Inhibition of DMBA-Induced Mouse Epidermal Carcinogenesis by Astaxanthin-Containing Egg Yolks (DMBA로 유발한 Mouse 피부암에 대한 Astaxanthin이 함유된 난황의 항암효과)

  • Anticarcinogenic activity of astaxanthin-containing egg yolks (designate AEY) was investigated for 7,12-dimethylbenz[a]anthracene (DMBA)-induced two stage mouse epidermal carcinogenesis. Female ICR mouse (6-7 weeks of age) were house in a humidity-and-temperature-controlled facility and subjected to feed and water ad libitum. AEY (10 mg/0.2 ml acetone) was painted on the back of mice 7 days, 3 days and 5 min before DMBA treatment (50 nmole/0.2 ml acetone). One week later after DMBA treatment, 6 ${\mu}g$ tetradecanoyl 12-phorbol 13-O-acetate (TPA) dissolved in 0.2 ml acetone was applied on the mouse twice weekly over a period of 22 weeks. No sample was given to control mice. Control egg yolk (CEY) and astaxanthin-containing oil (designate AO) from Phaffia rhodozyma were used as positive controls. Mouse treated with AEY exhibited 10 tumors per mouse whereas control mouse exhibited 15 tumors per mouse, the fact that 33% reduction of tumor per mouse by AEY treatment. Tumor incidence was also reduced to 15% by AEY treatment when compared to that of control group. Such effects were also seen in CEY and AO treatment groups, but leaser extent. AO gave reduction of food intake and body weights relative to those of AEY and CEY, indicating toxicity of AO. These results suggest that AEY exhibits anticarcinogenic activity for DMBA-induced mouse epidermal carcinogenesis.

Optimization of Drying Temperature and Time for Pork Jerky Using Response Surface Methodology

  • Response surface methodology (RSM) was applied to determine the optimum drying conditions for pork jerky. The physicochemical properties of pork jerky, such as final moisture content, water activity (Aw), pH, and shear force were investigated. In addition, sensory characteristics of pork jerky were evaluated and were used as a parameter for determining the optimum condition. Pork jerky samples were dried at different temperatures between 40 to $80^{\circ}C$ for the time ranged from 0 to 10 hr. The predicted values for moisture content, Aw, and shear force of dried pork samples were in good agreement with the experimental values with correlation coefficients ($R^2$) of 0.95, 0.96, and 0.97, respectively. Both drying temperature and time significantly (p<0.01) affected moisture content, Aw, pH, and shear force and their interactions were also significant at p<0.01 except for Aw. RSM showed the optimum drying conditions for pork jerky, based on moisture content, shear force, and sensory evaluation to be oC$65-70^{\circ}C$ for 7-8 hr.

Study on HPLC conditions for chemotaxonomy of Ganoderma species (영지버섯의 화학적 계통분류를 위한 HPLC분석 방법에 관한 연구)

  • This study was carried out to optimize the conditions for a chemotaxonomic classification of Ganoderma species. The mycelia of Ganoderma species were extracted with 100% MeOH, and the concentrated extracts were further purified and partitioned with column chromatography (HP20) and n-BuOH, respectively. From the result of high-performance liquid chromatography (HPLC), the constituents of the samples were efficiently separated with a Chemco Pak $C_{18}$ column ($250mm{\times}4.6mm$) by linear gradient elution using water and acetonitrile as mobile phase components at a flow rate of 0.8 ml/min and detector wavelength at 210 nm. However, the samples without purification or partition were not detected the characteristic peaks. This profile could be used to classify and identify the various Ganoderma species.

Hypoglycemic and Hypocholesterolemic Effects of Botryosphaeran from Botryosphaeria rhodina MAMB-05 in Diabetes-Induced and Hyperlipidemia Conditions in Rats

  • Botryosphaeran, a water-soluble exopolysaccharide of the ${\beta}-(1{\rightarrow}3;1{\rightarrow}6)$-D-glucan type that has been isolated from the culture medium of Botryosphaeria rhodina MAMB-05 grown in submerged fermentation using glucose as the sole carbon source, was previously demonstrated to be non-genotoxic in peripheral blood and bone marrow, and exhibited strong anticlastogenic activity. In the present study, the effects of botryosphaeran were investigated in streptozotocin-induced diabetic rats as well as in high-fat diet-fed hyperlipidemic Wistar rats. The plasma glucose level was reduced by 52% in the diabetic group of rats after administration of 12 mg botryosphaeran/kg body weight of the rats (b.w.)/day by gavage over 15 days. A reduction in the median ration intake was accompanied by an increase in the median body weight gain, as well as the efficiency of food conversion. These results demonstrate that botryosphaeran has protective effects by reducing the symptoms of cachexia in Diabetes mellitus. Botryosphaeran administered by gavage at a concentration of 12 mg botryosphaeran/kg b.w./day over 15 days also reduced the plasma levels of total cholesterol and low density lipoprotein-cholesterol by 18% and 27%, respectively, in hyperlipidemic rats. Based on these findings, we conclude that botryosphaeran possesses hypoglycemic and hypocholesterolemic properties in conditions of diabetes mellitus and hyperlipidemia, respectively, and may be used as an oral anti-diabetic agent.

