) 공격으로 발전하고 있다. 이러한 멀티벡터 공격은 DDoS 공격과 더불어 악성코드를 감염시켜, 내부 정보 유출 및 좀비PC를 만들어 DDoS 공격용에 활용될 경우에는 기존의 DDoS 공격 및 악성코드 감염에 대한 방어 전략으로는 한계점이 있다. 따라서 본 논문에서는 다양한 방법을 이용한 멀티벡터 공격을 효과적으로 방어하기 위한 Reverse Proxy Group과 PMS(Patch Management Server)를 제시하고자 한다.
외연적 유한요소법은 벡터처리에 적합한 구조를 가지고 있어 벡터컴퓨터를 이용하면 기존의 스칼라 컴퓨터에서보다 휠씬 빠르게 해석을 수행할 수 있다. 본 논문에서는 memory-to-memory방식의 벡터컴퓨터에서의 외연적 유한요소법의 효율적인 벡터화 방법을 제시하였다. 먼저 벡터컴퓨터의 구조적 특성과 무관하게 적용될 수 있는 일반적인 벡터화 기법을 고찰한 후 memory-to-memory방식의 벡터컴퓨터에 적합한 벡터화 기법을 개발하였다. 개발된 벡터화 기법의 유용성을 확인하기 위해 외연적 유한요소 프로그램인 DYNA3D를 memory-to-memory방식의 벡터컴퓨터인 HDS AS/XL V50에 이식한 결과 스칼라에 비해 2.4배 이상의 성능 향상을 얻을 수 있었다.
Viral and non-viral vectors have been used in the delivery of genetic materials into animal cells and tissues, with each approach having pros and cons. Non-viral vectors have many useful merits such as easy preparation, low immunity and size tolerance of a transgene when compared to those of viral vectors. Delivery specificity may be achieved by complex formation between receptor ligands and a non-viral vector. In the present study, non-viral vector systems are investigated in an effort to find a practical delivery means for gene therapy, Receptor-ligand interaction between transferrin-receptor and transferrin was utilized for efficient gene transfer into cancer cells. A plasmid vector, pcDNA3 (LacZ) was ligated with a small duplexed oligo fragment in which a Biotin- VN$^{TM}$ phosphoramidite was placed in the middle of the oligo. The plasmid vector labeled by biotin was then conjugated with biotin-labeled transferrin via streptavidin. This trimeric conjugates were delivered to a hepatoma cell line, HepG2. The delivery efficiency of the trimeric conjugate was 2-fold higher than that of cationic liposomes used for transfection of a plasmid vector. These results demonstrate that a plasmid vector can be efficiently transferred into cells by forming a trimeric complex of plasmid vector-linker-ligand.
In this paper, by managing the biometric data is changed with the passage of time, a systematic and scientifically propose a framework to increase the bio-vector generation efficiency of the smart health care. Increasing the development of human life as a medicine and has emerged smart health care according to this. Organic and efficient health management becomes possible to generate a vector when the biological domain to the wireless communication infrastructure based on the measurement of the health status and to take action in accordance with the change of the physical condition. In this paper, we propose a framework to create a bio-vector that contains information about the current state of health of the person. In the proposed framework, Bio vectors may be generated by collecting the biometric data such as blood pressure, pulse, body weight. Biometric data is the raw data from the bio-vector. The scope of the primary data can be set to active. As the collecting biometric data from multiple items of the bio-recognition vectors may increase. The resulting bio-vector is used as a measure to determine the current health of the person. Bio-vector generating the proposed framework, it can aid in the efficiency and systemic health of healthcare for the individual.
This paper presents an algorithm of the watermark insertion and extraction on the vector image. Most parts of the vector image consist of the array of the coordinate values. The vector watermarking method by Sakamoto et al [1] uses the mask within which all the coordinate values of all the vertices are changed depending on the value of the watermark. The proposed algorithm is the change of the vector image file instead of the change of the coordinate values on the vector image. We use the Drawing Exchange Format (DXF) files since the DXF file is one of the general files in the field of the vector image.
