예방접종 동시접종에 대한 접종의사의 인식 조사 (A Survey of Doctors' Awareness of Simultaneous Vaccination)

  • 목적: 예방접종을 시행하는 의사들에게 설문지를 이용해 동시접종 시행 여부와 안전에 대한 염려가 없는가를 조사하여, 동시접종 시행 시 문제점을 파악하고자 하였다. 방법: 조사자가 부산 6개 행정구 내에 있는 국가필수예방접종 사업 등록기관을 방문하여 의사들에게 설문을 실시하였다. 결과: 응답자 155명 중 144명(93%)이 동시접종을 시행하고 있었으며, 141명(91%)이 동시접종에 긍정적인 반응을 보였다. 동시접종을 시행하고 있는 144명 중 67명(47%)은 동시접종의 안전성에 대해서 확신을 가지지 못한 채 접종을 시행 중이었으며, 동시접종 시행 중 부작용을 경험한 86명 중 35명(41%)은 동시접종이 단독접종보다 부작용 빈도가 높을 수 있다고 생각하는 것으로 나타났다. 결론: 동시접종은 높은 비율로 시행 중인 것으로 나타났지만, 동시접종을 시행하는 의사들 중 안전성에 대해 확신을 가지지 못하는 비율이 높게 나타났다. 원활한 접종 시행을 위해 이런 의문을 해소할 수 있는 근거를 제시할 필요가 있다.

COVID-19에 대한 운동중재효과 분석 (Analysis of Physical Status on COVID-19: Based on Impacts of Physical Activity)

  • 본 연구는 COVID-19과 COVID-19 변이 바이러스 시기에 vaccination을 통한 집단면역달성을 목표로 하고 있지만, 백신의 종류에 따라서 다양한 부작용도 발생을 하여, 백신 이외에 평소 규칙적인 운동을 통한 백신 효과의 조그만 근거를 마련하고자 수행하게 되었다. 이러한 연구 연구목적을 달성하기 위해 본론에서는 COVID-19 vaccine의 이슈, COVID-19 블루, COVID-19과 스포츠이벤트, 및 COVID-19과 운동중재에 관해 분석하였다. 본 연구결과 COVID-19은 혈관을 우선적으로 공격하고 혈관 내막에 침투해 혈액순환을 방해하는 바이러스로 알려져 있다. 그리고 이어서 폐, 심장, 뇌 등을 공격한다. 심혈관 질환이 있는 환자들은 몸에서 바이러스를 몰아내는 능력이 떨어져 있는 이유로 이러한 바이러스의 공격대상이 될 수 있다. 혈관은 적혈구와 영양소만 이동하는 곳이 아니라 백혈구와 림프구들도 이동하므로 평소 불포화 지방산, 저탄수화물 식이요법을 포함하는 건강한 식이 습관, 금연, 일상생활도의 증가 및 규칙적인 운동 습관의 중요성이 중요하게 고려되어 진다. 이러한 관점에서 평소 혈관의 건강은 매우 중요하게 작용하고 이는 규칙적인 운동을 통하여 얻을 수 있는 것이다. 규칙적인 운동은 면역세포들, 면역글로불린, 다양한 사이토카인들의 순환을 돕고 특히, 폐로의 순환을 도와 염증인자들을 감소시키는 역할을 수행하고 병원균의 유입을 막아주는 역할을 한다. 특히, 혈관 내막에 침투해 혈액순환을 방해하는 것으로 알려져 있으며 폐, 심장, 뇌 등을 공격하는 COVID-19의 특성을 고려할 때 운동을 통한 건강관리는 바이러스 예방에 필수적이라고 할 수 있다. 현재 다양하고도 많은 백신의 사용은 팬데믹 시기에서 매우 희망적이지만 아나플락시스나 혈전 색전증 등의 다양한 부작용도 초래하므로 규칙적인 운동중재는 백신의 도움없이 세균이나 바이러스로부터 나를 지키는 면역력을 잘 유지하게 하는 것이 규칙적인 운동이 되는 것이다. 추후 이러한 분야에 관한 심도 있는 연구들이 수행되어져야 할 것으로 여겨진다.

