Investigation of Goats' Milk Adulteration with Cows' Milk by PCR

  • Goats' milk adulteration with cows' milk is becoming a big problem. In the past, the urea-polyacrylamide gel electrophoresis assay with different motility of ${\alpha}S1$-casein has been applied for the identification of cows' milk adulteration. The detection sensitivity is 1.0%. The aim of this study was to develop a faster and more sensitive method to detect cows' milk which may be present in adulterated goats' milk and goats' milk powder. The published primer was targeted at highly conserved regions in bovine mitochondrial DNA (a 271 bp amplicon). This amplicon was cloned and sequenced to further confirm bovine specific sequence. The chelex-100 was used to separate bovine somatic cells from goats' milk or goats' milk powder samples. Random sampling of different brands of goats' milk powder and tablets from various regions of Taiwan showed the adulterated rate was 20 out of 80 (25%) in goats' milk powders and 12 out of 24 (50%) in goats' milk tablets. With this system, as low as 0.1% cows' milk or cows' milk powder in goat milk or goat milk powder could be identified. This chelex DNA isolation approach provides a fast, highly reproducible and sensitive method for detecting the adulteration of goats' milk products.

  • A green and efficient method for the synthesis of 3,4-dihydropyrimidin-2(1H)-ones and -thiones through one-pot three-component reaction of ethyl acetoacetate, an aryl aldehyde, and urea or thiourea in acetonitrile using silica gel-supported polyphosphoric acid (PPA-$SiO_2$) as catalyst is described. Compared to the classical Biginelli reaction conditions, the present methodology offers several advantages such as high yields, relatively short reaction times, mild reaction condition and a recyclable catalyst with a very easy work up.

  • The compound 3-(2-Mercaptopyridine)phthalonitrile has been synthesized and characterized by IR, UV-vis, and X-ray single-crystal determination. The molecular geometry from X-ray determination of the title compound in the ground state has been compared using the Hartree-Fock (HF) and density functional theory (DFT) with the 6-31G(d) basis set. The calculated results show that the DFT and HF can well reproduce the structure of the title compound. The energetic behavior of the title compound in solvent media was examined using the B3LYP method with the 6-31G(d) basis set by applying the Onsager and polarizable continuum model. Using the TD-DFT and TD-HF methods, electronic absorption spectra of the title compound have been predicted and good agreement with the TD-DFT method and the experimental determination was found. The predicted nonlinear optical properties of the title compound are much greater than those of urea. Besides, molecular electrostatic potential of the title compound were investigated by theoretical calculations. The thermodynamic properties of the compound at different temperatures have been calculated and corresponding relations between the properties and temperature have also been obtained.

  • Alzheimer's disease (AD) is a devastating neurodegenerative disease that represents the most common form of dementia among the elderly population. The deposition of aggregated ${\beta}$-amyloid ($A{\beta}$) senile plaques in the human brain is a classic observation in the neuropathology of AD, yet an understanding of the mechanism of their formation remains elusive. $A{\beta}$ is formed through endoproteolysis of the amyloid precursor protein (APP) by ${\beta}$-secretase (BACE1, ${\beta}$-site APP-cleaving enzyme) and ${\gamma}$-secretase. In this study, BACE1 protein was successfully over-expressed, purified, and refolded and utilized in a binding study with hispidin. We developed a simpler refolding method using a urea gradient and size-exclusion gel filtration to purify an active BACE1 protein variant, in larger quantities than that reported previously, and measured the binding affinity of hispidin to the BACE1 protein variant through isothermal titration calorimetry.

  • Cerium-activated yttrium aluminate ($Y_3Al_5O_{12}:Ce^{3+}$) exhibiting a garnet structure has been widely utilized in the production of light emitting diodes (LEDs) as a yellow emitting phosphor. The commercialized yttrium aluminum garnet (YAG) phosphor is typically synthesized by a solid-state reaction, which produces irregular shape particles with a size of several tens of micrometers by using the top-down method. To control the shape and size of particles, which had been the primary disadvantage of top-down synthetic methods, we synthesized YAG:Ce nanoparticles with a diameter of 500 nm using a coprecipitation method under the atmospheric pressure without the use of template or special equipment. The precursor particles were formed by refluxing an aqueous solution of the nitrate salts of Y, Al, and Ce, urea, and polyvinylpyrrolidone (55 K) at $100^{\circ}C$ for 12 h. YAG:Ce nanoparticles were formed by the calcination of precursor particles at $1100^{\circ}C$ for 10 h under atmospheric conditions. The phase identification, microstructure, and photoluminescent properties of the products were evaluated by X-ray powder diffraction, scanning electron microscopy, absorption spectrum and photoluminescence analyses.

