Thin-film optical waveguide $K^{+}$-ion sensor using the evanescent field absorption (소산장 흡수를 이용한 박막 광도파로형 칼륨이온센서)

  • A thin film optical waveguide sensor has been developed to measure and analyze quantitatively some inherent optical properties of biochemical substances. In this paper, two different kinds of thickness of thin film waveguide were prepared by RF sputtering of Corning-7059 glass(n = 1.588 at ${\lambda}=\;514nm$, Ar laser) on Pyrex glass substrates. We made a sensing membrane coated on the thin film waveguide with the poly(vinyl chloride-co-vinyl acetate-co-vinyl alcohol) (91 : 3 : 6) copolymer membrane based on $H^{+}$-selective chromoionophore and $K^{+}$-selective neutral ionophore and then proposed the thin film opptical waveguide ion sensor which can select a potassium ion. This sensor based ell the absorbance change by utilizing chromoionophore and neutral ionophore, which changes their absorption spectrum in the UV-vis region upon complexation of the corresponding ionic species, have been reported. The sensitivity dependence of the proposed sensor on interaction length, waveguide thickness, and content of a chromoionophore was investigated. This sensor has the measurement range of $10^{-6}M{\sim}1M$ for $K^{+}$ concentration and 90% response time of duration within 1 min. Also, our thin film optical waveguide sensor using the evanescent field was investigated as compared with conventional transmission sensor or optode sensor by the optical fiber. The sensitivity of thin-film waveguide $K^{+}$ sensor is higher than that of the conventional transmission sensor. The proposed sensor is expected to be useful to biochemical, medical, environmental inspection and so on.

Antioxidant, Anti-diabetic, Anti-cholinesterase, and Nitric Oxide Inhibitory Activities of Fruiting Bodies of Agaricus brasiliensis (신령버섯 자실체 메탄올 추출물의 항산화, 항당뇨 및 Nitric Oxide의 저해 효과)

  • Agaricus brasiliensis, belonging to the family Agaricaceae of Agaricales, Basidiomycota, has been used for edible and medicinal purposes. This study was initiated to evaluate the antioxidant, anti-diabetic, and nitric oxide inhibitory activities of fruiting bodies of A. brasiliensis extracted with methanol. The HPLC analysis of phenolic compounds from the mushroom extracts identified 10 phenolic compounds including gallic acid, procatechuic acid, chlorogenic acid, (-)-epicatechin, vanillin, rutin hydrate, naringin, quercetin, formononetin, and biochanin-A. The free radical scavenging activities of methanol extract were lower than that of positive control, BHT. The chelating effects of methanol extract were significantly higher than those of the positive control, BHT at the all concentrations tested. The methanol extract exhibited the lower reducing power activities compared with the positive control at the 0.5~6.0 mg/mL concentration. The mushroom extract inhibited the ${\alpha}$-glucosidase activity by 54.48% and 78.43% at the 1.0 and 2.0 mg/mL while acarbose, the positive control, inhibited the ${\alpha}$-glucosidase activity by 51.77% and 81.81% at the same concentrations, respectively. Nitric oxide (NO) production in lipopolysaccahride (LPS) induced RAW 264.7 cells were inhibited by the methanol extracts in a concentration dependent manner. Therefore, it is concluded that fruiting bodies of A. brasiliensis contained natural antioxidant, anti-diabetic, and anti-inflammatory substances which can be useful for human health.

Hazard Assessment of Combustion Gases from Interior Materials (주요 건축 내장재의 연소가스 유해성 평가)

  • Toxic gases from five types of interior building materials were investigated according to Naval Engineering Standard (NES) 713. The materials were plywood, indoor wall coverings (wood wall plate members and pine wood), reinforced Styrofoam insulation, laminate flooring, and PVC. Specimens were measured using an NES 713 toxicity test apparatus to analyze the hazardous substances in combustion gas from the materials. We used the US Department of Defense standard (MIL-DTL, Military Standard) to calculate the toxicity index of the combustion gas. Emissions of $CO_2$ from all specimens did not exceed the NES 713 limit of 100,000 ppm. The amount of CO gas emissions from reinforced Styrofoam insulation was 6,098 ppm. 25 ppm and 49 ppm of formaldehyde were released from the reinforced Styrofoam insulation and PVC flooring, respectively. These values were less than the limit of 400 ppm. The highest emissions were from $NO_X$ emitted by plywood and were above the limit of 250 ppm. The toxicity index of the specimens were calculated as 5.19 for plywood, 4.13 for PVC flooring, 2.35 for reinforced Styrofoam insulation, 2.34 for laminate flooring, and 1.22 for indoor wall coverings (pine wood). Our research helps us to understand the properties of these five interior materials by analyzing the combustion gas and explaining the toxicity of constituents and the toxicity index. Also, it would be useful for giving fundamentals to guide the safe use of interior materials for applications.

