A 4-week energy balance study was conducted to estimate the energy expenditure (EE) of 7 high school age girl, 15 to 16 year age, by measurement of energy intakes and changes in body energy (BE) content (intake/balance technique), keeping their normal living pattern and eating behavior. Gross energy intake (GE) and fecal energy (FE) loss was measured by bomb calorimetry. Urinary energy (UE) loss was calculated from nitrogen excreted. Fat mass (FM) was determined from body density estimated from skinfold thickness. Mean constitutional ratio of carbohydrate, protein and fat for the total energy intake was 70.1$\pm$1.8%, 12.2$\pm$0.7% and 17.7$\pm$2.0%, respectively, Fecal energy loss was 2.8% proportion of the gross energy intake. Mean daily metabolizable energy estimated by subtract fecal and urinary energy loss was 2022 $\pm$ 50㎉. Total body energy change estimated from body composition change over 28days was increased 2400 $\pm$ 950㎉ . Mean daily energy expenditure was 1958$\pm$87㎉ (39$\pm$2㎉ /kg of body weight).
This study was conducted to investigate the effects of protein levels and casein/whey ratios on organ growth and protein metabolism in early weaned rats. Premature rats weaned by the 17th day were fed six semipurified synthetic, isocaloric and gel diets that contained three levels (low, medium and high) and two different combinations(casein/whey ; 80 : 20 or 20 : 80) of milk protein for 8 days. On the 25th day postpartum, frest weigth and DNA, RNA and milk protein contents in brain, liver, kidney and muscle were determined to ascertain organ and cellular growth. Futher, with a view to ascertain protein metabolism and renal functions, serum total protein, $\alpha$-amino N, urea N, and creatinine and creatinine and urinary urea N, creatinine and hydroxproline were determined. Total DNA contents of brain, liver and kidney, which may represent as an index of cell numbers in those organs were significantly decreased in the rats fed diets containing low level protein regardless of casein/whey ratio. However, as fat as the rats fed high protein diets were concerned, their fresh weight, protein contents and GFR of kidney were significantly increased. Furthermore, nitrogen components, $\alpha$-amino N, urea N and creatinie in serum and urine were also increassed. Another observation was that high casein/whey ratio significantly facilitated accumulation of porteins in muscle and kidney and urinary hydorxyproline excretion, not affecting the DNA content of those organs. This study showed that low(8%) or high(32%) contents of protein had less desirable effects either on protein metabolism or on organ cellular growth in prematurely weaned rats, whereas there were no effects on general growth and bone strength.
The influence of lead exposure on renal function was studied. Eighty nine lead exposed workers who worked in 2 storage battery factories, and seventy one control workers were chosen for this study. Blood lead(PbB) and zinc protoporphyrin in whole blood(ZPP) were selected as indicators of lead exposure. As indicators of renal function, urinary N-acetyl-$\beta$-D-glucosaminidase(NAG), blood urea nitrogen(BUN), serum creatinine(S-Cr), total protein in urine(U-TP),and serum uric acid(S-Ua) were selected. The results obtained were as follows: 1. While the mean values of lead exposure indicators of lead workers were significantly different from non-exposed ones, the mean values of NAG, U-TP, BUN and S-Cr of renal function indicators of exposed were also significantly different from non-exposed but their mean values were all within normal limits. 2. BUN, logarithmic U-TP, logarithmic NAG and S-Cr showed statistically significant correlation with PbB. 3. The proportion of workers whose values of renal function indicators were over the normal limits(NAG7.5 U/g Cr ; U-TP10.9 mg/dl ; BUN20 mg/dl ; S-Crl.2 mg/dl ; S-Ua7.0 mg/dl) by the level of lead absorption in terms of PbB and ZPP were calculated. The proportion of workers with over the normal limits of U-TP among total workers showed the dose-response relationship. When age is adjusted, U-TP showed significantly strong dose-response relationship with the level of PbB and ZPP.