Emission Characteristics of VOCs in Drying Process for Plywood Manufacturing (합판 제조용 목재 건조공정에서의 휘발성 유기화합물(VOCs) 배출특성)

  • Emission characteristics of volatile organic compounds (VOCs) were investigated in the flue gas emitted from wood drying process for plywood manufacturing. The moisture content of raw timber was average 48%, and its density was $831.55kg/m^3$. But the moisture content of dried wood is needed less than around 10%, thus the moisture contents of flue gas should be remarkably high(about 18.2 V/V%). Therefore, the vapor in flue gas is equivalent to 320 ton-vapor/day when 1100 ton-wood/day is treated in the wood drying process. The temperature of flue gas ranges from $140^{\circ}C\;to\;150^{\circ}C$ in each dryer stack with exception of the input site of wood(about $110^{\circ}C$). The velocity of flue gas in each stack ranges from 1.7 to 9.7m/sec. In order to assess the concentrations and attribution rate of odorous compounds, it was analyzed about 40 VOCs in the flue gases. It was found that the major odorous compounds were 8 compounds, and the concentrations of major VOCs(ppm) were as follows; benzene: $0.054{\sim}0.052$, toluene: $1.011{\sim}2.547$, ethylbenzene: $0.472{\sim}2.023$, m,p-xylene: $0.504{\sim}3.245$, styrene: $0.015{\sim}0.148$, o-xylene : $0.271{\sim}1.097$, ethanol: $11.2{\sim}32.5$, ${\alpha}$-pinene: $0.908{\sim}10.578$, ${\beta}$-pinene: $0.982{\sim}14.278$. The attribution rate of terpenes (${\alpha}$-pinene, ${\beta}$-pinene) was about 60.56%, and that of aromatics and alcohols was about 22.77%, and 16.67%, respectively. It is suggested that the adequate control device should be used to control both the water soluble and non-soluble compounds because both compounds were mixed in flue gas.

  • One of the most important factors that have a large influence on performance of the solar water heater system is performance of the solar collector, more detailedly, coating technology on the surface of the solar collector, which can provide high solar absorptance and low emittance. The core of the coating technology is to coat solar selective surfaces. In this study, various performance experiments are carried out using $Cr_2(SO_4)_3{\cdot}15H_2O$ coating technology. Here, IGBT(Insulated Gate Bipolar Transistor) of 5000A-15V was used as the surface processing rectifier which can stably output power and also can control voltage and current. The plating solution mainly contains black chrome$^{+3}$ concentration, H-y Conductivity, N-u Complex, NF Additive and NC-2 Wetter. Before applying the black chrome coating on the copper plate, optimal conditions are provided by using various preprocessing methods such as removal of fat, activation, electrolytic polishing, nickel strike, copper sulfate plating and bright neckel plating, and then the automatic continuous coating experiment are performed according to plating time and cathode current density. In the experiment, after the removal of fat, chemical polishing, nickel strike and activation processes as the preprocessing methods, the black chrome coating was performed in a plate solution temperature of $28^{\circ}C$ and a cathode current density of $18A/cm^2$ for 90 seconds. The thickness of chrome and nickel on the coated plate is $0.389{\mu}m$, $159{\mu}m$ respectively. As a result of the coating experiment, it showed the most excellent performance having a high solar absorptance of 98% and a low emittance of $5{\pm}1%$ when the black chrome surface had a thickness of $0.398{\mu}m$.

Effects of Pueraria radix in Water Extract on the Detoxification in Rat administered with Cadmium (카드뮴을 급여한 흰쥐에서 갈근 열수 추출액의 해독작용효과)

  • This study was designed to investigates the effects of Korean pueraris radix water extract in Cd(cadmium) administered rats. Forty male Sprague-Dawley rats weighing $100{\pm}10g$ were used for this experiment and divided into following 4 groups; control group, 3% pueraria radix in water extract group, 50 ppm Cd group, 50ppm Cd group with 3% pueraria radix in water extract group. The Cd administered rats were given 50 ppm of $CdCl_2\;{\cdot}\;2H_2O$ disolved in the distilled water. The Cd content in the rats tissue of Cd administered group was lower than in the rats tissue of Cd group with 3% pueraria radix in water extract group. Plasma levels of renin activity was increased by Cd administration group, compared with 3% pueraria radix in water extract group and Cd administred group. Glutamate oxaloacetate transaminase(GOT) and Glutamate pyruvate transaminase(GPT) were increased in Cd-administered group and lower in the 3% extracts of pueraria radix in water extract group. Lactate dehydrogenase(LDHase) was lower in the 3% extracts of pueraria radix-Cd group than in the Cd group. This results suggested that pueraria radix in water extract group, has a lowering effects on the accumulation of Cd and it is belived that the pueraria radix in water extract group has some protective effects to Cd administered in rats, but the mechanism of these effects was obscure.