Biosorption of Heavy Metal in Aqueous Solution by Heavy Metal Tolerant Microorganism Isolated from Heavy Metal Contaminated Soil (중금속으로 오염된 토양에서 분리한 중금속 내성 미생물의 수용액내 중금속 흡착)

  • This study was conducted to find out a useful bioremediation technology for heavy metal contaminated soil and water. We isolated strain CPB from heavy metal contaminated soil and evaluated the tolerance level and adsorption capacity of strain CPB to heavy metals (Strain is not determined yet). Strain CPB showed variable tolerance limit to different kinds heavy metal or concentrations of heavy metals. The growth of strain CPB was significantly inhibited by mixed heavy metals (Cd+Cu+Pb+Zn) than that of by single heavy metal. Strain CPB showed high binding capacity with Pb (Pb>Cd>Cu>Zn). In general, strain CPB showed high uptake of heavy metals such as Pb, Cd and Cu. It was observed that the capacity of heavy metal uptake from mixture of heavy metals was reduced in comparison with single heavy metal treatment. But total contents of heavy metal bound with cell in mixed heavy metal showed higher than in single heavy metal treatment. Heavy metal adsorption in cells was affected by several external factors, such as temperature and pH etc.. The optimum temperature and pH of the adsorption of heavy metal into cells were ca. $25{\sim}35^{\circ}C$ and pH ca. $5{\sim}7$, respectively. A large number of the electron dense particles were found mainly on the cell wall and cell membrane fractions, which was determined by transmission electron microscope. Energy dispersive X-ray spectroscopy revealed that the electron dense particles were the heavy metal complexes the substances binding with heavy metals.

Development of a Field Oxygenation Device and Its Practice in the Oxygen Depleted Water Mass (빈산소 수괴해역 용존산소 환경개선장치 개발과 현장 적용)

  • Oxygen depleted water mass can damage aquatic animals not only in direct way but also in indirect way by generating toxic substances including occurrence of hydrogen sulfide and ammonia which are also highly detrimental to animal life in the water mass. An oxygen dissolution device was developed, which makes turnover of the oxygen rich (over 20 mg/L) surface water down to the bottom where hypoxia is evident and tested the device in terms of oxygen recovery in the oxygen depleted bottom water. the device with turnover rates of $3.6\;m^2$/min at the liquid oxygen injection rate of 48~26.3 L/min could recover dissolved oxygen level to 7~25 mg/L at depth 7 m to lead to the dissolution level of over 90% by the supply of liquid oxygen. The running advantage of the device is that it does not require any auxiliary tank and higher energy for operation. Therefore, it can be highly useful device to relieve damages to the farmed animals in the oxygen depleted waters.

Dietary Factors for Secretary Digestive Enzyme from the Pancreas in the Chicken (계의 췌장소화효소 분비에 미치는 사료성분에 관한 연구)

  • The present study was done to investigate the mechanism of Pancreatic digestive enzyme secretion in response to dietary components in chicks. A simplefied pancreatic juice collection method, useful for a short-term experiment, was developed. By wing vein injection, it was shown that the increased trypsinogen and chymotrypsinogen, while neither other single amino acids nor glucose affected the secretion of enzymes, amylase, trypsinogen and chymotrypsinogen. Cholecystokinin (CCK) had an immediate effect on pancreatic enzyme secretion and this response was in a dose dependent fashion. The injection of CCK seemed to have selective stimulation favoring the secretion of chymotrypsinosen followed by amylase and trypsinogen. Simultaneous injection of single amino acid with CCK increased digestive enzyme secretion to various extents depending on the kind of amino acids whereas the injection of glucose with CCK did not affect when compared with that of CCK'alone. By varying doses, synergetic action of CCK plus amino acid on the secretion of pancreatic digestive enzymes was observed at 0.5mM for Val and 5mM for Arg. A further attempt was made to examine the effect of combined administration of amino acids with CCK on pancreatic enzyme secretion. The injected substances were an AAs mixture and combination of selected amino acids, i.e. Thr+Phe+Ile, Thr+Phe. Thr+Ile or Phe+Ile. When increases in enzyme outputs for the first 30 min were compared , it was shown that the responses of three enzymes, amylase, trypsinogen and chymotrypsinogen, brought about by the administration of the AAs mixture was almost entirely accounted for by the combined injection of Thr+Phe. Thus, it was well demonstrated that CCK and amino acids had a synergetic action on the secretion of a specific pancreatic digestive enzyme depending on a kind of amino acid injected.

Genetic Polymorphisms of UGT1A and their Association with Clinical Factors in Healthy Koreans