The experiment was aimed at studying the effect of ruminal $NH_3-N$ levels on ruminal fermentation, microbial population, urinary purine derivative excretion, digestibility and rice straw intake in swamp buffaloes. Five, 3 to 4 years old, rumen fistulated swamp buffaloes were randomly assigned according to a $5{\times}5$ Latin square design to rceive five different intraruminal infusions of $NH_4HCO_3$ (0, 150, 300, 450 and 600 g/d) on a continuous daily basis. Rice straw as a roughage was offered ad libitum while concentrate was given at 0.8% BW daily. The results were that as levels of $NH_4HCO_3$ increased, ruminal $NH_3-N$ concentrations increased from 7.1 to 34.4 mg%. The highest digestibility and voluntary straw intakes were found at 13.6 to 17.6 mg% ruminal $NH_3-N$ levels; straw intake was highest at 13.6 mg%. Total bacterial and protozoal counts linearly increased as the ruminal $NH_3-N$ increased and were highest at 17.6 mg%. Total urinary purine derivatives and allantoin excretion were highest (44.0, 37.4 mM/d) at 17.6 mg% ruminal $NH_3-N$. Highest total VFAs (115 mM) were obtained a 13.6 mg% ruminal $NH_3-N$ while blood urea nitrogen significantly increased as ruminal $NH_3-N$ increased. The results from this experiment suggest that optimum ruminal $NH_3-N$ in swamp buffaloes is higher than 13.6 mg%, for improving rumen ecology, microbial protein synthesis, digestibility and straw intake.
This study was designed to observe the change of body composition and nutrient metabolites and the excretion of minerals during complete fasting for 10 days in thirty women. Average loss of body weight was 7.98kg and body fat was gradually reduced after 9 days fasting, but the loss of lean body mass was reduced after 7 days fasting. Serum glucose level was sustained at constant level, but serum levels of blood urea nitrogen, free acid and $\beta$-hydroxybutyrate were significantly increased during fasting but decreased after re-feeding. Urinary excretions of 3-methylhistidine, total creatinine, and urea were high in the beginning of fasting but gradually decreased. Serum level of parathyroid hormone was significantly reduced by fasting but regained after re-feeding. Serum level of minerals was at the constant level throughout the experimental period. The urinary excretion of minerals(Ca, K, Mg, P) was significantly increased in the early stage of fasting and then reduced from 7 days, but the excretion of Zn was continuously increased until the late stage of fasting. These results showed that amino acid fatty acid released from the breakdown of muscle protein and body fat, respectively, were utilized for energy during fasting. Body weight and BMI were reduced because of the increased muscle protein breakdown and body water excretion during early stage of fasting, but the significant body fat loss was after 9 days fasting. Therefore, it could be suggested to fast for longer than 10 days if the reduction of body fat was planned by fasting, and recommed to exercise and ingest more vitamins and minerals to replace the excreted minerals during fasting. Paecilomyces sinclairiis (PS) is known as a functional food or human health supplement. However concerns have been raised about its kidney toxicity. This study was performed to investigate the kidney toxicity of PS by 13 week-oral administration to rats. Blood urea nitrogen (BUN), serum creatinine, and kidney damage biomarkers including beta-2-microglobulin (${\beta}2m$), glutathione S-transferase alpha (GST-${\alpha}$), kidney injury molecule 1 (KIM-1), tissue inhibitor of matrix metalloproteinase 1 (TIMP-1), vascular endothelial growth factor (VEGF), calbindin, clusterin, cystatin C, neutrophil gelatinase-associated lipocalin (NGAL) and osteopontin were measured during or after the treatment of PS. BUN, creatinine and kidney damage biomarkers in serum were not changed by PS. However, kidney cell karyomegaly and tubular hypertrophy were observed dose-dependently with higher severity in males. KIM-1, TIMP-1 and osteopontin in kidney and urine were increased dose dependently in male or at the highest dose in female rats. Increased urinary osteopontin by PS was not recovered at 2 weeks of post-exposure in both genders. Cystatin C in kidney was decreased at all treatment groups but inversely increased in urine. The changes in kidney damage biomarkers were more remarkable in male than female rats. These data indicate that the PS may provoke renal cell damage and glomerular filtration dysfunction in rats with histopathological lesions and change of kidney damage biomarkers in kidney or urine. Kidney and urinary KIM-1 and cystatin C were the most marked indicators, while kidney weight, BUN and creatinine and kidney damage biomarkers in serum were not influenced.
Urease plays an important role in the urea metabolism and the effect of urease activity on human and environment is enormous. For instance, urease acts as a virulence factor of the urinary and gastrointestinal tracts infections in human and animal, being involved in kidney stone formation, catheter encrusatation, pyelonephritis, ammonia encephalopathy, hepatic coma, and urinary tract infections. Widespread urease activity in soil induces a plant damage due to ammonia toxicity and pH increase. Therefore, urease activity regulation through urease inhibitors would lead to an enhanced efficiency of urea nitrogen uptake in plants and to the improved therapeutic strategies for ureolytic bacterial infections. To search for new inhibitory compounds on urease activity from herbs, MeOH extracts of herbs were screened. Among of them, the MeOH extracts of Areca catechu exhibited an excellent inhibitory effect on urease activity. Two compounds were isolated from the ethyl acetate fraction by the activity guided fractionation. Their chemical structures were identified as (+)-catechin(compound I) and allantoin(compound II) by spectroscopic evidence, respectively. Compound I showed a stronger inhibitory effect on urease activity than compound II.