딸의 자궁경부암 예방 관련 HPV 백신접종에 대한 어머니 수용도와 영향요인 (Factors Influencing Mothers' Acceptance of Human Papillomavirus Vaccination to Prevent Cervical Cancer in their Daughters)

  • Purpose: This study was done to examine mothers' acceptance and its influencing factors in daughters' human papillomavirus (HPV) vaccination to prevent cervical cancer. Methods: From July 20 to August 31, 2010, 220 mothers of unmarried daughters responded to self-administered questionnaires about their acceptance of HPV vaccination for their daughters and the optimal age for HPV vaccination. Descriptive statistics, univariate logistic and multiple logistic regression were used for data analysis with SPSS/WIN 12.0. Results: Mothers indicating their willingness to pay for their daughters to receive HPV vaccination accounted for 61.8%, and willingness with health insurance coverage, 84.5%. Mean optimal age for HPV vaccination was $19.78{\pm}3.96$) years. With self-payment the factor influencing mothers willingness was necessity of HPV vaccination (OR= 30.45, 95% CI=10.33~89.15). With health insurance coverage, income (OR=3.02, 95% CI: 1.19~7.62), necessity of HPV vaccination (OR=15.05, 95% CI=4.15~54.64), concern about HPV vaccine safety (OR=2.99, 95% CI=1.15~7.76), and experience of Pap test (OR=3.16, 95% CI=1.24~8.05) were factors influencing mothers willingness. Other influencing factors were optimal age for HPV vaccination, knowing about HPV (OR=7.66, 95% CI=2.19~26.82), and age of youngest daughter (OR=3.95, 95% CI=1.34~11.68). Conclusion: Financial support is anticipated for low income families in a primary approach to increase HPV vaccination. And focusing on earlier age and concerns about vaccination are necessary to gain mothers' acceptance.

넙치, Paralichthys olivaceus에 투여된 Edwardsiella tarda와 Streptococcus iniae에 대한 혼합백신의 예방효과와 성장에 미치는 영향 (The efficacy and influence on growth of olive flounder Paralichthys olivaceus vaccinated against Edwardsiella tarda and Streptococcus iniae)

  • 본 연구는 넙치, Paralichthys olivaceus에서 Edwardsiella tarda와 Streptococcus iniae에 대한 혼합백신의 효능과 안전성에 대해 조사하고자 하였다. 혼합백신을 주사한 넙치에서 2, 4, 6, 8주 째에 채혈하여 E. tarda와 S. iniae에 대한 항체가 를 microtitre 법으로 조사하였다. 백신 주사 후 3주째에 E. tarda와 S. iniae를 각각 인위감염 시켰을 때 백신구에서 81.0-92.8%의 높은 상대생존율을 보여 혼합백신에 의한 항병력의 증가가 확인되었다. 백신 주사 후 초기에 백신구가 대조구에 비해 체중 증가가 느렸으나 사육기간이 길어지면서 회복되어 대조구와 백신구 체중의 차이를 보이지 않았다. 이 결과는 백신 주사가 넙치의 성장에 영향을 주지 않고 에드와드병과 연쇄구균증을 예방할 수 있음을 보여준다.