  • The extraction of trace cobalt, copper, nickel, cadmium, lead and zinc in urine samples of organic and alkali metal matrix into chloroform by the complex with a dithizone was studied for graphite furnace AAS determination. Various experimental conditions such as the pretreatment of urine, the pH of sample solution, and dithizone concentration in a solvent were optimized for the effective extraction, and some essential conditions were also studied for the back-extraction and digestion as well. All organic materials in 100 mL urine were destructed by the digestion with conc. $HNO_3$ 30 mL and 30% $H_2O_2$ 50 mL. Here, $H_2O_2$ was added dropwise with each 5.0 mL, serially. Analytes were extracted into 15.0 mL chloroform of 0.1% dithizone from the digested urine at pH 8.0 by shaking for 90 minutes. The pH was adjusted with a commercial buffer solution. Among analytes, cadmium, lead and zinc were back-extracted to 10.00 mL of 0.2 M $HNO_3$ from the solvent for the determination, and after the organic solvent was evaporated, others were dissolved with $HNO_3-H_2O_2$ and diluted to 10.00 mL with a deionized water. Synthetic digested urines were used to obtain optimum conditions and to plot calibration-eurves. Average recoveries of 77 to 109% for each element were obtained in sample solutions in which given amounts of analytes were added, and detection limits were Cd 0.09, Pb 0.59, Zn 0.18, Co 0.24, Cu 1.3 and Ni 1.7 ng/mL, respectively. It was concluded that this method could be applied for the determination of heavy elements in urine samples without any interferences of organic materials and major alkaline elements.

Estimation of Long-term Water Demand by Principal Component and Cluster Analysis and Practical Application (주성분분석과 군집분석을 이용한 장기 물수요예측과 활용)

  • The multiple regression models which have two factors(population and commercial area) have been used to forecast the water demand in the future. But, the coefficient of population had a negative value because proper regional classification wasn't performed, and it is not reasonable because the population must be a positive factor. So, the regional classification was performed by principal component and cluster analysis to solve the problem. 6 regional characters were transformed into 4 principal components, and the areas were divided into two groups according to cluster analysis which had 4 principal components. The new regression models were made by each group, and the problem was solved. And, the future water demands were estimated by three scenarios(Active, moderate, and passive one). The increase of water demand ore $89.034\;m^3/day$ in active plat $49,077\;m^3/day$ in moderate plan, and $19,996\;m^3/day$ in passive plan. The water supply ability as scenarios is enough in water treatment plant, however, 2 reservoirs among 4 reservoirs don't have enough retention time in all scenarios.

Linearity Estimation of PET/CT Scanner in List Mode Acquisition (List Mode에서 PET/CT Scanner의 직선성 평가)

  • Purpose: Quantification of myocardial blood flow (MBF) using dynamic PET imaging has the potential to assess coronary artery disease. Rb-82 plays a key role in the clinical assessment of myocardial perfusion using PET. However, MBF could be overestimated due to the underestimation of left ventricular input function in the beginning of the acquisition when the scanner has non-linearity between count rate and activity concentration due to the scanner dead-time. Therefore, in this study, we evaluated the count rate linearity as a function of the activity concentration in PET data acquired in list mode. Materials & methods: A cylindrical phantom (diameter, 12 cm length, 10.5 cm) filled with 296 MBq F-18 solution and 800 mL of water was used to estimate the linearity of the Biograph 40 True Point PET/CT scanner. PET data was acquired with 10 min per frame of 1 bed duration in list mode for different activity concentration levels in 7 half-lives. The images were reconstructed by OSEM and FBP algorithms. Prompt, net true and random counts of PET data according to the activity concentration were measured. Total and background counts were measured by drawing ROI on the phantom images and linearity was measured using background correction. Results: The prompt count rates in list mode were linearly increased proportionally to the activity concentration. At a low activity concentration (<30 kBq/mL), the prompt net true and random count rates were increased with the activity concentration. At a high activity concentration (>30 kBq/mL), the increasing rate of the prompt net true rates was slightly decreased while the increasing rate of random counts was increased. There was no difference in the image intensity linearity between OSEM and FBP algorithms. Conclusion: The Biograph 40 True Point PET/CT scanner showed good linearity of count rate even at a high activity concentration (~370 kBq/mL).The result indicates that the scanner is useful for the quantitative analysis of data in heart dynamic studies using Rb-82, N-13, O-15 and F-18.