This study was performed to investigate the effect of protein intake on kidney development and function in growing rats. Fourty-two male Spraque-Dawley rats of weighing $97.5\pm1.9g$ were divided into 3 groups and given 5%, 15% or 50% casein diets for 6 weeks. Body weight gain was higher in the 50% group. The kidney weight was selectively affected more by the level of dietary protein compared to the other organs. DNA and RNA content were significantly higher in the 15% and 50% groups than in the 5% group but the differences disappeared when DNA and RNA were expressed per g of kidney weight. Protein and protein/g kidney content were increased with increasing level of protein in diet. GFR/animal and GFR/100gB. W. were significantly higher in the 50% group compared to the 5% and 15% groups. There was no differences in PAH clearence and RBF. Osmolality was not affected by dietary protein level. BUN and urinary nitrogen excretion were increased with the increasing dietary protein level. Although urinary Ca excretion was not significantly difference among 3 groups, the rats in the 5% group showed 30% less Ca excretion compared to the other groups. Above results suggest that dietary protein level has a great effect on the kidney weight and GFR in growing rats. Especially the hyperfiltration inhanced by high protein diet may accelerate the kidney senescence.

Background: Galgeun-tang used in traditional Korean medicine, is a mixture of the medicinal plants Cinnamomi Ramulus, Ephedrae Herba and Puerariae Radix, and has been prescribed for the treatment of various ailments, including fever. Although the use of traditional medicinal herbs to treat diseases has recently increased, their safety and toxicity profiles incompletely elucidated. Thus, we evaluated Galgeun-tang's toxicity in male and female Sprague-Dawley rats. Methods and Results: Galgeun-tang (1,000, 2,000 and 4,000 mg/kg) was orally administered to rats for 13 weeks, and then, they were maintained for 4 weeks without administration (recovery period). Their clinical signs, and hematological and urinary properties, were monitored. The results showed that Galgeun-tang administeration slightly increased serum creatinine, urea nitrogen and, aspartate aminotransferase levels. Additionally, 2,000 and 4,000 mg/kg Galgeun-tang significantly increased urinary bilirubn and protein levels of male and female rats, which were restored during the recovery period. Conclusions: The no-observed-adverse-effect level of orally administered Galgeun-tang was 4,000 mg/kg in both female and male rats, and no target organs were identified. In addition, 400 mg/kg was found to be the no-observed-effect level for toxicity under the study conditions.

Mao seed is a by-product of the wine and juice industry, which could be used in animal nutrition. The current study was designed to determine the effect of supplementation of mao (Antidesma thwaitesianum Muell. Arg.) seed meal (MOSM) containing condensed tannins (CT) on rumen fermentation, nitrogen (N) utilization and microbial protein synthesis in goats. Four crossbred (Thai Native${\times}$Anglo Nubian) goats with initial body weight (BW) $20{\pm}2kg$ were randomly assigned to a $4{\times}4$ Latin square design. The four dietary treatments were MOSM supplementation at 0%, 0.8%, 1.6%, and 2.4% of total dry matter (DM) intake, respectively. During the experimental periods, all goats were fed a diet containing roughage to concentrate ratio of 60:40 at 3.0% BW/d and pangola grass hay was used as a roughage source. Results showed that supplementation with MOSM did not affect feed intake, nutrient intakes and apparent nutrient digestibility (p>0.05). In addition, ruminal pH and ammonia nitrogen ($NH_3$-N) were not influenced by MOSM supplementation, whilst blood urea nitrogen was decreased quadraticly (p<0.05) in goats supplemented with MOSM at 2.4% of total DM intake. Propionate was increased linearly with MOSM supplementation, whereas acetate and butyrate were remained the same. Moreover, estimated ruminal methane ($CH_4$) was decreased linearly (p<0.05) when goats were fed with MOSM at 1.6% and 2.4% of total DM intake. Numbers of bacteria and protozoa were similar among treatments (p>0.05). There were linear decreases in urinary N (p<0.01) and total N excretion (p<0.01) by MOSM supplementation. Furthermore, N retention was increased linearly (p<0.05) when goats were fed with MOSM supplementation at 1.6% and 2.4% of total DM intake. Microbial protein synthesis were not significantly different among treatments (p>0.05). From the current study, it can be concluded that supplementation of MOSM at 1.6% to 2.4% of total DM intake can be used to modify ruminal fermentation, especially propionate and N utilization in goats, without affecting the nutrient digestibility, microbial populations and microbial protein synthesis.