Current Status, Challenges, Policies, and Bioethics of Biobanks

  • Many biobanks were established as biorepositories for biomedical research, and a number of biobanks were founded in the 1990s. The main aim of the biobank is to store and to maintain biomaterials for studying chronic disease, identifying risk factors of specific diseases, and applying personalized drug therapies. This report provides a review of biobanks, including Korean biobanks and an analysis of sample volumes, regulations, policies, and ethical issues of the biobank. Until now, the top 6 countries according to the number of large-scale biobanks are the United Kingdom, United States, Sweden, France, the Netherlands, and Italy, and there is one major National Biobank of Korea (NBK) and 17 regional biobanks in Korea. Many countries have regulations and guidelines for the biobanks, and the importance of good management of biobanks is increasing. Meanwhile, according to a first survey of 456 biobank managers in the United States, biobankers are concerned with the underuse of the samples in their repositories, which need to be advertised for researchers. Korea Biobank Network (KBN) project phase II (2013-2015) was also planned for the promotion to use biospecimens in the KBN. The KBN is continuously introducing for researchers to use biospecimens in the biobank. An accreditation process can also be introduced for biobanks to harmonize collections and encourage use of biospecimens in the biobanks. KBN is preparing an on-line application system for the distribution of biospecimens and a biobank accreditation program and is trying to harmonize the biobanks.

한탄바이러스 76-118을 이용한 치사 동물모델 확립 (Establishment of a Lethal Animal Model of Hantaan Virus 76-118 Infection)

  • Hantaan virus(HTNV) causes hemorrhagic fever with renal syndrome(HFRS) with a case fatality rate ranging from <1 to 15 % in human. Hantavax is a vaccine against the Hantavirus, which has been conditionally approved by the Ministry of Food and Drug Safety(MFDS). However, only 50 % of volunteers had neutralizing antibodies 1 year following the boost. Effective antiviral treatments against HTNV infection are limited. Hantaviruses generally cause asymptomatic infection in adult mice. On the other hand, infection of suckling and newborn mice with hantaviruses causes lethal neurological diesease or persistant infection, which is different from the disease in humans. The development of vaccines and antiviral strategies for HTNV has been partly hampered by the lack of an efficient lethal mouse model to evaluate the efficacy of the candidate vaccines or antivirals. In this report, we established a lethal mouse model for HTNV, which may facilitate in vivo studies on the evaluation of candidate drugs against HTNV. The median lethal dose value of HTNV was calculated by probit analysis of deaths occurring within two weeks. Five groups of ten ICR mice were injected intracranially with serial 2-fold dilutions (from 50 to 3.125 PFU/head) of HTNV. Mice injected with HTNV began to die at 8 days post-infection. The lethal dose required to kill 50 % of the mice (LD50) was calculated to be 2.365 PFU/head.

바리과 어류 3종(능성어 Hyporthodus septemfasciatus, 붉바리 Epinephelus akaara, 자바리 Epinephelus bruneus)에 대한 신경괴사증바이러스(Nervous Necrosis Virus, NNV)의 생백신 처리 효과 (Efficacy of live NNV immersion vaccination in three grouper species (Seven band grouper Hyporthodus septemfasciatus, Red spotted grouper Epinephelus akaara, Long tooth grouper Epinephelus bruneus))

  • In this study, we investigated safety and efficacy of a low temperature immunization protocol with NNV in red spotted grouper, Epinephelus akaara and long tooth grouper, Epinephelus bruneus and seven band grouper, Hyporthodus septemfasciatus. Further, growth rate between immunized and naïve fish was evaluated during the experiment to check side effect of immunization. Three grouper species were immunized by immersion method with live NNV at 105.0 TCID50/mL at 16.5℃ for 30 min and reared for 120 days at natural sea water temperature. To evaluate growth rate, total length and wet weight was measured 7 times after immunization. Immunized three grouper species were challenged by intramuscular inoculation with NNV at 104.2 TCID50/100 µL/fish. Immunization at low temperature with live NNV did not show any clinical symptoms of infection, mortality and inhibition of growth. After challenge, cumulative mortality of naïve seven band grouper, red spotted grouper, long tooth grouper were 45, 10, 20 %, respectively. However no mortality was observed at immunized groupers. Thus, it was demonstrated that immunization at low temperature with live NNV are able to protect three different species of groupers without inhibition of growth.