  • Glucuronidation by the uridine diphosphateglucuronosy-ltransferase 1A enzymes (UGT1As) is a major pathway for elimination of particular drugs and endogenous substances, such as bilirubin. We examined the relation of eight single nucleotide polymorphisms (SNPs) and haplotypes of the UGT1A gene with their clinical factors. For association analysis, we genotyped the variants by direct sequencing analysis and polymerase chain reaction (PCR) in 218 healthy Koreans. The frequency of UGT1A1 polymorphisms, -3279T>G, -3156G>A, -53 $(TA)_{6>7}$, 211G>A, and 686C>A, was 0.26, 0.12, 0.08, 0.15, and 0.01, respectively. The frequency of -118 $(T)_{9>10}$ of UGT1A9 was 0.62, which was significantly higher than that in Caucasians (0.39). Neither the -2152C>T nor the -275T>A polymorphism was observed in Koreans or other Asians in comparison with Caucasians. The -3156G>A and -53 $(TA)_{6>7}$ polymorphisms of UGT1A were significantly associated with platelet count and total bilirubin level (p=0.01, p=0.01, respectively). Additionally, total bilirubin level was positively correlated with occurrence of the UGT1A9-118 $(T)_{9>10}$ rare variant. Common haplotypes encompassing six UGT1A polymorphisms were significantly associated with total bilirubin level (p=0.01). Taken together, we suggest that determination of the UGT1A1 and UGT1A9 genotypes is clinically useful for predicting the efficacy and serious toxicities of particular drugs requiring glucuronidation.

Antimicrobial Effects and Antioxidative Activity of Baek-bu-ja(Aconiti koreani Rhizoma) by Extraction Solvent Ratio (추출용매 비에 따른 백부자(Aconiti koreani Rhizoma) 추출물의 항균효과 및 항산화효과)

  • For the purpose of developing natural antioxidant, the antioxidative activity and antimicrobial of phenolics isolated from Baek-bu-ja (Aconiti koreani Rhizoma) were determined. Optimum extracting condition for phenolics were water extracts. HPLC analysis showed that the four major phenolic metabolites were rosemarinic, protocatechuic, caffeic and chlorogenic acids. The water extracts of Baekbuja did not have antimicrobial activity against H. pylori; however, the ethanol extracts revealed higher antimicrobial activity. Electron donation ability on DPPH of Baekbuja ethanol extract was 20% higher than other ethanol extracts. The 2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid radical decolorization (ABTS) and antioxidant protection factor (PF) were determined with extracts from Aconiti koreani Rhizoma. 94% inhibition and 1.14 PF were shown on ABTS and antioxidant protection factor with 60% ethanol extracts. Also, TBARS (thiobarbituric acid reactive substances) showed $0.19\;{\mu}M$ in the control and $0.07\;{\mu}M$ in the 80% ethanol extracts. The result suggests that Baekbuja extract may be useful as potential sources of anti Helicobacter pylori, antioxidant.

The Antibiotic Effect of Acetic acid on Helicobacter pylori (초산에 의한 마우스 위의 Helicobacter pylori 살균효과)

  • Helicobacter pylori (H. pylori) is the main causal bacteria occurring stomach diseases such as gastritis and gastric ulcer. These bacteria are found in most adults' stomach. Especially, 60~90% of H. pylori is found in Korean stomach. As a lot of curing methods have been applied to remove H. pylori and certain effects have been done but it's impossible to remove it perfectly with the existing medicines for curing. Therefore, it's very urgent to develop a certain substance showing more excellent effect than the existing medicines. In this study, it wasfound that organic acids can be accessed easily, inserted into mouth for curing and has excellent sterilizing effect among the substances showing excellent antibiotic power. Among them, acetic acid showed the most excellent effect. To confirm the refraining power against H. pylori, we performed tests through in vitro contact testing methods by concentration and tsta. In the result, H. pylori were terminated perfectly in the solution of acetic acid more than 0.3% within 1 minute. With a base of the result of In vitro test, It was performed in vivo test. As the results, H. pyloriwere terminated perfectly on 0.2% solution of acetic acid from the result of confirming through urease test, ELISA method and RT-PCR. Therefore, the result of this research will be very useful information in developing the functional foodstuffs using acetic acid in order to terminate H. pylori on the people's stomach, who suffers from H. pylori.

Antioxidant effect of garlic and aged black garlic in animal model of type 2 diabetes mellitus

  • Hyperglycemia in the diabetic state increases oxidative stress and antioxidant therapy can be strongly correlated with decreased risks for diabetic complications. The purpose of this study is to determine antioxidant effect of garlic and aged black garlic in animal model of type 2 diabetes. The antioxidant activity of garlic and aged black garlic was measured as the activity in scavenging free radicals by the trolox equivalent antioxidant capacity (TEAC) assay. Three week-old db/db mice were fed AIN-93G diet or diet containing 5% freeze-dried garlic or aged black garlic for 7 weeks after 1 week of adaptation. Hepatic levels of lipid peroxides and activities of antioxidant enzymes were measured. TEAC values of garlic and aged black garlic were $13.3{\pm}0.5$ and $59.2{\pm}0.8{\mu}mol/g$ wet weight, respectively. Consumption of aged black garlic significantly decreased hepatic thiobarbituric acid reactive substances (TBARS) level compared with the garlic group which showed lower TBARS level than control group (p<0.05). Activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) of garlic and aged black garlic group were significantly elevated compared to the control group. Catalase (CAT) activity of aged black garlic group was increased compared with the control group. These results show that aged black garlic exerts stronger antioxidant activity than garlic in vitro and in vivo, suggesting garlic and aged black garlic, to a greater extent, could be useful in preventing diabetic complications.