본 논문에서는 영상의 프레임률을 증가시키기 위한 새로운 움직임 벡터 평활화 기법과 움직임 보상 기법에 대해 제안한다. 제안하는 움직임 벡터 평활화 방법은 적응적 가중 벡터 중간값 필터를 사용하는 기존 방법의 한계를 극복하기 위해 정확도를 반영하여 가중치를 개선함으로써 잘못된 움직임 평활화를 줄인다. 또한 기존 움직임 보상 기법인 OBMC로 보간된 영상의 화질을 향상시키기 위해 원래의 블록을 4등분하여 등분된 각 블록을 기준으로 움직임 벡터를 재추정하고 이를 통해 보간 프레임을 생성하는 방법을 제안하다. 모의실험 결과는 제안된 방법이 객관적 측면과 주관적 측면에서 기존 기법에 비해 우수한 화질을 제공할 수 있음을 보인다.
알코올 생산성이 높은 Zymomonas 균주의 기질 이용성을 넓히기 위한 목적으로 natural replicon을 포함하며 적당한 항생제 저항표지를 갖는 plasmid vector의 제조를 시도하였다. Z. mobilis ATCC10988에서 분리된 몇 개의 plasmid중 3.9kb의 적당한 크기를 갖는 pZM3를 선정하여 수종의 제한효소로 처리하여 절편의 크기에 따라 유전자 지도를 작성하였다. pZM 3의 replicon과 pBR 325의 chloramphenicol 저항유전자를 포함한 재조합 plasmid인 pHZ22를 개발하고 이 plasmid vector가 숙주세포인 Z. mobilis ATCC31821에서 독립적으로 replication됨을 확인하였다. 또 하나의 항생제 저항표지로서 RP4의 tetracycline 저항유전자를 분리하여 pHZ22에 도입함으로써 pHZT224를 제조하였는데 이 plasmid vector도 Zymomonas로 conjugation에 의해 전이되어 안정하게 유지 되었다. 본 연구를 통하여 개발된 plasmid vector는 Z. mobilis와 E. coli에 공히 작용하는 shuttle vector 로서 외부 유전자를 Zymomonas에 도입시킬 수 있는 유용한 유전자 운반체임이 확인되었다.
Ferritin light heavy chain (FLHC) gene는 일부 중금속과 결합, 저장 및 운반하여 무독화 시킬 수 있는 것으로 알려져 있다. Fe 관련 유전자인 FLHC유전자를 식물 발현용 promoter인 35S promoter와 Tnos를 사용하여 식물 형질전환용 vector를 재조합하였다. 식물세포형질전환용 binary vector는 상기 cassette vector가 조립이 매우 양호하며 border sequence를 가지고 있는 pRD400 binary vector를 사용하여 최종적으로 가나마이신 내성 유전자 (NPT II gene)와 tadpole ferritin heavy chain gene 및 human ferritin light chain gene를 함유하고 있는 binary vector를 재조합하였다. Binary vector의 아그로박테리움에 도입은 triparental mating 방법에 의하여 수행하여 AB배지 및 가나마이신 함유 배지에서 disarmed Ti-vector를 가지고 있는 Agrobacterium tumefaciens MP90/FLHC을 선발하였다. FLHC 유전자 도입된 식물형질전환용 binary vector를 이용하여 형질전환방법을 변형하여 많은 embryo를 유도하였으며 유도된 embryo들은 GA 10mg/L가 첨가된 배지에 지상부를 유도하였다. 형질전환체식물체의 정상적인 생장을 유도하기 위해 최적의 배양조건을 조사하였던 바, 비교적 1/3 MS배지에서 뿌리의 생장과 지상부의 생장이 균일하게 생장하는 경향을 보였으며, 뿌리와 줄기가 잘 발달된 약 7cm의 유식물체를 대량으로 증식하여, 모래와 흙이 1:1로 혼합된 토양에 옮겼다.