Bibliometric analysis on the evolution of knowledge structure of African swine fever

  • Since African swine fever (ASF) spread to East Asia, a fatal crisis has occurred in the global pig industry, because Asia is dominant in pig production. Although some studies conducted bibliometric analysis on ASF, few studies compared research networks, and identified subthemes by major keywords. To fill this gap, this study identified the knowledge structure network of the research, its influence, and core research themes by utilizing the bibliometric analysis of 337 ASF-related journal articles over 50 years from 1970 to 2020 on the Web of Science. The result indicated that papers are mainly published in the fields of veterinary science, virology, microbiology, infectious disease and applied microbiology, and in particular, the fields of veterinary science and virology showed unrivaled weights as they account for 73.40%. With regard to cooperative relationships, European countries such as the UK, Germany, Italy, and Denmark, centered on Spain, are actively contributing to the ASF research. China, France, Thailand, Japan, Vietnam, and South Korea are leading research cooperation, centering on the United States. In the early stage of the studies, major keywords appeared to be related to outbreaks, quarantine and diagnosis, and in the middle stage, the keywords were expanded to a wide range of pig diseases. Recently, the keywords are becoming more diverse towards antibodies, cross-border transmission and disease monitoring. Based on data on major keywords related to ASF, this study proposed discussions and implications for activating ASF research including genotype, protein, vaccine, diagnosis, defense against infection and epidemiological investigation.

DA-6034 ameliorates hepatic steatosis and inflammation in high fat diet-induced obese mice

  • Background: Nonalcoholic fatty liver disease (NAFLD) is characterized by an increase in hepatic triglyceride content and increased inflammatory macrophage infiltration through the C-C motif chemokine receptor (CCR) 5 pathway in the liver. DA-6034 (7-carboxymethyloxy-3',4',5-trimethoxy flavone), is a synthetic derivative of eupatilin that exhibits anti-inflammatory activity in inflammatory bowel disease. However, the effect of DA-6034 on the inflammatory response in NAFLD is not well elucidated. Therefore, we aimed to determine the effect of DA-6034 on hepatic steatosis and inflammation. Methods: Forty male C57BL/6J mice were divided into the following four groups: (1) regular diet (RD), (2) RD with DA-6034, (3) high fat diet (HFD), and (4) HFD with DA-6034. All mice were sacrificed 12 weeks after the start of the experiment. The effects of DA-6034 on macrophages were assessed using RAW 264.7 cells. Results: DA-6034 not only reduced hepatic triglyceride levels and lipid accumulation but also macrophage infiltration and proinflammatory cytokines in HFD-fed mice. According to fluorescence-activated cell sorter analysis, DA-6034 reduced the CD8+ T cell fraction in the liver of HFD-fed mice. DA-6034 also reduced CCR5 expression and the migration of liver macrophages in HFD-fed mice and inhibited CCR2 ligand and CCR4 ligand, which stimulated the migration of macrophages. Conclusion: Overall, DA-6034 attenuates hepatic steatosis and inflammation in obesity by regulating CCR5 expression in macrophages.

  • The global crisis caused by the coronavirus disease 2019 (COVID-19) led to the most significant economic loss and human deaths after World War II. The pathogen causing this disease is a novel virus called the severe acute respiratory syndrome coronavirus 2 (SARSCoV-2). As of December 2020, there have been 80.2 million confirmed patients, and the mortality rate is known as 2.16% globally. A strategy to protect a host from SARS-CoV-2 is by suppressing intracellular viral replication or preventing viral entry. We focused on the spike glycoprotein that is responsible for the entry of SARS-CoV-2 into the host cell. Recently, the US Food and Drug Administration/EU Medicines Agency authorized a vaccine and antibody to treat COVID-19 patients by emergency use approval in the absence of long-term clinical trials. Both commercial and academic efforts to develop preventive and therapeutic agents continue all over the world. In this review, we present a perspective on current reports about the spike glycoprotein of SARS-CoV-2 as a therapeutic target.