  • Nanosized $SnO_2$ particles were synthesized by homogeneous precipitation method using tin chloride ($SnCl_4{\cdot}5H_{2}O$) and urea ($CO(NH_2)_2$). The powders were heated at $500^{\circ}C$ and $600^{\circ}C$ for 2h. The crystal structure, microstructure, thermal behavior, specific surface area were analyzed using XRD, FE-SEM, TGA and BET, respectively. The initial resistance and the $H_2$ sensing properties were measured as a function of ${Sb_2}{O_3}$ and Pd doping concentrations. The resistance was decreased with the addition of ${Sb_2}{O_3}$ and the sensitivity for $H_2$ gas was increased with the addition of Pd. Thus, the optimum $H_2$ gas sensing property was obtained in the 0.25.mol% ${Sb_2}{O_3}$ and 1.w% added $SnO_2$ powders.

  • In this research, we investigated synthetic method of 2,6-DANT that can be used as a chain extender of urethane/urea and epoxy materials in a demilitarization method via chemical transformations considering environmental, economical aspects and stability of process. We was able to identify through GC monitoring that 2,4-DANT and 2,6-DANT were produced when we inject 'hydrazine monohydrate 3.3 eq. by TNT 1 eq.' with a fine metering pump for 30 minutes and then, reflux for an additional 2 hours. We was able to isolate only 2,6-DANT(99.3% purity and 45.0% yield) from mixture of 2,4-DANT and 2,6-DANT through the separating and refining methods using 2,6-DANT solubility in methanol and crystallinity of 2,6-DANT.

  • Optimal conditions for preparation of starch/PVA blends were investigated with the consideration of factors that may influence mechanical properties of the blends. $L_{27}(3^{13})$ experimental designs based on Taguchi method were performed and then tensile strength, strain at break, Young's modulus and tear strength of films of the blends were measured to determine the optimal conditions for preparation. Interaction effects for each factor were determined from analysis of variables. Results of $L_{27}(3^{13})$ experiments indicated that glycerol and urea were important process factors affecting optimization of the mechanical properties.

The Antimicrobial Activity of Medicinal Plants Extracts against Helicobacter Pylori (약용식물 추출물의 Helicobacter pylori에 대한 항균활성)

  • Antimicrobial activities were assayed through the hot-water extracts from 41 species of medicinal plants against Helicobacter pylori which is known as the ulcerogenic phathogen. Opuntia ficus-indica, Houttuynia cordata, Sinomenium acutum, and Coptis japonica showed the MIC at the concentrations less than 100 ppm, Pulsatilla koreana, Forsythia koreana, Rheum undulatum, and Perilla frutescens less than 200 ppm, Belamcanda chinensis, Arctium lappa, Cassia tora, Citrus tachibana, Siegesbeckia orientalis, and Caesalpinia sappan less than 300 ppm by the 2-fold dilution method. In disc method only three of them were confirmed to have antimicrobial activities which were increased in the order Perilla frutescens, Coptis japonica, Caesalpinia sappan. Three extracts were partitioned with chloroform, ethyl acetate and butanol in sequence and examined for the activity to inhibit H. pylori. The major ativities were observed in ethyl acetate fraction of Caesalpinia sappan, butanol fraction of Perilla frutescens, butanol and chloroform fraction of Coptis japonica. The partitioned fractions were found to have increased antimicrobial activities in all extracts. The experiments in which the extracts were added into urea R broth containing the crude urease derived from H. pylori resulted in the increase of pH and optical density at 560 nm to 8.15 and 1.7 respectively. Urease activity of H. pylori was inhibited over 80% by Caesalpinia sappan, Perilla frutescens, and Coptis japonica, of which Caesalpinia sappan suppressed up to 95%.

Manufacture of Wood Veneer-Bamboo Zephyr Composite Board: II. Effect of Manufacturing Conditions on Properties of Composite Board (목재 단판-대나무 제퍼 복합보드 제조: II. 복합보드의 성능에 미치는 제조조건의 영향)

  • This research was performed to investigate the feasibility of bamboo as a raw material for the manufacture of plywood. Wood veneer-bamboo zephyr composite boards (WBCB) were manufactured using keruing (Dipterocarpus sp.) veneers and hachiku bamboo (Phyllostacbys nigra var. henonis Stapf) using various adhesives, and the effect of the method and amount of resin spread on the mechanical properties of the composites were investigated. The WBCB manufactured using polymeric isocyanate (PMDI) showed the best mechanical properties, followed by phenol-formaldehyde resin (PF), phenol-melamine-formaldehyde resin, urea-melamine-formaldehyde resin, and urea-formaldehyde resin. However, considering the operation feasibility as well as mechanical properties, PF resin proved to be the appropriate adhesive for the practical purpose. As the amount of resin spread increased, the mechanical properties of 5-ply WBCB with 12 mm thicknesses manufactured using PF resin tended to increase, and more failure occurred at the interface between veneer and bamboo zephyr than at the interface among bamboo zephyrs. This result suggests that penetration of resin into bamboo zephyr could be the important factor. In this research, the appropriate amount of resin amount was $320g/m^2$. 5-ply WBCBs were manufactured using various methods of resin spread but the effect of the methods on the mechanical properties showed no little difference, which meant that the method of resin spread could be chosen considering the manufacturing conditions and operation feasibility.
